A series of yeast strains was tested in order to evaluate their catalytic potential in biocatalysis of one-pot indolizine’s synthesis. Yeast cultivation was performed in a submerged system at 28 °C for 72 h at 180 rpm. An assessment of the reagents’ toxicity on yeast viability and metabolic functionality concluded that the growth potential of three Yarrowia lipolytica strains were least affected by the reactants compared to the other yeast strains. Further, crude fermentation products (biomass and cell-free supernatant)—obtained by submerged cultivation of these yeasts—were used in multistep cascade reactions for the production of fluorescent indolizine compounds with important biologic activities. A whole–cell catalyzed multicomponent reaction of activated alkynes, α-bromo-carbonyl reagents and 4,4′-bipyridine, at room temperature in buffer solution led to the efficient synthesis of bis-indolizines 4a, 4b and 4c, in good-to-excellent yields (47%–77%). The metabolites of the selected Y. lipolytica strains can be considered effective biocatalysts in cycloaddition reactions and the high purity and bioconversion yields of the synthesized indolizines indicates a great potential of this type of “green” catalysts. Seeds of Triticum estivum L. were used to investigate the impact of the final products on the germination and seedling growth. The most sensitive physiological parameters suggest that indolizines, at the concentrations tested, have non-toxic effect on germination and seedling growth of wheat, fact also confirmed by confocal laser scanning microscopy images.

We describe the impact of the number and location of methoxy groups in the structure of chalcones on the speed and efficiency of their transformation by unconventional yeast strains. The effect of substrate concentration on the conversion efficiency in the culture of the Yarrowia lipolytica KCh 71 strain was tested. In the culture of this strain, monomethoxychalcones (2′-hydroxy-2″-, 3″- and 4″-methoxychalcone) were effectively hydrogenated at over 40% to the specific dihydrochalcones at a concentration of 0.5 g/L of medium after just 1 h of incubation. A conversion rate of over 40% was also observed for concentrations of these compounds of 1 g/L of medium after three hours of transformation. As the number of methoxy substituents increases in the chalcone substrate, the rate and efficiency of transformation to dihydrochalcones decreased. The only exception was 2′-hydroxy-2″,5″-dimethoxychalcone, which was transformed into dihydrochalcone by strain KCh71 with a yield comparable to that of chalcone containing a single methoxy group.

В публикации представлены результаты анализа органолептических и физико-химических показателей столовых сухих виноматериалов из винограда сорта Кокур белый (2015-2017 гг. урожая), произрастающего в Судакском районе Республики Крым, полученных с использованием штаммов дрожжей I-527 (Раса 47-К) и I-271 (Феодосия I-19) из Коллекция микроорганизмов виноделия Магарач и препаратов активных сухих дрожжей производства Martin Vialatte , Франция: Vitilevure KD , Vitilevure 58W3 , Vitilevure Quartz M05 . Все исследуемые виноматериалы соответствовали требованиям ГОСТ 32030-2013 для столовых сухих вин: объемная доля этилового спирта составляла в среднем 11,2-12,0 об., массовая концентрация сахаров 1,0-1,4 г/дм3, титруемых и летучих кислот 6,1-6,5 и 0,41-0,54 г/дм3 соответственно. В результате проведенных исследований установлено влияние коллекционных штаммов дрожжей и препаратов активных сухих дрожжей на образование вторичных продуктов брожения (высших спиртов, сложных эфиров, альдегидов и терпеновых соединений) и формирование аромата столовых сухих виноматериалов из крымского аборигенного сорта винограда Кокур белый. Использование штаммов дрожжей I-527 (Раса 47-К) и Vitilevure KD позволило получить виноматериалы с ярким сортовым ароматом цветочно-медового направления с фруктовыми оттенками и свежим, гармоничным вкусом. В этих виноматериалах отмечено высокое содержание сложных эфиров (58,9-62,3 мг/дм3) и терпеновых соединений (4,81-5,42 мг/дм3) по сравнению с другими исследуемыми виноматериалами. При использовании коллекционного штамма дрожжей I-271 (Феодосия I-19) были получены виноматериалы с приглушенным ароматом и полным, гармоничным вкусом. Использование препарата активных сухих дрожжей Vitilevure 58W3 привело к усилению пряных оттенков в аромате виноматериалов, а Vitilevure Quartz M05 к усилению медовых оттенков. Коллекционный штамм дрожжей I-527 (Раса 47-К) и препарат активных сухих дрожжей Vitilevure KD рекомендованы для производства столовых сухих виноматериалов из винограда сорта Кокур белыйThe paper synthesizes analytical data on organoleptic and physicochemical parameters of table base wines from Kokur beliy grapes (2015-2017 harvest) cultivated in the Sudaksky region of the Republic of Crimea, obtained using yeast strains I-527 (Rasa 47-К) and I-271 (Feodosia I-19) from the Collection of microorganisms for winemaking Magarach and dry active yeast preparations produced by Martin Vialatte, France: Vitilevure KD, Vitilevure 58W3, Vitilevure Quartz M05. All the studied base wines met the requirements of GOST 32030-2013 for table dry wines: the volume fraction of ethyl alcohol made on average 11.2-12.0 , total sugars 1.0-1.4 g/dm3, titrated and volatile acids 6.1-6.5 и 0.41-0.54 g/dm3, respectively. The study confirmed the impact of collection yeast strains and active yeast preparations on formation of secondary fermentation products (higher alcohols, esters, aldehydes and terpenic compounds) and aroma dvelopment of table dry base wines obtained from Crimean autochthonous grapevine cultivar Kokur beliy. The use of yeast strain I-527 (Rasa 47-К) and Vitilevure KD produced base wines characterized by rich varietal bouquet with floral and honey flavors and fruity overtones, and fresh, well-balanced taste. The base wines exhibited high ester (58.9-62.3 mg/dm3) and terpenic compounds content, as compared to other base wines analyzed. When collection yeast strain I-271 (Feodosiya I-19) was used, we obtained base wines characterized by muted nose, and full-bodied balanced taste. The use of active dry yeast preparation Vitilevure 58W3 enhanced spicy overtones in the aroma of the base wines, while Vitilevure Quartz M05 preparation strengthened honey hints. Collection yeast strain I-527 (Rasa 47-К) and active dry yeast preparation Vitilevure KD are recommended for use in production of table dry base wines from Kokur beliy grapes.

An assortment of redox-active phenazine compounds like pyocyanin with their characteristic blue-green colour are synthesized by Pseudomonas aeruginosa, Gram-negative opportunistic pathogens, which are also considered one of the most commercially valuable microorganisms. In this study, pyocyanin from Pseudomonas aeruginosa BTRY1 from food sample was assessed for its antibiofilm activity by micro titer plate assay against strong biofilm producers belonging to the genera Bacillus, Staphylococcus, Brevibacterium and Micrococcus. Pyocyanin inhibited biofilm activity in very minute concentrations. This was also confirmed by Scanning Electron Microscopy (SEM) and Confocal Laser Scanning Microscopy (CLSM). Both SEM and CLSM helped to visualize the biocontrol of biofilm formation by eight pathogens. The imaging and quantification by CLSM also established the impact of pyocyanin on biofilm-biocontrol mainly in the food industry.

Quality of plum jerkum is significantly associated to the profile of volatile compounds. Therefore, we decided to assess the impact of various fermentation types on selected properties of plum jerkums, especially compounds which contribute to the aroma of the finished product. We used the following yeast strains: S. cerevisiae S1, H. uvarum H2, and Ethanol RED (S. cerevisiae). Moreover, we considered spontaneous fermentation. S. cerevisiae and H. uvarum strains were isolated during the fermentation of Čačanska Lepotica or Węgierka Dąbrowicka (plum cultivars), respectively. As for fermentation type, spontaneous fermentation of H. uvarum H2 provided the best results. It could be associated to the fact that plum juices fermented with H. uvarum H2 presented the highest concentration of terpenoids, esters, or some higher alcohols. In the current paper, application of indigenous strains of yeasts resulted in the required oenological characteristics, e.g., highest fermentation efficiency and concentration of ethanol was determined in juices fermented with Ethanol RED (S. cerevisiae) and also with S. cerevisiae S1. Our results suggested that indigenous strains of yeasts present in plums demonstrate great potential for the production of plum jerkums of high quality.

The age of incidence of spinal cord injury (SCI) and the average age of people living with SCI is continuously increasing. However, SCI is extensively modeled in young adult animals, hampering translation of research to clinical applications. While there has been significant progress in manipulating axon growth after injury, the impact of aging is still unknown. Mitochondria are essential to successful neurite and axon growth, while aging is associated with a decline in mitochondrial functions. Using isolation and culture of adult cortical neurons, we analyzed mitochondrial changes in 2-, 6-, 12- and 18-month-old mice. We observed reduced neurite growth in older neurons. Older neurons also showed dysfunctional respiration, reduced membrane potential, and altered mitochondrial membrane transport proteins; however, mitochondrial DNA (mtDNA) abundance and cellular ATP were increased. Taken together, these data suggest that dysfunctional mitochondria in older neurons may be associated with the age-dependent reduction in neurite growth. Both normal aging and traumatic injury are associated with mitochondrial dysfunction, posing a challenge for an aging SCI population as the two elements can combine to worsen injury outcomes. The results of this study highlight this as an area of great interest in CNS trauma.

The rum industry is currently worth USD 16 billion, with production concentrated in tropical countries of the Caribbean and Asia-Pacific regions. The primary feedstock for rum production is sugar cane molasses, a by-product of sugar refineries. The main variables known to affect rum quality include the composition of the molasses, the length of fermentation, and the type of barrels and length of time used for aging the rum. The goal of this review is to provide an overview of the impact of these variables on rum quality, and to highlight current challenges and opportunities in the production of rum from molasses. In order to achieve this, we review the relevant contemporary scientific literature on these topics. The major contemporary challenges in the rum production industry include minimising the effects of variability in feedstock quality, ensuring the fermentation process runs to completion, preventing microbial contamination, and the selection and maintenance of yeast strains providing optimum ethanol production. Stringent quality management practices are required to ensure consistency in the quality and organoleptic properties of the rum from batch to batch. Further research is required to fully understand the influences of many of these variables on the final quality of the rum produced.

With the rise of a new generation of technology and industrial changes, the service-oriented manufacturing industry has become the direction of future development. With the background of new manufacturing, this paper constructs an economic development threshold model of employment density of consumer goods industry based on data from Shanghai and Tokyo from 2007 to 2016, and empirically analyzes the impact of the employment density of the consumer goods industry on urban economic development under different population densities. At the same time, by comparing the experience of Tokyo, the development status and prospects of Shanghai’s consumer goods industry are explored. The study found that the threshold of Tokyo’s consumer goods industry is 0.608. When population density is lower than this threshold, the consumer goods industry continues to promote the economic development of Tokyo; however, when the population density is higher than this threshold, the consumer goods industry begins to inhibit the economic development of Tokyo. The Shanghai consumer goods industry threshold is 0.329. Under the threshold, most of the consumer goods industry contributions to the economy are negative, but above the threshold, they begin to show a positive trend. The inflection point of the effect curve of Tokyo’s consumer goods industry on economic development has appeared, but the inflection point of Shanghai’s consumer goods industry has not yet appeared. Compared with Tokyo, the economic vitality of Shanghai’s consumer goods industry has not yet been fully released. With the continued increase of population density in Shanghai, the growth potential of the consumer goods industry is huge, and it is expected to reshape the flourishing age of Shanghai’s light industry brand.

AbstractVery little is known about the impact of flooding and ground saturation on ectomycorrhizal fungi (EcM) and increasing flood events are expected with predicted climate change. To explore this, seedlings inoculated with the EcM species Tuber aestivum were exposed to a range of flood durations. Oak seedlings inoculated with T. aestivum were submerged for between 7 and 65 days. After a minimum of 114-day recovery, seedling growth measurements were recorded, and root systems were destructively sampled to measure the number of existing mycorrhizae in different zones. Number of mycorrhizae did not display correlation with seedling growth measurements. Seven days of submersion resulted in a significant reduction in mycorrhizae numbers and numbers reduced most drastically in the upper zones. Increases in duration of submersion further impacted mycorrhizae numbers in the lowest soil zone only. T. aestivum mycorrhizae can survive flood durations of at least 65 days. After flooding, mycorrhizae occur in higher numbers in the lowest soil zone, suggesting a mix of resilience and recovery. The results will aid in furthering our understanding of EcM but also may aid in conservation initiatives as well as providing insight for those whose livelihoods revolve around the collection of EcM fruiting bodies or cropping of the plant partners.
