The fungal strain, Fusarium sp. LY019, was obtained from the Red Sea sponge Suberea mollis. Bioassay-directed partition of the antimicrobial fraction of the extract of the culture of the fungus provided two dimeric alkaloids, fusaripyridines A and B (1 and 2). The compounds possess a previously unreported moiety, 1,4-bis(2-hydroxy-1,2-dihydropyridin-2-yl)butane-2,3-dione. Further, the compounds display a highly oxygenated substitution pattern on the dihydropyridine moieties, representing an additional feature of the fusaripyridines. Fusaripyridines A and B are the first examples of natural products possessing 1,4-bis(2-hydroxy-1,2-dihydropyridin-2-yl)butane-2,3-dione backbone. Careful analyses of the one- and two-dimensional NMR and HRESIMS spectra of the compounds secured their structural mapping, while their absolute stereochemistry was established by analyses of their ECD spectra. The production of such dimeric alkaloids with an unprecedented moiety in the culture of Fusarium sp. LY019 supports further understanding of the biosynthetic competences of the cultured marine-derived fungi. Fusaripyridines A and B selectively inhibited the growth of Candida albicans with MIC values down to 8.0 µM, while they are moderately active against S. aureus, E. coli and HeLa cells.

AbstractThe ethyl acetate extracts prepared from the mycelia of three endophytic fungi Purpureocillium lilacinum, Aspergillus sp., and Fusarium sp., isolated from the roots of Rauvolfia macrophylla (Apocynaceae) were screened for their antiprotozoal activity in vitro against Plasmodium falciparum (NF54), Leishmania donovani, Trypanosoma brucei rhodesiense, and Trypanosoma cruzi. Amongst these extracts, the one from P. lilacinum showed potent antileishmanial activity against L. donovani (IC50 value of 0.174 μg mL−1) with good selectivity (SI=94.9) toward the L6 cell line, whereas the other extracts were inactive and not selective. The fractionation and purification of the active extract from P. lilacinum by column chromatography over silica gel yielded a new ergochromone derivative (1), together with six known compounds: (22E,24R)-stigmasta-5,7,22-trien-3-β-ol (2), (22E,24R)-stigmasta-4,6,8(14),22-tetraen-3-one (3), emodin (4), chrysophanol (5), aloe-emodin (6), and palmitic acid, whose structures were elucidated spectroscopically. Compound 1 was tested in vitro for its antiparasitic activities against the above listed parasites and for its antimicrobial activity against Staphylococcus aureus, Bacillus cereus, Listeria monocytogenes, Escherichia coli, Providencia stuartii, Klebsiella pneumoniae, and Pseudomonas aeruginosa. The compound displayed potent antileishmanial activity against L. donovani with an IC50 value of 0.63 μg mL−1 (0.87 μm) with good selectivity (SI=49.5) toward the L6 cell line. It also exhibited good antibacterial activity against three of the tested microbial strains B. cereus, E. coli ATCC879, and P. stuartii ATCC29916 with minimum inhibitory concentrations below 62.6 μg mL−1. Compound 1 is thus a promising active compound that could be investigated for antileishmanial and antimicrobial drug development.

Matrix porin, the ompF gene product of E. coli, has been the object of a electron crystallographic study of its pore geometry in an attempt to understand its function as a membrane molecular sieve. Three polymorphic forms have been found for two-dimensional crystals reconstituted in phospholipid, two hexagonal forms with different lipid content and an orthorhombic form coexisting with and similar to the hexagonal form found after lipid loss. In projection these have been shown to retain the same three-fold pore triplet geometry and analyses of three-dimensional data reveal that the small hexagonal and orthorhombic polymorphs have similar structure as well as unit cell spacings.

Chaperonins are a class of proteins characterized by their role as morphogenetic factors. They trantsiently interact with the structural components of certain biological aggregates (viruses, enzymes etc), promoting their correct folding, assembly and, eventually transport. The groEL factor from E. coli is a conspicuous member of the chaperonins, as it promotes the assembly and morphogenesis of bacterial oligomers and/viral structures.We have studied groEL-like factors from two different bacteria:E. coli and B.subtilis. These factors share common morphological features , showing two different views: one is 6-fold, while the other shows 7 morphological units. There is also a correlation between the presence of a dominant 6-fold view and the fact of both bacteria been grown at low temperature (32°C), while the 7-fold is the main view at higher temperatures (42°C). As the two-dimensional projections of groEL were difficult to interprete, we studied their three-dimensional reconstruction by the random conical tilt series method from negatively stained particles.

F.VIII R:Ag was determined by quantitative immunelectrophoresis (I.E.) with a prefabricated system. The prefabricated system consists of a monospecific f.VIII rabbit antiserum in agarose on a plastic plate for the one and two dimensional immunelectrophoresis. The lognormal distribution of the f.VIII R:Ag concentration in the normal population was confirmed (for n=70 the f.VIII R:Ag in % of normal is = 95.4 ± 31.9). Among the normal population there was no significant difference between blood donors (one blood donation in 8 weeks; for n=43 the f.VIII R:Ag in % of normal is = 95.9 ± 34.0) and non blood donors (n=27;f.VIII R:Ag = 94.6 ± 28.4 %). The f.VIII R:Ag concentration in acute hepatitis B ranged from normal to raised values (for n=10, a factor of 1.8 times of normal was found) and was normal again after health recovery (n=10, the factor was 1.0). in chronic hepatitis the f.VIII R:Ag concentration was raised in the majority of the cases (for n=10, the factor was 3.8). Out of 22 carrier sera 20 showed reduced, 2 elevated levels of the f.VIII R:Ag concentration. in 5 sera no f.VIII R:Ag could be demonstrated. The f.VIII R:Ag concentration was normal for n=10, reduced for n=20 and elevated for n=6 in non A-non B hepatitis (n=36). Contrary to results found in the literature no difference in the electrophoretic mobility of the f.VIII R:Ag was found between hepatitis patients sera and normal sera.

In order to develop new antimicrobial agents, a series of 3-formyl indole based Schiff bases were synthesized by reacting 3-formyl indole(indole-3-carboxaldehyde) with substituted aniline taking ethanol as solvent. The reaction was carried in the presence of small amount of p-toluene sulphonic acid as catalyst.All the synthesized compounds were characterized by IR, 1H-NMR spectral analysis. All the synthesized compounds were evaluated for antimicrobial activity against two gram positive bacterial strains (B. subtilisand S. aureus) and two gram negative bacterial strains (P. aeruginosaand E. coli) and one fungal strain (C. albicans). All the synthesized compounds were found to have moderate to good antimicrobial activity. The  standard drug amoxicillin, fluconazole were used for antimicrobial activity. Among the synthesized compounds, the maximum antimicrobial activity was shown by compounds GS04, GS07, GS08 and GS10.

Several 6- and 7-alkoxy-2-oxo-2H-chromene-4-carbaldehydes were prepared from corresponding alkyl ethers of 6- and 7-hydroxy-4-methyl-2-oxo-2H-chromen-2-ones by oxidation using selenium dioxide. 6- and 7-Alkoxy-4-methyl-2H-chromenes were obtained with yields of 57-85%. Corresponding 4-carbaldehyde derivatives were prepared with yields of 41-67%. Thiosemicarbazones of these aldehydes with D-galactose moiety were synthesized by reaction of these aldehydes with N-(2,3,4,6-tetra-O-acetyl-β-Dgalactopyranosyl) thiosemicarbazide with yields of 62-74%. These thiosemicarbazones were screened for their antibacterial and antifungal activities in vitro against bacteria, such as Staphylococcus aureus, Escherichia coli, and fungi, such as Aspergillus niger, Candida albicans. Several compounds exhibited strong inhibitory activity with MIC values of 0.78- 1.56 μM, including 8a (against S. aureus, E. coli, and C. albicans), 8d (against E. coli and A. niger), 9a (against S. aureus), and 9c (against S. aureus and C. albicans).

This chapter develops a Euclidean, two-dimensional, regions-based theory. As with the semi-Aristotelian account in Chapter 2, the goal here is to recover the now orthodox Dedekind–Cantor continuum on a point-free basis. The chapter derives the Archimedean property for a class of readily postulated orientations of certain special regions, what are called “generalized quadrilaterals” (intended as parallelograms), by which the entire space is covered. Then the chapter generalizes this to arbitrary orientations, and then establishes an isomorphism between the space and the usual point-based one. As in the one-dimensional case, this is done on the basis of axioms which contain no explicit “extremal clause”, and we have no axiom of induction other than ordinary numerical (mathematical) induction.

The paper presents the results of research on the use of ultrafiltration, using membranes of 200 and 400 kDa separation, for disinfection of municipal treated wastewater. The research was conducted on a fractional technical scale using real municipal treated wastewater from two large wastewater treatment plants treating most of the wastewater over the one-million polycentric Gdańsk agglomeration (1.2 million inhabitants). UF 200 kDa and UF 400 kDa processes enabled further improvement of the physical and chemical parameters of treated wastewater. Total phosphorus (to below 0.2 mg/L–UF 200 kDa, 0.13 mg/L–UF 400 kDa) and turbid substances (to below 0.2 mg/L, both membranes) were removed in the highest degree. COD was reduced efficiently (to below 25.6 mgO2/L–UF 200 kDa, 26.8 mgO2/L–UF 400 kDa), while total nitrogen was removed to a small extent (to 7.12 mg/L–UF 200 kDa and 5.7 mg/L–UF 400 kDa. Based on the reduction of indicator bacteria; fecal coliforms including E. coli (FC) and fecal enterococci (FE) it was found that the ultrafiltration is an effective method of disinfection. Not much indicator bacterial were observed in the permeate after processes (UF 200 kDa; FC—5 CFU/L; FE—1 CFU/L and UF 400 kDa; FC—70 CFU/L; FE—10 CFU/L. However, microscopic analysis of prokaryotic cells and virus particles showed their presence after the application of both membrane types; TCN 3.0 × 102 cells/mL–UF 200 kDa, 5.0 × 103 cells/mL–UF 400 kDa, VP 1.0 × 105/mL. The presence of potentially pathogenic, highly infectious virus particles means that ultrafiltration cannot be considered a sufficient disinfection method for treated wastewater diverted for reuse or discharged from high load wastewater treatment plants to recreational areas. For full microbiological safety it would be advisable to apply an additional disinfection method (e.g., ozonation).

In this article, a simple expression for the center of mass of a system of material points in a two-dimensional surface of Gaussian constant negative curvature is given. By using the basic techniques of geometry, we obtained an expression in intrinsic coordinates, and we showed how this extends the definition for the Euclidean case. The argument is constructive and serves to define the center of mass of a system of particles on the one-dimensional hyperbolic sphere LR1.

Abstract In a recent paper, Balthazar, Rodriguez and Yin found remarkable agreement between the one instanton contribution to the scattering amplitudes of two dimensional string theory and those in the matrix model to the first subleading order. The comparison was carried out numerically by analytically continuing the external energies to imaginary values, since for real energies the string theory result diverges. We use insights from string field theory to give finite expressions for the string theory amplitudes for real energies. We also show analytically that the imaginary parts of the string theory amplitudes computed this way reproduce the full matrix model results for general scattering amplitudes involving multiple closed strings.
