BACKGROUND The prevalence of obesity has increased substantially over recent decades and is associated with considerable health inequalities. Although the causes of obesity are complex, key drivers include overconsumption of highly palatable, energy-dense, and nutrient-poor foods, which have a profound impact on the composition and function of the gut microbiome. Alterations to the microbiome may play a critical role in obesity by affecting energy extraction from food and subsequent energy metabolism and fat storage. OBJECTIVE We report the study protocol and recruitment strategy of the PRedictors linking Obesity and the gut MIcrobiomE (PROMISE) study, which characterizes the gut microbiome in 2 populations with different metabolic disease risk (Pacific and European women) and different body fat profiles (normal and obese). It investigates (1) the role of gut microbiome composition and functionality in obesity and (2) the interactions between dietary intake; eating behavior; sweet, fat, and bitter taste perception; and sleep and physical activity; and their impact on the gut microbiome, metabolic and endocrine regulation, and body fat profiles. METHODS Healthy Pacific and New Zealand (NZ) European women aged between 18 and 45 years from the Auckland region were recruited for this cross-sectional study. Participants were recruited such that half in each group had either a normal weight (body mass index [BMI] 18.5-24.9 kg/m2) or were obese (BMI ≥30 kg/m2). In addition to anthropometric measurements and assessment of the body fat content using dual-energy x-ray absorptiometry, participants completed sweet, fat, and bitter taste perception tests; food records; and sleep diaries; and they wore accelerometers to assess physical activity and sleep. Fasting blood samples were analyzed for metabolic and endocrine biomarkers and DNA extracted from fecal samples was analyzed by shotgun sequencing. Participants completed questionnaires on dietary intake, eating behavior, sleep, and physical activity. Data were analyzed using descriptive and multivariate regression methods to assess the associations between dietary intake, taste perception, sleep, physical activity, gut microbiome complexity and functionality, and host metabolic and body fat profiles. RESULTS Of the initial 351 women enrolled, 142 Pacific women and 162 NZ European women completed the study protocol. A partnership with a Pacific primary health and social services provider facilitated the recruitment of Pacific women, involving direct contact methods and networking within the Pacific communities. NZ European women were primarily recruited through Web-based methods and special interest Facebook pages. CONCLUSIONS This cross-sectional study will provide a wealth of data enabling the identification of distinct roles for diet, taste perception, sleep, and physical activity in women with different body fat profiles in modifying the gut microbiome and its impact on obesity and metabolic health. It will advance our understanding of the etiology of obesity and guide future intervention studies involving specific dietary approaches and microbiota-based therapies. IntroductionChildhood and adolescence are the period of rapid physical and cognitive growth and development, requiring adequate nutrition. Malnutrition in the form of undernutrition or micronutrient deficiency or overweight/obesity affects the health, cognition and educational achievement of this age group. The objective of this study is to assess the prevalence of calcium, iron, zinc, selenium, folate, vitamin A, 25 hydroxy vitamin D and vitamin B12 deficiencies in the serum and haemoglobin, ferritin and lead levels and its association with reported dietary intake and cognitive abilities, in urban school going children aged 6–16 years in 10 cities of India.Methods and analysisA multicentric cross-sectional study will be conducted to recruit 2400 participants (240 per site) across India. Participants will be selected using random sampling and will be categorised into age groups of 6–11 years and 12–16 years, with equal distribution. Data on socioeconomic status, anthropometric measures and 3-day dietary intake and cognitive performance will be collected. Blood samples will be collected for biochemical analysis of micronutrients. Findings will estimate the prevalence of micronutrient deficiencies and their association with dietary habits and cognitive functioning.Ethics and disseminationStudy protocol has been reviewed and approved by institutional ethics committee of all 10 participating sites. Results will be shared and published in a peer-reviewed journal, so that the findings will be helpful for the stakeholders in planning nutritional interventions for targeted groups.Trial registration numberCTRI/2019/02/017783.

Background: Evidence shows that diet is associated with physical activity. However, there has been a lack of studies addressing it in a population of older adults. Therefore, the main purpose of this study was to explore associations between various frequencies of dietary intake and physical activity in older adults. Methods: In this cross-sectional study, the participants were 810 older adults aged ≥85. Frequency of dietary intake and the level of physical activity were assessed using the Elderly Diet Index score and International Physical Activity questionnaire. Mutual associations were examined using generalized estimating equations with a logistic regression model. Results: ”Optimal” intake of fish and seafood (OR (odds ratio) = 1.40; 95% CI (95 percent confidence interval) 1.01 to 2.00), fruits (OR = 2.10; 95% CI 1.45 to 3.02), legumes (OR = 1.73; 95% CI 1.19 to 2.50), olive oil (OR = 1.83; 95% CI 1.09 to 3.08) and bread (OR = 4.62; 95% CI 3.05 to 6.99) and the total Elderly Diet Index score (OR = 4.99; 95% CI 3.20 to 7.70) were associated with ”sufficient” physical activity. When all dietary components were entered simultaneously into the model, ”optimal” intakes of meat (OR = 1.73; 95% CI 1.10 to 2.71), fish and seafood (OR = 2.26; 95% CI 1.46 to 3.51), cereals (OR = 1.75; 95% CI 1.02 to 3.25), fruits (OR = 1.52; 95% CI 1.02 to 2.26), legumes (OR = 1.48; 95% CI 1.10 to 1.93), and bread (OR = 5.14; 95% CI 3.24 to 8.15) were associated with ”sufficient” physical activity. Conclusions: Our study shows that the total Elderly Diet Index score is the strongest predictor associated with ”sufficient” physical activity in a population of older adults. Thus, policies aiming to improve overall diet in order to achieve higher levels of physical activity are warranted.

Background: Physical inactivity and overweight are leading risk factors for global mortality, responsible for 6% and 5% of deaths worldwide due to chronic non communicable diseases. It is well established that regular moderate or vigorous-intensity exercise lowers the risks and symptoms associated with the co-morbidities of obesity. The stress involved in meeting responsibilities of becoming a physician may adversely affect the exercise habits of students. So, the current study aimed to study physical activity and its relation with body composition among under-graduate medical students.Methods: It was a cross-sectional study among 282 medical students. International Physical Activity Questionnaire (IPAQ) Short form was used to measure total physical activity (PA) and body composition parameters including Waist circumference, WHR, BMI, Body Fat% were assessed and analyzed using SPSS version 21.Results: Out of 282 medical students 33%, 45.3%, &21.3% were involved in low, moderate and high physical activity. ANOVAs between different levels of physical activities according to IPAQ categories showed significant difference for Waist Hip Ratio (F:3.8; p=0.02) and Body Fat percentage (F=5.7; p=0.00). There was an inverse and statistically significant correlation between physical activity and Waist circumference, Body Fat% and Waist –Hip Ratio.Conclusions: Medical Students involved in higher level of physical activity have significantly lower body Fat% level and WHR as compared to those belonging to lower and moderate activity group thus there is a need to actively promote physical activity among medical students.
