Abstract : Strychnos ligustrina stem has been empirically used by the people of West Nusa Tenggara and Bali in the treatment of malaria, tooth ache and diarrhea, but there is no scientific data that supports it. This study aims to determine and prove the antibacterial activity of Strychnos ligustrina methanol extract to pathogenic bacteria in vitro and in vivo. This research is an experimental study conducted by measuring the inhibition zone (mm) growth of pathogenic bacteria, determining minimum inhibitory concentration (MIC) and minimum killing concentration (MKC) in vitro, and determining the percentage of antibacterial activity of methanol extract of S. ligustrina stem in vivo. The experiment was conducted using 4 groups of concentrations of S. ligustrina stem methanol extract in an in vitro study of 25, 50, 75, and 100% with ciprofloxacin as a positive control and aquadest as a negative control. In vivo studies experiments were carried out using 6 treatment groups of test animals male mice Balb / c (Mus musculus). The in vitro test results showed that methanol extract of S. ligustrina stems was able to inhibit the growth of pathogenic bacteria with medium categories of clinical isolates of Staphylococcus aureus and categories of weaks to Klebsiella pneumonia and Escherichia coli isolates. The minimum inhibitory concentration (MIC) for S. aureus and K. pneumonia bacteria isolates was at a concentration of 25% while for E. coli isolates at a concentration of 30%. The methanol extract of the S. ligustrina stem has no killing power against the pathogenic bacteria tested. Antibacterial activity in vivo was able to inhibit the growth of S. aureus pathogenic bacteria by 6.60% (at 25% concentration), 8.62% (at 50% concentration), and 17.31% (at 100% concentration), against K. pneumonia was 11.85% (at 25% concentration), 51.21% (at 50% concentration), and 65.92% (at 100% concentration), against E. coliat 19.18% (at concentration 25%), 29.98% (at 50% concentration), and 40.88% (at 100% concentration). Methanol extract of S. ligustrina stem proved to have antibacterial activity in vitro and in vivo. Key words: Srychnos ligustrina, pathogenic bacteria, antibacterial, in vitro, in Vivo. Abstrak : Strychnos ligustrina secara empiris  telah digunakan oleh masyarakat Nusa Tenggara Barat dan Bali dalam pengobatan penyakit malaria, sakit gigi, dan diare, tetapi belum ada data ilmiah yang mendukung. Penelitian ini bertujuan untuk menentukan dan membuktikan aktivitas antibakteri ekstrak metanol batang bidara laut terhadap bakteri patogen secara in vitrodan in vivo. Penelitian ini merupakan penelitian eksperimental yang dilakukan dengan mengukur zona hambat (mm) pertumbuhan bakteri patogen, menentukan konsentrasi hambat minimum (KHM) dan konsentrasi bunuh minimum (KBM) secara in vitro, serta menentukan persentase aktivitas antibakteri ekstrak metanol batang bidara laut secara in vivo. Percobaan dilakukan menggunakan 4 kelompok konsentrasi ekstrak metanol batang bidara laut pada penelitian in vitro yaitu 25, 50, 75, dan 100% dengan ciprofloxacin sebagai kontrol positif serta aquadest sebagai kontrol negatif. Pada penelitian in vivo percobaan dilakukan menggunakan 6 kelompok perlakuan hewan uji mencit jantan galur Balb/c (Mus musculus). Hasil uji in vitro menunjukkan ekstrak metanol batang bidara laut mampu menghambat pertumbuhan bakteri patogen dengan kategori sedang terhadap Staphylococcus aureus isolat klinis dan kategori lemah terhadap Klebsiella pneumonia dan Escherichia coli isolat klinis. Nilai konsentrasi hambat minimum (KHM) untuk isolat bakteri S. aureus dan K. pneumoniae adalah pada konsentrasi 25% sedangkan untuk isolat E. coli pada konsentrasi 30%. Ekstrak metanol batang bidara laut tidak memiliki daya bunuh terhadap bakteri patogen yang diuji. Aktivitas antibakteri secara in vivo mampu menghambat pertumbuhan bakteri patogen S. aureus sebesar 6,60% (pada konsentrasi 25%), 8,62% (pada konsentrasi 50%), dan 17,31% (pada konsentrasi 100%), terhadap K. pneumonia sebesar 11,85% (pada konsentrasi 25%), 51,21% (pada konsentrasi 50%), dan 65,92% (pada konsentrasi 100%),   terhadap E. coli sebesar 19,18% (pada konsentrasi 25%), 29,98% (pada konsentrasi 50%), dan 40,88% (pada konsentrasi 100%). The volatile compounds obtained by hydrodistillation of the aerial parts of Rosmarinus tournefortii De Noé. growing wild in the occidental region of Algeria were analyzed by GC/MS. Thirty-six compounds were characterized representing 95.6% of the essential oil, with camphor (37.6%), 1,8-cineole (10.0%), p-cymene-7-ol (7.8%), and borneol (5.4%) as the major components. The antimicrobial activity was evaluated against three pathogenic bacteria: Gram-negative ( Escherichia coli and Pseudomonas aeruginosa) and Gram-positive ( Staphylococcus aureus). The minimum inhibitory concentration (MIC; mg/mL) was determined by sub-culture on Muller Hinton agar plates. The essential oil exhibited strong antibacterial activity against E. coli and P. aeruginosa, and was also active against Staphylococcus aureus.

The ethanol extracts of root, bark and leaf of Bridelia ferruginea was investigated for antibacterial activity against clinical isolate of Staphylococcus aureus and Escherichia coli. The extracts had significant antibacterial activity in vitro at concentration of 25 mg/ml, 50 mg/ml, 100 mg/ml and 200 mg/ml and in vivo at dose of 50 mg/kg and 100 mg/kg. The root extract in vitro had the highest zone of inhibition, followed by the bark extract for both Staphylococcus aureus and Escherichia coli. The concentration of 200 mg/ml had the highest zone of inhibition in vitro. The minimum inhibitory concentration (MIC) showed a decreasing inhibitory effect of the plant extracts for both Staphylococcus aureus and Escherichia coli as the concentration decreases with root having 3.125 mg/ml, bark having 6.25 mg/ml and leaf having 25 mg/ml for Staphylococcus aureus and Escherichia coli. Likewise, the minimum bactericidal concentration (MBC) showed decreasing bactericide effects with decrease concentration with root having 12.5 mg/ml, bark having 12.5 mg/ml and leaf having 25 mg/ml for Escherichia coli while root had 6.25mg/ml, bark had 12.5mg/ml and leaf had 25mg/ml for Staphylococcus aureus. The in vivo investigation showed that the root and bark extract exhibited antibacterial activity on both Staphylococcus aureus and Escherichia coli at doses of 100mg/kg and 50mg/kg; the root extract had higher activity than the bark and root/bark combined. The dose of 100 mg/kg had the highest colonies reduction for Staphylococcus aureus and Escherichia coli in vivo. Preliminary phytochemical screening of root, bark and leaves of Bridelia ferruginea revealed the presence of tannins, flavonoids, carbohydrates, cardiac glycoside (root, bark and leaves), saponins (root and bark). The presence of tannins, saponins, flavonoid, cardiac glycoside and carbohydrate in the bark and root extracts of the plant indicates that the bark and root extracts were pharmacological importance

Objectif : L’objectif de ce travail était d’évaluer in vitro l’activité antimicrobienne des extraits de feuilles et tige de Jatropha multifida sur la croissance de Candida albicans, Escherichia coli et Staphylococcus aureus, puis d’évaluer in vivo la toxicité de cette plante. Méthodologie et résultats : Les méthodes de diffusion en milieu gélosé et de microdilution en milieu liquide ont été utilisées pour évaluer l’effet antimicrobien. Une étude en subaigüe était réalisée afin d’explorer les effets toxiques de l’extrait aqueux des feuilles. Les résultats des tests antimicrobiens montrent une activité des extraits de feuilles et tige de J. multifida sur la croissance des souches utilisées avec des diamètres de zones d’inhibition allant de 8 à 25 mm et des concentrations minimales inhibitrices (CMI) variant de 0,039 mg/mL à 1,25 mg/mL à l’exception des souches de E. coli qui sont résistantes aux extraits de la tige. L’administration en subaigüe de l’extrait aqueux des feuilles de J. multifida à la dose de 600 mg/kg entraîne une perte significative de poids chez les souris. Conclusion et applications des résultats : Les extraits aqueux, éthanolique et hydroéthanolique des feuilles et tige de J. multifida possèdent d’activité antimicrobienne et pourraient être utilisés dans le traitement des Candidoses à C. albicans et des infections à S. aureus. Mais l’essai de toxicité subaigüe montre que l’extrait aqueux de la plante serait toxique. Des études toxicologiques approfondies restent donc nécessaires sur ces extraits afin de mieux élucider leur inocuité. But the subacute toxicity test shows that the aqueous extract of the plant would be toxic. Extensive toxicological studies therefore remain necessary on these extracts in order to better elucidate their safety. The antibacterial activity of geraniol and its effect in combination with ampicillin, amoxicillin and clarithromycin against Staphylococcus aureus, Escherichia coli and Helicobacter pylori was tested. The minimum inhibitory concentrations (MICs) and combinatory effects of geraniol against the bacteria were assessed by using the modified broth microdilution and checkerboard assay, respectively. The combinatory effect is expressed as fractional inhibitory concentration index (FICI). The MIC of geraniol against S. aureus, E. coli and H. pylori was found to be 11200, 5600, and 7325 μg/mL, respectively. A significant synergistic effect was observed with geraniol and ampicillin against S. aureus with FICI in the range 0.19 to 0.32. Geraniol and ampicillin exhibited a partial synergistic effect against E. coli. A similar effect was observed with geraniol and clarithromycin against S. aureus. A partial synergistic effect was observed with clarithromycin and geraniol against H. pylori with the FICI value in the range 0.86 to 0.89. An additive effect was observed with geraniol and amoxicillin combination against H. pylori. However, the amoxicillin and clarithromycin dose was reduced by thirty-two fold when combined with geraniol against H. pylori. The anti- H. pylori effect of geraniol with clarithromycin and amoxicillin could be of potential interest in the treatment of H. pylori infection and associated ulcers in humans. Further, geraniol, in combination with other antibiotics, has substantial therapeutic potential against S. aureus and E.coli infection.

ABSTRACT Expression of two genes of unknown function, Staphylococcus aureus scdA and Neisseria gonorrhoeae dnrN, is induced by exposure to oxidative or nitrosative stress. We show that DnrN and ScdA are di-iron proteins that protect their hosts from damage caused by exposure to nitric oxide and to hydrogen peroxide. Loss of FNR-dependent activation of aniA expression and NsrR-dependent repression of norB and dnrN expression on exposure to NO was restored in the gonococcal parent strain but not in a dnrN mutant, suggesting that DnrN is necessary for the repair of NO damage to the gonococcal transcription factors, FNR and NsrR. Restoration of aconitase activity destroyed by exposure of S. aureus to NO or H2O2 required a functional scdA gene. Electron paramagnetic resonance spectra of recombinant ScdA purified from Escherichia coli confirmed the presence of a di-iron center. The recombinant scdA plasmid, but not recombinant plasmids encoding the complete Escherichia coli sufABCDSE or iscRSUAhscBAfdx operons, complemented repair defects of an E. coli ytfE mutant. Analysis of the protein sequence database revealed the importance of the two proteins based on the widespread distribution of highly conserved homologues in both gram-positive and gram-negative bacteria that are human pathogens. We provide in vivo and in vitro evidence that Fe-S clusters damaged by exposure to NO and H2O2 can be repaired by this new protein family, for which we propose the name repair of iron centers, or RIC, proteins.

Penelitian ini bertujuan untuk menguji daya hambat dari ekstrak metanol beberapa jenis porifera terhadap pertumbuhan bakteri E. coli dan S. aureus. Pengujian aktivitas antibakteri menggunakan metode Kirby-Bauer, yaitu dilakukan dengan mengukur zona hambat di sekeliling cakram kertas. Ekstrak Haliclona sp dapat menghambat pertumbuhan S. aureus pada konsentrasi ekstrak 30%, 60% dan 90% dengan hasil pengukuran diameter zona hambat 13.50 mm, 20.67 mm dan 27.33 mm; serta menghambat pertumbuhan E. coli dengan diameter zona hambat 10.08 mm, 12.83 mm dan 14.17 mm. Daya hambat ekstrak Agelas sp terhadap S. aureus menunjukkan diameter zona hambat 8.33 mm dan hanya menunjukkan pada konsentrasi ekstrak 90%, sementara daya hambat ekstrak Agelas sp pada konsentrasi 30%, 60% dan 90% terhadap E. coli menunjukkan diameter zona hambat sebesar 7.67 mm, 10.17 mm dan 14.17 mm. Daya hambat Spheciospongia sp terhadap S. aureus dan E. coli hanya terlihat pada konsentrasi ekstrak sebesar 90% dengan diameter zona hambat adalah 8.42 mm dan 8.75 mm. Berdasarkan hasil yang diperoleh, dapat disimpulkan bahwa ekstrak Haliclona sp memiliki potensi aktivitas antibakteri yang dapat digunakan sebagai bahan dasar pembuatan obat antibiotik.This research aimed to test the inhibition capabilities of methanol extract from several kinds of Porifera on Escherichia coli and Staphylococcus aureus growth. The antibacterial activity test using the Kirby-Bauer method, which delivered by measuring the inhibition zone around paper disc. The extract of Haliclona sp can inhibit the S. aureus growth at 30%, 60% and 90% of extracts concentration with the measurement of inhibition zone diameters are 13.50 mm, 20.67 mm and 27.33 mm; also inhibit the E. coli growth with inhibition zone diameters are 10.08 mm, 12.83 mm and 14.17 mm. The inhibition capability of Agelas sp extract on S. aureus shows that the inhibition zone diameters is 8.83 mm and only appear at 90% of extracts concentration, meanwhile the inhibition capability of Agelas sp extract at concentration 30%, 60% and 90% on E. coli shows diameters of inhibition zone are 7.67 mm, 10.17 mm and 14.17 mm. The inhibition capability of Spheciospongia sp on S. aureus and E. coli only occurred at 90% of extracts concentration with inhibition zone diameters 8.42 mm and 8.75 mm. Based on the results, it can be assumed that extracts of Haliclona sp has a potential antibacterial activity that can be used as a basic ingredients for antibiotic medicine.

Abstract: The aim of this study is to investigate the antibacterial effect of endophytic fungi originated from the root of mangrove Rhizophora stylosa growing on Mangrove Plantation Area around DAS Simpang 5 Jl. Piere Tendean Manado. The method of this research was using combination of PDA and NA media. Two species of endophytic fungi were extracted from the root of R. stylosa, black mycelium fungi as isolate A and brown mycelium fungi as isolate B. The activities of both isolates were tested against pathogenic bacteria Staphylococcus aureus and Escherichia coli. The result showed the same inhibition zone of isolate A and B to S. aureus, while, isolate A showed bigger the inhibition zone in comparison to isolate B against E. coli. As a conclusion, the endophytic fungi taken from the root of R. stylosa have antibacterial activity toward S. aureus and E. coli. Keywords: endophytic fungi, Rhizophora stylosa, antibacterial, Staphylococcus aureus, Escherichia coli.   Abstrak: Penelitian ini bertujuan untuk melihat adanya efek antibakteri dari jamur endofit yang diambil dari akar bakau Rhizophora stylosa yang ditanam di sekitar Perairan Daerah Aliran Sungai Simpang Lima Jl. Piere Tendean Manado. Metode penelitian dilakukan dengan menggunakan kombinasi media Potato Dextrose Agar (PDA) dan Nutrient Agar (NA). Dari akar bakau R. stylosa diperoleh dua  jenis jamur endofit yaitu isolat A dengan karateristik miselium jamur berwarna hitam dan isolat B yang dengan karakteristik miselium berwarna coklat. Kedua isolat ini selanjutnya diujikan aktivitasnya terhadap bakteri Staphylococcus aureus dan Escherichia coli. Hasil penelitian menunjukkan isolat A dan B memberikan  zona hambat yang sama terhadap bakteri Staphylococcus aureus dan pada bakteri Escherichia coli isolat A menunjukkan diameter zona hambat yang lebih besar dibandingkan isolat B. Kesimpulan, Jamur endofit yang diisolasi dari akar bakau R. Stylosa memiliki efek antibakteri terhadap pertumbuhan bakteri S. aureus dan E. coli. Background: Eucheuma spinosum seaweed contain flavonoid, triterpenoid, alkaloids and polyphenol which has been widely used in antibacterial activity. Purpose(s):The aim of the research are to determine antibacterial activities of metanol extracts of E. spinosum against Staphylococcus aureus ATCC 25923 and Escherichia coli ATCC 25922; to formulate hand wash of seaweed of methanol extract E. spinosum that has antibacterial activity and has physical and chemical stability. Methods: Seaweed E.spinosum methanol extract was derived by maceration method. Antibacterial activities of the extract were tested by liquid dilution and solid dilution method. Hand wash was formulated by mechanical dissolved methods. Antibacterial activity of hand wash were tested by liquid dilution and solid dilution method. Physical and chemical stabilities were conducted by cycling test. Results:. These were showed through minimum inhibitory concentration (MIC) of ethanol extracts of E. spinosum against S. aureus ATCC 25923 at concentrations of 6% and E. coli ATCC 25922 at concentrations of 6%. The minimum bactericidal concentration (MBC) of ethanol extracts of E. spinosum against S. aureus ATCC 25923 at concentrations of 8% and E. coli ATCC 25922 at concentrations of 8%. Formulation of hand wash seaweed E. spinosum methanol extract at concentrations of 8% and 10%. The test of stabilities results of hand wash changes were organoleptic, viscosity, pH, and foaming ability were accordance to qualified standard. The antibacterial activity of hand wash contains seaweed E. spinosum methanol extract at concentration of  8% and 10% have bactericidal activity againts S. aureus ATCC 25923 and E. coli ATCC 25922. Conclusion: Overall, these results suggested that formula of hand wash contains E. spinosum metanol extracts have antibacterial properties against S. aureus ATCC 25923 and E. coli ATCC 25922.Keywords: antibacterial, Eucheuma spinosum, hand wash, physical and chemical stability Latar Belakang: Rumput laut Eucheuma spinosum mengandung flavonoid, triterpenoid, alkaloid, dan polifenol yang memiliki aktivitas antibakteri. Tujuan: Penelitian ini bertujuan mengetahui aktivitas antibakteri ekstrak metanol E. spinosum terhadap Staphylococcus aureus ATCC 25923 and Escherichia coli ATCC 25922; membuat sediaan sabun cuci tangan dari ekstrak metanol E. spinosum yang memiliki aktivitas antibakteri dan stabil secara fisika dan kimia. Metode: Ekstrak metanol E. spinosum diperoleh dengan metode maserasi. Uji aktivitas antibakteri ekstrak dilakukan dengan metode dilusi cair dan dilusi padat. Sabun cuci tangan diformulasi dengan metode pencampuran mekanik. Uji aktivitas antibakteri sabun cuci tangan dilakukan dengan metode dilusi cair dan dilusi padat. Uji stabilitas fisika kimia dilakukan dengan metodecycling test. Hasil: Konsentrsi Hambat Minimum (KHM) ekstrak metanol E. spinosum terhadap S. aureus ATCC 25923 adalah 6% dan terhadap E. coli ATCC 25922 adalah 8%. Ekstrak metanol E. spinosum dapat diformulasi menjadi sabun cuci tangan dengan konsentrasi 8% dan 10%. Uji stabilitas menunjukkan bahwa perubahan organoleptik, viskositas, pH, dan kemampuan membentuk busa masih berada dalam nilai yang dipersyaratkan. Uji aktivitas antibakteri sabun cuci tangan yang mengandung ekstrak metanol E. spinosum dengan konsentrasi 8% dan 10% memiliki aktivitas antibakteri terhadap ATCC 25923 and E. coli ATCC 25922. Simpulan: Formula sabu cuci tangan yang mengandung ekstrak metanol E. spinosum memiliki aktivitas antibakteri terhadap ATCC 25923 and E. coli ATCC 25922.Kata kunci: antibakteri, Eucheuma spinosum, sabun cuci tangan, stabilitas fisika kimia

Introducción. Las nanopartículas poliméricas constituyen una herramienta nanotecnológica que podría ayudar a combatir los microorganismos patógenos que han desarrollado resistencia a los antibióticos convencionales.Objetivo. Sintetizar nanopartículas de ácido poliláctico cargadas con ofloxacina y vancomicina, y determinar su actividad antibacteriana frente a Escherichia coli O157:H7 y Staphylococcus aureus resistente a la meticilina (SARM).Materiales y métodos. Las nanopartículas de ácido poliláctico cargadas con ofloxacina y vancomicina se sintetizaron utilizando el método de emulsión y evaporación de solvente. Se caracterizaron mediante dispersión de luz en modo dinámico, electroforesis Doppler con láser y microscopía electrónica de barrido (S-TEM). Se evaluó la actividad antibacteriana in vitro de las nanopartículas de ácido poliláctico con ofloxacina contra E. coli O157:H7 y nanopartículas de ácido poliláctico con vancomicina contra SARM, mediante el método de microdilución en caldo.Resultados. Se obtuvieron nanopartículas poliméricas con tamaños inferiores a 379 nm y carga superficial positiva de hasta 21 mV. Las nanopartículas cargadas con ofloxacina presentaron una concentración inhibitoria mínima (CIM50) de 0,001 μg/ml frente a E. coli O157:H7, valor 40 veces menor que la concentración de antibiótico libre necesaria para lograr el mismo efecto (CIM50=0,04 μg/ml). Para SARM, las nanopartículas mejoraron la potencia farmacológica in vitro de la vancomicina alexhibir una MIC50 de 0,005 μg/ml, comparada con la de 0,5 μg/ml del antibiótico libre.Conclusiones. Se mejoró el efecto antibacteriano de la ofloxacina y la vancomicina incorporadas en la matriz polimérica de ácido poliláctico. Las nanopartículas poliméricas constituirían una alternativa para el control de cepas bacterianas de interés en salud pública.

The antibacterial activity of three Cupressaceae plants (Thujaoccidentalis,ThujaorientalisandChamaecyparisobtusa) was tested against three bacteria using the agar diffusion method. The ether and ethylacetate fraction of crude methanol extract from the three plants showed potent antibacterial activity against the tested microorganisms. The result showed that Staphylococcus aureus revealed the most sensitivity among the tested bacteria. Thujaoccidentalisether fraction and Thujaorientalis hexane fraction exhibited the highest antibacterial activity against Staphylococcus aureus. E. coli was shown the highest MIC values compared to the other two tested bacteria, which indicates the lowest antibacterial activity against the bacterium. This study promises an interesting future for designing a potentially active antibacterial agent from the three Cupressaceae plants.
