The present study determined the antifungal activity of the root of extracts Argemone mexicana against 7 fungi using dichloromethane and methanol as solvent. Among these pathogens, 3 were dermatophytes and the rest 4 were nondermatophytes. The dichloromethane extract of Argemone mexicana exhibited moderate to good activity at concentration of 200 ?g/disc but at concentration of 50 ?g/disc, the same extract showed no activity against the tested fungi. The methanol extract were also inactive against the tested pathogens. In brine shrimp toxicity test, it was observed that LC50 value of the dichloromethane extract of Argemone mexicana was 22.35 ?g/ml. From these findings it is indicative that dichloromethane extract of Argemone mexicana is biologically active and may have antifungal principles that could be useful in fungal diseases.KYAMC Journal Vol. 4, No.-2, Jan 2014, Page 402-405

This study focuses on antibacterial, antioxidant and toxic potentials of Viscum album Linn, commonly known as European mistletoe associated with Acacia catechu (Khayer in Nepali). Methanol extract of the aerial parts of the Mistletoe was prepared by cold percolation method. The resulting extract was simultaneously subjected to phytochemical screening; anti-microbial activity; anti-oxidant potential and Brine shrimp toxicity test. The major biologically active phyto-constituents observed were alkaloids, glycosides, saponins, polyphenols, flavonoids, tannins, terpenoids and cardiac glycosides. Upon antibacterial activity screening, the plant extract was found to be highly effective against Pseudomonas aeruginosa with the zone of inhibition 16±1mm compared to 17±1mm of chloramphenicol (50 mcg). The antioxidant activity as EC50 value by DPPH (1,1-diphenyl-2-picrylhydrazyl) free radical scavenging activity was found to be 1.58 mg/ml while the ferric reducing capacity was measured to be 282.83±19.55 mg FeSO4.7H2O eqvt/g dry wt. of the extract during Ferric Ion Reducing Antioxidant Power (FRAP) Assay. The LC50 value for Brine Shrimp Toxicity Assay was found to be 31.62 ppm. This study shows the medicinal value of the mistletoe associated with Acacia catechu. Abstract : This study aims to determine the antidiabetic activity and toxicity of the acetone extract of Tristaniopsis merguensis Griff leaf. The antidiabetic test was the α-glucosidase inhibition method, while the toxicity test used the Brine Shrimp Lethality Test (BSLT) method. The acetone extract possessed antidiabetic activity with an IC50 value of 8.83 ± 0.31 (μg/mL). This value is not much different from the positive control of quercetin which has an IC50 value of 6.04 ± 0.14 (μg/mL). The characteristics of the FT-IR spectrum of acetone extract showed that Tristaniopsis merguensis leaf has the groups Ar-OH (phenolic), -OH (hydroxyl), C=O (ketone) and C=C (aromatic). Based on the toxicity test, the Tristaniopsis merguensis leaf acetone extract has an LC50 value of 959.25 ppm which means that the acetone extract is toxic. Therefore, the acetone extract of Tristaniopsis merguensis might be the potential agent of antidiabetic.Abstrak : Penelitian ini bertujuan untuk mengetahui aktivitas antidiabetes dan toksisitas dari ekstrak aseton daun Tristaniopsis merguensis Griff. Uji aktivitas antidiabetes ditentukan berdasarkan metode inhibisi enzim α-glucosidase, sedangkan toksisitas ditentukan berdasarkan metode Brine Shrimp Lethality Test (BSLT). Ekstrak aseton memiliki aktivitas antidiabetes dengan nilai IC50 8,83 ± 0,31 (μg/mL). Nilai ini tidak jauh berbeda dengan kontrol positif quersetin yang memiliki nilai IC50 6,04 ± 0,14 (μg/mL). Karakteristik spektrum FT-IR ekstrak aseton menunjukkan bahwa daun Tristaniopsis merguensis memiliki gugus fungsi Ar-OH (fenolik), -OH (hidroksil), C=O (keton) dan C=C (aromatik). Berdasarkan uji toksisitas, ekstrak aseton daun Tristaniopsis merguensis memiliki nilai LC50 sebesar 959,25 ppm yang berarti bahwa ekstrak aseton bersifat toksik. Oleh karena itu, ekstrak aseton dari Tristaniopsis merguensis berpotensi untuk dijadikan agen antidiabetes.

The bioactive compound of hyptolide has been isolated from an Indonesian plant Hyptis pectinata using various solvents. The pure compound was then used as a standard in quantitative analysis using HPLC and UV-Vis spectroscopy in ethanol (EEth), ethyl acetate (EEa), acetone (Eac) and hexane (Ehex) extract each earned 3.14%, 0.01%, 1.33% and 0.04% (dry weight per sample). The standard curve of hyptolide using UV spectroscopy has been obtained with the coefficient of relationship (r) of 0.997. However, the use of spectroscopy was not recommended for the standardization of hyptolide in the extract due to interference from other compounds that absorb the same wavelength. Furthermore, the toxicity test using the Brine Shrimp lethality test shows LC50 value of 92-181 ppm, which was in EEa compared to EEth, Eac, and Ehex.

The methanol extract of the powdered leaf of Eurya acuminata was investigated for isolation of secondary metabolites and two compounds were obtained by using VLC, column chromatography and TLC. The compounds were identified as phytol (1) and ?-sitosterol by extensive spectroscopic studies, including high field NMR analyses as well as co-TLC with authentic samples. The methanol extract of leaf of E. acuminata and its organic and aqueous soluble partitioning materials were evaluated for cytotoxic, thrombolytic and antimicrobial properties. In the cytotoxicity study the aqueous fraction of crude methanolic extract showed significant lethality towards brine shrimp having LC50 value 8.821 ?g/ml as compared to standard vincristine sulfate (0.404 ?g/ml). In the study for thrombolytic property, different extract of E. acuminata exhibited various thrombolytic activity ranging from 13.66 to 31.89 % as compared to standard streptokinase (46.51 %). Prasman leaves (Eupatororium triplinerve V) was well documented to have anti cancer benefit in Indonesian traditional medicine history. However, there were no scientific studies including toxicological assessment on the plan extract. The purpose of this study was to investigate the toxicological effect of some fractions of Prasman leaves methanol extract. Phytochemical screening by the Farnsworth method on powder and some fraction of the methanol extract were conducted followed by toxicity test using the “Brine Shrimp Lethality” test (BSLT) method. In the current study results, the phytochemical screening showed the presence of flavonoid, saponin, coumarin, tannin, steroid and volatile oil. LC50 of the n-hexane fraction 238.66 µg/mL, ethyl acetate fraction 24.42 µg/mL,  and n-butanol 64.10 µg/mL.Keywords : BSLT, Toxicity test, Eupatororium triplinerve

AbstrakEfek toksisitas pinang yaki (Areca vestaria) sebagai tumbuhan obat potensial perlu diuji untuk mengetahui ambang batas penggunaannya. Penelitian ini bertujuan untuk menguji tingkat toksisitas ekstrak batang pinang yaki terhadap larva Artemia salina Leach. dengan metode Brine Shrimp Lethality Test. Ekstrak diencerkan dengan konsentrasi 200, 400, 600 dan 800 ppm masing-masing untuk supernatan rendaman pertama (larutan A),supernatan rendaman ke dua (larutan B) dan supernatan rendaman ke tiga (larutan C) . Hasil penelitian menunjukkan bahwa ekstrak batang pinang yaki mempunyai efek toksisitas dengan nilai LC50 sebesar 398,28 ppm untuk larutan A, 390,84 ppm untuk larutan B dan 438,53 ppm untuk larutan C.Kata kunci: batang Areca vestaria,LC50, uji toksisitasAbstractToxicity effect of pinang yaki (Areca vestiaria) as potential medicinal plant should be evaluated to know its application threshold. Extract was diluted to concentration of 200, 400, 600 and 800 ppm each for solution A (first soaking supernatant), solution B (second soaking supernatant)and solution C (third soaking supernatant). Different extracts of the leaves and barks of Zizyphus rugosa and Zizyphus oenoplia were studied for their antibacterial, antifungal, and b-glucuronidase inhibitory activities. The methanol extract of Z. rugosa bark showed significant antibacterial activity against Streptococcus pyogens, Staphylococcus aureus and Pseudomonas aerogenes whereas the methanol extract of leaves demonstrated moderate activity against Salmonella typhi. The chloroform and methanol extracts of Z. oenoplia showed good activity against a few bacteria strains. The chloroform extracts of the barks and leaves of Z. rugosaalso showed antifungal activity. The methanol and ethyl acetate extracts of the bark of Z. rugosa revealed significant b-glucuronidase inhibitory activity. Lupeol, betuline, betulinaldehyde and betulinic acid, isolated from Z. rugosa, also showed good activity against a few bacteria. Since 1970s, aplysiatoxins (ATXs), a class of biologically active dermatoxins, were identified from the marine mollusk Stylocheilus longicauda, whilst further research indicated that ATXs were originally metabolized by cyanobacteria. So far, there have been 45 aplysiatoxin derivatives discovered from marine cyanobacteria with various geographies. Recently, we isolated two neo-debromoaplysiatoxins, neo-debromoaplysiatoxin G (1) and neo-debromoaplysiatoxin H (2) from the cyanobacterium Lyngbya sp. collected from the South China Sea. The freeze-dried cyanobacterium was extracted with liquid–liquid extraction of organic solvents, and then was subjected to multiple chromatographies to yield neo-debromoaplysiatoxin G (1) (3.6 mg) and neo-debromoaplysiatoxin H (2) (4.3 mg). They were elucidated with spectroscopic methods. Moreover, the brine shrimp toxicity of the aplysiatoxin derivatives representing differential structural classifications indicated that the debromoaplysiatoxin was the most toxic compound (half inhibitory concentration (IC50) value = 0.34 ± 0.036 µM). While neo-aplysiatoxins (neo-ATXs) did not exhibit apparent brine shrimp toxicity, but showed potent blocking action against potassium channel Kv1.5, likewise, compounds 1 and 2 with IC50 values of 1.79 ± 0.22 µM and 1.46 ± 0.14 µM, respectively. Therefore, much of the current knowledge suggests the ATXs with different structure modifications may modulate multiple cellular signaling processes in animal systems leading to the harmful effects on public health.

ABSTRAK Penentuan nilai LC50 dari ekstrak metanol daun liwas menggunakan metode Brine Shrimp Lethality Test (BSLT) terhadap hewan indikator udang Artemia salina menunjukkan nilai LC50 dari  A. salina bersifat sangat toksik.Nilai LC50 yang didapatkan sebesar 15,696 mg/L. Nilai LC50 ditentukan dengan menggunakan metode SPSS 20.0.Ekstrak metanol daun liwas mengandung kandungan senyawa metabolit sekunder seperti flavonoid, tanin dan saponin sebagai hasil uji fitokimia. Kata kunci: BSLT, Daun Liwas, Metabolit Sekunder,LC50.   PHYTOCHEMICAL ANALYSIS AND DETERMINING LC50 VALUE OF LIWAS LEAVE METHANOL EXTRACT ABSTRACT Determination of LC50 of methanol extract of leaves liwas using Brine Shrimp Lethality Test (BSLT) against animal indicator shrimp Artemia salina showed LC50 values of  A. salina is highly toxic. LC50 values were obtained at 15.696 mg / L. LC50 values determined using SPSS 20.0. Liwas leaf methanol extract contains compounds secondary metabolites such as flavonoids, tannins and saponins as phytochemical test results.
