Objective: The objective of this study was to determine if there were any harmful effects of monosodium glutamate (MSG) on the liver of Wistar albino rats chronically at three different doses, namely, low, mid, and high doses equivalent to human consumption doses in developing countries. Methods: The Wistar albino rats (n=24) were divided into four groups, namely control, Low dose MSG (180 mg/kg), Mid dose MSG (360 mg/kg), and High dose MSG (720 mg/kg). At the end of the experimental period (120 days), animal blood was collected retro-orbitally to analyze the liver enzymes such as aspartate transaminase (AST), alanine transaminase (ALT), alkaline phosphatase (ALP), Total protein, Albumin, and Total Bilirubin in blood serum. Lipid profiles, namely, Triglycerides, low-density lipoprotein (LDL), high-density lipoprotein (HDL), and Total cholesterol were subjected to analysis using blood serum. Results: Significant increase (p<0.05) in AST, ALT, ALP, and total bilirubin in serum of MSG induced low, mid, and high dose groups when compared to control group were recorded. There was a significant increase (p<0.05) in LDL, decrease in HDL, increase in total cholesterol and triglycerides of MSG-induced animal groups. Conclusion: The effects of MSG on serum liver enzymes and lipid profiles in this present animal study were not severely alarming even though the dosage was chronic which opens further discussion on the controversies revolving around MSG.

Objective: To evaluate the liver protective effect of Pancha Lavana Dravagam (PLD) against Paracetamol induced hepatotoxicity in wistar albino rat models. Methods: The hepatoprotective activity of PLD was evaluated using paracetamol induced liver damage in rats. Wistar albino rats were divided into five groups of six animals each. Paracetamol 1gm/kg bw, p.o. was given to produce liver toxicity. The normal control was given the vehicle (water 1ml/kg bw, p.o). Two test groups with PLD 1ml/kg, 2ml/kg bw, p.o. were tested for hepatoprotective potential. Silymarin 50mg/kg bw, p.o. was given as standard drug. All these drugs were administered for 7 days. On 8th day, the animals were sacrificed and blood was collected from retro-orbital plexus and analyzed for serum enzymes like Serum Glutamate Pyruvate Transaminase (SGPT), Serum Glutamate Oxaloacetate Transaminase (SGOT), Serum Alanine Phosphate (ALP), Total Bilirubin, Total Proteins and liver was excised for histopathological analysis. Results: In toxicant control group, paracetamol produced liver toxicity due to decrease in glutathione (GSH) by oxidative stress and mitochondrial dysfunction of hepatic cells. It resulted in an increase of serum liver enzymes like SGPT, SGOT, ALP and Total Bilirubin. This increased serum liver enzymes were reduced significantly in the test drug PLD treated groups and Standard group. The histology of liver tissues was also improved in PLD treated groups when compared to the toxicant group. Conclusion: Since, no scientific evidence is available to claim the hepatoprotective effect of PLD, in vivo studies were conducted. It demonstrated that it has a potent hepatoprotective effect against the paracetamol induced hepatotoxicity by suppression of the reactive oxygen species and increasing the anti-oxidant glutathione in liver cells.

Objective: The study was intended to explore whether Monosodium glutamate (MSG) induces oxidative stress on the liver of Wistar albino rats when fed chronically at three different doses, namely, low, mid, and high doses identical to human consumption doses in growing countries. Methods: The acclimatized Wistar albino rats (n=24) were randomly selected and grouped into four groups, namely Control, Low dose MSG (180 mg kg), Mid dose MSG (360 mg/kg), and High dose MSG (720 mg/kg). The animals were orally administered MSG for 120 days. After completion of the experimental period (120 days), euthanized animal liver was homogenized to investigate the oxidative stress marker enzymes such as Superoxide Dismutase (SOD), Glutathione Peroxidase (GPx), Catalase (CAT), and Myeloperoxidase (MPO). Results: The MPO showed a significant increase (p<0.05) in liver homogenate of all MSG induced groups when compared to control group. The SOD, CAT, and GPx activity deteriorated (p<0.05) in monosodium induced groups contrasting to the control group. Conclusion: The effects of MSG on oxidative stress markers on liver homogenate in the current study exhibited erratic abnormal changes in oxidative stress markers of monosodium induced groups which contemplate the harmful effects of MSG consumed chronically. The further studies should confirm the genetic basis of oxidative stress damage and transform the safety regulations of MSG consumption throughout the world.

Dyslipidemia and increases in plasma homocysteine usually occur at end-stage renal disease; both are recognized as risk factors for atherosclerosis. Folate administration reduces homocysteine concentration. In this study we determined the effect of a high dose of folic acid (40 mg intravenous injection three times a week) on plasma and red blood cell lipid profiles in twelve chronic renal failure patients on regular hemodialysis. Fasting blood samples were taken at the beginning of the study (baseline) and after 21, 42, and 64 days of treatment. Folic acid supplementation decreased plasma homocysteine. Plasma triglyceride levels decreased whereas polyunsaturated fatty acid values increased after 21 days; then they returned to baseline levels at the end of treatment. Total cholesterol and low-density lipoprotein (LDL) cholesterol were higher than those of the baseline during all the study, whereas high-density lipoprotein (HDL) cholesterol was reduced. In erythrocyte membranes, folic acid therapy enhanced cholesterol/phospholipid ratios and the fluorescence anisotropy of diphenyl-hexatriene. We conclude that large doses of folic acid produce a favorable effect, reducing plasma homocysteine levels and protecting patients from atherosclerosis. However, as this therapy induces significant alterations in both plasma and erythrocyte membrane lipid profiles, plasma lipid values should be controlled throughout the treatment of patients with renal failure.

The effects of atorvastatin and carboxymethylated β-glucan (CMG) on the lipoprotein-cholesterol (LP-C) and lipoprotein-triglyceride (LP-TG) fractions and subfractions at the early stage of murine hyperlipidemia, and its pleiotropic anti-inflammatory effects, were studied. Atorvastatin and CMG were administered in ICR male mice with acute lipemia induced with a single injection of poloxamer 407 (P-407). A novel small-angle X-ray scattering method for the determination of fractional and subfractional composition of LP-C and LP-TG was used. In P-407-treated animals, there was a drastic increase of total cholesterol and especially TG. Atorvastatin decreased both the total cholesterol and TG, but not to control levels. CMG primarily decreased TG and was not as potent as atorvastatin. P-407 increased atherogenic LDL-C (IDL-C and LDL1–3-C subfractions) and very low-density lipoprotein-C (VLDL-C) (VLDL1–2-C and VLDL3–5-C subfractions) fractions, with an increase of the total anti-atherogenic HDL-C fraction (HDL2-C subfraction). Atorvastatin treatment of lipemia was followed by a decrease in the total LP-C, total LDL-C (LDL1–3-C subfraction), and the LDL1–3-TG subfraction. Additionally, atorvastatin treatment resulted in an increase in the serum matrix metalloproteases activity both in control and P-407-treated mice. In general, high-dose atorvastatin therapy exerts its lipid-lowering and pleiotropic effects in the early stages of acute lipemia induced in mice by treatment with P-407.

Lipid lowering effect of polyherbal formulations using eight different plants was evaluated in triton and diet induced hyperlipidemic models of wistar albino rats. Formulations such as Tablet, Syrup and Suspension inhibited the elevation in serum cholesterol and triglyceride levels on Triton WR 1339 administration rats. The formulations at the same dose level significantly attenuated the elevated serum total cholesterol and triglycerides with an increase in high-density lipoprotein cholesterol in high-fat diet-induced hyperlipidemic rats. The standard drug Niacin showed slightly better effects. The treatment with herbal formulations produced 30-35 percentage improvement in oral glucose tolerance. Similarly all the formulations also reduced the elevated C-reactive protein which is a marker of Hyperlipidemia. In histopathological study it was found that treatment of polyherbal formulation significantly reduced the plaque size in aorta compared with HFD treated control group. The outcome of the study reveals the lipid lowering activity of polyherbal formulations in dyslipidaemic conditions by interfering with the biosynthesis of cholesterol and utilization of lipids.

Introduction: Hypertension and dyslipidemia are the two coexisting and synergizing major risk factors for cardiovascular diseases. The cellular constituents of blood affect the volume and viscosity of blood, thus playing a key role in regulating blood pressure. Overweight and obesity are key determinants of adverse metabolic changes including an increase in blood pressure. The aim of this study was to evaluate lipid profiles and hematological parameters in hypertensive patients at Debre Markos Referral Hospital, Northwest Ethiopia. Methods: Laboratory-based cross-sectional study was conducted in 100 eligible hypertensive patients at the hospital. The required amount of blood was withdrawn from the patients by healthcare professionals for immediate automated laboratory analyses. Data were collected on socio-demographic factors, anthropometric measurements, blood pressure, lipid profiles, and hematological parameters. Result: The mean serum levels of triglyceride, total cholesterol, and low-density lipoprotein were significantly higher than their respective cut-off values in the hypertensive patients. Besides, 54%, 52%, 35%, and 11% of the hypertensive patients had abnormal low-density lipoprotein, total cholesterol, triglyceride, and high-density lipoprotein levels, respectively. Higher levels of low-density lipoprotein, hemoglobin, and red blood cell count were observed in the hypertensive patients whose blood pressure had been poorly controlled than the controlled ones ( p < 0.05). Waist circumference had a significant positive association with the serum levels of total cholesterol and white blood cell count ( p < 0.05). Conclusion: Hypertensive patients had a high prevalence of lipid profile abnormalities and poorly controlled blood pressure which synergize in accelerating other cardiovascular diseases. Some hematological parameters such as red blood cell count are also increased as do the severity of hypertension.

Objective: The objective of this study is to assess the activity of Ulva lactuca polyphenols fraction in protecting the myocardial infarction induced by a high dose of isoproterenol. Methods: This study was carried out using Wistar albino rats divided into six groups. Group 1 was the normal group. Groups 2, 3, 4, 5, and 6 received isoproterenol (85 mg/kg) i.p. twice at an interval of 24 h on the 14th and 15th day. In addition to isoproterenol, Group 3 received verapamil (5 μmol/kg) as a single dose intravenously on the 14th day 10 min before isoproterenol and Groups 4, 5, and 6 received U. lactuca polyphenols fraction at the doses of 50, 100, and 200 mg/kg, respectively, for 14 days. On the 16th day, serum and heart samples were harvested from the animals and the cardioprotective and antioxidant activities were assessed by studying the levels of cardiac functional heart marker enzymes, lipid profile, reduced glutathione, and antioxidant enzymes. Results: U. lactuca polyphenols fraction, at the tested doses, restored the levels of all serum markers and enzymes (aspartate aminotransferase, alanine aminotransferase, creatine phosphokinase, lactate dehydrogenase, cholesterol, triglycerides, and reduced glutathione) and heart homogenate antioxidant markers (reduced glutathione, superoxide dismutase, catalase, and glutathione peroxidase) significantly, in dose-dependent manner. Conclusion: This study suggests that U. lactuca polyphenols fraction has a cardioprotective effect against isoproterenol-induced myocardial infarction and possess antioxidant activities.

Edible insects have emerged as an inexpensive alternative source of protein for reducing the burden of malnutrition worldwide. However, there is a dearth of evidence on its efficacy, and thus, the aim of this study was to investigate the effect of edible insect consumption on the nutritional status of female Wistar albino rats. The study assessed the subchronic effect of palm weevil larvae (PWL) and orange-fleshed sweet potato- (OFSP-) enriched biscuits (fortified biscuits (FB), plain biscuits (PB), biscuits fortified with PWL (PWB), and biscuits fortified with OFSP only (SPB)) as a model to predict the potential of PWL to improve the nutritional status of pregnant women in Ghana. Twenty-five female Wistar albino rats were randomly assigned to five experimental groups to receive one of the five feed supplements for 28 days. After which, the effects of treatment on haematological and biochemical parameters including lipid profile were assessed. No significant differences were observed with haematological (Hb) parameters. However, total cholesterol levels of the FB, PB, PWB, and SPB were significantly higher than in the N group. Apart from elevated total cholesterol concentrations, biscuits fortified with PWL had no adverse effects and can be a nutritious snack for maintaining acceptable HB levels.

Background The prevalence of smoking in adolescentstends to increase. Smoking is associated with a higher risk ofdyslipidemia.Objective To compare the lipid profiles of tobacco-smoking andnon-tobacco-smoking male adolescents.Methods We performed a cross- sectional study in three vocationalhigh schools in Yogyakarta from January to April 2011. Dataon smoking status, duration of smoking and number cigarettesconsumed per day were collected by questionnaires. We randomlyselected 50 male smokers and 50 male non-smokers as the studysubjects.Results Mean differences between smokers and non-smokerswere 44.5 (95%CI 28. 7 to 60.1) mg/dL for triglyceride levels; 8.0(95% CI 1.0 to 14.9) mg/dL for low density lipoprotein (LDL)cholesterol; 11.8 (1.1 to 22.4) mg/dL for total cholesterol and -5.7mg/dL (95% CI -8.8 to -2.6) for high density lipoprotein (HDL)cholesterol. Mean differences (95% CI) between smokers whohad engaged in smoking for > 2 years and those who had smokedfor :S:2 years were -18.1 (95% CI -33 .9 to -2.3) mg/dL for totalcholesterol; -49.4 (95% CI -67.2 to -3 1.5) mg/dL for triglycerides.Mean differences between those who smoked > 5 cigarettes/dayand :s:5 cigarettes per day were -18 .4 (95% CI -32.8 to -4.1) mg/dL for total cholesterol and -29.1 (95% CI -53.6 to -4.6) mg/dLfor triglycerides.Conclusion Smoking more than 5 cigarettes/day significantlyincreases total cholesterol, LDL cholesterol, and triglyceridelevels, as well as reduces HDL cholesterol levels; while smokingmore than 2 years significantly increases total cholesterol andtriglyceride levels
