Background: Development of new preventive agents for dental caries is needed. One of the candidates for preventive agents from natural products is Soursop leaf. The present study aimed to determine the effect of Soursop leaf oil on the cultured epithelial and fibroblast cells.Methods: In this experimental study, Soursop leaf essential oils were provided, and their e?ect was discovered on epithelial and fibroblast cells line using MTT assay. The MTT assay was conducted to measure the activity of enzymes that reduce MTT and switch it to formazan dye creating a purple colour. Using a microplate reader, the optical density was measured at 550 nm and the absorbance value directly represented relative cell numbers.Results: Data compilation and analysis were done using one-way analysis of variance. Soursop leaf essential oils exhibited variable noxious e?ects on cultured cells. The present study shows that epithelial cell death was less than 30% at the concentration 2.5 �l/ml while the percentage of fibroblast cell death was less than 30% at smaller concentrations of 1.25 �l/ml. Through an increase in the concentration of Soursop leaf essential oils, the toxicity of these materials substantially increased (p<0.05)Conclusion: Soursop leaf essential oils at certain concentrations may cause epithelial and fibroblast cell death.

A B S T R A C TIntroduction. A CO 2 incubator is an essential tool for the initiation of theproliferation of primary culture cells or cell lines. In principle, this tool works bykeeping the sample cell line at an optimum temperature of 37 o C and 5% carbondioxide supply. The ability of the CO 2 incubator to maintain temperature and supplyof 5% carbon dioxide are essential points in the development of the CO 2 incubator.This study is an attempt to convince the potential of Sriwijaya CO 2 Incubator inmaintaining the proliferation ability of cultured cells in an in vitro study. Methods.This study is an experimental pre-post test that explores the percentage of viabilityof primary culture cells (fibroblasts) before and after incubation in CO 2 incubators.The object of this study was fibroblast cells obtained from the prepuce of patientswho performed circumcision. Results. Fibroblast cell proliferation in CO 2 incubatorsshows an increase in the number of fibroblast proliferation which can be seen withthe increasing number of cells visualized by inverted microscopy. Conclusion.Sriwijaya CO 2 incubator has the potential to be used in in vitro research to triggerthe growth and proliferation of fibroblast cells.

Novel psychoactive substances (NPS) are synthetic substances belonging to diverse groups, designed to mimic the effects of scheduled drugs, resulting in altered toxicity and potency. Up to now, information available on the pharmacology and toxicology of these new substances is very limited, posing a considerable challenge for prevention and treatment. The present in vitro study investigated the possible mechanisms of toxicity of two emerging NPS (i) 4′-methyl-alpha-pyrrolidinoexanophenone (3,4-MDPHP), a synthetic cathinone, and (ii) 2-chloro-4,5-methylenedioxymethamphetamine (2-Cl-4,5-MDMA), a phenethylamine. In addition, to apply our model to the class of synthetic opioids, we evaluated the toxicity of fentanyl, as a reference compound for this group of frequently abused substances. To this aim, the in vitro toxic effects of these three compounds were evaluated in dopaminergic-differentiated SH-SY5Y cells. Following 24 h of exposure, all compounds induced a loss of viability, and oxidative stress in a concentration-dependent manner. 2-Cl-4,5-MDMA activates apoptotic processes, while 3,4-MDPHP elicits cell death by necrosis. Fentanyl triggers cell death through both mechanisms. Increased expression levels of pro-apoptotic Bax and caspase 3 activity were observed following 2-Cl-4,5-MDMA and fentanyl, but not 3,4-MDPHP exposure, confirming the different modes of cell death.

Aim:Endodontic retreatment is a procedure that removes the lling materials from the root canals followed by their cleaning, shaping and obturation. This in-vitro study aimed to compare and evaluate the ability of various essential oils as solvents in dissolving gutta-percha, epoxy resin, and zinc oxide eugenol (ZOE) cements. Materials and methods: A total of 28 cylindrical specimens in each group ZOE, epoxy resin, 28 ISO size 40 gutta-percha cones were prepared and divided into four groups for immersion in the different solvents, i.e. lemon oil, citronella oil, lavender oil, and TCE(Tetrachloroethylene (control)) for 5 minutes. The obturating materials dissolution in the solvents were obtained by the difference between the pre-immersion original weight and the post-immersion weight on a digital analytical scale. Data were statistically analysed by a paired t-test and Analysis of Variance (ANOVA) (p < 0.05). Results: Order of efcacy of dissolution of essential oils was found to be lemon oil > Citronella oil > Lavender oil and was highly signicant (p < 0.01). In all the solvents, Gutta-percha showed maximum dissolution (Fvalue:149.56) followed by ZOE(89.07 ) and resin sealer least (23.86). Conclusion: It can be concluded that the lemon oil can be used as a solvent for dissolving obturating materials.

To conduct a study regarding the antibacterial activity of essential oils against bacteria causing Caries. Essential oils are distillates of the volatile compounds of a plant’s secondary metabolism and may act as photoprotective agents. Their curative effect has been known since antiquity. It is based on a variety of pharmacological properties which are specific for each plant species. The mouth contains a variety of oral bacteria, but only a few species of bacteria are believed to cause dental caries. Antibacterial activity of the three essential oils, Rosemary oil, Holy basil oil, Thyme oil was screened against Streptococcus mutans, using disc diffusion technique. The rosemary oil was more effective against Streptococcus mutans with a zone of inhibition of 52 mm diameter (at concentration 200 µl), Rosemary oil showed a zone of inhibition of 44 mm diameter and with thyme oil, the zone diameter was 30 mm. The results of this study showed that the essential oils at different concentrations exhibited antibacterial activity against the bacterial species tested.

Objective: This aim of this study was at compare the fibroblast cytotoxicicty of four restorative materials - a conventional glass ionomer cement (GC Fuji Type II GIC), a ceramic reinforced glass ionomer cement (Amalgomer), a giomer (Beautifil II) and a resin composite (Filtek Z350) at three different time periods (24, 48 and 72 hours). Method: The succinyl dehydrogenase (MTT) assay was employed. Cylindrical specimens of each material (n=15) were prepared and stored in Dulbecco's modified Eagle medium, following which L929 fibroblasts were cultured in 96 well plates. After 24 hours of incubation, the MTT assay was performed to detect the cell viability. The method was repeated after 48 and 72 hours. The impact of materials and exposure times on cytotoxicity of fibroblasts was statistically analyzed using two way ANOVA (P=0.05). Results: Both time and material had an impact on cell viability, with giomer demonstrating the maximum cell viability at all time periods. The cell viability in the giomer group was significantly different from all other materials at 24 and 72 hours (P&lt;0.05), while at 48 hours giomer was significantly different only with resin composite (P&lt;0.05). Conclusions: Giomers showed better biocompatibility than conventional and ceramic reinforced glass ionomer cements and, resin composite. Ceramic reinforced glass ionomer demonstrated superior biocompatibility compared to conventional glass ionomer.

Essential oils from the dried spikes ofNepeta tenuifolia(Benth) are obtained by steam distillation. Pulegone was identified as the main component in the spikes ofN. tenuifoliathrough analysis, with greater than 85% purity obtained in this study. The essential oils are extremely active against all Gram-positive and some Gram-negative reference bacteria, particularlySalmonella enterica,Citrobacter freundii, andEscherichia coli. The minimum inhibitory concentration was found to be between 0.08 and 0.78% (againstS. enterica), 0.39 and 0.78% (againstC. freundii), and 0.097 and 0.39% (againstE. coli), whereas the minimum bactericidal concentration varied in range from 0.097% to 1.04%. In general, the essential oils show a strong inhibitory action against all tested reference strains and clinical isolates. However, the antibacterial activity of EOs against bothPseudomonas aeruginosareference strains and clinical isolates was relatively lower than other Gram-negative pathogens. The essential oils ofN. tenuifoliaalso displayed bactericidal activities (MBC/MIC < 4) in this study. These findings reflect the bactericidal activity of the essential oils against a wide range of multidrug-resistant clinical pathogens in an in vitro study. In addition, we propose the fragmentation pathways of pulegone and its derivatives by LC-ESI-MS/MS in this study.

The objective of this study was to evaluate in vitro the effects of three essential oils (EO) [cinnamon leaf (250 mg L-1), garlic oil (100 and 250 mg L-1), and juniper berry oil (20 mg L-1)] and two EO compounds (EOC) [anethol (20 mg L-1) and p-cymene (20 mg L-1)] on proteolytic, deaminative and methanogenic activities of mixed ruminal bacteria. Concentrations of total VFA were similar (P > 0.05) among treatments. With the exception of cinnamon and garlic oils, which reduced (P < 0.05) the proportion of propionate, the other EO and EOC had no effect on the proportions of individual VFA, compared with the control. Proteolytic activity of ruminal bacteria was unaffected (P > 0.05) by treatments; however, bacterial deaminative activity and NH3 concentration were increased (P < 0.05) by the addition of EO (except for cinnamon leaf oil and garlic oil at 250 mg L-1) and EOC. Except for anethol, methanogenic activity of ruminal bacteria was reduced (P < 0.05) by EO and EOC, which was reflected by a marked decrease in methane concentration. This study shows that at the concentrations evaluated, anethol, garlic oil (100 mg L-1), juniper berry oil, and p-cymene may not be beneficial to improve efficiency of N utilization in ruminants because they enhance deaminative activity, while cinnamon and garlic oil (250 mg L-1) could be good alternatives to antibiotics because they reduce methanogenic activity of ruminal bacteria.
