This research was aimed to investigate the pathogenicity of Vibrio fluvialis 24SK in humpback grouper (Cromileptes altivelis) based on its Lethal Dosage 50 (LD50). V. fluvialis 24SK was isolated from ren of humpback grouper cultured in floating net cage at Brackishwater Aquaculture Development Center (BADC) Situbondo, with vibriosis signs. The bacterium was cultured in Tryptone Soy Broth (TSB) medium dissolved in trisalt solution (KCl, 0.75 g/l; MgSO4.7H2O; 14.2 g/l; NaCl, 18.4 g/l), incubated at 300C for 24 h. Infection was carried out by interperitoneal injection to humpback grouper (8-9 cm of total length) at 102, 104, 106, and 108 cfu/fish. Control fishes were injected with 0.2 ml trisalt solution. Disease sign and mortality of fishes were observed every eight hour for 40 days. LD50 was calculated based on Dragstedt Behrens method (Hubert, 1980). Result indicated that infection of the bacteria at 106 and 108 cfu/fish caused sub-acute disease signs, such as haemorhagic on operculum, base of fins (pinnae pectorales, pinnae abdominales, pinna analis), and also head and abdomen, while infection at 102 and 104 cfu/fish caused chronic disease signs, such as haemorhagic on fins base which was followed by necrotic on fins and skin tissue in prolonged time. Histopathologically, infection of the bacteria caused atrophy on the gills, infiltrations of lymphocyte, heterofel and plasma cell on the gills and fins base, vacuolar degeneration on the liver, and also present the bacteria colony on the fins base and intestine tissues. V. fluvialis 24SK has LD50 at (1,1±0,5)x107 cfu/fish.

This research was conducted to isolate, identify, and characterize pathogenic Vibrio spp., causative agents of vibriosis in grouper at Brackishwater Aquaculture Development Center (BADC), Situbondo. Twenty-nine isolates were isolated from wound, gills, eyes, liver, and ren of grouper with vibriosis on Thiosulphate Citrate Bile Sucrose Agar (TCBSA) medium. Koch Postulate Test was conducted to determine pathogenic Vibrio spp., by intraperitoneal injection to humpback grouper (Cromileptes altivelis) (9-10 cm of total length) at 106 cells/fish in triplicates. Results indicated that the pathogenic Vibrio spp., causative agents of vibriosis in grouper at BADC, Situbondo were identified to be V. alginolyticus, V. anguillarum, V. metchnikovii, V. vulnificus, V. fluvialis, V. furnisii, and V. parahaemolyticus. Infection of the pathogenic Vibrio spp. caused acute mortality within 17-46 hours with specific disease signs like haemorhagic on fins (pinnae pectorales, pinnae abdominales, pinna analis) and also on the body. These results suggested that vibriosis should be monitored and controlled properly and quickly.

AbstrakPelatihan industri telah dilakukan untuk mempelajari dan memahami teknik budidaya dari Kerapu Cantang (Epiphenelus sp.). Teknik budidaya kerapu Cantang merupakan salah satu faktor yang akan mempengaruhi tingkat pertumbuhannya. Pengambilan sampel ikan dilakukan selama 21 hari di Pusat Pengembangan Budidaya Perikanan Air Payau, Situbondo, Jawa Timur. Hasil penelitian ini menunjukkan bahwa pertumbuhan kerapu Cantang meningkat signifikan sebesar 2,29 gram/hari. Pengelolaan pakan dan kualitas air yang baik dianggap sebagai faktor utama yang berpengaruh pada pertumbuhan Kerapu Cantang selama proses budidaya. AbstractAn industrial training has been conducted to learn and understand the culture techniques of Cantang Grouper (Epiphenelus sp.). The culture technique of Cantang Grouper is one of the factors that affect its growth. The fish sampling was carried out for 21 days at Brackish Water Aquaculture Development Center, Situbondo, East Java. The result of this study demonstrated that growth of Cantang Grouper was increased significantly of 2.29 gram/day. Good management of feed and water quality are considered as the main factors that gave effect on the growth of Cantang Grouper during the cultivation process.

Some benthic minute amphipods have been found at abalones’rearing tank, at Lombok Marine Aquaculture Development Center, Sekotong, West Nusa Tenggara, Indonesia. The minute amphipod community was found along with Gracillaria verrucosa which was harvested from Sekotong fishpond. There is some vigilance that it might be a pest that interferes the production of abalones. This research aims to reveal the type of the minute amphipod at the rearing tank. Morphological characters were examined to identify the Amphipod. Some slide was made to recognise and identify minute apparatus of the Amphipod. A hundred of Amphipod was reared in a small container to see their immediate behavior such as feeding or reproduction or other/s or its product. Some video were used to record for 30” to 60” to see the behavior of Amphipods. Result show that at least four types of Gammaridea present at the abalone culture pond. Some Amphipod was descent to abalone for some time, hop and perch to Gracillaria. Some Amphipods build a nest from pieces of Gracillaria in a tube form. There are two types of nest, firstly, small tube which were attached to the substrate and secondly, bigger tube which were float with the Gracillaria.

ABSTRACT Mycobacterium marinum causes a systemic tuberculosis-like disease in a large number of poikilothermic animals and is used as a model for mycobacterial pathogenesis. In the present study, we infected zebra fish (Danio rerio) with different strains of M. marinum to determine the variation in pathogenicity. Depending on the M. marinum isolate, the fish developed an acute or chronic disease. Acute disease was characterized by uncontrolled growth of the pathogen and death of all animals within 16 days, whereas chronic disease was characterized by granuloma formation in different organs and survival of the animals for at least 4 to 8 weeks. Genetic analysis of the isolates by amplified fragment length polymorphism showed that M. marinum strains could be divided in two clusters. Cluster I contained predominantly strains isolated from humans with fish tank granuloma, whereas the majority of the cluster II strains were isolated from poikilothermic species. Acute disease progression was noted only with strains belonging to cluster I, whereas all chronic-disease-causing isolates belonged to cluster II. This difference in virulence was also observed in vitro: cluster I isolate Mma20 was able to infect and survive more efficiently in the human macrophage THP-1 and the carp leukocyte CLC cell lines than was the cluster II isolate Mma11. We conclude that strain characteristics play an important role in the pathogenicity of M. marinum. In addition, the correlation between genetic variation and host origin suggests that cluster I isolates are more pathogenic for humans.

AbstractBackgroundSystematic analysis of the determinants of choice of a treatment modality aids to the understanding of decision process of healthcare utilization. The revealed preference of a single modality may differ according to the nature of disease. Existing studies have not integrated possible causal factors in a model with respect to diseases. This study identifies major determinants and formulates their integral effect on choice of a particular modality on acute and chronic diseases in accordance to socio-behavioural model.MethodologyA cross-sectional study on 300 samples using a 30-point questionnaire, developed in Likert scale and dichotomous scale. Possible determinants are tested on choice of CAM in case of acute disease and of chronic disease separately.ResultsRevealed single modality treatment preference (of CAM) varies widely between acute disease (13%) and chronic disease (58.67%). Bivariate associations are significant for gender (For, overall CAM preference, p=0.001, acute disease, p<0.001, chronic disease, p=0.024), Disease burden (overall and chronic: p<0.001, acute: p=0.008) and previous CAM usage (overall and chronic: p<0.001, acute: p=0.016). Social factor individually has significant influence on choosing CAM both acute (OR=1.096, p<0.001) and chronic disease (OR=1.036, p<0.001). Ideation of philosophical need factor, guided by philosophical congruence with CAM (OR=1.047, p<0.001) is a novel finding of this study. While with multiple logistic regression male gender (p=0.03), social factor (p<0.001), perception of CAM efficacy (p=0.02) and negative ideation about CAM cost-effectiveness (p=0.002) are found to be important in Acute disease; choosing CAM in chronic disease is guided by female gender (p=0.001), making decision in-group (p=0.001), low disease burden (p<0.001), philosophical need factor (p=0.001), and perception of CAM efficacy (p<0.001).ConclusionDemographic, social, cognitive and philosophical factors are important determinants of choosing CAM as a treatment modality over conventional medicine, but they act differently on CAM preference in acute and chronic diseases.

AbstractThis study was carried out to investigate the effect of Asian seabass (Lates calcarifer) culture in floating net cages at the Brackishwater Aquaculture Development Center, Situbondo. Asian seabass were fed with commercial fish pellet (KPA) and fresh fish (trash fish) for 21 days. Seabass fed with trash fish and commercial fish pellet displayed the growth response to the average weight and length. The fish grew from an initial weight of 230g, 238g, and 244g in 21 days and the average length of seabass changed from 23 cm, 25 cm and 28 cm respectively. The fish were fed with 3-7% body weight of fish and the growth was monitored in 21 days. The results showed that trash fish can increase the significant growth response in Asian seabass. AbstrakStudi ini dilaksanakan untuk mengetahui pengaruh budidaya kakap putih Asia (Lates calcarifer) di keramba jarring apung di pusat pengembangan budidaya air payau, Situbondo. Kakap putih Asia diberi pelet ikan komersial (KPA) dengan ikan segar (limbah ikan) selama 21 hari. Kakap putih makan dengan limbah ikan dan pelet ikan komersial menunjukkan respon pertumbuhan rata-rata dari berat dan panjang. Pertumbuhan ikan dari berat awal 230g, 238g dan 244g pada hari ke- 21 dan terjadi perubahan panjang rata-rata kakap putih masing-masing menjadi 23 cm, 25 cm dan 28 cm. Ikan diberi makan dengan 3-7% dari berat badan ikan dan pertumbuhan diamati dalam 21 hari. Hasil penelitian menunjukkan bahwa limbah ikan dapat meningkatkan respon pertumbuhan yang signifikan di kakap putih Asia.

We investigated 17 patients (12 males and 5 females, ages 2 to 57 years old) with posttransfusion non-A, non-B hepatitis to determine relationships between clinical courses and hepatitis C virus (HCV) markers. The patients were grouped according to time course of abnormal serum alanine aminotransferase (ALT) levels into three categories (chronic biochemical disease, biochemically resolved chronic disease, and acute disease). Latest serum samples (1.3 to 10.8 years after blood transfusion) were used to detect antibodies against C100–3 antigen (anti-HCV) by enzyme-linked immunosorbent assay and HCV sequences by polymerase chain reaction (PCR) assay. Of the 17 patients, 13 patients (76.5%) were anti-HCV positive and 8 patients (47.1%), including one anti-HCV negative case, were positive for HCV RNA. In total, 14 patients (82.4%) were positive for either HCV markers. With respect to clinical course, HCV RNA was detected in six of eight patients (75%) with chronic biochemical disease, and in two of five patients (40%) with biochemically resolved chronic disease. HCV RNA was not detectable in convalescent sera from four patients with acute disease. These results show that there is a relationship between clinical status and HCV viremia, but that normal liver function tests do not always represent the clearance of the virus. Viremia in two patients with normal ALT level suggests that hepatitis is not only caused by viral cytopathic effects, but also by immunologic reactions against virus- infected cells. Thus, PCR is useful in determining the persistence of HCV infection as well as to diagnose anti-HCV negative HCV infection.

Abstract Shirt-sleeve magnification (holding a slide over a white sleeve) and low-power magnification serve as useful adjuncts in the general categorization of noninfectious medical lung disease. This article divides medical lung disease into chronic and acute, where the temporality is determined first by clinical circumstances and then confirmed by histopathology. The low-power patterns of various lung diseases overlap, sometimes greatly. Nevertheless, classic examples of chronic disease can be sorted as linear, lobular filling, nodular dispersed, nodular lymphangitic, or cystic patterns at shirt-sleeve or low-power magnification. Classic examples of acute disease generally produce a solidifying pattern at shirt-sleeve or low-power magnification, which can be followed by a determination as to whether alveolar filling is principally fibrotic or principally fluid or cells at higher magnification. This article gives a simple system for the categorization of medical lung disease by this approach, with an emphasis on the most common diseases to be encountered in a general surgical pathology practice. In our experience, this system also proves useful in arriving at some therapeutic decisions.
