Petunia hybrida is a worldwide most popular annual bedding ornamental plants and economically profitable potted plants. Potting media is most important factor which plays a key role of root growth, vegetative growth and quality production dependent on a relatively small volume of the medium. Hence present study was conducted to evaluate the best potting media to enhance flowering performance of Petunia hybrida. The experiment was conducted at the plant house located at botanical garden, Gampaha by using Petunia variety of F1 multi. Fenice.

Petunia × hybrida (Solanaceae) includes several hybrids that are grown as ornamental plants and are very much appreciated for their long-lasting flowering period. During the spring of 2009, extensive wilting was observed on 2-month-old potted plants of Petunia × hybrida cv. Sanguna Lilac Vein grown in a commercial glasshouse near Albenga (northern Italy). First symptoms included stem necrosis and darkening and withering of leaves. Wilt occurred a few days after the appearance of the first symptoms. Infected plants were characterized by the presence of soft, watery tissues. In the presence of high relative humidity, lesions became covered with a whitish mycelium and irregular, dark gray sclerotia (3.0 to 6.5 × 2.0 to 5.0 mm, average 4.8 × 3.3 mm) were produced on the mycelium. Diseased tissue was surface sterilized for 1 min in 1% NaOCl and plated on potato dextrose agar (PDA) amended with 100 mg/l of streptomycin sulfate. Sclerotinia sclerotiorum (Lib.) de Bary (3) was consistently recovered from infected stem pieces. Sclerotia produced on PDA measured 2.0 to 6.0 × 1.5 to 5.0 mm (average 3.9 × 3.1) mm. The internal transcribed spacer (ITS) region of rDNA was amplified with primers ITS4/ITS6 and sequenced. BLAST analysis (1) of the 548-bp segment showed a 100% homology with the sequence of S. sclerotiorum. The nucleotide sequence has been assigned GenBank Accession No. GQ375746. Pathogenicity of one isolate obtained from sclerotia of infected plants was confirmed by inoculating three 90-day-old plants belonging to cv. Sanguna Lilac Vein transplanted in 22-cm-diameter pots in a glasshouse in a sphagnum peat/pomix/pine bark/clay (50:20:20:10) mix. Inoculum that consisted of 2 g/liter of substrate of wheat kernels infested with mycelium and sclerotia was placed in the soil and around the base of each plant. Three noninoculated plants served as controls. Plants were maintained in greenhouse at temperatures ranging between 22 and 26°C and relative humidity >90%. The inoculation trial was carried out twice. All inoculated plants developed leaf yellowing by 20 days after soil infestation. White, cottony mycelium and dark sclerotia developed on stems and at the base of all inoculated plants. Eventually, infected plants wilted. Control plants remained symptomless. S. sclerotiorum was reisolated from the stems of inoculated plants. To our knowledge, this is the first report of S. sclerotiorum on Petunia × hybrida in Italy. Isolated rabbit hearts were perfused with rabbit red cells suspended in Ringer solution. A small volume of perfusate was recirculated for 10 min at Pco2 of 33.4 +/- 0.9 or 150.8 +/- 7.5 mmHg. Hypercapnia resulted in an increase in perfusate HCO3- concentration that was smaller than that observed when isolated perfusate was equilibrated in vitro with the same CO2 tensions (delta HCO-3e = 1.6 mM, P less than 0.01). This difference is consistent with a net movement of HCO3- into or H+ out of the mycardial cell, and cannot be accounted for by dilution of HCO3- in the myocardial interstitium. Recirculation of perfusate through the coronary circulation at normal Pco2 for two consecutive 10-min periods was not followed by changes in perfusate HCO3- concentration. A high degree of correlation (r = 0.81) was observed between intracellular HCO-3e concentration and the corresponding delta HCO-3e in individual experiments. The results suggest that transmembrane exchange of H+ or HCO3- is a buffer mechanism for CO2 in the myocardial cell.

Under what circumstances did Canadian municipalities, charged with ensuring the existence of services which encouraged the development of real property, come to be involved in the field of culture and leisure? By examining the process through which the Municipal Library and the Botanical Garden were established in Montreal, this article attempts to answer this question. Because these projects constituted precedents in relation to the usual areas of Canadian municipal intervention, they provoked vigorous debate. Each in their own way, the projects served to call into question the definition of public culture and the democratization of knowledge. They also raised the equally contentious issue of the role of municipal government in the fields of leisure and culture.

Like many other filamentous ascomycetes, Fusarium graminearum contains two genes named CPK1 and CPK2 that encode the catalytic subunits of cyclic AMP (cAMP)-dependent protein kinase A (PKA). To determine the role of cAMP signaling in pathogenesis and development in F. graminearum, we functionally characterized these two genes. In addition, we generated and characterized the cpk1 cpk2 double and fac1 adenylate cyclase gene deletion mutants. The cpk1 mutant was significantly reduced in vegetative growth, conidiation, and deoxynivalenol production but it had increased tolerance to elevated temperatures. It was defective in the production of penetration branches on plant surfaces, colonization of wheat rachises, and spreading in flowering wheat heads. Deletion of CPK1 had no effect on perithecium development but the cpk1 mutant was defective in ascospore maturation and releasing. In contrast, the cpk2 mutant had no detectable phenotypes, suggesting that CPK2 contributes minimally to PKA activities in F. graminearum. Nevertheless, the cpk1 cpk2 double mutant had more severe defects in vegetative growth and rarely produced morphologically abnormal conidia. The double mutant, unlike the cpk1 or cpk2 mutant, was nonpathogenic and failed to form perithecia on self-mating plates. Therefore, CPK1 and CPK2 must have overlapping functions in vegetative growth, differentiation, and plant infection in F. graminearum. The fac1 mutant was also nonpathogenic and had growth defects similar to those of the cpk1 cpk2 mutant. However, deletion of FAC1 had no effect on conidium morphology. These results indicated that CPK1 is the major PKA catalytic subunit gene and that the cAMP-PKA pathway plays critical roles in hyphal growth, conidiation, ascosporogenesis, and plant infection in F. graminearum.

The article deals with one of the most relevant topics of the modern city and ecology - the problem of landscaping in Volgograd. The city is characterized by an industrial urbanism, with a high concentration of population, saturation of production facilities and vehicles, which contributes to a high level of negative impact on the environment. The pace of greening of the city lags behind the pace of construction of the residential sector, and the existing green spaces do not meet the sanitary and hygienic, aesthetic requirements. The prospects of this problem are determined, the existing green spaces are considered, data on the climatic conditions of the city and the ecological situation are given. Improvement in the field of landscaping is associated with the scientifically sound placement of green spaces, taking into account all the factors that affect the growth and development of plants in the urban environment. Unfortunately, due to the uneven distribution of the assortment of trees and shrubs, the species composition of the central streets and squares of the city is extremely poor. The study focuses on the role of the botanical garden in urban landscaping and aims to create recommendations for the sorting of tree and shrub plants for urban landscaping. As a result of the analysis of the assortment of ornamental plants of the open ground of the botanical garden, the article proposed species and varieties of woody and shrubby plants that have passed the introduction tests for more than 5-10 years. Recommendations for sorting are aimed at improving the ecological situation of the city and greening the recreation areas of citizens. The proposed list of tree and shrub species cannot fully solve the problem of greening the city, but it will help to significantly enrich the composition of tree and shrub plants.

ABSTRACT The Bacillus subtilis spoIIIJ gene, which has been proven to be vegetatively expressed, has also been implicated as a sporulation gene. Recent genome sequencing information in many organisms reveals that spoIIIJ and its paralogous gene, yqjG, are conserved from prokaryotes to humans. A homologue of SpoIIIJ/YqjG, the Escherichia coli YidC is involved in the insertion of membrane proteins into the lipid bilayer. On the basis of this similarity, it was proposed that the two homologues act as translocase for the membrane proteins. We studied the requirements for spoIIIJ and yqjG during vegetative growth and sporulation. In rich media, the growth of spoIIIJ and yqjG single mutants were the same as that of the wild type, whereas spoIIIJ yqjG double inactivation was lethal, indicating that together these B. subtilis translocase homologues play an important role in maintaining the viability of the cell. This result also suggests that SpoIIIJ and YqjG probably control significantly overlapping functions during vegetative growth. spoIIIJ mutations have already been established to block sporulation at stage III. In contrast, disruption of yqjG did not interfere with sporulation. We further show that high level expression of spoIIIJ during vegetative phase is dispensable for spore formation, but the sporulation-specific expression of spoIIIJ is necessary for efficient sporulation even at the basal level. Using green fluorescent protein reporter to monitor SpoIIIJ and YqjG localization, we found that the proteins localize at the cell membrane in vegetative cells and at the polar and engulfment septa in sporulating cells. This localization of SpoIIIJ at the sporulation-specific septa may be important for the role of spoIIIJ during sporulation.

The production of potted ornamental plants is very important in the Albenga Region of northern Italy, where plants are grown for export to central and northern Europe. During fall 2000 and spring 2001, sudden wilt of tussock bellflower (Campanula carpatica Jacq.) and butterfly flower (Schizanthus × wisetonensis Hort.) was observed on potted plants in a commercial greenhouse. Initial symptoms included stem necrosis at the soil line and yellowing and tan discoloration of the lower leaves. As stem necrosis progressed, infected plants growing in a peat, bark compost, and clay mixture (70-20-10) wilted and died. Necrotic tissues were covered with whitish mycelia that produced dark, spherical (2 to 6 mm diameter) sclerotia. Sclerotinia sclerotiorum was consistently recovered from symptomatic stem pieces of both plants disinfested for 1 min in 1% NaOCl and plated on potato dextrose agar amended with streptomycin sulphate at 100 ppm. Pathogenicity of three isolates obtained from each crop was confirmed by inoculating 45- to 60-day-old C. carpatica and Schizanthus × wisetonensis plants grown in containers (14 cm diameter). Inoculum that consisted of wheat kernels infested with mycelia and sclerotia of each isolate was placed on the soil surface around the base of previously artificially wounded or nonwounded plants. Noninoculated plants served as controls. All plants were maintained outdoors where temperatures ranged between 8 and 15°C. Inoculated plants developed symptoms of leaf yellowing, followed by wilt, within 7 to 10 days, while control plants remained symptomless. White mycelia and sclerotia developed on infected tissues and S. sclerotiorum was reisolated from inoculated plants. To our knowledge, this is the first report of stem blight of C. carpatica and Schizanthus × wisetonensis caused by S. sclerotiorum in Italy. The disease was previously observed on C. carpatica in Great Britain (2) and on Schizanthus sp. in the United States (1). References: (1) D. F. Farr et al. In September 1999, royal ferns (Osmunda regalis L.) at a South Carolina wholesale nursery were found to be infected by foliar nematodes. Lesions were brown, vein-limited, and often fan shaped. As severity increased, affected leaflets became totally necrotic. Nematodes were extracted by excising and dicing symptomatic leaf sections and placing them in water for up to 24 h. Ten adult nematodes from each of two fern plants were examined microscopically and determined to be Aphelenchoides fragariae (Ritzema Bos) Christie. This is the first report of this nematode infecting royal fern. In August 1996, leaves from several cultivars of Hosta spp. with yellow to tan, vein-limited lesions were submitted from The South Carolina Botanical Garden (Clemson, SC) to the Clemson University Plant Problem Clinic for diagnosis. Nematodes were extracted and examined as described above and identified as A. fragariae. This is the first report of this nematode infecting Hosta spp. in South Carolina. Since 1996, foliar nematodes have been recovered from hostas at several wholesale nurseries in South Carolina. Aphelenchoides spp. also have been detected previously in commercially produced ornamental plants in South Carolina, including a Begonia sp. in 1988; Polygonum bistorta L. ‘Super-bum’ (snakeweed) in 1997; and a Polystichum sp. (holly fern) in 1997. All plants exhibited angular or vein-limited, necrotic lesions typical of foliar nematode infections.
