Bacteria of the genus Enterococcus are mainly commensals building natural microflora in the digestive tract of birds and mammals. They belong to the potentially pathogenic microorganisms. Among poultry, infections caused by enterococci were reported in chickens, turkeys, ducks and ostriches. The aim of this study was to evaluate the occurrence of enterococci in poultry in Poland, including identification of enterococcus species composition and determination of the age of birds. The analysis was based on data obtained from 2014 – 2015 from Division of Avian Diseases at Warsaw University of Life Sciences-SGGW and four veterinary laboratories in Poland: Lab – Vet, Tarnowo Podgórne; SLW Biolab, Ostróda; Vetdiagnostica, Solec Kujawski; Vet – Lab Brudzew. Seven enterococcal species were isolated from broiler chickens (CB), commercial layers (CL), and broiler breeder flocks (BB), nine from all poultry types (chickens, turkey, ducks and geese). The most often isolated enterococci from CB were E. faecalis (57%) > E. cecorum (7%) > E. faecium (5.2%) > E. hirae (3.6%) > E. gallinarum (2.5%) > E. casseliflavus (0.7%) > E. durans (0.2%). Seven Enterococcus species were isolated from sources associated with poultry, most often E. faecalis > E. faecium > E. cecorum > E. hirae. The differences in the occurrence of particular enterococcal species were observed between CB, BB and CL. The mean age at the time of isolation of E. cecorum was approx.: 3.6 weeks in CB, 27.5 weeks in BB, 33.3 weeks in CL, 12.9 weeks in turkeys, 3.6 weeks in ducks, 39.5 weeks in geese. E. faecalis and E. faecium dominated in samples obtained from hatching eggs, dead-in-shell embryos and from samples related to poultry environment. In conclusion, this study indicates the high prevalence of bacteria of the Enterococcus genus in poultry. The present findings demonstrate the differences in Enterococcus species between poultry groups, including with regard to age. In total 10 enterococcal species (E. faecalis, E. cecorum, E. hirae, E. faecium, E. gallinarum, E. casseliflavus, E. durans, E. avium E. thailandicus, E. aquimarinus) were detected in poultry, poultry environmental samples, hatching eggs and dead-in-shell embryos. Enterococcus faecalis and E. cecorum were found in all above-mentioned sources.

Abstract Due to high prevalence of anthelmintic resistance (AR), complementary approaches to anthelmintic use to control gastrointestinal nematodes (GIN) in sheep are necessary. Copper oxide wire particles (COWP) are effective against Haemonchus contortus but not other GIN, and FAMACHA aids to minimize further development of AR. The objective was to determine the efficacy of COWP alone or in combination with levamisole, albendazole, or a 3-way combination to control GIN known to have AR to these anthelmintics. Naturally infected Katahdin lambs were selectively dewormed based on FAMACHA© and/or packed cell volume (PCV). If FAMACHA© scores were 3, 4, or 5, lambs received 1 g COWP alone (n = 120), COWP and levamisole (n = 47) or albendazole (n = 6), or a 3-way combination (n = 10), respectively. Untreated lambs were included (CON; n = 31). Feces and blood were collected on days 0 (day of deworming) and 14 for determination of fecal egg counts (FEC) and FEC reduction (FECRED) and PCV. Data were analyzed using repeated measures in a mixed model. The mean FECRED was greater for the combination of levamisole and COWP than COWP alone (P < 0.001). In a mixed GIN population, COWP in combination with levamisole increased the efficacy of FECRED of H. contortus and other GIN genera present, offering greater GIN management in the presence of resistance to these anthelmintics.

A study on Infectious Bursal Disease (IBD) was conducted on broiler chicks (N=200+70) of strains: Cobb-500 and Hubbard classic at Laxmipur district of Bangladesh. The blood sample were collected at day old chicks (1st day), pre-vaccinated flocks (11th day) and post-vaccinated flocks (26th day), to measure the antibody titer by indirect ELISA method. Maternal derived antibody (MDA) mean titer of day old chicks (N=40) were found 9621.65 ± 780.78 and 4232.60 ± 301.66 for Cobb-500 and Hubbard classic respectively. MDA mean titer was measured at day eleven chicken (N=60) before vaccination and found 1963.00 ± 143 and 984.16 ± 126.4037 respectively. Of these two titers; Cobb-500 strain was found to be protective (>1000) where Hubbard classic was below protective (<1000) titer level. The two flocks of Cobb-500 and Hubbard classic strains of broiler chickens were vaccinated with intermediate (Bursine-2) and intermediate plus (IBD Blen) vaccine respectively at day twelve and serum was harvested on day twenty six. The mean serum titers were found 131.30 ± 36.04 and 7413.54 ± 569.39 in Cobb-500 and Hubbard classic respectively, where mean titer level of Cobb-500 was below the minimum protective level but Hubbard classic strain was developed 7413.54, protective level. These results were indicating that the maternal derived antibody titer persists above 1000 level hindering the development of antibody titer in vaccinated flocks. In order to investigate the relationship between carbon dioxide tensions in the venous blood of young chickens and ascites susceptibility, one hundred dayold chickens from two pure broiler breeder lines differing in susceptibility to ascites syndrome were obtained and reared at low environmental temperature. Weekly, blood samples were taken for the determination of blood gas parameters and plasma thyroid hormone levels. Dead birds were examined for lesions of ascites daily and all survived birds were autopsied at the end of the trials. In cold conditioning, the cumulative incidence of right ventricular hypertrophy (RVH) and ascites was 78% (39/50) in fast-growing (line A) and 50% (25/50) in slow-growing (line B) chickens, confirming that line A chickens had higher susceptibility to ascites than line B chickens. At 12 days of age, the mean pCO2tension was significantly (P < 0.01) higher in the venous blood of line A than line B chickens. During the experiment, the majority of line A and line B chickens showing RVH and ascites exhibited pCO2tensions equal to, or exceeding 50 mmHg at 12 days of age. Additionally, fast-growing chickens showed significantly lower plasma T3 and T4 concentrations than slow-growing chickens. In conclusion, our data indicate that carbon dioxide pressure in the venous blood of early postnatal broiler chickens could be used as a valuable predictor in the selection of birds susceptible to pulmonary hypertension and ascites. The function of thyroid hormones should also be taken into consideration in further selection procedures for meat-type chickens.

The purpose of the study was to determine the features of the development of osteoporotic changes in bones in patients with liver fibrosis against the background of non-alcoholic steatohepatosis. Materials and methods. 79 patients with non-alcoholic steatohepatosis and liver fibrosis were examined. 38 surveyed persons were women, 41 were men. The average age of the patients was 42.5±5.8 years. All patients were excluded from comorbidities for inclusion in the study. For female patients, it was imperative to maintain menstrual function. The degree of liver fibrosis was established based on 2D shear wave elastometry in SWE mode. The patients included to study have liver fibrosis F1 - F3 according to METAVIR. Determination of bone mineral density was performed using a DEXXUM T X-ray densitometer by dual energy absorptiometry. Results and discussion. In patients with fibrotic changes in the liver, a decrease in bone mineral density was found in 49.3%. Osteopenia and osteoporosis in women occurred in 47.4%, which was significantly more frequent than among men – 26.8%, p = 0.014. There was a significant relationship between the duration of the presence of steatohepatosis and the incidence of osteoporosis and osteopenia: among patients with a duration of steatohepatosis of less than 7 years, a decrease in bone mineral density was noted in 62.5% of cases, and with a duration of more than 7 years – already in 89.3% of patients (χ2 = 5.5; p = 0.011). Among patients with liver fibrosis F3 METAVIR, a decrease in bone mineral density was observed in 85.7%. Among patients with liver fibrosis F1-2 METAVIR, osteoporosis and osteopenia were found in 46% (p = 0.0008). In patients with METAVIR F3 fibrosis, osteoporotic changes were observed in 85.2% of cases. An isolated decrease in the mineral density of the lumbar spine was diagnosed in 12 (30.7% of patients with a decrease in bone mineral density). In 9 patients (23%), there was a combination of a decrease in mineral density of the spine with a decrease in mineral density of the hip, the mean T score of the vertebrae was -2.5±0.2, the mean T score of the femoral neck was 2.1±0.3. In patients with F1-F2 fibrosis METAVIR, bone mineral density decreased mainly to the level of osteopenia, isolated osteopenia of the lumbar spine was diagnosed in 18 (46%) patients, there was no decrease in hip mineral density in this group of patients, the mean criterion of T vertebrae was 1.5±0.15. Conclusion. Patients with steatohepatosis and hepatic fibrosis are characterized by a high prevalence of osteoporosis and osteopenia. The risk of developing this complication is higher in patients with fibrosis stages F3 METAVIR; the decrease in bone mineral density in patients with steatohepatosis and liver fibrosis is influenced not only by population risk factors, but also by the duration of the presence of steatohepatosis; the severity of liver fibrosis METAVIR affects the localization of changes in the bone tissue. In patients with degrees of fibrosis F1 - F2, trabecular bone tissue is affected, and in patients with degrees of fibrosis F3, trabecular and cortical bone tissue is equally affected

SummaryImpedance plethysmography (IPG) has high sensitivity and specificity in patients with symptomatic deep vein thrombosis (DVT) while it fails to detect asymptomatic DVT. The aim of this study was to determine whether the features of thrombi such as location, size and occlusiveness could explain the different accuracy of IPG in symptomatic and asymptomatic DVT patients. One-hundred and seventeen consecutive outpatients with a clinical suspicion of DVT and 246 consecutive patients undergoing hip surgery were admitted to the study. In symptomatic patients IPG was performed on the day of referral, followed by venography, while in asymptomatic patients IPG was performed as a surveillance programme, followed by bilateral venography.A venography proved DVT was observed in 37% of the symptomatic patients and 34% of the asymptomatic limbs. A significantly higher proportion of proximal DVTs was found in symptomatic patients than in asymptomatic patients (78% vs 46%; p = 0.001). The mean Marder score, taken as an index of thrombus size, was significantly higher in symptomatic patients than in asymptomatic patients (19.0 vs 9.6; p = 0.0001). A significantly higher proportion of occlusive DVTs was observed in symptomatic than in asymptomatic patients (69% vs 36%; p = 0.001).We conclude that the unsatisfactory diagnostic accuracy of IPG in asymptomatic DVT is due to the high prevalence of distal, small and non occlusive thrombi. Such thrombi are unlikely to cause a critical obstruction of the venous outflow and therefore to produce a positive IPG.

SummaryThe plasma tissue factor (TF) concentration was correlated to factor VII concentration (FVIIag) and factor VII activity (FVIIc) in 498 healthy volunteers ranging in age from 17 to 64 years. Immunoassays using monoclonal antibodies (mAbs) were developed for the determination of TF and FVIIag in plasma. The mAbs and the test systems were characterized. The mean value of the TF concentration was 172 ± 135 pg/ml. TF showed no age- and gender-related differences. For the total population, FVIIc, determined by a clotting test, was 110 ± 15% and the factor VIlag was 0.77 ± 0.19 μg/ml. FVII activity was significantly increased with age, whereas the concentration demonstrated no correlation to age in this population. FVII concentration is highly correlated with the activity as measured by clotting assay using rabbit thromboplastin. The ratio between FVIIc and FVIIag was not age-dependent, but demonstrated a significant difference between men and women. Between TF and FVII we could not detect a correlation.

SummaryThe plasminogen content was determined by a casein method in plasma and serum from 20 normal volunteers. The mean plasminogen content was found to be 10.1 ACU (the arbitrary caseinolytic unit defined in such a way that using a 3% casein solution and a digestion time of 20 min. at 37°C, 10 ACU gave an extinction of 0.300). No difference between serum and plasma regarding the plasminogen content was found.Plasminogen was determined in drained and drained plus washed clots prepared from 2 ml plasma. The highest values found in the drained clots were 0.9 ACU/clot and 0.2 ACU/clot in the drained plus washed clots.Plasminogen was also determined in drained and drained plus washed clots prepared from plasma with added purified plasminogen. The plasminogen was recovered in the washing fluid. According to these tests, then, purified added plasminogen is washed out of the clots.The plasminogen content of 20 thrombi obtained post mortem was also determined. The mean value was found to be 0.7 ACU/cm thrombus. Judging from our results, the “intrinsic clot lysis theory” is not the main mechanism of clot dissolution.

ABSTRACT An improved method for the determination of serum triiodothyronine (T3) has been developed. After addition of a tracer amount of the hormone, T3 was extracted from 1 ml serum under conditions of pH and ionic strength which favoured T3 extraction (89%) over thyroxine (T4) extraction (58%). Chromatography of the extracted material on Sephadex LH-20 separated T3 completely from residual T4. The T3 eluate was dried, then re-dissolved in 0.5 ml NaOH 0.04 n. To 0.2 ml duplicate aliquots, a standard amount of TBG was added for the competitive protein analysis. After one hour incubation at 4°C, separation of bound from free T3 was achieved on small Sephadex G-25 columns. Overall recovery was 67 ± 10.8% and correction for the loss was made. The solvent blank was 37 ± 27 (sd) ng/100 ml. Accuracy of measurement of known quantities of T3 added to serum was 98.4%. The coefficient of variation within the assay was 6.2% and between the assays it was 11.4%. The limit of detection (0.1 ng) corresponded to a concentration of 25 ng/100 ml. T4 added to serum did not interfere with T3 determination until high non-physiological values were reached. The mean ± sd serum T3 in 54 euthyroid subjects was 153 ± 58 ng/100 ml and in 24 hyperthyroid patients it was 428 ±186 ng/100 ml; 4 out of the 24 hyperthyroid values were within 2 sd of the mean euthyroid group. All the values found in the euthyroid group were well above the limit of detection of the method.

The estimation of uncertainty of such averaging requires knowledge of probability distribution of the function form of their calculations. The developed solution, leading to the recurrent determination of the probability distribution function for the estimation of the mean value of noise levels and its variance, is shown in this paper.
