Objective: Evaluate the antioxidant, phytochemicals and antibacterial screening of leaves extracts of Peristrophe bicalyculata.Methods: The antibacterial activity against the respiratory tract pathogens i.e., Staphylococcus aureus (MTCC 1144), Streptococcus pneumoniae (MTCC 655), Streptococcus pyogenes (MTCC 442), Pseudomonas aeruginosa (MTCC 2474) and Klebsiella pneumoniae (MTCC 4030) was examined by the method agar well diffusion and the minimum inhibitory concentration (MICs) was determined by the method of twofold serial dilution. Broad spectrum antibiotic erythromycin was used as positive control and dimethyl sulphoxide (DMSO) used as negative control. The qualitative method was adapted for phytoconstituents screening and antioxidant activity of plant extract was examined by DPPH free radical scavenging method.Results: The results showed that the chloroform (CHF) extract has a higher degree of antibacterial potency then the other extract. The zone of inhibition showed by chloroform extract against tested bacteria ranged between 9.3±0.59 mm to 26.6±0.66 mm, respectively. MICs values were recorded between 6.25 mg/ml to 25 mg/ml against all the test organisms. Phytoconstituents analysis of P. bicalyculata extract exposed the presence of alkaloids, flavonoids, glycosides, steroids, saponins and tannins. The methanolic extract of P. bicalyculata % inhibition of DPPH radical is up to 86.33%. The P. bicalyculata (Methanolic extract) gives best antioxidant activity than another extract.Conclusion: This investigation ropes a good answer to the use of P. bicalyculata as a natural antioxidant and in herbal medicine as a support for the development of new drugs and phytomedicine in the foundation for its use in remedial of respiratory infectious diseases.

Muntingia calaburaL. is a tropical plant species that belongs to the Elaeocarpaceae family. The present study is aimed at determining the hepatoprotective activity of methanol extract ofM. calaburaleaves (MEMC) using two models of liver injury in rats. Rats were divided into five groupsn=6and received 10% DMSO (negative control), 50 mg/kg N-acetylcysteine (NAC; positive control), or MEMC (50, 250, and 500 mg/kg) orally once daily for 7 days and on the 8th day were subjected to the hepatotoxic induction using paracetamol (PCM). The blood and liver tissues were collected and subjected to biochemical and microscopical analysis. The extract was also subjected to antioxidant study using the 2,2-diphenyl-1-picrylhydrazyl-(DPPH) and superoxide anion-radical scavenging assays. At the same time, oxygen radical antioxidant capacity (ORAC) and total phenolic content were also determined. From the histological observation, lymphocyte infiltration and marked necrosis were observed in PCM-treated groups (negative control), whereas maintenance of hepatic structure was observed in group pretreated with N-acetylcysteine and MEMC. Hepatotoxic rats pretreated with NAC or MEMC exhibited significant decreaseP<0.05in ALT and AST enzymes level. Moreover, the extract also exhibited good antioxidant activity. In conclusion, MEMC exerts potential hepatoprotective activity that could be partly attributed to its antioxidant activity and, thus warrants further investigations.

In an attempt to further establish the pharmacological properties ofBauhinia purpurea(Fabaceae), hepatoprotective potential of methanol extract ofB. purpurealeaves (MEBP) was investigated using the paracetamol- (PCM-) induced liver toxicity in rats. Five groups of rats (n=6) were used and administered orally once daily with 10% DMSO (negative control), 200 mg/kg silymarin (positive control), or MEBP (50, 250, and 500 mg/kg) for 7 days, followed by the hepatotoxicity induction using paracetamol (PCM). The blood samples and livers were collected and subjected to biochemical and microscopical analysis. The extract was also subjected to antioxidant study using the 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging assay with the total phenolic content (TPC) also determined. From the histological observation, lymphocyte infiltration and marked necrosis were observed in PCM-treated groups (negative control), whereas maintenance of the normal hepatic structural was observed in group pretreated with silymarin and MEBP. Hepatotoxic rats pretreated with silymarin or MEBP exhibited significant decrease (P<0.05) in ALT and AST enzyme level. Moreover, the extract also exhibited antioxidant activity and contained high TPC. In conclusion, MEBP exerts potential hepatoprotective activity that could be partly attributed to its antioxidant activity and high phenolic content and thus warrants further investigation.

The methanolic extract of Duabanga grandiflora (Roxb. ex DC.) Walpers leaves was investigated to explore in-vitro anti-arthritic, thrombolytic and antioxidant potential and subsequent detection of preliminary phytochemicals. It is used for treating various diseases by folk practitioners and rural people. Initially, BSA protein denaturation method was employed to evaluate in-vitro anti-arthritic potency, where the extract exhibited potent anti-arthritic activity with 73.52±1.01% inhibition of BSA denaturation at a dose of 1000 μg/ml compared to the standard dichlofenac sodium (83.66±1.03% inhibition at 1000 μg/ml dose). In-vitro thrombolytic efficacy was determined using streptokinase (positive control) and water (negative control), and the average lysis of blood clot was found to be 31.12±1.09%, which is indicative towards the thrombolytic effect of the extract. Antioxidant activity was evaluated by the DPPH radical scavenging method followed by the total phenolic and flavonoid content determination. The crude methanolic extract revealed the IC50 value of 129.31μg/ml and 204.89μg/ml against DPPH and standard ascorbic acid, respectively. The total phenolic content and total flavonoid content of the plant leaves were observed to be 52.81±1.74μg/ml and 25.77±1.01μg/ml, respectively. The results indicate that the leaves of D. grandiflora possess favorable anti-arthritic activity, moderate thrombolytic and excellent antioxidant activity, which is demonstrating the presence of alkaloids, phenols, flavonoids and many other biologically important phyto-constituents that needs further exploration. Verbena officinalis Linn is a traditionally known medicinal plant which is used against a number of diseases including inflammatory conditions. In this study its antioxidant activity (reducing powers, 2, 2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activities), ferric reduction activity potential (FRAP), total flavonoid concentration and antimicrobial activities of 80%, 90%, 100% methanol and chloroform extracts of V. officinalis Linn root and 90% and100% methanol leaf extracts were determined. Its antioxidant activity increases with increase in amount of extract (10% to 40%v/v). Total flavonoid content (TFC) varied from 73.32±0.002 mgQE/100g of dry weight (90% methanol) to 42.39±0.032 mgQE/100g dry weight (chloroform), 2,2-diphenyl-1-picrylhydrazyl (DPPH), radical scavenging activity (%) was varied between 87.39% (90% methanol) to 45.57% (chloroform) while Ferric reducing antioxidant power was observed between 372.93±0.04 mgAAE/100 g extract (90% methanol) to 129.41±0.026 mgAAE/100 g chloroform in the root extract. The methanolic extract of the leaf showed less antioxidant activity than the methanolic extract of the root. Crude extracts of V. officinalis root showed various degree of antimicrobial activity towards drug resistance microbial pathogens. Growth inhibition tests against bacterial pathogens demonstrated concentration dependence. Moreover, gram positive bacteria were more susceptible to V. officinalis root extract when compared to gram negative bacteria. In general V. officinalis root and leave extracts possess strong antioxidant and antimicrobial activities.

AbstractThe chemical composition, antimicrobial and antioxidant activity of Eryngium palmatum, an endemic plant species from the Balkan Peninsula, were investigated. The flavonoids apigenin (9.5±0.3 mg g−1) and apigenin 7-O-glucoside (2.4±0.1 mg g−1) were determined in a methanol extract of aerial parts using HPLC analysis. The methanol extract of roots contained catechin (5.0±0.1 mg g−1), epicatechin (2.9±0.1 mg g−1), chlorogenic acid (1.6±0.0 mg g−1), gallic acid (0.9±0.0 mg g−1) and rosmarinic acid (0.9±0.2 mg g−1). GC-FID and GCMS analysis of a chloroform extract of aerial parts showed that the main volatile constituents were falcarinol, linoleic acid, hexadecanoic acid and methyl linoleate (comprising 32.6%; 24.4%; 19.9; 13.2% of the volatile fraction, respectively), while octanoic acid, tetradecanol and dodecanol dominated in the chloroform extract of the roots (34.9%; 25.8%; 22.2% of the volatile fraction, respectively). Investigation of antimicrobial activity by broth microdilution showed that the methanol and chloroform extracts of aerial parts and roots exerted a significant effect (MIC 3.5–15.6 μg mL−1) against tested Gram-positive and Gram-negative bacteria. The methanol extracts of aerial parts or roots exerted moderate ferric reducing antioxidant power, DPPH radical scavenging activity and hydroxyl radical scavenging activity.

Pisang Ambon (Musa paradisiaca L.) is one type of bananas usually consumed by Indonesian people. Besides its flesh which has high nutrition, its peels also has antioxidant activity. Antioxidants has the ability to reduce oxidative damage in people’s body with diabetes mellitus. Therefore, this study aimed to determine the antioxidant activity of the aqueous  extract of Pisang Ambon peels and to determine it’s effective dose as an antidiabetic agent in hyperglycemic mice. This study used male mice which all have been intravenously induced with alloxan at a dose of 50 mg/kgBW. They were then divided into five groups. The first two groups got Na CMC 0.5% (negative control) and glibenclamide 0.65 mg/kgBW (positive control), while the other three got  the aqueous  extract of Pisang Ambon peels successively at doses of 400, 800, and 1200 mg/kgBW. The data were statistically analyzed using ANOVA (Analysis of Variance) at 95% confidence interval with parameter of blood glucose levels difference between before and after treatment. The results showed that the aqueous extract of Pisang Ambon peels had antidiabetic activity at an effective dose of 400 mg/kgBW in hyperglycemic mice which was comparable to glibenclamide

This study determined the antihyperglycemic and antioxidant activity of nanoemulsion extracts of M. affine leaves in alloxan-induced rats. This research used 24 male Wistar rats around three months old which grouped as normal (untreated), negative control (treated with carboxymethyl cellulose sodium/Na-CMC,), positive control (treated with glibenclamide), and various concentration (30, 60, and 90%) of nanoemulsion extract of M. affine leaves groups. The extract of M. affine leaves had an antioxidant activity with IC50 5.30 ppm, categorized as a very strong antioxidant. Furthermore, the administration of this extract decreased glucose levels in antihyperglycemic rats.  We concluded that M. affine leaves extract potential as antioxidants and be developed as an ingredient for diabetic drugs.

It was reported that 8-hydroxygenistein (8-OHG) was synthesized by methylation, bromination, methoxylation, and demethylation using cheap and readily available biochanin A as raw material. All synthesized products were structurally confirmed by ultra-high-performance liquid chromatography (UHPLC), infrared spectroscopy, mass spectrometry, 1H-nuclear magnetic resonance (NMR), and 13C-NMR. In addition, we examined the antioxidant capacity of 8-OHG using 6 different methods such as 1,1-diphenyl-2-picrylhydrazyl radical scavenging, 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonate) radical (ABTS) scavenging, nitric oxide radical (NO) scavenging, superoxide radical (O2 −•) scavenging, reducing power assay, and total antioxidant activity using ascorbic acid (VC) as a positive control. Compared with VC, 8-OHG exhibited higher total antioxidant activity and stronger scavenging activity on ABTS, NO, and O2 −•. These results indicate that 8-OHG is an excellent antioxidant agent and may be effective in preventing damage induced by free radical.

<p><strong>Objective: </strong>To evaluate the <em>in vitro </em>antioxidant activity of liquorice (<em>Glycyrrhiza glabra) </em>against H<sub>2</sub>O<sub>2</sub> induced oxidative stress in HepG2 cell line.</p><p><strong>Methods: </strong>Antioxidant activity of methanolic extracts of <em>Glycyrrhiza glabra</em> was investigated by measuring total phenolic content using folin-ciocalteu reagent (FCR), free radical scavenging activity by DPPH and ferric reducing antioxidant power (FRAP). The presence of phenolic compounds and flavonoids in the extract was confirmed by Liquid Chromatography-Mass Spectrometry (LC-MS) analysis. Furthermore, the protective effect of methanolic extract of <em>Glycyrrhiza glabra</em> against oxidative stress induced by H<sub>2</sub>O<sub>2 </sub>in HepG2 cells was investigated by MTT assay. HepG2 cells were exposed with five different treatments viz. liquorice, H<sub>2</sub>O<sub>2</sub>, ascorbic acid, H<sub>2</sub>O<sub>2</sub>+liquorice and H<sub>2</sub>O<sub>2</sub>+ascorbic acid, to explore the effect of the extract on malondialdehyde (MDA) production, catalase activity, and glutathione reductase levels.<strong></strong></p><p><strong>Results: </strong>The total phenolic content estimated in <em>Glycyrrhiza glabra </em>extract was found to be 241.47 µg per 1000 µg/ml of methanolic extract. It was found that as the concentration of the extract was increased both the free radical scavenging activity and ferric ion reducing power was also found to increase. LC-MS analysis confirmed the presence of eight different phenolic compounds in the methanolic extract which are possibly contributing to the antioxidant activity exhibited by the extract. It was also observed that liquorice treated HepG2 cells showed lower MDA and higher glutathione and catalase levels as compared to only H<sub>2</sub>O<sub>2 </sub>treated HepG2 cells where increased MDA production, decreased glutathione reductase and catalase production was observed.</p><p><strong>Conclusion: </strong>Our results thus conclude that, the methanolic extract of <em>Glycyrrhiza glabra </em>can be used as natural supplements in various disease conditions where oxidative stress has been reported. <strong></strong></p><p> </p>

Objectives: The objectives of this study were to determine the antioxidant activity and cardioprotective activity of bangun-bangun leaves ethanolic extract.Methods: Bangun-bangun leaves ethanolic extract was obtained by maceration process. The antioxidant activity test was performed by 2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) free radical scavenging method with various concentrations of extract. The absorbance was measured by visible spectrophotometric method and calculated the inhibitory concentration (IC50) value for antioxidant activity analysis. Cardioprotective activity test was performed by measuring the cardiac troponin T (cTnT) level, creatine kinase-muscle/brain (CK-MB) level, and histology of the heart tissue. Animals induced with doxorubicin at the 8th day and the 9th day, bangun-bangun leaves ethanolic extract was administered from the 1st day to the 9th day with various doses of extract.Results: Bangun-bangun leaves ethanolic extract had IC50 value of 57.79 μg/mL. Difference dose of bangun-bangun leaves ethanolic extract shows difference cardioprotective activity. Bangun-bangun leaves ethanolic extract at dose 300 mg/kg bw did not differ significantly to the positive control group and normal group. The higher the dose of an extract the greater the decrease in cTnT and CK-MB levels and increase protection against heart damage.Conclusion: Bangun-bangun leaves ethanolic extract had strong antioxidant and had cardioprotective activity.
