As current technology cannot cure cancer completely, prevention becomes the main choice. To prevent the development of cell transformation into cancer cells, polyphenols that are widely found in cocoa beans (Theobroma cacao) can be used. IFNγ plays an important role in immunity against cancer. This study aims to see the ability of cocoa beans ethanol extract to increase the number of IFNγ expression in Balb/c mice undergoing cell transformation. This study used three groups Balb/c (n=4), namely K1 (normal control), K2 (negative control: injected with benzopyrene without ethanol extract of cocoa beans), and K3 (treatment: injected with benzopyrene, given 4mg/30gBW/po/day ethanol extract of cocoa beans). The mice were biopsied, and IFNγ expression was examined by immune histochemical method. The results showed that IFNγ expression increased significantly in K3. It can be concluded that ethanol extract of cocoa beans could increase IFNγ expression in Balb/c mice undergoing cell transformation.

Physalia utriculus is one of the invertebrate marine biota that is often found in Indonesia. Some symptoms of venoming due to jellyfish stings cause pain, itching, and hemolysis. In Indonesia, 13 cases of jellyfish stings were reported in 2005-2009 with three people dying in Java, Bali, and Bangka. Cocoa beans (Theobroma cacao L.) contain fat, carbohydrates, proteins, and polyphenol compounds that are useful as antioxidants. Polyphenols in the form of epicathechins, catechins, and procyanidins serve to provide protection to hemolysis. The purpose of this study was to determine the potential of ethanol extract of cacao (Theobroma cacao L.) in inhibiting the damage of erythrocyte induced by Physalia utriculus in vitro. This study used 28 samples of erythrocytes divided into seven groups, namely the normal control group, negative controls, and treatment with cocoa ethanol extract 0.2%, 0.1%, 0.04%, and 0.02%. Each subsequent group induced venom Physalia utriculus. The results showed that the average speed of erythrocyte lysis in the treatment group by giving cocoa ethanol extract 0.2%, 0.1%, 0.04%, and 0.02% respectively (seconds ± standard deviation) was 858,25 ± 94,44; 1.000,5 ± 159,93; 678,5 ± 19,71; and 1.006 ± 159,50. The mean speed of erythrocyte lysis in the negative control group was 1,025 ± 164.63 and the positive control group with the administration of N-Acetylcystein can last up to one hour after administration of venoms. Test for normality and homogeneity shows that data is normally distributed and homogeneous. One Way Annova analysis shows the significance value of p <0.05, then a post hoc analysis test was performed with the Bonferoni method to find out the differences in significance in each group. In this study it can be concluded that the administration of cocoa ethanol extract has no potential to inhibit erythrocyte damage that has been venomed by Physalia utriculus in vitro. This study was aims to exploit the potential waste of cocoa pod (Theobroma cacao L.) as a natural acaricide in ticks (Boophilus microplus). The study was conducted with a two-variable completely randomized design (CRD) with two repetitions. The sample does not differentiate between sex, body weight, and length of about 0.8 - 1 cm. Each 5 ticks were treated as follows: treat 1 extract of 10% cocoa pod husk; treatment 2 20% cocoa pod skin extract; treatment 3 30% cocoa pod husk extract; treatment 4 negative control; treat 5 positive controls. In the first variable the tick was treated with methanol extract, while in the second variable the ethanol extract was treated with 10%, 20%, 30% respectively, negative control and positive control. In treatment I used 25 ticks plus treatment II 25 ticks. Each treatment was repeated to avoid data bias so that a total of 100 ticks were used. The results showed that the cocoa pod husk extract had the best killing power at a concentration of 30% with methanol solvent at 6.21 hours while ethanol solvent at 5.91 hours. The use of extraction materials has not been able to compensate for the use of synthetic materials that can kill in minutes.

Red Gedi (Abelmoschus moschatus Medik) is a plant found only in tropical climate areas such as Africa and Asia. Some people’s experience in Manado stated that red gedi plants can be used to treat various types of disease, one of them was kidney stones. Therefore, this study aimed to determine the activity and the effective dose of red gedi leaf extract as an inhibitor of kidney stone formation. The extract was prepared by maceration using ethanol 96%. Inhibitory activity test of kidney stone formation was done by firstly dividing the labrats into 6 groups. Each group consisted of 3 male rats. Both the first group (Normal control) and the second group (Negative control) weregiven Na.CMC 0.5%; the third group (Positive control) was given Batugin Elixir at a dose of 2.7 ml/kgBW; while the fourth, fifth, and sixth were respectively given red gedileaf extract at doses of 50, 100, and 150 mg/kgBW. After 2 hours, all treatment groups were induced with0.75% of ethylene glycol and 2% of ammonium chloride except for the normal control. Ratio of kidney weight and the level of calcium were then measured and calculated. Data obtained were statistically analyzed using ANOVA (Analysis Of variance) and continued with LSD (Least Significantly Difference). It can be concluded that ethanol extract of red gedileaves had inhibitory activity of kidney stones formation in all dose variation and the most effective one was at a dose of 150 mg/kgBW

There was estimated 150 million envenomation cases due to jellyfish stings occur globally every year. 100 from 10,000 jellyfish species in the world known to be dangerous, one them is Physalia utriculus. The aim of this study was to determine the potential of cocoa (Theobroma cacao L.) ethanol extract toward nematocyst tubul firing inhibition of jellyfish (Physalia utriculus) toxin in vitro. The method was true experimental design with post test only control group design. The study divided into 8 groups: 1 normal control, 1 positive control, 1 negative control, and 5 treatments grups by giving cocoa ethanol extract with concentration 20%, 2%, 0.2%, 0.02%, 0.002%. The observation was made by calculating the percentage number of firing nematocysts. The result showed average percentage of firing nematocyst in the normal control group was 42.50 ± 3.18, positive control group was 37.97 ± 5.57, negative control group was 52.44 ± 2.98, and treatment group which given with cocoa ethanol extract 20%, 2%, 0.2%, 0.02%, 0.002% were 48.24 ± 5.37; 40.62 ± 7.10; 29.45 ± 5.39; 37.60 ± 9.78; 41.11 ± 3.92, respectively. The One Way Annova statistical results test show significance value 0.001 (p≤0.05). The conclusion of this study was the cocoa ethanol extract with concentration 0.2% has most potential to inhibit the jellyfish (P. utriculus) nematocyst tubule firing in vitro. Purpose: To investigate the neuroprotective and memory-boosting properties of ethanol extract of Vateria indica bark on scopolamine-mediated defects in learning and memory in young mice. Methods: The ethanol extract of V. indica bark was prepared via Soxhlet extraction and subjected to qualitative and quantitative phytochemical assessment. The acute toxicity of the extract was also evaluated in mice. Six groups of 3-month-old Swiss albino mice (6 per group) were used: normal control, negative control, piracetam group, 250 mg/kg V. indica extract alone group, 500 mg/kg V. indica extract alone group, 250 mg/kg V. indica extract + scopolamine group and 500 mg/kg V. indica extract + scopolamine group. The mice were pretreated with piracetam (standard nootropic drug) or varied doses of the extract for 14 days prior to induction of amnesia. With the exception of normal control group, amnesia induction using scopolamine (3 mg/kg) i.p. on day 14 at 1½ h after the last extract dose. Mice in normal and negative control groups received 0.5 % tragacanth orally at a dose of 10 mL/kg. Cognitive deficit was assessed using elevated plus maze (EPM), step-down avoidance, and Morris water maze (MWM) tests. Results: Qualitative phytochemical screening of V. indica bark extract showed flavonoids, phenolics, glycosides, tannins, carbohydrate, saponins and steroids. The total phenol and total flavonoid contents were 580.96 ± 0.95 mg GAE/g extract and 66.89 ± 0.56 mg RE/g extract, respectively. The mice tolerated the extract up to 5000 mg/kg bwt. They all survived during and after the acute toxicity study and no significant changes in appearance or general behavior were noticed. The extract significantly enhanced learning and memory, and improved spatial recognition in scopolamine-induced amnesic mice (p < 0.05). Acetylcholinesterase (AChE) activity and levels of dopamine and noradrenaline were markedly higher in negative control mice than in normal control, but were significantly reduced after pretreatment with ethanol extract of V. indica bark (p < 0.05). The results of histopathological examination provided evidence in support of the protective effect of the extract on hippocampal and cortical neurons. Conclusion: Pretreatment with ethanol extract of V. indica bark confers neuroprotection and enhances memory in young amnesic mice. Therefore, the extract of the plant can potentially be develpoed for the management of degenerative brain conditions. Kasus penyakit hati semakin meningkat seiring penggunaan senyawa hepatotoksin salah satunya karena penggunaan parasetamol dengan dosis berlebih. Hal tersebut dapat meningkatkan produksi radikal bebas sehingga memicu terjadinya stress oksidatif yang dapat menimbulkan kerusakan jaringan yang ditandai dengan peningkatan kadar Malondialdehyde (MDA). Stress oksidatif dapat diatasi dengan antioksidan dari berbagai tanaman. Kulit kayu manis memiliki aktivitas antioksidan dengan nilai IC50 53ppm dan daun pandan wangi 39,7%  Penelitian ini bertujuan untuk mengetahui aktivitas kombinasi ekstrak etanol daun pandan wangi dan kayu manis dalam menurunkan kadar MDA. tikus yang diinduksi parasetamol. Penelitian menggunakan metode eksperimental, dilakukan selama 9 hari dengan 30 ekor tikus jantan dibagi menjadi 6 Kelompok, yaitu: Normal diberi aquadest, Kontrol Positif diberi silimarin 100 mg/kgBB, Kontrol Negatif diberi CMC-Na 0,05%, serta 3 kelompok lainnya diberi kombinasi ekstrak daun pandan wangi:kayu manis berturut-turut dosis I (25:75), dosis II (50:50), dosis III (75:25). Semua kelompok diinduksi parasetamol 2,5 g/kgBB pada hari ke-7  setelah 30 menit perlakuan, kecuali kelompok normal. Pada hari ke 9 dilakukan pengukuran kadar MDA dengan metode TBARs menggunakan spektrofotometri. Pemberian kombinasi ekstrak etanol daun pandan wangi dan kayu manis dapat menurunkan kadar MDA dengan kombinasi dosis yang paling optimal adalah 75:25 berdasarkan statistik dengan nilai signifikan 0,000<0,05 dibandingkan dengan kelompok negatif.    Cases of liver disease have increased with the use of hepatotoxin compounds, one of which is due to the use of paracetamol with excessive doses. This can increase the production of free radicals so that it triggers oxidative stress which can cause tissue damage which is characterized by increased levels of Malondialdehyde (MDA). Oxidative stress can be overcome with antioxidants from various plants. Cinnamomum burmanii has antioxidant activity with IC50 value of 53ppm and Pandanus amarrylifolius 39.7%. This study aims to determine the combined activity of ethanol extract of Pandanus amarrylifolius and Cinnamomum burmanii  in reducing MDA levels. Paracetamol-induced rats. Research using experimental methods, conducted for 9 days with 30 male rats divided into 6 groups, namely: Normal given aquadest, Positive Control were given silimarin 100 mg / kgBB, Negative Control was given CMC-Na 0.05%, and 3 other groups were given a combination of Pandanus amarrylifolius extract: Cinnamomum burmanii dose I (25:75), dose II (50:50), dose III (75:25). All groups induced paracetamol 2.5 g / kgBB on the 7th day after 30 minutes of treatment, except the normal group. On the 9th day MDA levels were measured using the TBARs method using spectrophotometry. Giving a combination of Pandanus amarrylifolius and Cinnamomum burmanii ethanol extract can reduce MDA levels with the most optimal dose combination is 75:25 based on statistics with a significant value of 0,000<0.05 compared with the negative group.

Background: Natural product, osthol has been found to have important biological and pharmacological roles particularly having inhibitory effect on multiple types of cancer. Objective: The unmet needs in cancer therapeutics make its derivatization an important and exciting field of research. Keeping this in view, a whole new series of diverse analogues of osthol (1) were synthesized. Method: All the newly synthesized compounds were made through modification in the lactone ring as well as in the side chain of the osthol molecule and were subjected to anti-proliferative screening through 3-(4,5-Dimethylthiazol-yl)-diphenyl tetrazoliumbromide (MTT) against four different human cancers of diverse origins viz. Colon (Colo-205), lung (A549), Leukemia (THP- 1) and breast (MCF-7) including SV40 transformed normal breast epithelial cell (fR-2). Results: Interestingly, among the tested molecules, most of the analogs displayed better antiproliferative activity than the parent Osthol 1. However, among all the tested analogs, compound 28 exhibited the best results against leukemia (THP1) cell line with IC50 of 5µM.Compound 28 induced potent apoptotic effects and G1 phase arrest in leukemia cancer cells (THP1). The population of apoptotic cells increased from 13.8% in negative control to 26.9% at 8μM concentration of 28. Compound 28 also induced a remarkable decrease in mitochondrial membrane potential (ΛΨm) leading to apoptosis of the cancer cells. Conclusion: A novel series of molecules derived from natural product osthol were synthesized, wherein compound 28 was found to be most effective against leukemia and with 10 fold less toxicity against normal cells. The compound induced cancer inhibition mainly through apoptosis and thus has a potential in cancer therapeutics.

<p>Penelitian ini bertujuan untuk melihat pengaruh pemberian ekstrak etanol kulit petai (Parkia speciosa Hassk) terhadap penurunan kadar glukosa darah mencit jantan yang diinduksi aloksan. Hewan percobaan dibagi atas 5 kelompok diantaranya kelompok kontrol negatif, kelompok kontrol positif,dosis I (280 mg/kgBB mencit), dosis II (560 mg/kg BB mencit), dosis III (840 mg/kg BB mencit). Penelitian dilakukan selama 21 hari. Persentase penurunan kadar glukosa darah mencit jantan setelah diberikan ekstrak etanol kulit petai pada hari ke-21 adalah dosis I (77,52 %) lebih besar dibandingkan dengan dosis II (69,5 %) dan dosis III (73,37 %). Data yang diperoleh dianalisis dengan uji Two Way Anova dengan program SPSS 17. Hasil penelitian ini menunjukkan bahwa pemberian ekstrak etanol kulit petai untuk tiga variasi dosis menyatakan perbedaan yang bermakna secara statistik terhadap penurunan kadar glukosa darah mencit jantan.</p><p><em>Petai (Parkia speciosa Hassk) has a compound β-sitosterol and stigmasterol that have efficacy to decreased blood glucose levels. This study aimed to determine the effect of ethanol extract of petai peel for decrease blood glucose levels of male mice induced by alloxan. Experimental animals were divided into 5 groups including negative control group, positive control group, the first dose (280 mg/kg in mice), the second dose (560 mg/kg in mice), the third dose (840 mg/kg in mice). The study was conducted for 21 days. After 21 days, the result found that the percentage of blood glucose levels after the male mice given the ethanol extract of petai peel was, the first dose (77.52%) biger than the second dose (69.5%) and the third dose (73.37%). The data obtained were analyzed by Two Way ANOVA using SPSS 17. The results showed that have signicantly difference between three dose variation of ethanol extract of petai peel in blood glucose levels.</em></p>

Abstract Background Cancer is one of the most considerable concerns because of increasing the death rate all over the world. Recent studies have disclosed that plant extracts exhibit anticancer activity through various mechanisms. Xanthium strumarium has been used by Vietnamese in herbal medicines to support the medication of infirmities. This study is to consider the secondary metabolites, antioxidant and anticancer capacities of extract from the aerial parts (stems and leaves) of X. strumarium (AP-XS). Methods AP-XS was analyzed for the presence of phytochemicals via qualitative chemical tests and determined total polyphenol and flavonoid contents. DPPH (1,1-diphenyl-2-picrylhydrazyl) quenching assay and sulforhodamine B (SRB) assay were selected to investigate antioxidant capacity and anti-proliferative activity, respectively. Besides, acridine orange-ethidium bromide (AO-EB) dual staining was applied to evaluate the ability to induce apoptosis on HepG2 cancer cells. Results Results of present study indicated that AP-XS contains the main phytochemicals such as flavonoids, tannins, saponins, alkaloids, and triterpenes. Ethanol extract had highest content of polyphenol (84.86 mg gallic acid equivalent/g dry mass), and exhibited the great total antioxidant property (IC50 = 184.13 μg/mL) and anti-proliferative activity on HepG2 cancer cells (IC50 = 81.69 μg/mL). Furthermore, the characteristics of apoptosis including shrinkage of the cell and apoptotic bodies were found following 60 h of AP-XS extract treatment through AO-EB dual staining. Conclusion The data suggest that AP-XS extract had antioxidant potential and anti-proliferative effect. The anti-proliferative property was considered to have an association with a rising of apoptosis. These results were reliable for further research on X. strumarium as a source of phytochemicals with anticancer activity potential for cancer therapeutics.

Background and Objective: We have previously reported that BRCA2 N372 H i.a.1342A>C heterozygous variation presented in platinum-resistant patients. This study aimed to further investigate the mechanism of BRCA2 N372 H mutation in the development of platinum resistance in ovarian cancer. Methods: The BRCA2 N372 H i.a.1342A>C was synthesized and used to exchange 1 wildtype allele followed by sequencing to confirm the mutant allele sequence. Plasmids were constructed and transfected into the OVCAR-3 cells after lentiviral packaging. BRCA2 N372 H mRNA was detected by qPCR. BRCA2 protein was assessed by immunoblotting. Binding of the BRCA2 to Rad51 was detected by immunofluorescence staining. Sensitivity of the cells to cisplatin treatment was assessed with CCK-8 assay. Results: It was found that expression of BRCA2 protein in ovarian cancer cells transfected with BRCA2 N372 H i.a.1342A>C gene (2.177 ± 0.003) was significantly increased compared to that of the cells transfected with lenti-EGFP only (1.227 ± 0.003, P < 0.001). Binding of the BRCA2 and Rad51 proteins was significantly increased in the cells with BRCA2 N372 H i.a.1342A>C mutation (3.542 ± 0.24) than that in the cells transfected with lenti-EGFP (1.29 ± 0.32) or empty cells (1.363 ± 0.32, P < 0.001). Cell viability significantly increased in the cells transfected with BRCA2 N372 H mutant gene. The IC50 value was significantly higher in the cells transfected with BRCA2 N372 H mutant gene (1.963 ± 0.04) than that of the cells transfected with lenti-EGFP (0.955 ± 0.03, P < 0.01) or empty cells (1.043 ± 0.007, P < 0.01). Conclusion: Over expression of mRNA and protein of BRCA2 was detected in the cells with BRCA2 N372 H i.a.1342A>C mutation but not in the lentivirus negative control (lenti-EGFP) or the cells without transfection (empty cells), which may lead to resistance to platinum-based drugs in ovarian cancer cells through homologous recombination repair pathway.
