Polymers have always played a critical role in the development of novel drug delivery systems by providing the sustained, controlled and targeted release of both hydrophobic and hydrophilic drugs. Among the different polymers, polyamides or poly(amino acid)s exhibit distinct features such as good biocompatibility, slow degradability and flexible physicochemical modification. The degradation rates of poly(amino acid)s are influenced by the hydrophilicity of the amino acids that make up the polymer. Poly(amino acid)s are extensively used in the formulation of chemotherapeutics to achieve selective delivery for an appropriate duration of time in order to lessen the drug-related side effects and increase the anti-tumor efficacy. This review highlights various poly(amino acid) polymers used in drug delivery along with new developments in their utility. A thorough discussion on anticancer agents incorporated into poly(amino acid) micellar systems that are under clinical evaluation is included.

The Ocular drug delivery system (ODDS) is the prominently challenging system faced by pharmaceutical researchers. Ophthalmic preparations are available in buffered, sterile and isotonic solutions. For the ocular delivery of drugs, various types of dosage forms are prepared and dispensed. As the drops are easier for the administration likewise more prescribed dosage form is the eye drop solution. For obtaining prolonged therapeutic effect ointment, suspensions and gelled systems are also used. The presence of various barriers as anatomical, physiological and physiochemical barriers makes difficulties in delivery of drugs in at the intended sites. Scientists invented alternate delivery routes to direct access at intended target sites. Second invention involves development of novel drug delivery systems providing better permeability, treatability and controlled release at target site. The liposomal delivery is beneficial because they have the ability of envelopment and both hydrophobic and hydrophilic drugs are suitable for delivery to both the anterior and posterior segment of the eye. Therefore, the uses of this alternative approach become quite a necessary. This formulation of novel devices will definitely help to the overcome ocular barriers and side effects with conventional topical drops. Current reviews on the conventional formulations of ocular delivery and their advancements followed by current nanotechnology based on the formulation developments. The recent incident with other ocular drug delivery planning consists of in situ gels, implants, contact lens and nano wafers are discussed. Drug delivery at ophthalmic route has been proven significant advancement for the future perspectives.

Background: A semisolid gel-like preparation constitutes with the cross-linked polymers may easily swell in the presence of water and incorporate the amount of drug substance known as ‘Hydrogel.’ Objective: To prepare the review on the hydrogels, leading ennoblements, classification, mechanism, recent patents, and evaluation parameters. Methods: This review accrued by taking the help of online and offline journals, books, and other sources. This field includes new developments in the field of inventions in the novel drug delivery of hydrogels. Results: The impact of the formulation of hydrogels is attracting the focus of the researchers expeditiously because of their nature, three-dimensional structure, compatibility with the other excipients was found enormously useful in the biomedical applications and pharmaceutical industries, less time consuming, better sustained and prolonged the effect. Currently, hydrogels growing expeditiously for the intensive work done by the use of natural, synthetic, and semi-synthetic polymers for the transformation of hydrophobic and hydrophilic drugs into hydrogels as compared to develop the conventional gels. Conclusion: The journey of hydrogel goes through with the numerous inventions research papers, reports, publications, and many other sources. The development arouses with the hydrogels in the use of wound dressing, making diapers, contact lens, etc. Now a day it’s pivotally growing in the field of tissue engineering scaffolds, preparation of hydrophobic drugs into hydrogels which is a critical challenge in prompt conditions.

Zedoary turmeric oil and its main active ingredient β-elemene are novel plant-derived anticancer agents with long-term clinical application history and low toxicity, which have been approved by the Chinese SFDA to treat different tumors including cancers of the brain, ovary, prostate, breast, lungs, liver, colon, and other tissues. Unfortunately, their hydrophobic properties, poor stabilities and low bioavailabilities seriously hamper their applications in clinic. Therefore, more attention should be paid to develop novel drug delivery systems for zedoary turmeric oil and β-elemene to enhance their overall quality. Recently, increased research has been carried out on a nanoparticle drug delivery system of zedoary turmeric oil and β-elemene to solve their poor aqueous solubilities and low bioavailabilities in vivo with much remarkable achievements springing up in the last decade. This review presents the novel nanoparticle formulations of zedoary turmeric oil and β-elemene and introduces the possible future prospects of their further study.

Oral route is presently the gold standard in the pharmaceutical industry where it is regarded as the safest, most economical and most convenient method of drug delivery resulting in highest patient compliance. Over the past three decades, orally disintegrating tablets (FDTs) have gained considerable attention due to patient compliance. Usually, elderly people experience difficulty in swallowing the conventional dosage forms like tablets, capsules, solutions and suspensions because of tremors of extremities and dysphagia. In some cases such as motion sickness, sudden episodes of allergic attack or coughing, and an unavailability of water, swallowing conventional tablets may be difficult. One such problem can be solved in the novel drug delivery system by formulating “Fast dissolving tablets” (FDTs) which disintegrates or dissolves rapidly without water within few seconds in the mouth due to the action of superdisintegrant or maximizing pore structure in the formulation. The review describes the various formulation aspects, superdisintegrants employed and technologies developed for FDTs, along with various excipients, evaluation tests, marketed formulation and drugs used in this research area.

Psoriasis is one of the most stern and persistent autoimmune disease which affects about 1-3% population around the globe.  It influences an individual’s life quality by hindering them physically, socially and emotionally. The hyperproliferation of keratinocytes leads to excessive growth of skin cell which causes scaling and formation of plaques. Psoriasis can be mild moderate or severe depending on the percentage of skin area affected. Based on severity of the disease, location and the type of psoriasis, the treatment approaches can vary. Treatment regimens for psoriasis include topical therapies, systemic therapies and phototherapy. Conventional drug delivery approaches face various disadvantages therefore a need for better novel drug delivery system is required. The review paper focuses at providing a broad aspect of psoriasis and the novel treatment approaches which can help in tackling the disease.

Pharmaceutical technology has developed various newer modes of novel drug delivery aspects. Modifications in the previously existing drug delivery methods have led to various newly innovated technologies serving as a safe and effective means of improvement over the existing ones. Novasome technology is one of the new innovations of liposomes which have solved many of the problems related to liposomal drug delivery system. It offers a seven bilayer membrane which has the ability to incorporate both water soluble and insoluble drugs. It has an excellent entrapment efficiency which provides better medication. Formulation of novasomes is achieved in a high shear device. Due to its numerous advantages, novasomes have been used extensively in various fields like cosmetics, chemical, personal care, foods, pharmaceuticals and agrochemicals.
