The purpose of this work was to determine the protein composition of whole saliva in 50 year-old patients an d older with manifested and terminal stages of periodontal disease, in correlation with control group of young healthy persons. Experimental and control group consisted of 30 patients. The periodontal status was determinate by using appropriate periodontal index (Sillness-Loe). Investigations were led by electrophoretic method on aragose gel which was used as a routine method for investigation of human serum proteins. Investigating in very alkaline conditions (pH-H) as well as acid conditions (pH=5.9) resulted in separating alkaline and acid glycoprotein fractions in whole saliva. Results showed that there were more separated fractions within basic conditions than within acid conditions. The fraction identification was done by using relative molecule mass determination method by Weber and Osborne. Molecule mass fractions at basic conditions in comparison with the healthy sample (Sz) indicated the destruction of big molecular weight alkaline proteins as well as a significant presence of acid fractions, of little molecule masses up to 60 000 (St), consequences due to aging processes in human organism. It can be concluded that changes in protein composition of whole saliva could point out to the direction and development of periodontal disease as well as changes concerning aging of human organism.

Gastrointestinal cancers (GI) are one of the most abundant types of cancers among the world population, though statistical data indicate that in eastern Asia these types of cancer occur 4 times more often than in Western Europe. Absence of treatment of bacterial infections, obesity, and lack of vegetable food in a diet can be the case of GI cancer. All pathologies are inevitably connected to the changes in cell cycle, abnormal protein amount and their dysfunction. Serum proteins are widely used as an additional source of information about body condition, also changes in protein composition can point out the mechanism of disease development and effectiveness of treatment. In the presented work we studied protein composition of GI cancer patients in different stages of cancer development, after and before chemotherapy and compared these data to protein composition of healthy control group of voluntaries. Treatment of patients was performed according the guidelines appropriate for the GI cancer. Association of the effectiveness of treatment at the different stages of chemotherapeutic courses and changes of protein composition of blood serum has been assessed. Proteins composition was studies by SDS-PAGE electrophoresis and densitometry analysis. Experimentally gained molecular and statistical information exposed the most vulnerable groups of proteins affected by chemotherapeutic agents indicating targets for searching new biomarkers for treatment effectiveness. Research involving human patients performed in accordance with the requirements of the Council of Europe Convention on Human Rights and Biomedicine, Biomedical Research, as well as the UNESCO Declaration of Bioethics and Human Rights.

Introduction and objective: Chronic periodontitis is the most common form of periodontal disease. Progression of the disease is due to high levels of pro-inflammatory cytokines. Statins are a class of lipid-lowering drugs that used for cardiovascular disease and stroke. Statins has potential anti-inflammatory effect by blocking intermediate metabolites of the mevalonate pathway. The aim of this study was to evaluate the influence of Lovastatin and Simvastatin in improve the parameters of chronic periodontitis in the population of Khorasan Razavi province. Methods and materials: 40 subjects with chronic periodontitis were selected and informed consent was obtained from participants. Participants were divided into control and experimental groups and we scaling for patients of both groups, then control group without high blood cholesterol and treatment of patients was done without statin drugs and in case groups, patients with blood cholesterol higher than 240 mg/dl with Lovastatin 20 mg/day during 3 month treated. Periodontal indices; such as probing pocket depth (PPD), GI, PI, CAL and bleeding on probing in patients with chronic periodontitis were measured by the examiner before and after treatment in the control group and the experimental group. Also this index were measured 3 months after treatment in both groups by the same person examiner as Blind and dataes were analyzed by statistical software. Results: In this study, Mean ± SD of age was 7.93 ± 43.8 in the control group and the experimental group was 7.72 ± 47.8 and of the 40 patients in the study, 18 were males and 22 were females. Our study showed that the index of GI, CAL, BOP and probing depth were significantly different between the two groups after the intervention (P less than 0.05) and only after the intervention PI index was not significantly different between the two groups (P>0.05). Conclusion: Our findings showed that statins may improve periodontal index in patient with periodontal disease. This is probably because that statins increase bone regeneration and reduced inflammatory parameters such as CRP, MMP-9, TNF-α and the intermediate products.

Abstract Oral manifestations of common variable immunodeficiency (CVID) are rare, have rarely been studied and have given controversial results. There are few data about IgA, IgG, and IgM antibody salivary levels in the literature, and there are few papers about the clinical impact of antibody deficiencies and CVID on the oral health of such patients. The aim of this study was to measure serum and salivary IgA, IgG, and IgM levels in CVID participants and controls, and to associate immunoglobulin levels with caries and periodontal disease. This was a case-control study involving 51 CVID individuals and 50 healthy controls. All participants underwent examination for dental caries and periodontal disease. Blood and whole saliva samples were collected on the same day of the oral examination. Serum IgA, IgM, and IgG levels were measured by turbidimetry and salivary IgA, IgM, and IgG titers were assessed by enzyme-linked immunosorbent assay. Incidences of caries and gingivitis were significantly higher in the CVID group than in the control group (p<0.05). Salivary and blood IgA and IgM titers were significantly reduced in the CVID group, but there was no association of salivary immunoglobulin levels with periodontal disease or with caries incidence (p>0.05 for both). Although CVID was associated with increased susceptibility to caries and gingivitis, it was not associated with low salivary levels of IgA and IgM.

There has been considerable interest in the serum fructosamine assay as a measure of glycated serum proteins. We have measured serum fructosamine in three groups of patients—those with uraemia; those with multiple myeloma; and those with acute inflammatory conditions—none of whom were known to have diabetes. Serum fructosamine was significantly higher in the uraemic group than in the other two, and also than in a control group. When allowance was made for prevailing serum albumin levels fructosamine was shown to be increased in the acute inflammatory group also. There was a significant correlation between random plasma glucose and serum fructosamine only when fructosamine was adjusted for prevailing albumin levels. In control and uraemic subjects there was a significant positive correlation between serum fructosamine and albumin levels, whereas in the myeloma group there was a negative correlation with serum protein. These data would suggest the need to take into account serum albumin levels and protein composition if serum fructosamine is accurately to reflect short-term integrated glycaemia.

The aim of this cohort study was to determine whether periodontitis and gingivitis are associated with impaired salivary antioxidant status and increased oxidative injury. One hundred and twenty-nine patients attending a routine dental check-up were recruited for the study. Periodontal disease status was characterized using the Community Periodontal Index of Treatment Needs (CPITN) system. Total salivary antioxidant capacity and salivary ascorbate, urate and albumin were determined in a sample of whole unstimulated saliva. Protein carbonyl concentrations were determined as an index of oxidative injury. Patients in the lowest tertile of CPITN score exhibited decreased salivary delivery of antioxidants and specifically urate than patients in the upper tertile. Poor periodontal health was associated with increased concentrations of protein carbonyls in saliva. Women had significantly lower total antioxidant status than men, regardless of periodontal health. Periodontal disease is associated with reduced salivary antioxidant status and increased oxidative damage within the oral cavity.

Smoking is associated with significantly increased risk of diseases of organs and tissues of the oral cavity, especially periodontal pathology and cancer of the mouth.The aim of the study – to investigate the prevalence, structure and clinical course of periodontal tissue diseases in people prone to regular active smoking.Materials and Methods. A study of 120 visitors who had undergone preventive examinations in the Clinic of Zbarazh Central District Municipal Hospital exposed to regular active smoking was performed. To assess the degree of inflammation in periodontal tissues we used marginally-papillary-alveolar index, periodontal index, the Green-Vermilyon index.Results and Discussion. Dental examination of persons exposed to regular active smoking, revealed periodontal disease in 102 individuals (85.0 %), intact periodont – 18 persons (15.0 %). At the same time, investigation of 30 control individuals showed periodontal disease in 12 persons (40.0 %), clinically healthy periodontal tissue – in 18 persons (60.0 %). PMA index of individuals exposed to regular active smoking, treated as severe gingivitis and was significantly higher than data of the control group in 2.5 times. Average numerical values of periodontal index in patients exposed to regular active smoking answered moderate periodontitis and significantly were higher than data of the control group in 4 times. Oral hygiene status in 62 % of those exposed to regular active smoking, was poor, 7 % – very poor, 22 % – satisfactory, 9 % – good. Average numerical values of Green-Vermilyon index in smokers was significantly higher than data of the control group in 3.1 times.Conclusions. Thus, smoking contributes to increasing of diffuse periodontal diseases and deepening of pathological process.

ABSTRACT Background Periodontitis is multifactorial in nature. The effects of fluorosis as an environmental factor are not evaluated and researched to designate it as a risk factor similar to smoking. The primary objectives of this study were to investigate whether there is any association between fluorosis and periodontal disease and to assess salivary oxidative stress in fluorosed and nonfluorosed patients with periodontitis contributing to periodontal disease. Aim This is a case—control study with an aim to investigate whether fluorosis acts as a risk factor for periodontal disease and to assess salivary oxidative stress in fluorosed and nonfluorosed patients with periodontitis contributing to periodontal disease. Materials and methods About 295 systemically healthy patients were divided into test group (n = 154 fluorosed subjects) and control group (n = 141 nonfluorosed subjects) and assessed for their periodontal status. Clinical parameters recorded were plaque index, gingival bleeding index (GBI), and Jackson's fluorosis index to assess the degree of fluorosis, and community periodontal index was to assess the periodontal status. Biochemical analysis of saliva was done for assessment of malondialdehyde (MDA) levels, superoxide dismutase (SOD) levels, and total antioxidant (TAOC) levels. Results The plaque and GBI scores were found similar in fluorosed and nonfluorosed groups. Gingivitis was significantly higher in nonfluorosed than in fluorosed group; in contrast, periodontitis was significantly higher in fluorosed group than in nonfluorosed group. Gingivitis appeared to decline as the fluoride status worsened, while periodontitis showed an increasing gradient from lower fluoride score to higher fluoride score. As the degree of fluorosis increased, periodontitis also increased. The salivary antioxidant levels were found similar in both the groups. Background: Periodontitis is a chronic inflammatory disease of periodontal tissue, characterized by persistent inflammation of the connective tissue and alveolar bone destruction. Patients with periodontal disease show the differences in the composition of saliva. Newer diagnostic tools based on analysis of body fluids such as saliva, GCF and serum are found to be useful for diagnosis as well as monitoring the disease activity. Thus, aim of the study was to estimate the concentration of salivary glycoconjugates (sialic acid, total protein) and salivary ROS in unstimulated whole saliva of subjects with chronic periodontitis and to compare the concentration with healthy/gingivitis subjects.Methods: The study sample consisted of 60 subjects (33 males and 27 females) with age ranging from 30-60 years. A detailed case history was taken from all the subjects and periodontal disease parameters (bleeding on probing, probing pocket depth and clinical attachment levels) were recorded at baseline and subjects were divided into 2 groups. Group I- control group (healthy/gingivitis subjects) and Group II -test group (chronic periodontitis). Saliva samples were collected from the subjects and stored at -700 ºC.  Periodate Resorcinol Assay was done to estimate the levels of sialic acid, Biuret test was done to assess the levels of total protein and  d-ROMs test was done to assess the level of ROS. Statistical analysis was done using students unpaired ‘t’ test and Pearsons correlation test. Results: It was found that the levels of salivary glycoconjugates and ROS are increased in subjects with chronic periodontitis when compared to healthy/gingivitis subjects. Thus it can reflect the clinical status of gingival and periodontal tissues.Conclusions: Estimation of the levels of glycoconjugates and ROS may be used as one of the reliable biomarkers to assess the severity of periodontal disease and to monitor the disease progression.

Abstract Periodontal disease is highly prevalent and affects 10%–15% of the world population. Salivary markers of oxidative stress and antioxidant status represent promising tool for research of oral diseases. Given the importance of reactive oxygen species (ROS) in the pathogenesis of periodontal disease, the aim of this study was to determine the association between oxidative stress parameters and periodontal disease gingivitis in adolescents. The study included a consecutive sample of 80 male and female participants referred to the Institute of Dentistry, Kragujevac, Serbia, as a result of periodontal problems or for routine control, aged 18 to 21, with definitive dentition as an inclusion criterion. Patients were divided into three groups depending on their gingival inflammation status. The first group was control group (n=20) with no gingival inflammation, the second group was with mild gingival inflammation (n=19), the third group was with moderate gingival inflammation (n=21) and the fourth group was with severe gingival inflammation (n=20). Oxidative stress parameters were measured in unstimulated whole saliva samples (superoxide anion radical, hydrogen peroxide, nitric oxide, index of lipid peroxidation, reduced gluthatione, catalase and superoxide dismutase). We have found increased levels of hydrogen peroxide and reduced glutathione in the saliva of patients with moderate levels of gingival inflammation, while the other markers were not significantly affected. In conclusion, oxidative stress plays a central role in the pathogenesis and the determination of oxidative and antioxidative levels could be a potent tool in controlling the development of gingivitis.
