Trees are able to colonize, establish and survive in a wide range of soils through associations with ectomycorrhizal (EcM) fungi. Proper functioning of EcM fungi implies the differentiation of structures within the fungal colony. A symbiotic structure is dedicated to nutrient exchange and the extramatricular mycelium explores soil for nutrients. Eventually, basidiocarps develop to assure last stages of sexual reproduction. The aim of this study is to understand how an EcM fungus uses its gene set to support functional differentiation and development of specialized morphological structures. We examined the transcriptomes of Laccaria bicolor under a series of experimental setups, including the growth with Populus tremula x alba at different developmental stages, basidiocarps and free-living mycelium, under various conditions of N, P and C supply. In particular, N supply induced global transcriptional changes, whereas responses to P supply seemed to be independent from it. Symbiosis development with poplar is characterized by transcriptional waves. Basidiocarp development shares transcriptional signatures with other basidiomycetes. Overlaps in transcriptional responses of L. bicolor hyphae to a host plant and N/C supply next to co-regulation of genes in basidiocarps and mature mycorrhiza were detected. Few genes are induced in a single condition only, but functional and morphological differentiation rather involves fine tuning of larger gene sets. Overall, this transcriptomic atlas builds a reference to study the function and stability of EcM symbiosis in distinct conditions using L. bicolor as a model and indicates both similarities and differences with other ectomycorrhizal fungi, allowing researchers to distinguish conserved processes such as basidiocarp development from nutrient homeostasis.

Ectomycorrhizae formed by the symbiotic interaction between ectomycorrhizal fungi and plant roots play a key role in maintaining and improving the health of a wide range of plants. Mycorrhizal initiation, development, and functional maintenance involve morphological changes that are mediated by activation and suppression of several fungal and plant genes. We identified a gene, Lbras, in the ectomycorrhizal fungus Laccaria bicolor that belongs to the ras family of genes, which has been shown in other systems to be associated with signaling pathways controlling cell growth and proliferation. The Lbras cDNA complemented ras2 function in Saccharomyces cerevisiae and had the ability to transform mammalian cells. Expression of Lbras, present as a single copy in the genome, was dependent upon interaction with host roots. Northern analysis showed that expression was detectable in L. bicolor 48 h after interaction as well as in the established mycorrhizal tissue. Phylogenetic analysis with other Ras proteins showed that Lbras is related most closely to Aras of Aspergillus nidulans.

AbstractA metasurface hologram combines fine spatial resolution and large viewing angles with a planar form factor and compact size. However, it suffers coherent artifacts originating from electromagnetic cross-talk between closely packed meta-atoms and fabrication defects of nanoscale features. Here, we introduce an efficient method to suppress all artifacts by fine-tuning the spatial coherence of illumination. Our method is implemented with a degenerate cavity laser, which allows a precise and continuous tuning of the spatial coherence over a wide range, with little variation in the emission spectrum and total power. We find the optimal degree of spatial coherence to suppress the coherent artifacts of a meta-hologram while maintaining the image sharpness. This work paves the way to compact and dynamical holographic displays free of coherent defects.

The feeding habits of myctophid larvae of Symbolophorus californiensis were examined in the southern transition region of the western North Pacific where the main spawning and nursery grounds of S. californiensis are formed. This species is a key component of the pelagic ecosystems of this region, and their larvae attain one of the largest sizes among myctophids. To analyse gut contents larvae, including most life history stages after yolk-sac absorption (3.7 to 22.2 mm body length (BL)), were collected in the upper 100 m layer in 1997 and 1998. Feeding incidence was higher during the day than at night (53.1–92.3% versus 0–5.6%), and daytime feeding incidence increased gradually with larval growth. Larvae fed mainly on copepods of various developmental stages. Larvae of S. californiensis showed an ontogenetic change in their diet: larvae ≤7.9 mm BL (i.e. preflexion stage) fed mainly on copepod eggs and nauplii, while the larvae ≥8 mm BL consumed mainly calanoid copepodites such as Pseudocalanus and Paracalanus spp. In the largest size-class (16–22.2 mm BL), the furcilia stage of euphausiids was also an important prey item. There was an increase in the average prey size with growth in larvae ≤11.9 mm BL, while the number of prey eaten positively correlated with growth in larvae ≥12 mm BL. The trophic niche breadth also increased with larval growth, which would ensure a wide range of available food resources for the larger size-class larvae.

Abstract During oogenesis, oocytes gain competence and subsequently undergo meiotic maturation and prepare for embryonic development; trimethylated histone H3 on lysine-4 (H3K4me3) mediates a wide range of nuclear events during these processes. Oocyte-specific knockout of CxxC-finger protein 1 (CXXC1, also known as CFP1) impairs H3K4me3 accumulation and causes changes in chromatin configurations. This study investigated the changes in genomic H3K4me3 landscapes in oocytes with Cxxc1 knockout and the effects on other epigenetic factors such as the DNA methylation, H3K27me3, H2AK119ub1 and H3K36me3. H3K4me3 is overall decreased after knocking out Cxxc1, including both the promoter region and the gene body. CXXC1 and MLL2, which is another histone H3 methyltransferase, have nonoverlapping roles in mediating H3K4 trimethylation during oogenesis. Cxxc1 deletion caused a decrease in DNA methylation levels and affected H3K27me3 and H2AK119ub1 distributions, particularly at regions with high DNA methylation levels. The changes in epigenetic networks implicated by Cxxc1 deletion were correlated with the transcriptional changes in genes in the corresponding genomic regions. This study elucidates the epigenetic changes underlying the phenotypes and molecular defects in oocytes with deleted Cxxc1 and highlights the role of CXXC1 in orchestrating multiple factors that are involved in establishing the appropriate epigenetic states of maternal genome.

AbstractEthanol-induced transcriptional changes underlie important physiological responses to ethanol that are likely to contribute to the addictive properties of the drug. We examined the transcriptional responses of Caenorhabditis elegans across a timecourse of ethanol exposure, between 30 min and 8 h, to determine what genes and genetic pathways are regulated in response to ethanol in this model. We found that short exposures to ethanol (up to 2 h) induced expression of metabolic enzymes involved in metabolizing ethanol and retinol, while longer exposure (8 h) had much more profound effects on the transcriptome. Several genes that are known to be involved in the physiological response to ethanol, including direct ethanol targets, were regulated at 8 h of exposure. This longer exposure to ethanol also resulted in the regulation of genes involved in cilia function, which is consistent with an important role for the effects of ethanol on cilia in the deleterious effects of chronic ethanol consumption in humans. Finally, we found that food deprivation for an 8-h period induced gene expression changes that were somewhat ameliorated by the presence of ethanol, supporting previous observations that worms can use ethanol as a calorie source.

A versatile method to access differentially substituted 1,3- and 1,4-diamines via a nickel-catalyzed three-component 1,2-carboamination of alkenyl amines with aryl/alkenylboronic ester nucleophiles and N–O electrophiles is reported. The reaction proceeds efficiently with free primary and secondary amines without needing a directing auxiliary or protecting group, and is enabled by fine-tuning the leaving group on the N–O reagent. The transformation is highly regioselective and compatible with a wide range of coupling partners and alkenyl amine substrates, all performed at room temperature. A series of kinetic studies support a mechanism in which alkene coordination to the nickel catalyst is turnover-limiting.

Background: Although guidelines for fitting hearing aids for children are well developed and have strong basis in evidence, specific protocols for fitting and verifying technologies can supplement such guidelines. One such technology is frequency-lowering signal processing. Children require access to a broad bandwidth of speech to detect and use all phonemes including female /s/. When access through conventional amplification is not possible, the use of frequency-lowering signal processing may be considered as a means to overcome limitations. Fitting and verification protocols are needed to better define candidacy determination and options for assessing and fine tuning frequency-lowering signal processing for individuals. Purpose: This work aims to (1) describe a set of calibrated phonemes that can be used to characterize the variation in different brands of frequency-lowering processors in hearing aids and the verification with these signals and (2) determine whether verification with these signal are predictive of perceptual changes associated with changes in the strength of frequency-lowering signal processing. Finally, we aimed to develop a fitting protocol for use in pediatric clinical practice. Study Sample: Study 1 used a sample of six hearing aids spanning four types of frequency lowering algorithms for an electroacoustic evaluation. Study 2 included 21 adults who had hearing loss (mean age 66 yr). Data Collection and Analysis: Simulated fricatives were designed to mimic the level and frequency shape of female fricatives extracted from two sources of speech. These signals were used to verify the frequency-lowering effects of four distinct types of frequency-lowering signal processors available in commercial hearing aids, and verification measures were compared to extracted fricatives made in a reference system. In a second study, the simulated fricatives were used within a probe microphone measurement system to verify a wide range of frequency compression settings in a commercial hearing aid, and 27 adult listeners were tested at each setting. The relation between the hearing aid verification measures and the listener’s ability to detect and discriminate between fricatives was examined. Results: Verification measures made with the simulated fricatives agreed to within 4 dB, on average, and tended to mimic the frequency response shape of fricatives presented in a running speech context. Some processors showed a greater aided response level for fricatives in running speech than fricatives presented in isolation. Results with listeners indicated that verified settings that provided a positive sensation level of /s/ and that maximized the frequency difference between /s/ and /∫/ tended to have the best performance. Conclusions: Frequency-lowering signal processors have measureable effects on the high-frequency fricative content of speech, particularly female /s/. It is possible to measure these effects either with a simple strategy that presents an isolated simulated fricative and measures the aided frequency response or with a more complex system that extracts fricatives from running speech. For some processors, a more accurate result may be achieved with a running speech system. In listeners, the aided frequency location and sensation level of fricatives may be helpful in predicting whether a specific hearing aid fitting, with or without frequency-lowering, will support access to the fricatives of speech.

The palps of all stages of Ixodes trianguliceps are provided with a ventral plate below the basal article; formerly this plate was thought to be the first palpal article. This article in the larva and nymph is produced forward into a spur, but in the female tick this spur is incorporated into the basis capituli as the sella. The hypostomes of I. trianguliceps and I. canisuga are redescribed to clear up existing inaccuracies.Ticks in which the rostrum does not extend beyond the apex of the first palpal article are found on birds, and this probably represents a primitive condition. Those with palpal spurs, which may or may not be fused with the basis capituli, are found on birds (not in Britain) or on mammals of the mouse size group, and those where the rostrum is produced beyond the first palpal article occur on a wide range of large and small animals. The longer and more heavily toothed digits of ticks appear to be associated with a wide host range and vice versa. The structure of the digit may also influence the choice of attachment sites by ticks on their hosts because the microstructure of the skin varies in different parts.Variations in size and form of the scuta of some British ticks are described, and the mean growth rate is ascertained from this data. The information shows that the material of I. ricinus and I. hexagonus is homogeneous, and that specific differences occur in size, shape, the position and type of dermal ducts and in the relation between scutal and alloscutal bristles.The morphology of Gené's organ in I. hexagonus is described. It consists of a basal sac-like portion surmounted by four horns and lined with a cuticle beneath an epithelium. The gland is a proliferation of the epithelium and located near the bifurcation of the base into the horns. A watery refractile fluid, secreted by the gland, accumulates between the epidermis and the cuticle in the horn-like extensions. Proximally the thick endocuticle and epidermis lie close together, and two cuticularized rods penetrate the endocuticle for about half-way along the stalk. The rods arise from the postero-dorsal margin of the basis capituli. Muscles, arising from the free ends of the rods, pass back to near the hind-margin of the scutum; they retract the basis capituli after egg laying and indirectly assist in the retraction of Gené's organ. A suggested mechanism for everting Gené's organ in I. hexagonus is outlined.During feeding the opisthosomatic cuticle of all developmental stages of the tick is much stretched. In the larva, where the cuticle is thin, this is effected by the flattening of the epicuticular pleats, but nymphs and females have, in addition, two longitudinal folds alongside the body which stretch to a far greater extent than do the epicuticular pleats. Similar folds are present between the hard ventral plates and the scutum and between the epimeral and median plates in the male. Coupled with the large size of the male digit, this suggests that morphologically, at least, males can imbibe blood.The growth of the leg segments of I. hexagonus is not constant from stage to stage, and as a result the shortest ‘leg-length’ of the female exceeds the longest ‘leg-length’ of the male.
