Cyanobacteria toxins and toxin producers in nine drinking water reservoirs in Taiwan

Cyanobacteria are present in many drinking water reservoirs in the world, and some of them may produce microcystins, anatoxin-a and other natural toxins: In this study, two groups of algal toxins: microcystins and anatoxin-a, and associated toxin producers were investigated. Water samples from nine major drinking water reservoirs and seven associated water purification plants were collected. An HPLC and an LC/MS were employed to detect the concentrations of five microcystins (microcystins-LR, RR, YR, LW, LF) and anatoxin-a. Molecular biotechnology methods were used to sequence Microcystis sp. gene from the cyanobacteria bloom from two of the reservoirs. Monitoring results suggested that microcystins were present in all the drinking water reservoirs studied, and some of them had concentration higher than the WHO guideline of microcystins-LR (1 μg/L). Unlike microcystins, anatoxin-a was only found in four reservoirs, mostly in King-Men island. After comparing the DNA sequence with existing cyanobacteria reference database, Microcystis aeruginosa and other Microcystis sp. were confirmed present in the two reservoirs tested.

AbstractThis study aimed to determine the quality of drinking water supplied in different types of food stalls in Jashore Municipality, Bangladesh. A total of 35 water samples were collected from different tea stalls, street side fast food stalls, normal restaurants and well-furnished restaurants. The water quality was evaluated by determining the distinct physical, chemical and biological parameters. The results revealed that the water used in the food stalls and restaurants for drinking purpose was in desired quality in terms of turbidity, electrical conductivity, pH, total dissolved solids, nitrate (NO3−), sulfate (SO42−), phosphate (PO43−), chloride (Cl−), sodium (Na) and potassium (K) concentrations. The values were within the permissible limit proposed by the Bangladesh Bureau of Statistics and the World Health Organization. Concentrations of calcium (Ca) and magnesium (Mg) found in several samples were higher than the World Health Organization standard. Iron (Fe) concentrations were higher than the permissible limit of the World Health Organization. Only 46% exceeded the permissible limit of Bangladesh Bureau Statistics. The threatening result was that the samples were contaminated by fecal coliform, indicating that the people of Jashore Municipality may have a greater chance of being affected by pathogenic bacteria. The drinking water provided in the street side fast food stalls was biologically contaminated. The findings demonstrate that the drinking water used in food stalls and restaurants of Jashore Municipality did not meet up the potable drinking water quality standards and therefore was detrimental to public health.

This protocol outlines the all the steps necessary to become a GenomeTrakr data contributor. GenomeTrakr is an international genomic reference database of mostly food and environmental isolates from foodborne pathogens. The data and analyses are housed at the National Center for Biotechnology Information (NCBI), which is a database freely available to anyone in the world. The Pathogen Detection browser at NCBI computes daily cluster results adding the newly submitted data to the existing phylogenetic clusters of closely related genomes. Contributors to this database can see how their new isolates are related to the real-time foodborne pathogen surveillance program established in the USA and a few other countries, and at the same time adding valuable new data to the reference database. ------ Although originally published as a Chapter in Methods and Protocols, Foodborne Bacterial Pathogens, the protocol has since been adapted and split into four separate protocols all of which are contained in this collection.

<p>Forested watersheds are critical sources of the majority of the world’s drinking water. Almost one-third of the world’s largest cities and two-thirds of cities in the United States (US) rely on forested watersheds for their water supply. These forested regions are vulnerable to the increasing incidence of large and severe wildfires due to increases in regional temperatures and greater accumulation of fuels. When wildfires occur, increases in suspended sediment and organic carbon can negatively affect aquatic ecosystem health and create many costly challenges to the drinking water treatment process. These effects are often largest in the first year following a wildfire. While past research has shown the likelihood of source water impacts from wildfire, the magnitude of effects remains uncertain in most regions. In our study, we will quantify the projected short-term effects of three large (>70,000 ha) wildfires on key water quality parameters (sediment and organic carbon) in two important forested source watersheds in the Cascade Range of Oregon, US. We calibrated and validated a modified Soil and Water Assessment Tool (SWAT) to simulate streamflow, sediment loads and transport, as well as in-stream organic carbon processes for a historical period prior to wildfire. The calibrated model parameters were then modified based on literature values and burn severity maps to represent post-fire conditions of the three large wildfires. The parameter adjustments for simulating wildfire will be validated with post-fire water quality field samples from the wildfires. We will present estimations of future water quality impacts in the burned watersheds under different precipitation conditions at a daily scale for the first year following the wildfires, which will provide testable hypotheses. Additionally, we will determine catchment characteristics most critical in determining the post-fire water quality response. This work will help predict the magnitude of effects from these historic wildfires, which can inform forest and drinking water management decision making.</p>

<p>Although cyanobacterial blooms and cyanotoxins represent a worldwide-occurring phenomenon, there are large differences among different countries in cyanotoxin-related human health risk assessment, management practices and policies. While national standards, guideline values and detailed regulatory frameworks for effective management of cyanotoxin risks have been implemented in many industrialized countries, the extent of cyanobacteria occurrence and cyanotoxin contamination in certain geographical regions is under-reported and not very well understood. Such regions include major parts of tropical West and Central Africa, a region constisting of more than 25 countries occupying an area of 12 million km<sup>2</sup>, with a total population of 500 milion people. Only few studies focusing on cyanotoxin occurrence in this region have been published so far, and reports dealing specifically with cyanotoxin contamination in drinking water are extremely scarce. In this study, we report seasonal data on cyanobacteria and microcystin (MC) contamination in drinking water reservoirs and adjacent treatment plants located in Ghana, West Africa. During January-June 2005, concentrations of MCs were monitored in four treatment plants supplying drinking water to major metropolitan areas in Ghana: the treatment plants Barekese and Owabi, which serve Kumasi Metropolitan Area, and the plants Kpong and Weija, providing water for Accra-Tema Metropolitan Area. HPLC analyses showed that 65% samples of raw water at the intake of the treatment plants contained intracellular MCs (maximal detected concentration was 8.73 µg L<sup>-1</sup>), whereas dissolved toxins were detected in 33% of the samples. Significant reduction of cyanobacterial cell counts and MC concentrations was achieved during the entire monitoring period by the applied conventional water treatment methods (alum flocculation, sedimentation, rapid sand filtration and chlorination), and MC concentration in the final treated water never exceeded 1 µg L<sup>-1</sup> (WHO guideline limit for MCs in drinking water). However, cyanobacterial cells (93-3,055 cell mL<sup>-1</sup>) were frequently found in the final treated water and intracellular MCs were detected in 17% of the samples (maximal concentration 0.61 µg L<sup>-1</sup>), while dissolved MCs were present in 14% of the final treated water samples (maximal concentration 0.81 µg L<sup>-1</sup>). It indicates a borderline efficiency of the water treatment, thus MC concentrations in drinking water might exceed the WHO guideline limit if the treatment efficiency gets compromised. In addition, MC concentrations found in the raw water intake might represent significant human health risks for people living in areas with only a limited access to the treated or underground drinking water.</p>
