Seaweed (EucheumaCottonii J. Agardh) is one of the biological resources found in coastal and marine areas. Seaweed contains secondary metabolites such as flavonoids, alkaloids, polyphenols and tannins which can reduce cholesterol levels in the blood. This study aims to determine the effect of seaweed ethanol extract on reducing cholesterol and obesity levels by comparing plasma cholesterol levels, body weight, and weight of vital organs (liver, kidneys and jantumg) of obese rats, mice given seaweed ethanol extract with normal mice . Rats were grouped randomly into 6 groups. group one was given 0.5% CMC Na, group two was given high fat diet food, group three was given simvastatin, group four was given seaweed ethanol extract 100 mg / kgbb, group five was given ethanol extract of seaweed 200 mg / kgbb and group six was given ethanol extract of seaweed 300 mg / kg with a long suspension of extract for 14 days. Measurement of cholesterol levels in the blood using the CHOD-PAP method, body weight and organs carried out by weighing. The results showed that seaweed ethanol extract has the effect of inhibiting an increase in plasma cholesterol levels and has anti-obesity potential at a dose of 300 mg / kgbb.

Telah dilakukan penelitian untuk mempelajari pengaruh pemberian ekstrak etanol dan heksana dari daun gedi merah terhadap kadar kolesterol plasma tikus wistar yang diberi pakan aterogenik. Penelitian ini menggunakan 20 ekor tikus jantan, berumur 2-3 bulan dengan berat 130-200 g, yang dibagi dalam 4 kelompok perlakuan secara acak, yaitu kelompok K diberi pakan beras jagung, kelompok K- diberi pakan aterogenik, sedangkan kelompok P1 dan P2 diberi ekstrak etanol dan heksana dengan dosis 20 mg/KgBB. Setiap perlakuan terdiri dari 5 ekor tikus yang dilakukan 5 kali pemeriksaan kadar kolesterol dengan metode biosensor amperometrik dengan menggunakan alat ukur NESCO GCU. Data yang diperoleh dari pemeriksaan dianalisis menggunakan ANOVA satu arah <(0,005). Hasil penelitian menunjukkan bahwa ekstrak heksana dan ekstrak etanol daun gedi merah dengan dosis 20 mg/kgBB dapat menurunkan kadar kolesterol sebesar 72% atau sekitar 12,8 mg/dL dan 14% atau sekitar 2,4 mg/dL, dibandingkan dengan dengan kelompok K-.The research had been done with the purpose were to study the influence of granting the ethanol and hexane extract of red gedi leaves on Wistar rats plasma cholesterol levels which were fed by atherogenics. This study used about 20 male rats, Wistar rats aged 2-3 months with a weight 130-200 g were divided into 4 groups randomly, for the grop of K were fed corn rice, group of K- were fed atherogenics, while the P1 and P2 were given ethanol and hexane extract with a dose of 20 mg/KgBW. Each treatment consisted of 5 rats were performed 5 times cholesterol checks with amperometric biosensor method using a measuring instrument NESCO GCU. Data obtained from the examination were analyzed using one-way ANOVA <(0.005). The results showed that the hexane extract and red gedi leaf ethanol extract at a dose of 20 mg / kg body weight can low down the cholesterol levels by 72% or approximately 12.8 mg / dL and 14%, or about 2.4 mg / dL, compared with the group K -.

 Objective: This study was carried out to investigate the potential of Dunaliella salina microalgae to ameliorate obesity induced by high-fat diet (HFD) in male Wistar rats.Methods: Fifty rats weighing 150–160 g were fed HFD for 12 weeks. The rats were randomly divided into five groups of ten rats each. Obese rats were orally administered D. salina ethanolic extract (150 mg/Kg body weight), and orlistat as standard drug (12 mg/Kg body weight), for 6 weeks.Results: Treatment of obese rats with both D. salina and orlistat had a significant effect in reducing body and liver weights as well as visceral fat, inhibiting pancreatic lipase activity, decreased lipid profile, and increased fecal fat and ameliorating liver function enzymes activity, insulin, blood glucose, and leptin levels. Besides, food intake was insignificantly increased as a result of D. salina and orlistat treatments compared with normal control rats.Conclusion: It could be concluded that D. salina rich in β-carotene significantly reduced body weight gain and ameliorated several metabolic pathways implicated in obesity and its related complication. Hence, further intensive study must be carried out to formulate D. Salina extracts to apply as a promising natural anti-obesity nutraceutical drug.

Objective: The aim of this study was to investigate hypoglycemic and hypolipidemic effects of Annona muricata leaf ethanol extract. This study also investigated phytochemical analysis of the extract and improvement in the islet of Langerhans.Methods: Hypoglycemic and hypolipidemic effects were evaluated using alloxan induced diabetic rats. Twenty-five Wistar rats devided into five groups, that are normal group, control group that induced by alloxan 125 mg/kg body weight and given CMC (carboxymethyl cellulose), and three treatment group that induced by alloxan and given Annona muricata leaf ethanol extract (AMLEE) with 50, 100 and 200 mg/kg body weight. Blood glucose and total cholesterol levels were measured before and after alloxan induction, and 21 d after AMLEE treatment. At the end of the study, all of the animals experiment were sacrificed for histopathological examination.Results: Phytochemical analysis revealed that flavonoids, tannins, saponins, phytosterols and phenols were present in ethanol extract of Annona muricata L. leaf. At the end of the study, blood glucose and total cholesterol levels in all AMLEE treated group were decreased significantly (p<0.05). Final blood glucose level in the groups that given AMLEE 50 mg/kg body weight (86.7±14.6 mg/dl) was almost the same with that of the normal group (91.25±28.38 mg/dl). The result of the histopathological examination is not showed an improvement of the islet of Langerhans in AMLEE treated groups.Conclusion: In conclusion, the ethanol extract of Annona muricata L. leaf have hypoglycemic and hypolipidemic effects. However, there was no improvement in the islet of Langerhans damage.

Red dragon fruit (Hylocereus polyrhizus) peel contains anthocyanin, fiber and vitamin C, so it can be used to improve lipid profile in dyslipidemia. The peel of the dragon fruit is not durable, so in this study we used freeze-dried dragon fruit peel. The aim of this study was to prove that the administration of the red dragon fruit peel in a dose of 0.72 g/200 g BW, 1.08 g/200 g BW, and 1.44 g/200 g BW of rat per day for 28 days may improve lipid profile in male wistar strain white rats with high-fat diet. Lipid profiles were studied by examining of total cholesterol, LDL cholesterol, HDL cholesterol, and triglycerides. This was a pure experimental study using randomized post test only control group design. This study used experimental animal of 30 male wistar strain white rats which were divided into five groups. Measurements of total, LDL, and HDL cholesterol levels were done by CHOD-PAP method, while triglyceride level was measured with GPO-PAP method. Data were analyzed statistically by One Way Anova test. The results of this study indicated that giving the peel of red dragon fruit as much as 0.72 g lowered total cholesterol levels, 1.08 g lowered total and LDL cholesterol levels, and 1.44 g lowered total cholesterol, LDL cholesterol, triglyceride levels, and raised HDL cholesterol level. Red dragon fruit peel can be used alternatively to improve lipid profile in dyslipidemia.

Background The apolipoprotein E-deficient (apoE-/-) mouse is a well-established atherosclerotic model with impaired lipoprotein clearance and development of vessel plaques. However, the small size of the mouse limits its use as an animal model in longitudinal positron emission tomography (PET) imaging studies of atherosclerosis. Recently, apoE-/- rats have become available. This study addresses the suitability of the apoE-/- rat as model for atherosclerotic PET imaging. Methods Ten male apoE-/- rats and ten male control rats (apoE+/+) (age 10+/-1 weeks), each fed with a Western diet, were injected at baseline week 4, 12, 26 and 51, with 60 MBq of [18F]2-fluoro-2-deoxy-D-glucose. Plasma cholesterol, body weight and fat were measured. 3h after injection, a computed tomography (CT) and a 20-min PET scan were made. After the final scan, aortic tissue was collected for histological staining. Results Cholesterol levels started to increase after 4 weeks in the apoE-/- rats, whereas in the apoE+/+ rats levels stayed stable. Body weight and body fat increased more rapidly in the apoE-/- rats but were similar in both strains at the end of the study. SUVmean and max in the aortic arch and abdominal aorta were significantly higher (p &lt; 0.001) in apoE-/- versus apoE+/+ rats at weeks 12, 26 and 51. Oil red O staining showed lesions in 20.0 % of the surface of the aortic arch in the apoE-/- rats. Hardly no fatty streaks were detected in the apoE+/+ rats (2.29%). More histology data is being analyzed. Conclusion Plasma cholesterol levels were elevated in apoE-/- rats. PET imaging demonstrated differences in [18F]-FDG uptake in the aortic arch and abdominal aorta after 12, 26 and 51 weeks. Combined, these data demonstrate that apoE-/- rats represent a useful preclinical model for the non-invasive assessment of atherosclerosis in longitudinal studies.

Background: Basil and Chia seeds contain higher nutritive values like vitamin, carbohydrates, Omega-3 oil and other dietary fibers. With all these rich dietary benefits these seeds regulate necessary health conditions and maintain body weight. Ocimumbasilicum (Basil) plant have been known to contain properties of weight loss, better digestion and other health benefits. Aim: To check the Basil seed against hyperlipidemia in mice with Chia seeds. Methodology: In this research, the effect of both seeds extract on body weight and plasma lipid profile were estimated in Albino mice after raising their cholesterol levels by high fatty diet. The experiments were performed in different groups like normal control, standard control, hyperlipidemia group and four groups of diet supplemented chia or basil seeds with two different doses. Results: The biochemical analysis revealed that the supplementation of Basil seeds (400mg/kg/day) significantly lowered the levels of total plasma cholesterol, lipoproteins and triacylglycerol. Moreover, histopathological analysis of vital organs like kidneys, heart reported no toxicity. Conclusion: Extracts of Chia and Basil seeds have shown controlling effects over the given parameters in the blood and weights of the animals and these may have potential to control high fat diet-induced hyperlipidemia when taken as dietary supplements. Experiments were performed in the genetically hypertensive Koletsky rats and in their lean siblings at the age of two and three months. In the study of development of glycide and lipid abnormalities animal represents control for itself. At the age of two months Koletsky obese rats show relative to their lean controls elevation of plasma triglycerides (males +184%, females +152%) and insulin (males +169%, females +201%). During one month plasma triglycerides elevated in lean males +9%, in lean females 0%, but in obese males +21%, in obese females +139%.Considering insulinemia similar results were obtained. Thus during one month insulin elevates in lean males +19%, in lean females +23%, but in obese males +80%, in obese females +144%. During one month glucose intolerance is elevated as well only in obese rats. Total plasma cholesterol during period of one month shows no changes in both substrains of rats.Similar picture can be found in basal glycemia.In all groups of rats no changes were registered except one, i.e., obese females show decrease.Considering the substrain differences in basal glycemia then at age of one as well as two months obese of both sexes show elevation. As to the body weight at the age of two as well as three months there is increase in obese rats. The changes of body weight during one month are expressively higher in obese rats.

Chronic exposures to high-fat diets are linked to neuropathological changes that culminate in obesity-related cognitive dysfunction and brain alteration. Learning, memory performance, and executive function are the main domains affected by an obesogenic diet. There are limited effective therapies for addressing cognitive deficits. Thus, it is important to identify additional and alternative therapies. In African traditional medicine, Gnidia glauca has putative efficacy in the management of obesity and associated complications. The use of Gnidia glauca is largely based on its long-term traditional use. Its therapeutic application has not been accompanied by sufficient scientific evaluation to validate its use. Therefore, the current study sought to explore the modulatory effects of dichloromethane leaf extracts of Gnidia glauca on cognitive function in the high-fat diet- (HFD-) induced obese rats. Obesity was induced by feeding the rats with prepared HFD and water ad libitum for 6 weeks. The in vivo antiobesity effects were determined by oral administration of G. glauca at dosage levels of 200, 250, and 300 mg/kg body weight in HFD-induced obese rats from the 6th to the 12th weeks. The Lee obesity index was used as a diagnostic criterion of obesity. The Morris water maze was employed to test spatial learning and memory retention in rats. The results indicated that Gnidia glauca showed potent antiobesity effects as indicated in the reduction of body weight and obesity index in extract-treated rats. Moreover, Gnidia glauca exhibited cognitive-enhancing effects in obese rats. The positive influences on cognitive functions might be attributed to the extracts’ phytochemicals that have been suggested to confer protection against obesity-induced oxidative damage, reduction of central inflammation, and increased neurogenesis. The therapeutic effects observed suggest that Gnidia glauca might be an alternative to current medications for the symptomatic complications of obesity, such as learning and memory loss. Further studies are therefore needed to establish its toxicity profiles.

Objectives: Drugs can cause undesired effects on the fetus during pregnancy, especially embryonic/organogenesis which could lead to defects in the fetus because some types of drugs can penetrate the placenta and will undergo biotransformation into a highly reactive compound that has the potential to become a teratogenic compound. The aim of this research was to examine the teratogenic effect of bintangur leaves (Calophyllum soulattri Burm. F) ethanol extract to Sprague Dawley strain white rats. Methods: The white rats are divided into four treatment groups: Control group was given carboxymethyl cellulose Na 1%, comparison group was given trimethoprim 360 mg/kg BW, C. soulattri leaves ethanol extract (CLE) 100 mg/kg BW, and CLE 500 mg/kg BW. The treatment was administrated since organogenesis period. Cesarian section was performed to pregnant rat at the 20th day to separate the fetuses. Observation covered body weight of pregnant rats, fetal biometric, morphological malformation, and skeletal formation. Results: CLE 100 mg/kg BW and 500 mg/kg BW did not cause any change in the number of a living fetus, body weight, and length of fetuses like the comparison group. Both doses of CLE shown have a normal skeletal formation. Resorption was found in the comparison group and CLE 100 mg/kg BW with the percentage was 65.21% and 6.67%. It was found that there is no significant difference (p<0.05) between both doses of CLE compared to control group. Conclusion: From the results, it is concluded that CLE did not have the teratogenic effect.
