Despite the significance of n-3 long-chain polyunsaturated fatty acid (n-3 LC-PUFA) in fish oil in promoting the maturation of female broodstocks, the detailed mechanism of the effect on the expression of hepatic reproduction-related genes is still unclear. In this study, transcriptome sequencing was used to analyze the effect of the higher dietary n-3 LC-PUFA level on gene expression in the liver of adult females spotted scat. Two-year-old female spotted scat (average weight, 242.83 ± 50.90 g) were fed with diets containing 8% fish oil (FO) or 8% soybean oil (SO) for 40 days. The fatty acid profile in the serum, liver, and ovary was analyzed, and high proportions of n-3 LC-PUFA were observed in the FO group. The final average fish body weight and gonadosomatic index were similar between the FO and SO groups. The serum vitellogenin (Vtg) and hepatosomatic index (HSI) of the FO group were significantly higher and lower than that of the SO group, respectively. Comparatively, the liver transcriptome analysis showed 497 upregulated and 267 downregulated genes in the FO group. Among them, the expression levels of three estrogen-regulated genes (i.e., Vtga, Vtgb, and Zp4) were significantly higher in the FO than in the SO group. This expression pattern could be related to the upregulation of Hsd17b7 (the key gene for the synthesis of liver steroid hormone) and the downregulation of the Hsp90 (the estrogen receptor chaperone). The expression levels of Foxo1a and Lep, which are involved in the lipid metabolism, decreased significantly in the FO group, which may be related to the lower level of HSI in the FO group. The genes related to liver LC-PUFA absorption and transport, Fabp2 and Mfsd2ab, were significantly upregulated in the FO group, indicating that fish actively adapt to high-fish-oil diets. In brief, high-fish-oil diets can influence the expression of genes related to liver n-3 LC-PUFA metabolism and reproduction, inhibit the accumulation of liver fat, and promote the liver health and gonad development. This study will contribute to clarifying the mechanism of dietary n-3 LC-PUFA on promoting reproductive development in teleost fish.

The principle aim of this study is to elucidate the relationship between the fatty acid desaturase 2 gene (fads2) expression pattern in peripheral blood cells (PBCs) and liver of gilthead seabream (GSB), Sparus aurata broodstock in order to determine the possible use of fads2 expression as a potential biomarker for the selection of broodstock. This selection could be utilized for breeding programs aiming to improve reproduction, health, and nutritional status. Passive Integrated Transponder (PIT)-tagged GSB broodstock (Male-1.22 ± 0.20 kg; 44.8 ± 2 cm and female-2.36 ± 0.64 kg; 55.1 cm) were fed a diet containing low levels of fish meal and fish oil (EPA 2.5; DHA 1.7 and n-3 LC-PUFA 4.6% TFA) for one month. After the feeding period, fads2 expression in PBCs and liver of both male and female broodstock were highly significantly correlated (r = 0.89; p < 0.001). Additionally, in male broodstock, liver fads2 expression was significantly correlated (p < 0.05) to liver contents in 16:0 (r = 0.95; p = 0.04) and total saturates (r = 0.97; p = 0.03) as well as to 20:3n–6/20:2n–6 (r = 0.98; p = 0.02) a Fads2 product/precursor ratio. Overall, we found a positive and significant correlation between fads2 expression levels in the PBCs and liver of GSB broodstock. PBCs fads2 expression levels indicate a strong potential for utilization as a non-invasive method to select animals having increased fatty acid bioconversion capability, better able to deal with diets free of fish meal and fish oil.

Previous studies have shown that it is possible to increase the ability of marine fish to produce long-chain polyunsaturated fatty acid from their 18C precursors by nutritional programming or using broodstock with a higher fatty acyl desaturase 2 (fads2) expression. However, those studies failed to show the effect of these interventions on the expression of the fads2 gene in the developing egg. Moreover, there were no studies on the temporal expression of the fads2 during ontogeny in the gilthead sea bream (Sparus aurata). In order to determine the changes in expression of fads2 during ontogeny, gilthead sea bream broodstock with a high (HRO) or low (LRO) fads2 expression fed a diet previously used for nutritional programming, or a fish oil-based diet (LFO) were allowed to spawn. The samples were taken at the stages of spawning, morula, high blastula, gastrula, neurula, heart beating, hatch and 3 day-old first exogenous feeding larvae to determine fads2 expression throughout embryonic development. The results showed the presence of fads2 mRNA in the just spawned egg, denoting the maternal mRNA transfer to the developing oocyte. Later, fads2 expression increased after the neurula, from heart beating until 3-day-old larvae, denoting the transition from maternal to embryonic gene expression. In addition, the eggs obtained from broodstock with high fads2 expression showed a high docosahexaenoic acid content, which correlated with the downregulation of the fads2 expression found in the developing embryo and larvae. Finally, feeding with the nutritional programming diet with the partial replacement of fish oil by rapeseed oil did not affect the long chain polyunsaturated fatty acid (LC-PUFA) contents nor fads2 expression in the gilthead sea bream developing eggs.

The aim of this study was to investigate the alteration in growth performance and lipid metabolism during the development of yellow fat in pigs. A total of 30 pigs (9.23 ± 0.21 kg) were assigned to three treatments: (1) low fish oil (LFO), basal diet + 2% fresh fish oil; (2) high fish oil (HFO), basal diet + 8% fresh fish oil; and (3) oxidized fish oil (OFO), basal diet + 8% OFO (peroxide value = 250 meqO2 kg−1). Pigs fed HFO and OFO diets showed yellow staining of fat and decreased growth performance, including average daily gain, average daily feed intake, and final body weight (P < 0.01). The oxidized lipid markers malondialdehyde, yellowness b* of backfat, perirenal fat, and abdominal fat were markedly increased in the pigs fed with HFO and OFO (P < 0.05). Furthermore, following HFO feeding, pigs showed significant decreases in n-6 polyunsaturated fatty acid, n-6/n-3 polyunsaturated fatty acid ratio and mRNA expression levels of CCAAT-/enhancer-binding protein alpha, fatty acid synthase, lipoprotein lipase, and hormone-sensitive lipase in backfat (P < 0.01). Overall, pigs with yellow-fat trait showed decreased growth performance and altered lipid metabolism by the high fish oil feeding.

Abstract The length-weight relationship (LWR) plays an important role in fishery management as it can be used to estimate the average weight of a specific length group. The results of the current study revealed that the mean values of regression coefficient b for both sexes of Schizothorax labiatus (McClelland) in the Jhelum River indicated positive allometric growth, while in the Sindh River, the b value for males indicated positive allometric growth and for females negative allometric growth. The value of coefficient of determination r2 for both sexes of S. labiatus was equal to or greater than 0.90 in both water bodies, except for females in the Sindh River. Higher significant (P < 0.05) average values of Fulton’s condition factor (K) and the hepatosomatic index (HSI) for both sexes of S. labiatus were noted in the Jhelum River than in the Sindh River. In contrast, higher significant (P < 0.05) values of the gonadosomatic index (GSI) were recorded in the Sindh River. The data generated during the current study provide basic information for researchers and fish biologists for the conservation and sustainable management of this commercially important food fish species in the Kashmir Valley.
