Histological and Physiological Study of the Effect of Silver Nanoparticles and Omega-3 on Asthma of Male Mice Induced by Ovalbumin

In the current study ninety one of male mice weighting (25-30 g) aged (15-17) weeks at the animal house faculty of science / university of Kufa during the period from January 2017 to September 2017. This study included some physiological and histological criteria to evaluate the protective role of omega-3(2 and 3 mg/kg) and silver nanoparticles (5 and 10 mg/kg) against asthma that induced by ovalbumin. The animals experimental are divided into 16 groups (n= 6 mice per each group) for duration of one and two months. The results showed significant increase (p less than 0.05) in the leukocyte count (eosinophil, neutrophil, lymphocyte, monocyte) in asthma group as compared with control group .Also ,the results showed significant decrease (p less than 0.05) in the leukocyte count in the treated group of omega-3 and silver nanoparticles for both concentration as compared with asthma group .The result sowed significant increase(p less than 0.05) in the serum level of periostin and Galectin -3 and Interleukin -33 in asthma group as compared with control group. The histological study of lung tissue revealed that induced the tissue with ovalbumin caused necrosis, degeneration and increase of mucous in bronchiole as well as acute inflammation around bronchioles while the effect of ovalbumin in trachea tissue were sluphing, necrosis and degeneration around the epithelium of bronchioles.

Objectives: To find out the effects of noise stress on variations in number of ovarian follicles in mice. Study Design: Randomized control trial, laboratory based. Place and Duration of Study: Anatomy department, Army Medical College Rawalpindi, it was conducted from April to May 2014. Material and Method: 20 adult mice (BALBc strain) female weighing 25-27 grams were taken for this study on day first of their estrous cycle. Two groups comprising of 10 rats each were made. Control group was given name tag of group A and was kept in animal house under normal heathy environment for one month. Noise stress of 100 decibels for 06 hours per day for one month was given to experimental group B. Mice were dissected after 1 month. Mice right ovaries were examined for size, shape, color and weight. The specimen were processed and embedded. Hematoxyline and Eosin stained slides were prepared for histological study. Follicular count (primary, secondary and tertiary follicles) was done and noted. For statistical analysis SPSS 20 was used. The tests applied was independent sample’s t test for descriptive statistics. Results: There was a significant decrease in primary, secondary/antral and graafian follicular count in the ovary of Group B (Experimental). Conclusion: The noise stress prominently affected the follicular count in ovaries of noise stress treated group.

<p>To investigate the protective role of broccoli powder “<em>Brassica Oleracea Italica</em>” against continuous ingestion of escitalopram antidepressant drug induced hepatotoxicity in Swiss albino male mice.</p><p>Mice were divided into different groups. Group1: Normal control (0.9% NaCl), Group 2: Escitalopram drug treated only (20 mg/kg), Group 3: Broccoli powder with Escitalopram drug treated (200 mg/kg + 20 mg/kg), Group 4: Olive oil vehicle control, Group 5: Carbon tetrachloride (CCl<sub>4</sub>) referenced as positive control (33 mg/kg), Group 6: Broccoli powder with CCl<sub>4</sub> treated (200 mg/kg + 33 mg/kg). The effect of these groups on liver tissue was studied after three different time periods for 4, 8 and 12 weeks.</p>The results showed that the treatment with escitalopram drug displayed significantly increased serum SGOT, SGPT, ALP level and alter liver antioxidant enzymes level (LPO, SOD and GSH) that are comparable with CCl<sub>4</sub>intoxicated group considered as positive control. Comparing escitalopram drug treated group with group that received both broccoli powder and escitalopram drug displayed a significant decrease in serum SGOT, SGPT, ALP levels and restored the level of antioxidant enzymes. The protective effect of broccoli powder on escitalopram drug induced hepatotoxicity was also supported by histopathological studies.<p> </p>

Introduction and Aim: Oxidative stress plays a very important role in endosulfan-induced toxic effects on reproductive organs. Vitamin C is a potent antioxidant which plays an important role in decreasing oxidative stress. The present study was aimed to investigate the protective role of vitamin C against endosulfan-induced testicular toxicity in Wistar rats. To investigate a protective effect of vitamin C against endosulfan induced toxicity on biochemical changes. Materials and Methods: Seventy male neonatal Wistar rats were divided into  seven groups. The group  I was taken as the control group, the endosulfan-treated were grouped into II (3 mg/kg body weight (BW) and group III (6 mg/kg BW), Group IV (9 mg/kg BW) and Group V (12 mg/kg BW). Group VI (9 mg/kg BW) and group VII (12 mg/kg BW) were pretreated with vitamin C (20 mg/kg BW) for 60 days. After  the experimental procedures, the testicular weight, lactate dehydrogenase (LDH) enzyme and testosterone in plasma, LDH, steroidogenic enzymes 3?-HSD and 17?-HSD in testis were evaluated. One-way ANOVA was used to determine the statistical significance. Results: Significant improvement in the testicular weight (P<0.05) , LDH (P<0.05) levels both in plasma and testis, increase in testosterone(P<0.001) and steroidogenic enzyme levels(P<0.001) was observed in the group pretreated with vitamin C treated group when compared to the endosulfan treated group. Conclusion: Vitamin C decreases the toxic effect of endosulfan on testis. The present action might be  due to its antioxidative properties.

Purpose. To explore the regulatory effects of liraglutide on the kidney and liver through the miR-34a/SIRT1 pathway with related factors in diabetic nephropathy (DN) rats. Methods. DN rats were randomly divided into two groups ( n = 10 ) and were injected with liraglutide or normal saline twice a day. The 24-hour urine microalbumin content and biochemical index levels were measured. qRT-PCR was performed to detect the expression of miR-34a in the kidney and liver tissues. The levels of SIRT1, HIF-1a, Egr-1, and TGF-β1 in kidney and liver tissues were determined using qRT-PCR, western blot, and immunohistochemistry. Electron microscopy and HE staining were used to observe the ultrastructure and pathological changes. Results. Liraglutide treatment in DN rats decreased blood glucose, 24-hour urine microalbumin, TC, TG, LDL-C, UA, Cr, UREA, ALT, and AST levels and increased the level of HDL-C ( P < 0.05 ). Compared with the control group, the miR-34a levels were significantly decreased in kidney and liver tissues followed by liraglutide treatment ( P < 0.05 ). The levels of SIRT1 in the liraglutide group are significantly higher than those in the control group with the kidney and liver tissues ( P < 0.05 ). Conversely, the contents of HIF-1a, Egr-1, and TGF-β1 were significantly lower in the liraglutide group than in the control group ( P < 0.05 ). Electron microscopy showed that the kidney of the liraglutide-treated group exhibited minor broadening of the mesangial areas, fewer deposits, and a well-organized foot process. HE staining revealed that the kidney of the liraglutide-treated rats had a more regular morphology of the glomerulus and Bowman sac cavity and lighter tubular edema. Additionally, the liraglutide-treated DN rats had a clear hepatic structure, a lower degree of steatosis, and mild inflammatory cell infiltration. Conclusion. Liraglutide, through its effect on the miR-34a/SIRT1 pathway, may have a protective role in the kidney and liver of DN rats.

ABSTRACT We investigated the protective role of ceftriaxone plus sulbactam with VRP1034 (Elores) on hematological, lipid peroxidation, antioxidant enzymatic activities and Cd levels in the blood and tissues of cadmium exposed rats. Twenty-four male rats were divided into three groups of eight rats each. The control group received distilled water whereas group II received CdCl2 (1.5 mg/4 ml/body weight) through gastric gavage for 21 days. Group III received CdCl2 and was treated with ceftriaxone plus sulbactam with VRP1034 for 21 days. The hematological, biochemical, lipid per-oxidation levels and enzymatic parameters were measured in plasma and tissues (brain, liver and kidney) of all groups. The Cd, Zn and Fe levels were measured in blood and tissues of all groups. Our findings showed significantly decreased cadmium (p<0.001), malonaldialdehyde (p<0.001) and myloperoxidase (MPO) levels along with significantly increased hemoglobin (p<0.01), RBC (p<0.05), hematocrit (p<0.05) levels and all antioxidant enzymatic activities (SOD, CAT, GR, GPx) in plasma and tissues of ceftriaxone plus sulbactam with VRP1034 treated group as compared to cadmium exposed group. Delta aminolevulinate dehydratase (δ-ALAD) activity was significantly (p<0.001) increased in the blood of ceftriaxone plus sulbactam with VRP1034 treated group as compared with cadmium exposed group. The levels of hepatic and renal parameters were significantly (p<0.001) decreased in ceftriaxone plus sulbactam with VRP1034 treated group as compared to cadmium exposed group. These findings indicate that ceftriaxone plus sulbactam with VRP1034 acts as a potent free radical scavenger and exhibits metal chelating properties that reduce free radical mediated tissue injury and prevent dysfunction of hepatic and renal organs during metal intoxication.

The declining rate of male fertility is a growing concern. Tributyltin (TBT) is a well-known endocrine disruptor (ED), that induces imposex in female gastropods and is widely used in various industrial applications. The aim of this study was to evaluate the toxic effects of TBT on the testes of adult albino rats and the possible role of omega-3. Forty two adult male albino rats were divided into five groups; control group (Group I) and four experimental groups: omega-3 treated group, TBT treated group, TBT & omega-3 treated group and follow up group. At the end of the study, the rats were subjected to biochemical, histological, immunohistochemical staining for Ki-67 and seminal examinations. Our results clarfied that TBT induced a significant decrease in testosterone, FSH, LH and serum glutathione peroxidase levels and a significant increase in the serum Malondialdehyde as compared to the control group. Tributyltin induced disorganization and shrinkage of seminiferous tubules, apoptosis, cellular damage and marked reduction in the germinal epithelium. A significant decrease in the cell proliferation and arrested spermatogenesis were also detected. Seminal analysis of TBT group showed a significant affection of all parameters as compared to other groups. Omega-3 ameliorated all of these hazardous effects. Follow up group still showed toxic effects. In conclusion, TBT has a toxic effect on the testis. Increased testicular oxidative stress, cellular damage and arrest of spermatogenesis with attenuation in antioxidant defenses are all contributing factors. Omega-3 can protect against TBT induced reproductive toxicity.

Objective: Cobalt nanoparticles (NPs), especially cobalt oxide NPs (Co3O4 NPs) are attracting unique shaped NPs that are used in different biomedical applications and medicine. Different in vitro studies report their toxic and carcinogenic effect but limited in vivo studies were present on its genotoxic potential. The present study was aimed to evaluate the genotoxic potential of Co3O4 NPs on bone marrow cells and sperms and the protective role of omega-3 in male albino mice.Methods: Animals were segregated into four groups that were orally treated for 3 consecutive days, Group 1: Negative control; Group 2: Omega-3 (250 mg/kg); Group 3: Co3O4 NPs (20 mg/kg); and Group 4: Combined group (250 mg/kg Omega-3 and Co3O4 NPs 20 mg/kg).Results: The present results show that Co3O4 NPs administration significantly increased number of micronucleated polychromatic erythrocytes (PCEs)/1000 PCEs, sperm abnormalities, and DNA damage, significantly decreased sperm motility and concentration in comparison to negative control group. However, Omega-3 administration in the combined group modulates the genotoxic potential of Co3O4 NPs in comparison to Co3O4 NPs group.Conclusion: The present study reports the genotoxic potential of Co3O4 NPs in vivo and assesses the protective role of Omega-3 administration due to its antioxidant effect.

Staphylococcus aureus infection causes oxidative stress in neutrophils. The immune cells use reactive oxygen species (ROS) for carrying out their normal functions while an excess amount of ROS can attack cellular components that lead to cell damage. The present study was aimed to test the protective role of nanoconjugated vancomycin against vancomycin-sensitive Staphylococcus aureus (VSSA) and vancomycin-resistant Staphylococcus aureus (VRSA) infection induced oxidative stress in neutrophils. VSSA- and VRSA-infection were developed in Swiss mice by intraperitoneal injection of 5×106 CFU/mL bacterial solutions. Nanoconjugated vancomycin was treated to VSSA- and VRSA-infected mice at its effective dose for 10 days. Vancomycin was treated to VSSA and VRSA infected mice at similar dose, respectively, for 10 days. The result reveals that in vivo VSSA and VRSA infection significantly increases the level of lipid peroxidation, protein oxidation, oxidized glutathione level, and nitrite generation and decreases the level of reduced glutathione, antioxidant enzyme status, and glutathione-dependent enzymes as compared to control group; which were increased or decreased significantly near to normal in nanoconjugated vancomycin-treated group. These finding suggests the potential use and beneficial protective role of nanoconjugated vancomycin against VSSA and VRSA infection induced oxidative imbalance in neutrophils.

Background: The effect of zinc supplementation on birth weight, litter size, total erythrocyte count (TEC), hemoglobin estimation, total leukocyte count (TLC), blood glucose level, abnormal sperm count and histopathology of testis in mice was evaluated. Methods: 45 albino mice were randomly divided into 3 equal groups viz. control group, A; 10 mg zinc/kg feed treated group, B; 20 mg zinc/kg feed treated group, C. Each group was comprised of 10 male mice and 5 female mice. Then they are allowed to breed for about 25 days. After breeding males were withdrawn from each group and treated with zinc for 30 days. Then male mice were transferred back to female cage and were allowed to breed for 25 days. Results: Mice of group B and C showed a significant increase (p<0.01) in total erythrocyte count (TEC), total leukocyte count (TLC), hemoglobin content, litter size and decreased abnormal sperm count. No significant increase in body weight, even a decrease in glucose content was recorded in group B and birth weight was found increased in group C. In the histopathological study, no significant change found with different doses of zinc supplementation except reactive cell infiltration and slight tissue degeneration in the mice fed with 20 mg zinc supplement/kg feed was recorded. Conclusion: Supplementing zinc @ 10-20 mg/kg feed was found to enhance hematological parameters, increase litter size and decrease abnormal sperm count in mice.

Purpose: Contrast-induced nephropathy (CIN) is the third cause of hospital-acquired renal failure and is associated with significant morbidity and mortality. Several studies have revealed the protective role of omega-3 in prevention and treatment of some kidney injuries. This study was conducted to examine the effect of omega-3 supplementation on the markers of renal function and to evaluate its potential in the prevention of CIN in patients undergoing elective percutaneous coronary intervention (PCI). Methods: In this double-blind, randomized clinical trial, 85 eligible patients scheduled for PCI was randomly divided into omega-3 (a single 3750 mg dose of omega-3 as well as routine hydration therapy within 12 hours before PCI) or control (placebo plus routine hydration therapy) groups. Serum creatinine (SCr) and cystatin C levels were measured at baseline and 24 hours after PCI. Results: Our results indicated that post- PCI cystatin C levels were significantly decreased in the omega-3 group compared to the control group (P < 0.001). Although less upward manner was seen in the level of 24-hour creatinine in the omega-3 group, it did not reach the significance level (P = 0.008). Conclusion: The positive effect of omega-3 on cystatin C levels showed that it may have a protective role in the prevention of CIN in post-PCI patients with normal kidney function. However, to better assess this effect, it is highly recommended to design future studies with higher doses and longer duration of therapy with omega-3 plus long-term follow up.
