Background: Sargassum polycystum C. Agardh has potent antioxidant and anti-inflammatory properties. However, its anti-allergic effect has not yet been reported. In this study, we investigated the anti-allergic effects of sulfated polysaccharide of S. polycystum (SPSP) in Dinitrofluorobenzene (DNFB)- induced allergic contact dermatitis animal model. Methods: SPSP was extracted through hot water extraction method and was subjected to compositional analyses. For the allergic contact dermatitis (ACD) model, symptoms were induced by the topical application of 0.5% DNFB on the shaved ventral skin of mice. SPSP (500, 1000, and 2000 mg/kg) and Prednisolone were orally administered for seven days after sensitization. Elicitation was performed seven days later with 0.2% DNFB. After this, ear thickness was measured at baseline and 24 hours post elicitation using a dial thickness gauge. Serum of mice was obtained 24 hours post elicitation, and the level of IFNγ and TNF Results: SPSP afforded 33.6% carbohydrates, 23.7% sulfate, 7.5% protein, and 1.5% uronic acid contents. The effect is attributed to polysaccharide-protein complex present in the extract, but further studies are needed to establish the exact mechanism of action of SPSP in the treatment of the disease.

Probiotics seem to have promising effects in the prevention and treatment of allergic conditions including atopic dermatitis (AD) and food allergy. The purpose of this multicenter randomized placebo-controlled trial was to evaluate the effectiveness of a probiotic preparation comprising Lactobacillus rhamnosus ŁOCK 0900, Lactobacillus rhamnosus ŁOCK 0908, and Lactobacillus casei ŁOCK 0918 in children under 2 years of age with AD and a cow’s milk protein (CMP) allergy. The study enrolled 151 children, who—apart from being treated with a CMP elimination diet—were randomized to receive the probiotic preparation at a daily dose of 109 bacteria or a placebo for three months, with a subsequent nine-month follow-up. The primary outcomes included changes in AD symptom severity assessed with the scoring AD (SCORAD) index and in the proportion of children with symptom improvement (a SCORAD score decreased by at least 30% in comparison with that at baseline). After the three-month intervention, both the probiotic and placebo groups showed a significant (p < 0.0001) decrease in SCORAD scores, which was maintained nine months later. The percentage of children who showed improvement was significantly higher in the probiotic than in the placebo group (odds ratio (OR) 2.56; 95% confidence interval (CI) 1.13–5.8; p = 0.012) after three months. Probiotics induced SCORAD improvement mainly in allergen sensitized patients (OR 6.03; 95% CI 1.85–19.67, p = 0.001), but this positive effect was not observed after nine months. The results showed that the mixture of probiotic ŁOCK strains offers benefits for children with AD and CMP allergy. Further research is necessary to assess the effect of probiotic supplementation on the development of immune tolerance. (ClinicalTrials.gov NCT04738565)

We report here the draft genome sequence of the bacteriocin producer Enterococcus faecium strain L-3, isolated from a probiotic preparation, Laminolact, which is widely used in the Russian Federation. The draft genome sequence is composed of 74 contigs for a total of 2,643,001 bp, with 2,646 coding genes. Five clusters for bacteriocin production were found.

The purpose of this study was to prepare a novel paclitaxel (PTX) microemulsion containing a reduced amount of Cremophor EL (CrEL) which had similar pharmacokinetics and antitumor efficacy as the commercially available PTX injection, but a significantly reduced allergic effect due to the CrEL. The pharmacokinetics, biodistribution,in vivoantitumor activity and safety of PTX microemulsion was evaluated. The results of pharmacokinetic and distribution properties of PTX in the microemulsion were similar to those of the PTX injection. The antitumor efficacy of the PTX microemulsion in OVCRA-3 and A 549 tumor-bearing animals was similar to that of PTX injection. The PTX microemulsion did not cause haemolysis, erythrocyte agglutination or simulative reaction. The incidence and degree of allergic reactions exhibited by the PTX microemulsion group, with or without premedication, were significantly lower than those in the PTX injection group (P<.01). In conclusion, the PTX microemulsion had similar pharmacokinetics and anti-tumor efficacy to the PTX injection, but a significantly reduced allergic effect due to CrEL, indicating that the PTX microemulsion overcomes the disadvantages of the conventional PTX injection and is one way of avoiding the limitations of current injection product while providing suitable therapeutic efficacy.

The combined effect of homobiotics, probiotics based on recombinant strains of g. Bacillus, & enrofloxacin on the dynamics of body weight gain in geese was studied. According to the principle of steam-analogs, one control and five experimental groups of 10 goslings in each group were formed. Gosyatam experimental groups used homobiotics vetom 15.1; vetom 13.1 incombination with enrofloxacin followed by replacement with a probiotic preparation Vetom 1.2. The absolute body weight and average daily gain in geese under the influence of the studied drugs increases. The severity of the effect depended on the pharmacological composition of the drugs used. The maximum absolute weight gain was observed when using the drugs according to the scheme: vetom 13.1 at doses of 25–50 mg/kg for 16 days, with further use of a 10% enrofloxacin solution at a dose of 0.5 ml/kg for 5 days and in The experiment period was 1.2 at a dose of 50 mg/kg for 16 days. After the cessation of the use of drugs, the growth rate increased for 30 days when prescribing the drugs according to the scheme: veto 13.1 at a dose of 50 mg/kg for 16 days and with further introduction of homobiotic veto 1.2 into the bird in a dose of 50 mg/kg for 16 days. The maximum average daily gain in live weight of geese was observed in the period from the 16th to the 32nd day with the administration scheme: homobiotic vetom 13.1 at a dose of 25 mg/kg of body weight 1 time per day for 5 consecutive days, then every other day for 16 days, then daily 10 % solution of enrofloxacin at a dose of 0.5 ml/kg of body weight for 5 days, then vetom 1.2 at a dose of 25 mg/kg once a day for 16 days in a row. And the maximum average daily gain in the post using period of the drug was observed on the 32nd day of the study according to the scheme: homobiotic veto 15.1 at a dose of 50 mg/kg of body weight 1 time per day for 16 days, then daily with vetom 1.2 at the same dose 1 once a day for 16 days. The studied drugs in the applied doses did not have a toxic effect on the physiological state of geese.

Introduction. Gliadin proteins are one of the gluten fractions. They are soluble in alcoholic solution and divided into four groups (α + β, γ, ω1.2, and ω5-gliadins). In this paper gliadins were extracted from wheat flour, and optimal conditions for their separation were determined. Study objects and methods. The separation was performed by capillary gel electrophoresis on Agilent apparatus, CE 7100 (a capillary with an inner diameter of 50 μm, a total length of 33 cm, and an effective length of 23.50 cm). In order to determine the optimal conditions, different solvent concentrations (50, 60, and 70% ethanol), capillary temperatures (20, 25, 30, 35, and 40°C), and electrode voltages (–14.5, –16.5, –17.5 and –18.5 kV) were applied. Migration time and relative concentration of each protein molecules within gliadin fractions in the electrophoregram were analysed using Agilent ChemStation Software. Results and discussion. The optimal conditions for gliadin separation were: solvent 70% (v/v) ethanol, capillary temperature of 25°C, and electrode voltage of –16.5 kV. Under these conditions, the total proteins were indetified as Xav = 23.50, including α + β gliadin fraction (Xav = 7.50 and relative concentration RC = 28.29%), γ-gliadins (Xav = 5.00, RC = 26.66%), ω1.2-gliadins (Xav = 4.33, RC = 14.93%), and ω5-gliadins (Xav = 6.67, RC = 30.98%). Conclusion. The results of the research can be of fundamental importance in the study of gluten proteins and the influence of technological procedures on their change and the possibility of reducing the allergic effect of gluten during processing.

The effect of the probiotic preparation Vetom 1.2 on the basis of Bacillus subtilis VKPM B-10641, Bacillus amyloliquefaciens VKPM B-10642 and Bacillus amyloliquefaciens VKPM B-10643 in different doses on the correlation interactions between the parameters of turkey hemoglobin exchange, blood erythrocytes and blood red blood cells direct and total bilirubin, total protein and serum albumin was studies. Correlations were calculated using Spearman formula, the reliability of correlations was checked by Student. The formulation was used for turkeys once a day for 30 days in doses of 12.5; 25; fifty; 75 and 100 mg / kg body weight. It was found that Vetom 1.2 in minimal doses leads to an inverse correlation between iron and total bilirubin, direct bilirubin, total protein and albumin, an increase in the concentration of the formulation leads to the disappearance of this effect with a gradual subsequent decrease in this indicator. Vetom 1.2 in doses up to 50 mg / kg leads to an inverse correlation between the concentrations of hemoglobin and direct bilirubin, total protein and serum albumin, at a dose of 75 mg / kg there is a direct correlation between these indicators, and at a dose of 100 mg / kg returns a negative correlation. The probiotic preparation Vetom 1.2 leads to a direct dependence of the concentration of bilirubins (total and direct) in serum on the concentrations of hemoglobin and red blood cells when used in most doses, except 75 mg / kg, which reduces the correlation.
