Polyaniline (PANI) slim film was set up by electrochemical polymerization strategy at room temperature in a standard three-electrode cell from (0.1M) aniline monomer and (0,5M) from Sulfuric acid in the presence of distilled water. The development of PANI film was portrayed by Voltammetric studies, SEM, XRD, and FTIR. Voltammetric studies were performed in 0.5 M acidic aqueous solutions using H2SO4. The XRD design demonstrated that the diffraction top at 2θ = (30˚). The FTIR spectroscopy spectra give particular and unmistakable bonds at 3500, 1572.52, 1302.53, 831.98, and 592.85 cm-1.

Electrochemical polymerization of aniline was carried on platinum (Pt) wire electrode in sulfuric acid (H2SO4) solution by cyclic voltammetry using a conventional three-electrode cell at room temperature (20°C). The effects on the electrodeposition of the monomer concentration, anodic potential and potential scan rate are discussed. The conductive layer of emeraldine base polyaniline (Pani) was prepared in this work by repeating potential cycling between -0.24 and 0.9 V/SCE at 50 mV/s in 1 M H2SO4 solution containing 0.1 M aniline monomer.

Polyaniline (PANI) nanofibers with diameters ranging from 60 nm to 190 nm were successfully synthesized by rapid mixing polymerization method at room temperature. The effects of synthesis conditions such as concentration of hydrochloric acid (HCl), ammonium persulfate (APS), and aniline monomer (Ani) on the morphology of PANI nanofibers were investigated using scanning electron microscopy (SEM). FTIR and UV-Vis analysis were performed to characterize the PANI nanofibers. The effect of polymerization time and electrical properties of PANI nanofibers was also discussed. A rapid mixing polymerization method is found to be advantageous in producing bulk quantities of nanofibers.

Use of the electron microscope to examine wet objects is possible due to the small mass thickness of the equilibrium pressure of water vapor at room temperature. Previous attempts to examine hydrated biological objects and water itself used a chamber consisting of two small apertures sealed by two thin films. Extensive work in our laboratory showed that such films have an 80% failure rate when wet. Using the principle of differential pumping of the microscope column, we can use open apertures in place of thin film windows.Fig. 1 shows the modified Siemens la specimen chamber with the connections to the water supply and the auxiliary pumping station. A mechanical pump is connected to the vapor supply via a 100μ aperture to maintain steady-state conditions.

The intestinal roundworm of swine is pinkish in color and about the diameter of a lead pencil. Adult worms, taken from parasitized swine, frequently were observed with macroscopic lesions on their cuticule. Those possessing such lesions were rinsed in distilled water, and cylindrical segments of the affected areas were removed. Some of the segments were fixed in buffered formalin before freeze-drying; others were freeze-dried immediately. Initially, specimens were quenched in liquid freon followed by immersion in liquid nitrogen. They were then placed in ampuoles in a freezer at −45C and sublimated by vacuum until dry. After the specimens appeared dry, the freezer was allowed to come to room temperature slowly while the vacuum was maintained. The dried specimens were attached to metal pegs with conductive silver paint and placed in a vacuum evaporator on a rotating tilting stage. They were then coated by evaporating an alloy of 20% palladium and 80% gold to a thickness of approximately 300 A°. The specimens were examined by secondary electron emmission in a scanning electron microscope.

In cotton seeds the radicle has 12% moisture content which makes it possible to prepare freeze-fracture replicas without fixation or cryoprotection. For this study we have examined replicas of unfixed radicle tissue fractured at room temperature to obtain data on organelle and membrane structure.Excised radicles from seeds of cotton (Gossyplum hirsutum L. M-8) were fractured at room temperature along the longitudinal axis. The fracture was initiated by spliting the basal end of the excised radicle with a razor. This procedure produced a fracture through the tissue along an unknown fracture plane. The warm fractured radicle halves were placed on a thin film of 100% glycerol on a flat brass cap with fracture surface up. The cap was rapidly plunged into liquid nitrogen and transferred to a freeze- etch unit. The sample was etched for 3 min at -95°C to remove any condensed water vapor and then cooled to -150°C for platinum/carbon evaporation.
