Danau Panggang is a unique habitat for fish species used for food and economic purposes that live in it. This study aims to identify several important and endangered fish species in Danau Panggang using the DNA sequence method and water quality in Danau Panggang. The DNA sequence was then aligned with the RefSeq from the GenBank data library by using the BLAST program of NCBI to find the closest identity associated with several types of fish samples being analyzed. The sequence analysis of the DNA show that some fish species analyzed have specific identifications :Trichopodus pectoralis, Phalacronotus apogon, Belodontichthys dinema, Anabas testudineus, and  Leptobarbus hoevenii. Based on the results of the phylogenetic tree, it is also found that the relationship between fish taxons in flooded swamps of Danau Panggang is close to one another.

Keban Dam Lake is the second largest dam lake in Turkey. In addition, it has the feature of being one of the most important dam lakes in our country in terms of both aquaculture potential and electricity generation. In this study, fish samples belong to Capoeta trutta caught from Örencik region of Keban Dam Lake were used. The concentrations of some heavy metals (copper, iron, zinc, chromium, nickel, cadmium, arsenic and mercury) in muscle of Capoeta trutta and the potential health risk from consumption of this fish species were examined. In addidion, the relationship between the heavy metal levels and some biological aspects of fish (weight, length and sex) were determined. As a result of the study, it was determined that the most and the least accumulated heavy metals in the muscle tissue of Capoeta trutta were zinc and cadmium respectively. The concentration of heavy metals showed differences according to weight, length and sex of fish. The results were found indicated that heavy metal levels in the muscle tissue of Capoeta trutta are below the tolerable values recommended by EPA, WHO and FAO. Therefore, there is not any risk for human consumption of this fish species.

Riverbed substrate is important to many riverine organisms, however, it is normally difficult to study the substrate intensively especially in large and fast flowing rivers. The Chinese sturgeon Acipenser sinensis is a critically endangered fish species, it spawns on a specific riverbed substrate. In this paper, the hydroacoustic and underwater optical video technologies were integrated to study the riverbed substrate at the only remaining spawning area of the fish. Sixteen samples were firstly studied to verify the relationship between acoustic backscattering characteristics (hardness and roughness) and video observed characteristics (embeddedness and medium diameter of the grain). Then eight vertical mobile hydroacoustic surveys were conducted to acoustically map the 16 km spawning reach. The results revealed that the heterogeneous mix of boulder/cobble and sand was the dominant substrate at the spawning reach. However, the embeddedness and medium diameter of the grain varied within the area which tend to affect the spawning site selection of the fish. This study indicated that the combination of hydroacoustic and optical video technologies could be very useful to study the riverbed substrate in river ecological issues.

Lake Lebo Taliwang is one of the lakes in the West Nusa Tenggara with high freshwater fish species diversity. However, the species identification of freshwater fish species from Lake Lebo Taliwang using DNA barcoding is very limited. Therefore, the objective of this study was to identify seven samples of freshwater fish species collected from Lake Lebo Taliwang based on COI mitochondrial gene as a DNA barcoding marker and establish library COI sequences of Indonesian freshwater fish. We are using a standard DNA analysis and data obtained from this study was then examined using Nucleotide BLAST and the phylogenetic tree was analyzed using the Neighbour-Joining (NJ) method with Kimura 2 Parameter (K2P) model. The results revealed that among the seven samples of freshwater fishes collected from the Lake Lebo Taliwang, three samples were identified as Anabas testudineus, the other three samples were Barbodes binotatus and one sample was Trichopodus trichopterus. The level of similarity of these freshwater fish samples referred to the database from the GenBank and BOLD was between 98-100%. The NJ tree supports the clade of each species identified in this study. This occurrence indicated that DNA barcoding by using the COI mitochondrial gene was proven to be able to identify the freshwater fish samples accurately.


Consumers all over the world are increasingly becoming aware of the health and nutrition status of fish and fishery products. There have develop some preference for fish species and even their processing methods. This study aims to investigate the effects of two drying methods (smoking and oven drying) on the biochemical components and organoleptic properties of two less preferred food fishes abundant in the study area. The fish samples Mormyrus rume and Labeo coubie were purchased from fresh landings of fishermen at Ahaha beach. The biochemical assay of the moisture, protein, fat, carbohydrate, ash, calcium (Ca), magnesium (Mg), phosphorus (P), iron (Fe), sodium (Na) and potassium (K) were conducted in the labouratory using standard methods. The proximate composition of raw M. rume was determined as moisture 70.38%, protein 17.43%, carbohydrate 1.13%, fat 5.93% and ash 2.77% while oven dried were 18.78, 63.85, 3.37, 8.73 and 7.58% respectively. All the proximate parameters and gross energy level investigated were higher (p < 0.05) in M. rume except the moisture content of dried samples of L. coubie. The mineral content were in the order Ca < Mg < Fe < Na < P and < K in both species and were higher in M. rume except Na. The drying methods showed increase in the proximate and mineral compositions in the order raw < smoking and < oven drying except moisture content that decreased respectively is both species. Organoleptic properties revealed that dried samples (smoked and oven) were not significantly different (p < 0.05) in taste, flavour, texture and overall acceptance, however while oven dried had better colour (4.33), smoked dried taste (4.63) better. The drying methods shows concentration of the required nutrients in human diets and were also found to be most palatable by the panellist. To improve fish nutrient quality smoke and oven drying should be encourage.

Ciguatera fish poisoning (CFP) is one of the most frequently reported seafood poisoning diseases. It is endemic to the tropical region and occurs most commonly in the regions around the Pacific Ocean, Indian Ocean, and Caribbean Sea. The principal toxins causing CFP are ciguatoxins (CTXs). In the Pacific region, more than 20 analogs of CTXs have been identified to date. Based on their skeletal structures, they are classified into CTX1B-type and CTX3C-type toxins. We have previously reported species-specific and regional-specific toxin profiles. In this study, the levels and profiles of CTXs in fish present in the tropical western Pacific regions were analyzed using the liquid chromatography–tandem mass spectrometry (LC–MS/MS) technique. Forty-two fish specimens, belonging to the categories of snappers, groupers, Spanish mackerel, and moray eel, were purchased from various places such as Fiji, the Philippines, Thailand, and Taiwan. Only the fish captured from Fijian coastal waters contained detectable amounts of CTXs. The toxin levels in the fish species found along the coastal regions of the Viti Levu Island, the main island in Fiji, and the toxin profiles were significantly different from those of the fish species present in other coastal regions. The toxin levels and profiles varied among the different fish samples collected from different coastal areas. Based on the toxin levels and toxin profiles, the coast was demarcated into three zones. In Zone-1, which covers the northern coast of the main island and the regions of the Malake Island and Korovau, CTXs in fish were below the detection level. In Zone-2, CTX3C-type toxins were present in low levels in the fish. CTX1B-type and CTX3C-type toxins co-occurred in the fish present in Zone-3. The toxin profiles may have reflected the variation in Gambierdiscus spp.

AbstractDespite high effort for food traceability to ensure safe and sustainable consumption, mislabeling persists on seafood markets. Determining what drives deliberate fraud is necessary to improve food authenticity and sustainability. In this study, the relationship between consumer’s appreciation and fraudulent mislabeling was assessed through a combination of a survey on consumer’s preferences (N = 1608) and molecular tools applied to fish samples commercialized by European companies. We analyzed 401 samples of fish highly consumed in Europe and worldwide (i.e. tuna, hake, anchovy, and blue whiting) through PCR-amplification and sequencing of a suite of DNA markers. Results revealed low mislabeling rate (1.9%), with a higher mislabeling risk in non-recognizable products and significant mediation of fish price between consumer´s appreciation and mislabeling risk of a species. Furthermore, the use of endangered species (e.g. Thunnus thynnus), tuna juveniles for anchovy, and still not regulated Merluccius polli hake as substitutes, points towards illegal, unreported and/or unregulated fishing from African waters. These findings reveal a worrying intentional fraud that hampers the goal of sustainable seafood production and consumption, and suggest to prioritize control efforts on highly appreciated species.

This study is focused on evaluating the trace metal levels in water and tissues of two commercial fish species Arius thalassinus and Pennahia anea that were collected from Kapar and Mersing coastal waters. The concentrations of Fe, Zn, Al, As, Cd and Pb in these coastal waters and muscle, liver and gills tissues of the fishes were quantified. The relationship among the metal concentrations and the height and weight of the two species were also examined. Generally, the iron has the highest concentrations in both water and the fish species. However, Cd in both coastal waters showed high levels exceeding the international standards. The metal level concentration in the sample fishes are in the descending order livers > gills > muscles. A positive association between the trace metal concentrations and weight and length of the sample fishes was investigated. Fortunately the level of these metal concentrations in fish has not exceeded the permitted level of Malaysian and international standards.

Understudying the presence of bacteria and fungi population on freshwater fishes, provides the knowledge and understanding of their diversity and potential roles they may play in the health of the aquatic and terrestrial ecosystems. The present study was conducted to evaluate the diversity of bacteria and fungi species of identified fish samples from River Mijawal, Nasarawa State. The fishes were sampled from the only landing for three months (October to December, 2019) for this study. During the sampling periods, thirteen (13) different fish species were identified; they were  swabbed with sterilized swab stick from the skin surface for bacteria and fungi identification. The Dominant fish species were Tilapia zillii, Labeo senegalensis, Mormyrus rume and Mormyrus tapines. The total number of thirteen (13) bacterial and five (5) fungi species were isolated and  identified. Eight (8) of the bacteria isolates, were gram negative bacteria (Pseudomonas sp, Aeromonas sp, Serratia mercescenes, Proteus sp, Salmonella spp, Escherichia coli, Klebsiella sp, and Enterobacter sp.); whilst the other five (5) were gram positive bacteria (Bacillus sp, Micrococcus sp, Staphylococcus sp, Enterococcus sp and Listeria monocytogenes).The five isolated fungi species were Aspergillus niger, Rhizopus sp, Penicillium  sp, Trichoderma viridae and Geotrichum candidum. The frequency of occurrence of the isolated bacteria indicated that Staphylococcus sp had the highest frequency of occurrence (16.67%) while Serratia mercescens had the least occurrence (2.78%). The highest number of occurrence of the isolated fungi species was observed in Rhizopus sp while the least was recorded in Geotricum candidum. It was observed that Tilapia zillii and  Mormyrus rume had the most frequent bacteria and fungi isolates. It can therefore be deduced from the results that fish samples identified from River Mijawal has high bacteria and fungi diversity that may be pathogenic to man. Enhancer sequences control gene expression and comprise binding sites (motifs) for different transcription factors (TFs). Despite extensive genetic and computational studies, the relationship between DNA sequence and regulatory activity is poorly understood and enhancer de novo design is considered impossible. Here we built a deep learning model, DeepSTARR, to quantitatively predict the activities of thousands of developmental and housekeeping enhancers directly from DNA sequence in Drosophila melanogaster S2 cells. The model learned relevant TF motifs and higher-order syntax rules, including functionally non-equivalent instances of the same TF motif that are determined by motif-flanking sequence and inter-motif distances. We validated these rules experimentally and demonstrated their conservation in human by testing more than 40,000 wildtype and mutant Drosophila and human enhancers. Finally, we designed and functionally validated synthetic enhancers with desired activities de novo.
