Lactobacillus acidophilus (L. acidophilus) is one of the etiological agents for dental caries and dominant in the deep carious lesion. L. acidophilus has also been identified in persistent root canal infection and also related to the failure of endodontic treatment. Photodynamic therapy is a therapeutic process involving the combination of a nontoxic photosensitizer and a light source. The excited photosensitizer reacts with reactive oxygen species (ROS), which induce injury and death of the microorganism. This study aimed to prove the effect of irradiation time of photodynamic therapy to the number of L. acidophilus. Fortytwo Eppendorf tubes were treated with 0.5 ml L. acidophilus distributed into seven groups. Group 1 as the control group received no treatment. Groups 2, 3, 4, 5, 6 and 7 were treated with a combination of 0.5 ml toluidine blue O (TBO) as a photosensitizer and 630 nm photoactivated (Fotosan®) exposure time for 10, 20, 30, 40, 50 and 60 sec. Then, all were stored in an incubator of 37ºC for 48 h. Later, the colony-forming unit (CFU) was counted for each group. There were significant differences in the number of L. acidophilus in CFU of the various irradiation times. The longer the photodynamic therapy irradiation was, the lesser the number of live L. acidophilus became. At 50 sec and 60 sec irradiation, none of the L. acidophilus was found alive.

Gentamicin can induce damage to the kidney because of the accumulation of epithelial cells in the cortex. This accumulation can cause an increase in Reactive Oxygen Species (ROS), which in turn can trigger an increase in free radicals which causes a decrease in kidney function, which can be seen with Blood Urea Nitrogen (BUN) and sCr (serum creatinine) parameters. Garlic (Allium sativum L.) contains several bioactive compounds, including alliin, allisin, ajoene, and allil cysteine. The content of allisin has activity as an antioxidant. This study used a nephrotoxic mouse modelling method with 100 mg/kgBW of gentamicin induction intraperitoneally, then BUN and sCr levels were checked 24 hours after the 10th day induction. This study aimed to determine the effect of garlic ethanol extract in preventing the decline of renal function in rats induced by gentamicin with a parameter of BUN and sCr. The animal test were divided into 5 treatment groups, normal control group, negative controls, and three treatment groups at a dose of 50, 120, and 300 mg/kgBW for 10 days of concurrent treatment between extracts with gentamicin. The results of the study showed that garlic ethanol extract with a dose of  50, 120, 300 mg/kgBW was able to prevent a decrease  in kidney function due to the administration of gentamicin significantly when compared with negative controls (p 0,05).

Objective We investigated the effects of bumetanide alone and in combination with dexamethasone on facial nerve regeneration in rats with facial paralysis. Study Design A prospective controlled animal study. Setting An animal laboratory. Subjects and Methods Facial paralysis was induced in 32 Wistar rats that we then divided into 4 groups: group 1, control; group 2, bumetanide; group 3, dexamethasone; group 4, bumetanide and dexamethasone. Electroneurography was performed 1, 2, and 4 weeks later, and nerve regeneration was evaluated by electron and light microscopy and Western blotting in week 4. Results Regarding the comparison between preoperative values and week 4, the latency difference in group 1 (1.25 milliseconds) was significantly higher than those of groups 2 to 4 (0.56, 0.34, and 0.10 milliseconds, respectively; P = .001). The latency increment in groups 2 and 3 was higher than that of group 4 ( P = .002 and P = .046) in week 4, whereas groups 2 and 3 did not differ significantly ( P = .291). Amplitude difference was not statistically significant from week 4 among all groups (all P > .05). The number of myelinated axons was significantly higher in all treatment groups than in the control group ( P = .001). Axon number and intensity were significantly higher in group 4 as compared with groups 2 and 3 ( P = .009, P = .005). Conclusion After primary neurorrhaphy, dexamethasone and bumetanide alone promoted nerve recovery based on electrophysiologic and histologic measures. Combination therapy was, however, superior.

Background: The success of endodontic treatment can be achieved when pathogenic bacteria are eliminated from the root canal and periapical tissue resulting in healing of such tissue. One of the bacteria located in root canals is Staphylococcus aureus (S. aureus) reportedly found to be in severe periapical abscesses. Photodynamic therapy is one current technology that can help eliminate microorganisms without causing damage to human body cells. Average of research has been conducted using different tools and bacteria to evaluate the effects of exposure time used in photodynamic therapy on the number of bacteria. Purpose: The research reported here aimed to determine the correlation between the exposure time of photodynamic therapy and the number of S. aureus bacteria. Methods: The S. aureus bacteria used in this research were divided into seven treatment groups: a control group and six treatment groups with respective exposure times of 10, 20, 30, 40, 50 and 60 seconds. All of the bacteria were administered a photosensitiser and radiated according to the treatment intended for each group. They were then planted in nutrient agar and incubated for 48 hours. The colonies of bacteria formed were calculated using the Quebec colony counter and subsequently analyzed by means of both Kruskal Wallis and Mann Whitney U tests. Results: After calculating the number of bacterial colonies, the average number of Staphylococcus aureus bacteria in the non-irradiated group was 119 CFU/ml, 29 CFU/ml in the group with a 10-second exposure time, 20 CFU/ml in the group with a 20-second exposure time, 13 CFU/ml in the group with a 30-second exposure time, 7 CFU/ml in the group with a 40-second exposure time, but none in the groups with exposure times of 50 or 60 seconds. Conclusion: The longer the photodynamic therapy exposure time, the greater the number of S. aureus bacteria eliminated. An exposure time of 50 seconds was found to be sufficient to exterminate all S. aureus bacteria present.

Cancer is one of the world health problems. Free radical could damage cells and lead to cancer. Antioxidant compound could be found in some vegetable such as phenols and flavonoids. Flavonoids and phenol significantly decrease micronucleus formation on mitotic chromosomes. This present study aim to explore the inhibitory effect of Bombay onion extracts (BOE) on mitotic chromosomes induced by cyclophosphamide. Twenty-five mice (body weight 20–35 g) were divided into 5 groups, with 5 mice in each group. Normal control group (NCG) were given CMC 5 mg/mice/7 days. Mutagen control group (MCG) that is previously given as NCG, 4 hours later were given cyclophosphamide at a dose of 50.0 mg/kg bw intraperitoneally. Three treatment groups, group 1 (TG-1), TG-2, and TG-3, were given BOE with doses of 100; 200; and 400 mg/kg bw for 7 days, respectively. After 7 days, all three groups were treated similarly with MCG. Thirty hours later, all groups were put to death and femur bone marrows were analyzed to count the number of micronucleus. The results showed that the number of micronucleus in every 200 polychromatic erythrocyte cells at mitotic chromosomes for NCG is 29.8 ± 2.387, MCG with a value of 120.8 ± 5,718, TG-1 showed 94.8 ± 7.049, TG-2 is 68.8 ± 3.421, and TG-3 which is 30.8 ± 0.837. TG-3 showed a similar result with NCG (p> 0.05).Keywords : mitotic chromosomes, Bombay Onion Extracts, femur bone marrow, micronucleus

Abstract Objectives The aim of this study was to evaluate the efficacy of dalbavancin against MRSA biofilm-related infection in orthopaedic implants in vivo. Methods One MRSA strain isolated from human osteomyelitis was used to promote biofilm formation on the surface of screws. The implants were inserted in the proximal tibia under general anaesthesia. Thirty-nine Wistar rats were divided into three groups [control group (no treatment), Group 1 (7 days of treatment) and Group 2 (14 days of treatment)]; both treatment groups were administered dalbavancin intraperitoneally and euthanized after treatment. cfu of bacteria present in both the tibia and the implant were quantified. The infection severity was assessed by histopathology and scored from 0 (no infection) to 4 (severe infection). Results The high number of cfu/g and cfu/mL present in the control group indicated a well-established infection. There was a significant reduction in cfu in rats treated with dalbavancin both in the tibia (2.8 × 105 cfu/g) and the implant (1.1 × 106 cfu/mL) in Group 1 (1.8 × 103 cfu/g and 2.4 × 105 cfu/mL, respectively) and in Group 2 (8.2 cfu/g and 8.2 × 103 cfu/mL, respectively). Most animals from the control group presented an infection scored as 3 (severe). At the end of the experiment, most rats from Groups 1 and 2 presented an infection scored as 2 (moderate) and 0 (no infection), respectively. Conclusions Although there was a marked decrease in cfu number, signs of biofilm-induced infection prevailed after 14 days of treatment. Further studies should be carried out to evaluate the potential of dalbavancin in the treatment of bone and orthopaedic implant-associated MRSA infections.

Background. The treatment of phlegmon of the maxillofacial area and neck at the present moment remains difficult and insufficiently effective. Aim. To develop a method of using photodynamic therapy (PDT) for the treatment of phlegmon of the maxillofacial area and neck and to evaluate its effectiveness. Material and methods. A total of 92 patients with phlegmons of the maxillofacial area and neck were under observation. They were divided into 3 groups: group 1 – “control group 1” (30 patients) – where traditional treatment was used, group 2 – “control group 2” (30 patients) – in which low-intensity laser radiation was used in addition to topical treatment and group 3 – “experimental” (32 patients) – where PDT was used for topical treatment. Results. The data of clinical, cytological and bacteriological studies show that the local application of PDT has an anti-inflammatory effect and stimulates the reparative processes in the wound, and shortens the treatment period for patients. Conclusion. PDT improves the results of complex treatment of patients with phlegmons of the maxillofacial area and neck. Its application leads to the purification of a purulent wound on the 3rd (3;4) day, allows to apply secondary sutures on the 7th day and to shorten the period of inpatient treatment in comparison with the control groups from 3 to 9 days.

This study investigated therapeutic effect of ethanol extract of Anthocleista vogelii stem bark (EEAV) in the treatment of jaundice on paracetamol-induced hepatotoxicity in adult wistar rats. A total of 30 wistar rats weighing between 160-200 g were distributed into 6 groups comprising group 1 (normal control), group 2 (paracetamol 2 g/Kg bw), group 3 (paracetamol 2 g/Kg bw + Silymarin 100mg/Kg bw) and groups 4-6 (paracetamol 2 g/Kg bw each + 250, 500, & 1000 mg/Kg bw of EEAV, respectively). Experiment lasted for a period of 16 days. Paracetamol induction elevated liver indices and enzyme activities. However, administration of EEAV significantly reduced (p≤0.05) these effects in groups 3-6 when compared to group 2. A significant elevation (p≤0.05) was reported in creatinine, urea, sodium (Na+) and chloride (Cl-) when group 2 was compared with group 1. Haematological study showed that packed cell volume (PCV), haemoglobin (Hb) and red blood cell (RBC) levels significantly increased (p≤0.05) in treatment groups 3-6 when compared to group 2. The catalase (CAT) activity increased significantly (p≤0.05) in groups 3, 4 and 6 when compared to group 2. Liver histology showed normal hepatocytes architecture with normal central vein in group 1. Group 2 revealed a histologically distorted liver tissue while there was regeneration of hepatic cells in groups 3-6. The study revealed that EEAV could serve as potent herbal therapeutic agent in the treatment of jaundice triggered by paracetamol induction in adult wistar rats.
