In vitro antioxidant activity test and determination of phenolic and flavonoid content of Moringa oleifera pulp and seeds

Moringa oleifera is a plant that is empirically widely used as a medicine to treat hyperglycemia, anti-inflammatory, antidiabetic, antimicrobial, anticancer, antioxidant, gastric ulcer, skin disease, fever, fatigue, hysteria, thrush, bladder, and bronchitis. Moringa oleifera contains secondary metabolites such as flavonoids, tannins, terpenoids, alkaloids, and phenolics. This study aimed to investigate the antioxidant activity and levels of phenolic and flavonoids in the extracts and fractions of moringa pulp and seeds. The pulp and seeds of Moringa were extracted using the maceration method and fractionated using the liquid-liquid extraction method. Antioxidant activity was measured using the 2,2-diphenyl-1-picrylhydrazyl (DPPH) method. Phenolic content was measured using the Folin-Ciocalteau method. Meanwhile, the flavonoid content was determined using the AlCl3 colorimetric method with UV-Vis spectrophotometry. The ethyl acetate fraction of moringa pulp and seeds had the strongest antioxidant activity with IC50 values of 5.97 µg/mL and 5.08 µg/mL, respectively. Ethyl acetate fraction also has high phenolic and flavonoid content in the pulp and seeds of moringa with a phenolic content of 43.956 mg GAE/g sample for pulp and 9.429 mg GAE/g sample for moringa seeds. The flavonoid content of 94.257 mg QE/g sample for pulp and 93.719 mg QE/g sample, with the highest correlation between phenolic compounds and flavonoids on antioxidant activity, was obtained in moringa pulp with the highest correlation (R2 = 0.6514) for phenolic and R 2 = 0.7531) for flavonoids. In conclusion, the ethyl acetate fraction of moringa pulp and seeds can be further developed as an antioxidant and functional food.

Free radical is any molecular species that have unpaired free electrons in their outer orbital shell that make radicals highly reactive, resulting in pathogenesis conditions such as cellular injury, premature aging, cancer, hepatic disorders, neurodegenerative diseases, cardiovascular disease, and kidney disease. One source of natural antioxidant is jackfruit. The purpose of this research was to determine the phenolic and flavonoid contents in the extracts and fractions of jackfruit peel and their potential as antioxidants. Jackfruit peel powder was extracted from maceration. The total phenolic content was determined by the Folin-Ciocalteu method. Meanwhile, flavonoid content was determined using the aluminium chloride complex colorimetric method. Measurements of antioxidant activity were conducted using the 2,2-diphenyl-1-picrylhydrazyl (DPPH). The ethyl acetate fraction had high phenolic and flavonoid contents, which were 49.667±1.508 g GAE/100 g of sample and 70.199±0.374 g of quercertin equivalent/100 g. The ethyl acetate fraction had the strongest antioxidant activity with IC50 value of 4.539±0.201 µg/ mL and correlation value (R2 ) of 0.5881 for phenols and R2 of 0.7241 for flavonoids. Ethyl acetate fraction of jackfruit peel is very potential to be developed as a natural antioxidant and functional food.

A raw extract and four extractive fractions were obtained from Cedrus brevifolia (Cyprus cedar) bark. They were all studied regarding the phenolic content and profile using spectrophotometry and HPLC-DAD-ESI-MS. The antioxidant activity was investigated using in vitro assays: DPPH and ABTS radicals scavenging and reducing power assays. The ethyl acetate fraction had the highest total phenolic and proanthocyanidin contents; a taxifolin-O-hexoside, catechin, epicatechin and procyanidin oligomers (three dimers, two trimers) were identified in this fraction. The ethyl acetate fraction was found to possess the highest DPPH and ABTS radicals scavenging effects (EC50=13.9 ± 0.3 and 2.3 ± 0.0 μg/mL, respectively) and reducing capacity (EC50=9.1 ±0.1 μg/mL). Antioxidant effects were highly correlated with total phenolic and proanthocyanidin contents (r=0.89-0.99). These results suggest that Cedrus brevifolia bark is a new source of antioxidants.

Kersen (Muntingia calabura L) fruits have the potential to be used as an active ingredient in sunscreens because of phenolic and flavonoid content that can absorb UV rays. This study aims to determine the percentage of erythema/pigmentation transmission and SPF value as parameters for sunscreen activity.  Kersen fruits were extracted by maceration using 96% ethanol. The ethanol extract of Kersen Fruits was also fractionated to separate the components of the active compounds based on the polarity level using n-hexane, ethyl acetate, and ethanol as solvents. The test was carried out using the in vitro method by measuring the ability of the material to absorb ultraviolet light at a wavelength of 292.5-372.5 nm. This research was conducted at concentrations of 100, 200, 400, 600, and  800 µg/mL for ethanol, lyophilisate, n-hexane, and ethanol fractions, while the ethyl acetate fraction concentrations are 50, 100, 150, 200, 250 µg/mL. The results showed the best value at the ethyl acetate fraction concentration of 250 µg/mL with % Te of 5.28 and % Tp of 28.65 and the SPF value of 16.54. Based on the % Te and Tp, the ethyl acetate fraction exhibited protection against erythema and pigmentation with the category of extra protection and based on the SPF value with the category of ultra protection.

This experiment aims to determine the correlation of total phenolic and flavonoid content of jati putih leaves fraction (Gmelina arborea Roxb.) towards Antioxidant activity .  Sample was extracted by maceration method using ethanol 70% to obtain the ethanol extract (EE), followed by liquid-liquid extraction method to obtain fraction of ethyl acetate (EA) and n-Hexane (EH). The phytochemical screening  and determination of total phenolic and flavonoid content were done by colorimetric method. Antioxidant activity were done by DPPH, FRAP and ABTS methods. Phytochemical screening showed positive results for flavonoids, phenolic and saponins.  The largest total phenolic content was found on EA (11,59 µg/ml ± 0,3 %b/b EAG) and the largest total flavonoid content was on EA (3,88 µg/ml ± 0,02 %b/b EK). The total phenolic and flavonoid content of Jati putih leaves has a correlation with antioxidant activity. The coefficient correlation of activity on reducingDPPH radical was 56,7% (total of phenolic content) and 57,8% (total of flavonoid content) and on iron reduction power in FRAP method  was 99,9% (total of phenolics and flavonoids content). The relationship with the activity in reducing radical ABTS obtained coefficient correlation of 57,0% and 58,1% for total phenolic and flavonoids contents, respectively.

Antioxidant activity, total phenolic and flavonoid content of several indigenous species of ferns in East Kalimantan, Indonesia. Biodiversitas 20: 576-580. This study aimed to determine the total phenolic and flavonoids content and antioxidant activity of ethanol extract of several indigenous species of ferns in East Kalimantan. Total phenolic content was determined by Folin-Ciocalteau method and flavonoid content was measured by colorimetric method. Antioxidant activity was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) method. The result of photochemical screening indicated that the leaves of Plagiogyria pycnophylla, Plagiogyria glauca, and Stenochlaena palustris contained alkaloid, flavonoid, tannin, saponin, and steroid while Acrostichum aureum contained flavonoid, tannin, saponin and steroid. The result showed extract Acrostichum aureum had the highest total phenolic content (366.4573 ± 2.2117 mg GAE.g-1), flavonoid content (228.6087 ± 2.2548 mg QE.g-1), and very strong antioxidant activity with IC50 value 29.5303 ppm. There is positive correlation between total phenolic content, flavonoid with antioxidant activity.

Objective: This study was to investigate the phytochemical screening, total phenolic content (TPC), and antioxidant activity of fractions of water, ethyl acetate, and n-hexane from Dendrophthoe pentandra (L.) Miq. and mistletoe of Moringa oleifera Lam.Methods: Simplicia extracted with 96% ethanol using maceration method. Liquid-liquid extraction was used for fractionation with water, ethyl acetate, and n-hexane as solvent. Phytochemical screening, determination of TPC, and antioxidant activity were performed against these fractions. Antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl method. The TPC was measured by Folin–Ciocalteu reagent and use gallic acid (GA) as a standard.Results: Water fraction contains saponin and polyphenols. Ethyl acetate fraction contains alkaloids, flavonoids, saponins, terpenoids, and polyphenols. N-hexane fraction only contains alkaloids and terpenoids. The TPC and antioxidant activity of fractions of water, ethyl acetate, and n-hexane were 9.42±1.16%, 21.35±1.86%, and 67.40±0.82% mg GA equivalent/g fraction and 29.46 μg/ml±0.99%, 7.08 μg/ml±0.39%, and 10.90 μg/ml±1.05%, respectively.Conclusion: Fraction of ethyl acetate exhibited the highest antioxidant activity and TPC. This study showed that there were significant correlations between TPC and antioxidant activity at significance level p<0.05.


Dates consisting of 90% flesh have been known to have various benefits, but the remaining 10% of them in the form of seeds has not been utilized optimally. The date palm contains a fairly high polyphenol compound, which can act as an antioxidant. This study aims to determine the antioxidant activity of methanol extract and fractionation of the extract of dates using the in vitro method of DPPH (1,1-diphenyl-2-picrilhidrazil). The date seeds are made into simplicia, macerated with methanol, and then the extracted methanol is gradually fractionated by n-hexane, ethyl acetate, n-butanol, and water. Each fraction and extract is phytochemically screened using the standard methods and its antioxidant activity is determined by DPPH method. Methanol extract, ethyl acetate, n-butanol, and water fractions contain tannins, flavonoids, alkaloids, and saponin, while the n-hexane fraction not contain phytochemical compound based on the phytochemical screening. The ethyl acetate fraction has the strongest antioxidant activity with the lowest IC50 value, that is equal to 3.72 ± 0.44 μg/mL, followed by methanol extract of 4.71 ± 0.64 μg/mL, n-butanol fraction of 6.62 ± 0.08 μg/mL, water fraction of 53.00 ± 18.96 μg/mL, and n-hexane fraction of 250.19 ± 9.69 μg/ mL, while the IC50 value of vitamin C as the positive control is 4.29 ± 0.74 μg/mL. Therefore, date seeds can be a source of natural antioxidants

Prunus domestica L. is a member of the Rosaceae family that shows many biological activities including antioxidant, antimicrobial, antihaemolytic, anti-inflammatory, hepatoprotective activity and many other activities. In the current study, we evaluated nutritive value, phytochemical screening, total phenolic content and antioxidant activity by DPPH and FRAP method for the different extracts obtained by successive soxhlet extraction using the different solvents based on their polarity. Results show that it is a good source of energy. Phytochemical screening revealed the presence of many secondary metabolites which include alkaloids, carbohydrates, glycosides, protein, steroids and terpenoids, fixed oils and fat as well as phenolic compounds. The highest total phenolic content was found in the ethyl acetate fraction. Highest antioxidant activity by DPPH method is reported in ethyl acetate fraction (IC50 =1837.399±0.377µg/ml) while the ferric reducing antioxidant power was maximum for diethyl ether (56.032±0.985µM/ml FRAP value = 0.325±0.002).

<p><strong>Objective: </strong>The current study was to deal the isolation and identification of secondary metabolites from <em>Polygonum equisetiforme</em> and evaluation of antioxidant activity of its extract.</p><p><strong>Methods: </strong>The methanol-water extract (7:3) of the air-dried aerial parts of <em>Polygonum equisetiforme</em> was fractionated and separated to obtain the isolated compounds by different chromatographic techniques. Structures of these compounds were elucidated by UV and 1D⁄2D H⁄ C NMR spectroscopy and compared with the literature data. The crude extract was evaluated for <em>in vitro</em> antioxidant activity using the 2,2 diphenyl dipicryl hydrazine (DPPH) method.</p><p><strong>Results: </strong>Ten secondary metabolites were isolated from <em>Polygonum equisetiforme</em> in this study. Of which three new flavonoids named as 3,5,7,2’,5’ pentahydroxyflavone 3-<em>O</em>-b-D-glucopyranoside (1), 3,5,7,2’,5’ pentahydroxyflavone 3-<em>O</em>-b-D-glucopyranoside 8 C-sulphated (2) and quercetin 3-<em>O-β</em>-D-glucucorinde 6''-methyl ester 8-sulphated (3) as well as quercetin 3-<em>O-β</em>-D-glucucorinde methyl ester (4), quercetin 3-<em>O-</em>β-D-glucopyranoside (5), quercetin 7-<em>O-β-</em>D-glucopyranoside (6),<em> </em>quercetin(7)<sub>, </sub>myricetin (8), <em>P</em><sub>-</sub>methoxy gallic acid methyl ester (9) and gallic acid (10). The antioxidant potential of <em>P. equisetiforme</em> extract was evaluated by investigating it's total phenolic and flavonoid content and DPPH radical scavenging activity whereby the extract showed significant antioxidant activity (IC<sub>50 </sub>= 37.45 μg/ml). The total phenolic and flavonoid content was found to be 130.79±5.502 and 45.8±1.63 μg/ml, respectively.</p><p><strong>Conclusion: </strong><em>Polygonum equisetiforme</em> is a promising medicinal plant, and our study tends to support the therapeutic value of this plant as an antioxidant drug.</p>

Paku atai plant (Angiopteris ferox Copel), specifically the tuber has been used empirically as an anticancer and antidote materials by Dayaknese in West Kutai, with limited scientific study. Thus, this research had been conducted to determine and evaluate the total phenolic, flavonoid content of this plant along with the antioxidant and toxicity profile. The research used several extracts’ solvents including extraction with ethanol (crude extract, CE) and subsequently partitioned with n-hexane (HF); ethyl acetate (EF) and ethanol aqueous (EAF). The resulted crude extract and fractions were then analyzed through colorimetric method to determine the phenolic and flavonoid total; with DPPH and FRAP to observe the antioxidant activity; and using BSLT method to evaluate the toxicity activity. The results showed that the EF fraction provided the strongest antioxidant activity with IC50 value of 13.79 μg/mL and iron reduction with FRAP value of 387.5±6.41 µM/g. However, a high correlation was observed between the antioxidant and the total phenolic content (r2 = 0.970 - 0.974) but little correlation in total flavonoid (r2 = 0.345 - 0.373). Furthermore, the EF (19.56±7.35) showed the highest toxicity activity followed by CE (22.42±2.10), HF (39.52±7.38), and EAF (41.75±5.10). Therefore, the paku atai tuber can be potentially developed as a natural antioxidant and anticancer material.
