A therapeutic strategy to improve wound healing has become an increasingly important medical task due to the rising incidence of adiposity and type II diabetes as well as the proceeding population aging. In order to cope with the resulting burdens, new strategies to achieve rapid and complete wound healing must now be developed. Accordingly, the development of a bioactive wound dressing in the form of a messengerRNA (mRNA)-bearing poly(lactide-co-glycolide acid) (PLGA) coating on surgical suture is being pushed further with this study. Furthermore, the evaluation of the polymer-based transfection reagent Viromer RED has shown that it can be used for the transfection of eukaryotic cells: The mRNA gets properly complexed and translated into a functional protein. In addition, the mRNA-PLGA coating triggered the expression of the keratinocyte growth factor (KGF) in HaCat cells although KGF is not expressed under physiological conditions. Moreover, transfection via surgical sutures coated with mRNA does not affect the cell viability and a proinflammatory reaction in the transfected cells is not induced. These properties make the mRNA-PLGA coating very attractive for the in vivo application. For the future, this could mean that through the use of mRNA-coated sutures in surgical wound closure, cells in the wound area can be transfected directly, thus accelerating and improving wound healing.

In this study, a novel Polycaprolactone suture assembled with Tadalafil was investigated to improve wound healing processes via vascular stimulation. Tadalafil/Polycaprolactone (TP) suture was developed by the electrospinning method. The designed suture was characterized by SEM, mechanical properties assessments, tensile strength measurements and the drug release study. For in vivo tests, rats were classified into two study groups. An incision was made on their back skin and they were sutured with TP suture and Polycaprolactone suture as control. Rats were sacrificed at 7 days following surgery for histopathological examinations with Hematoxylin and Eosin staining. Results of Tensile test demonstrated that the lowest tensile strength belonged to 3 and 4 % wt and the highest tensile strength belonged to 1 and 2 % wt of TP suture. The rate of Tadalafil release showed that the highest drug release was related to 3 and 4% wt which were about 125 to 210 μg during 15 days. The histopathology revealed that the number of blood vessels, collagen fibers, fibroblast, polymorphonuclear leukocytes, and epithelization was remarkable in Tadalafil/Polycaprolactone group during 7-day. A novel Tadalafil/Polycaprolactone suture improved the processes of wound healing by releasing the Tadalafil drug around the sutured wound and can be used in medical applications.

Objective: The extracellular matrix (ECM) is one of the most important elements in wound healing. Absence or dysfunction of the ECM may impair wound healing. The application of acellular dermal matrix (ADM) as a substitute for ECM has been suggested. This study investigated the clinical application and wound healing effects of a paste-type ADM in patients presenting with hard-to-heal wounds due to various causes. Method: Patients with a hard-to-heal wound for >1 month, from September 2017 to February 2019, were included in this study. After debridement, the paste-type ADM was applied, at zero (baseline), two and four weeks. After application of the paste-type ADM, a conventional dressing was applied using polyurethane foam. Wound size, the formation of granulation tissue, re-epithelialisation, complete healing and adverse events were recorded at zero (baseline), one, two, four, eight and 12 weeks after the initial treatment. Results: A total of 18 patients took part (eight male, 10 female, mean age of 56±16.16 years). The mean wound area decreased from 17.42±10.04cm2 to 12.73±7.60cm2 by week one (p<0.05), to 10.16±7.00 by week two (p<0.0005), to 5.56±5.25 by week four (p<0.0001), to 2.77±5.15 by week eight (p<0.0001) and to 2.07±4.78 by week 12 (p<0.0001). The number of patients with >75% re-epithelialisation increased from two (11.1%) at two weeks to five (27.8%) at four weeks, to 11 (61.1%) at eight weeks and to 13 (72.2%) at 12 weeks. The number of patients showing complete wound healing was two (11.1%) at four weeks, nine (50.0%) at eight weeks and 12 (66.7%) at 12 weeks. No adverse events were reported during treatment. Conclusion: The paste-type ADM used in this study is a viable option for facilitating wound healing; it can shorten hospitalisation, and promote a faster recovery and return to normal life activities.

Tilapia skin is being already use in humans and wild animals present burning wounds and showed a great result. The objective is to evaluate if tilapia skin used as an occlusive curative improves equine wound healing in two horses present chronic wound. Both animals are males, adults, both of breed Mangalarga Marchador South America. Every seven days wound we measured, photographed, biopsied for histopathological analysis, cleaned and tilapia curative was changed. Image J software was used to measure wound area. Tilapia skin as an occlusive biological factor seemed to improve healing process, wounds present an area reduction and clinical improvement during 35 days treatment, even though is still waiting for complete wound healing. In equine tilapia skin curative seemed to speed up healing process and allowed reduced curative change from every two days to once a week. This implies in decrease animal´s stress, less pain and treatment cost reduction since we used less bandage amount. Beside that tilapia skin industrial waste. Furthermore, it allowed avoid using antibiotics, which reduces environment pollution and there´s no antibiotic resistance issues.

Abstract Background Citrus reticulata is one of the most cultivated fruit with great benefits for humans in the world. Citrus reticulata peel has several biological activities within them hypoglycemic, hepatoprotective, antimicrobial and antioxidant. The present study emphasizes on the in vivo wound healing and in vitro antimicrobial and antioxidant activities of Citrus reticulata peel extract. Methods Forty albino mice (23–29 g) of either sex were divided into four groups. The test groups were treated with experimental ointment (0.5% and 10% of methanolic extract), negative control were treated with Vaseline and the positive control were treated with silver sulfadiazine. Burn wounds were induced on dorsal area of mice bodies. Wound area measurement was carried out every day during 22 days. Biochemical screening was performed to identify possible compounds. Antioxidant and antimicrobial activities were also determined. Results Significant wound healing activity was observed with topical application of Citrus reticulata peel extract. Wound area reduction at day 16 of treatment was 100% for both treated groups (0.5% and 10%) when compared to positive and negative control it was 100% and 98.32% respectively at day 22. Furthermore higher rate of wound contraction (100% on 16th day) was observed for both treated groups. The result of biochemical screening showed that C. reticulata peel is characterized by highest amount of total polyphenols (13.19 mg/g), flavonoids (4.07 mg/g), vitamin C (13.20 mg/g), carotenoids (0.032 mg/g) and lowest content of macronutrients (Proteins: 0.40%, reducing sugars: 7.21%; lipids: 1.5%). Additionally C. reticulata peel exhibited remarkable antioxidant activity using DPPH and phosphomolybdate methods as well as the extract possess antimicrobial effect against pathogen bactria. Conclusion The findings from this research indicated that Citrus reticulata peel extract is effective in inhibiting the growth of pathogen bacteria and could be of therapeutic potentials for wound healing.

AbstractThis study investigated the efficacy of Omega-7 isolated from the sea buckthorn oil (Polyvit Co., Ltd, Gangar Holding, Ulaanbaatar, Mongolia) in ovine burn wound healing models. In vitro, proliferation (colony-forming rate) and migration (scratch) assays using cultured primary ovine keratinocytes were performed with or without 0.025% and 0.08% Omega-7, respectively. The colony-forming rate of keratinocytes in the Omega-7 group at 72 and 96 h were significantly higher than in the control (P < 0.05). The percentage of closure in scratch assay in the Omega-7 group was significantly higher than in the control at 17 h (P < 0.05). In vivo, efficacy of 4% Omega-7 isolated from buckthorn oil was assessed at 7 and 14 days in grafted ovine burn and donor site wounds. Telomerase activity, keratinocyte growth factor, and wound nitrotyrosine levels were measured at day 14. Grafted sites: Un-epithelialized raw surface area was significantly lower and blood flow was significantly higher in the Omega-7-treated sites than in control sites at 7 and 14 days (P < 0.05). Telomerase activity and levels of keratinocyte growth factors were significantly higher in the Omega-7-treated sites after 14 days compared to those of control (P < 0.05). The wound 3-nitrotyrosine levels were significantly reduced by Omega-7. Donor sites: the complete epithelialization time was significantly shorter and blood flow at day 7 was significantly higher in the Omega-7-treated sites compared to control sites (P < 0.05). In summary, topical application of Omega-7 accelerates healing of both grafted burn and donor site wounds. Omega-7 should be considered as a cost-efficient and effective supplement therapy for burn wound healing.

Background and objectives: The present research was designed to evaluate the chemical composition of Lavandula stoechas essential oil (EOLS) as well as the in vivo wound-healing property. The chemical composition of EOLS was identified by gas chromatography mass spectrometry. Nineteen compounds of EOLS were reported. Linalool was identified as the major chemical compound (24.87%), followed by linalyl acetate (19.10%). EOLS showed a high content of oxygenated compounds (63.54%). In vivo wound healing activity of the topical cream prepared from EOLS (0.5% w/w) was assessed using a circular excision wound model. The wound area (mm2) in all animal groups was estimated and measured on day 0, 4, 8, 11, and 16. Results: The EOLS formulation cream (0.5% v/w) showed the highest effect on wound models when compared to reference Madecassol® (Asiaticoside). On days 4, 11, and 16, wound contractions were 26.4%, 78%, and 96.3% for the EOLS-treated group, and 8.5%, 64.1%, and 86.1% for the vehicle cream-treated group. Animals treated with EOLS cream showed a significant decrease in the epithelization period, wound area, and scar thickness, whereas the rate of wound contraction significantly increased. This is the first such report to be published. Histological analyses were also consistent with the results of the excision experimental method. Treatment with EOLS cream formulation resulted in decreased inflammation and an increased rate of tissue perfusion and proliferation as well as remodeling, along with re-epithelization. Conclusions: Our results support the use of EOLS in the development of pharmaceuticals for the management of wounds, and/or inflammatory-related diseases. Additional studies are needed to elucidate and explain the exact mechanism of its pharmacological activity.

Suturing the wound provides temporary mechanical support enabling natural tissue healing. Understanding the inherent material properties of the suture materials facilitates clinical adjustments in the rate of degradation to ensure proper wound healing. This review aimed to summarize the analysis of the factors that contribute to the tensile strength of surgical sutures and the implications of these factors in wound healing. Our initial search criteria used keywords Tensile or Strength or suture material and returned 494 potentially related articles, these were narrowed to just seven key articles pertaining to the analysis of the primary outcome variable which are reviewed in detail here. The physical properties of these materials are substantially affected by the biological conditions of the model system during the period of analysis. Nevertheless, under in vivo conditions decisions on suture materials should take comorbidities like dietary habits, smoking, oral hygiene maintenance into account as they can all have a significant impact on the prevailing physiological conditions at the wound site.

Background. We compared the efficacy ofP. granatum(P) flower extract with that of silver sulfadiazine (SSD) for treating thermal burn injuries in rats.Methods. Ten Wistar rats in each group were topically given base cream, normal saline, cream containing 1% SSD, or creams containing 5% or 10%Punica granatumflower extract. The treatments were administered once daily until complete wound healing was observed. The wound area and healing time were assessed. In addition, percentage wound contraction and histopathological characteristics such as neovascularization and collagen formation were determined. The tannin content inP. granatumextract was determined.Results. The decrease in the average size of wounds on day 15 of the treatment was higher in rats treated with creams containingP. granatumextract than in rats treated with cream containing SSD (2.8±0.9 cm2versus8.4±3.2 cm2). The wounds completely healed on day 25 of the treatment in rats treated with creams containingP. granatumflower extract compared with those in rats treated with the other agents.Conclusion. These results indicated thatP. granatumflower extract promoted wound healing in rats and could be used for managing burn injuries.
