CHARACTERIZATION OF MICROSPOROGENESIS IN THE TETRAPLOID FORM WHICH INITIAL FOR APPLE TREE SELECTION

One of the priority directions in apple breeding at VNIISPK is breeding with polyploidy using for the development of triploid apple cultivars with high commercially valuable traits. Tetraploid forms are an intermediate in the creation of apple varieties with a triple set of chromosomes, so the study of the state of the generative sphere is important in assessing them as donors of diploid gametes in breeding with polyploidy using. Data on the study of meiosis in microsporogenesis in tetraploid apple form 34-21-36 [30-47-88 (Liberty×13-6-106 (s.s. Suvorovetz)) (4x) × Krasa Sverdlovska (2x)] obtained in the laboratory of apple breeding, are presented in the article. The disorders at all successive stages of meiosis were observed. At different stages of division there were run-ins and emissions of chromosomes in the cytoplasm of microsporocyte, lags and emissions, presence of micro-nuclei and nuclei in excess of the number. In the metaphase-1, the microspocyte was observed at the same time with two types of disorders in the same cell (run-ins +emissions). At the stage of tetrads, polyads were formed (pentads, hexads, heptads, octads, nanads). The amount of disorder types varied at different stages of microsporogenesis from 1 to 5. Despite the presence of disorders during meiosis, the most part of microsporocytes in the tetraploid apple form had correct pictures of division and in 36.1% of cases the process of microspogenesis ended with the formation of normal tetrads, decaying after maturation into normal microspores containing a diploid set of chromosomes. On the basis of the reduction division it was concluded that tetraploid apple form 34-21-36 (4x) was suitable as a pollinator in intervalent crosses.

As a result of the long-term research, several of the most promising donors of diploid gametes were selected among apple cultivars and elite seedlings. Among the cultivars, these are Antonovka Ploskaya (2-4-4-4x), Melba tetraploid (4x), Papirovka tetraploid (2-4-4-4x) and Wealthy tetraploid (2-4-4-4x), while among the seedlings these are 13-6-106 (Suvorovetz seedling) (4x) and 30-47-88 (Liberty × 13-6-106) (4x + Vf). New apple cultivars of summer maturation Avgusta, Dariona, Maslovskoye and Osipovskoye have already been released with the participation of Papirovka tetraploid (2-4-4-4x) and included in the State register of breeding achievements admitted for use. New zoned cultivars Alerksandr Boyko, Bezhin Lug, Vavilovskoye and Ministr Kisilev have been released with the participation of Wealthy tetraploid (2-4-4-4x). Triploid apple cultivars Orlovsky Partizan, Patriot and Mars have been released with the participation of 13-6-106 (Suvorovetz seedling) as a donor of diploid gametes. Orlovsky Partizan and Patriot are zoned, while Mars is passing the state testing. 30-47-88 (Liberty × 13-6-106) (4x + Vf) is of particular value as both a donor of diploid gametes and a donor of scab immunity (Vf). Priorities in apple breeding at VNIISPK are the development of the immune to scab (24 scab immune cultivars are zoned) and columnar cultivars (5 cultivars are zoned: Vostorg, Girlianda, Poezia, Priokskoye and Orlovskaya Yesenia). We have already developed triploid apple cultivars that have immunity to scab. It is tempting to create triploid cultivars that are immune to scab and columnar ("three in one"). The creation of such cultivars will allow to provide more regular fruiting in the orchard and fruits of high commodity and consumer qualities (due to triploidy), improve the environmental situation in the orchard and its surroundings (due to immunity to scab), and significantly increase the yield (due to column-shaped varieties). We hope that in the coming years such cultivars will be created, since there are already selected apple seedlings with such a combination of valuable features.

Russian Research Institute for Fruit Crop Breeding (VNIISPK) is the main supplier of new valuable apple cultivars to update the assortment. For the last 30 years more than 50 apple cultivars meeting the requirements of modern gardening have been developed at VNIISPK and included in the State Register of Breeding Achievements. The time of single breeders is a thing of the past, and in connection with the ever-increasing requirements for varieties, researchers of different specialties should work at their creation. A well-coordinated interdisciplinary team of researchers and technicians work in VNIISPK at the development of new apple varieties. In addition to breeders, researchers in physiology, biochemistry, processing of fruits, cytoembryology, genetics and agronomic practice are involved in the work. Along with other areas of selection, apple breeding with polyploidy using was started at the Institute in 1970. To date, 19 triploid apple varieties have been created for the first time in the world at our Institute from intervalent crosses of diploid x tetraploid type. 11 of them are included in the State Register of Breeding Achievements Admitted for Use (zoned). It has experimentally been confirmed that triploid apple cultivars differ from diploids in higher weight of fruit and their marketability and are not inferior to diploids in the attractiveness of the appearance and taste of the fruit. Apple breeding for immunity to the most harmful scab was started at our institute in 1977, to date, 25 scab immune apple cultivars have been developed and zoned, 9 of them are triploids. The first domestic scab immune Imrus was zoned in Central Russia in 1996. We have shown that diploid and triploid varieties immune to scab are not inferior to non-immune varieties by fruit weight. The first in Russia and world 9 triploid resistant to scab apple varieties are Vavilovskoe, Zhilinskoe, Mars, Prazdnichnoe, Spasskoe, Trener Petrov including have been zoned Aleksandr Boyko, Yablochnyy Spas, Maslovskoe in our opinion should significantly enrich the range of varieties of Russia midland apple. The paper gives a comparative assessment of commodity and consumer qualities of new diploid and triploid fruit varieties of apple selection of All-Russian Research Institute of Fruit Crops Breeding.

ABSTRACT Climate is the set of average atmospheric conditions that characterizes a region. It directly influences the majority of human activities, especially agriculture. Climate classification systems (CCSs) are important tools in the study of agriculture, enabling knowledge of the climatic characteristics of a region. Thus, we aimed to perform the climatic characterization of the State of Paraná using the methods proposed by Köppen and Geiger (1928), modified by Trewartha (1954) (KT), Thornthwaite (1948) (TH) and Camargo (1991) and modified by Maluf (2000) (CM), using data from the European Center for Medium-Range Weather Forecast (ECMWF) model. The results of spatial interpolation (virtual stations) were performed using the Kriging method in spherical shape with one neighbour and resolution of 0.25°. The CCSs displayed the ability to separate the warm and dry from cold and wet regions. The most predominant climates were Cfa (temperate humid with hot summers), C1rA'a' (sub-humid with little water deficiency, megathermal) and ST-UMi (humid subtropical with dry winter), according to KT, TH and CM, respectively. CM is an intermediate CCS between KT and TH.

Physical and chemical properties of 6 apple varieties were measured to gain the variations. The qualities of apples are important for the consumer. These quality attributes can be described by colour, texture and physical attributes such as size and shape. Individual sugars, organic acids, and vitamin C were quantified in six apple cultivars with same soil, climatic conditions and altitudes. The results showed physical and chemical characterization existed different variance in apple varieties, the variance ranged from 6.1% to 109.05%. Fructose was the most dominant sugar in the different apple cultivars, followed by glucose and sucrose, while malic acid was the principal organic acid.

In this study, a metabolomic investigation was presented to correlate single polyphenolic compounds in apple pulp with quality characteristics such as antioxidant activity and content of phenolic compounds and anthocyanins in apple skin. Since the concentration of these compounds is influenced by environmental factors, the twenty-two apple cultivars originate from the same site. The polyphenolic compounds were analyzed by ultra-high-performance liquid chromatography coupled with triple quadrupole mass spectrometry (UHPLC-QqQ-MS/MS). The antioxidant activity, phenolic content, and anthocyanins were evaluated on the sunny and the shady sides of apple skin by spectrometric assays. In old apple varieties, the measured parameters were higher than in the commercial and red-fleshed varieties. By contrast, the profile of flavan-3-ols and anthocyanins was variable amongst commercial and red-fleshed varieties. The partial least square (PLS) method was applied to investigate the association between the skin proprieties and the metabolic profile of the pulp. The highest coefficients of determination in prediction (Q2) were obtained for compounds quantified in old cultivars. These results provided information to define the old apple varieties as a reliable group based on the pathway of the antioxidant compounds and anthocyanins content. Our results show the possibility to find cultivars with promising health features based on their content of polyphenols suitable for commercialization or breeding.

The basic moments of a process of obtaining apple tetraploids as donors of diploid gametes for apple breeding with polyploidy using are demonstrated. In industrial terms, triploids are of the greatest importance. The manifested effect of heterosis leads to the improvement of many characteristics - higher resistance to diseases, pests and adverse environmental conditions, greater autogamy than in diploids, less pronounced periodicity of fruiting, larger fruits and a convenient crown for harvesting. Triploids are developed as a result of crosses 2n × 3n or 2n × 4n. Tetraploids are necessary for more successful apple breeding with polyploidy using. For industry they are not of great importance but they are of interest as donors of diploid non-reduced gametes and allow to make the selection process more directional. One of the methods of experimental polyploidy is the induction of polyploids using mutagenes. The germs were treated with colchicines at concentrations 0.1% - 0.4% during 24 and 48 hours. According to the morphology the obtained plants were divided into five groups. Colchicine concentrations 0.3% and 0.4% during 48 hours of the treatment had a disastrous impact on the development of germs. As a result of the cytological analysis, tetraploids and chimeras were revealed, which were obtained from the seeds from the open pollination of Orlik and Svezhest (treatment variants – 0,1% colchicines solution and 24 and 48 hours of exposition), as well as from the seeds obtained as a result of the Svezhest × Bolotovskoye crossing. Tetraploids had a normal growth but they differed in large leaves, while chimeras were of low size with normal leaves and internodes.

Abstract:Description Alternative: Abstract: The students 'reading ability is low, it certainly affects the students' writing ability. Implementation of the School Literacy Movement at the stage of habituation to reading, the lack of assistance in extracurricular reading clubs that are scheduled at the State Junior High School 5 Malang. This study aims to foster students' character through the culture of school literacy that embodies the Implementation of the School Literacy Movement so that students of SMP Negeri 5 Malang become lifelong learners. This study uses a qualitative approach with a descriptive type that describes the School Literacy Movement in State Junior High School 5 Malang by using data collection techniques of observation, interviews, and documentation. The results showed that: 1) The habituation phase was carried out by way of students bringing reading books from home or borrowing books to the library. At this stage a class reading corner was prepared, reading 15 minutes before learning began, and a literacy journal; 2) The Development and Learning Phase increases the school resources especially at the State Junior High School 5 Malang, namely the existence of a reading corner in each class, an increase in the number of books, a 30-minute reading club, and a product of the School Literacy Movement.Keywords: School Literacy Movement, Habituation, Development, and Learning Abstrak: Kemampuan membaca peserta didik tergolong rendah pasti berpengaruh terhadap kemampuan menulis peserta didik. Implementasi Gerakan Literasi Sekolah pada tahap pembiasaan minat baca, kurangnya pendampingan pada ekstrakurekuler club baca yang di agendakan di Sekolah Menengah Pertama Negeri 5 Malang.  Penelitian ini bertujuan untuk menumbuhkembangkan budi pekerti peserta didik melalui pembudayaan literasi sekolah yang mewujudkan dalam Implementasi Gerakan Literasi Sekolah supaya peserta didik SMP Negeri 5 Malang menjadi pembelajar sepanjang hayat. Penelitian ini menggunakan pendekatan kualitatif dengan jenis deskriptif yang mendeksripsikan Gerakan Literasi Sekolah di Sekolah Menengah Pertama Negeri 5 Malang dengan  menggunakan teknik pengumpulan data observasi, wawancara, dan dokumentasi. Hasil penelitian menunjukan bahwa: 1) Tahap Pembiasaan dilaksanakan dengan cara peserta didik membawa buku bacaan dari rumah atau meminjam buku ke perpustakaan. Pada tahap ini sudah disiapkan pojok baca kelas, membaca 15 menit sebelum pembelajaran dimulai, dan jurnal literas; 2) Tahap Pengembangan dan Pembelajaran meningkatkan sumber daya sekolah khusnya di Sekolah Menengah Pertama Negeri 5 Malang yaitu adanya pojok baca di masing-masing kelas, penambahan jumlah buku, adanya club baca 30 menit, dan hasil produk Gerakan Literasi Sekolah. Recent detailed simulations have shown that an insufficiently accurate characterization of the contamination of unresolved polarized extragalactic sources can seriously bias measurements of the primordial cosmic microwave background (CMB) power spectrum if the tensor-to-scalar ratio r ∼ 0.001, as predicted by models currently of special interest (e.g., Starobinsky’s R2 and Higgs inflation). This has motivated a reanalysis of the median polarization fraction of extragalactic sources (radio-loud AGNs and dusty galaxies) using data from the Planck polarization maps. Our approach, exploiting the intensity distribution analysis, mitigates or overcomes the most delicate aspects of earlier analyses based on stacking techniques. By means of simulations, we have shown that the residual noise bias on the median polarization fraction, Πmedian, of extragalactic sources is generally ≲0.1%. For radio sources, we have found Πmedian ≃ 2.83%, with no significant dependence on either frequency or flux density, in good agreement with the earlier estimate and with high-sensitivity measurements in the frequency range 5–40 GHz. No polarization signal is detected in the case of dusty galaxies, implying 90% confidence upper limits of Πdusty ≲ 2.2% at 353 GHz and of ≲3.9% at 217 GHz. The contamination of CMB polarization maps by unresolved point sources is discussed.

This study was conducted with the aim of developing fruit spirits by utilizing old (autochthonous) apple and pear cultivars that can be attractive to both consumers and producers. Consumers of spirits could enjoy the unique flavor, and producers could gain an opportunity for brand development. In total, eight old apple cultivars (Sarija, Žuja, Samoniklica, Prijedorska zelenika, Bobovec, Masnjača, Lijepocvjetka, and Šarenika) and three pear cultivars (Budaljača, Krakača, and Kalićanka) from Bosnia and Herzegovina were used for the spirits production and for characterizing the flavor of distillates. Golden Delicious was used as a representative of commercial apple cultivar. The aroma profile was conducted through the identification of minor volatile organic compounds (VOCs) and the sensory perception of spirits. Analysis of the VOCs was performed by gas chromatography mass spectroscopy (GC/MS) techniques after enrichment via solid-phase microextraction (SPME). Sensory evaluation was performed by 12 trained panelists. Overall, 35 minor volatile compounds were found in spirits: 13 esters, 7 alcohols, 6 acids, 5 terpenes, and 4 aldehydes. Significant differences were detected in the distribution and quantity of the VOCs, which were fruit cultivar-dependent. Spirits made from Šarenika apple cultivar showed the largest amount of all acids, especially short- and medium-chain fatty acids; however, this richness was not correlated with pleasant sensory attributes. Spirits obtained from Prijedorska zelenika and Masnjača apple cultivars had the best sensory attributes. Budeljača and Krakača pears are promising cultivars as flavoring in spirits production.
