A series of hydroxytyrosol (HT) derivatives were synthesized by modification of alcohol hydroxyl group of HT, twenty-five target compounds were obtained and characterized by NMR and HRMS. The antioxidant activities of those compounds were evaluated in three different assays. Except 3e and 3y, all other compounds demonstrated significant 2,2and#39;-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) free radical cation scavenging activity ranging from IC50 3.4 to 24.4 μM, which were more potent than L-ascorbic acid (IC50=24.8 μM). Compounds 3b-3d, 3f-3k, 3m-3x were better than Trolox (18.3 M). Moreover, the ferric reducing antioxidant power (FRAP) of all compounds were discovered to be more potent than L-ascorbic acid (40.7 mmol/g), except 3e, all other compounds (141.5-202.1 mmol/g) were better than Trolox (94.7 mmol/g). Compounds 3a-3d, 3f-3j, 3l-3m, 3o, 3q, 3t, 3v-3y exhibited more potent hydroxyl radical scavenging activity (IC50=245.1-475.1 M) than L-ascorbic acid (554.4 M) and Trolox (500.4 M). Compounds 3q, 3t and 3y exhibited more potent -Glucosidase inhibition activity (39.1-52.4 M) than Acarbose (60.9 M). Compounds 3a, 3d, 3f-3m, 3s-3t, 3v-3y showed some acetylcholinesterase inhibition activities, compounds 3a, 3d, 3f-3j, 3l-3m, 3o-3p, 3s-3t, 3w showed some butyrylcholinesterase inhibition activities.

The antioxidant activities of crude extract ofPhaulopsis fascisepalaleaf were evaluated and compared with α-tocopherol and BHT as synthetic antioxidants and ascorbic acid as natural-based antioxidant.In vitro, we studied its antioxidative activities, radical-scavenging effects, Fe2+-chelating ability and reducing power. The total phenolic content was determined and expressed in gallic acid equivalent. The extract showed variable activities in all of thesein vitrotests. The antioxidant effect ofP. fascisepalawas strongly dose dependent, increased with increasing leaf extract dose and then leveled off with further increase in extract dose. Compared to other antioxidants used in the study, α-Tocopherol, ascorbic acid and BHT,P. fascisepalaleaf extract showed less scavenging effect on α,α,-diphenyl-β-picrylhydrazyl (DPPH) radical and less reducing power on Fe3+/ferricyanide complex but better Fe2+-chelating ability. These results revealed thein vitroantioxidant activity ofP.fascisepala.Further investigations are necessary to verify these activitiesin vivo.

Ten indeterminate tomato cultivars differing in fruit shape and typology, grown simultaneously in a greenhouse from Oltenia (Southwestern Romania) and harvested at red ripe stage, were investigated for the contents in ascorbic acid, lycopene, β-carotene, total phenolics, minerals (K, Na, Ca and Mg) and trace elements (Fe, Cu, Mn, Cr, Zn and B). In addition, their antioxidant activities were determined. Significant differences were found among tomato cultivars in all studied antioxidant compounds, as well as in the antioxidant activity. Ascorbic acid ranged from 91.9 to 329.7 mg kg-1 fw, lycopene ranged from 19.7 to 49.0 mg kg-1 fw, while total phenolic compounds varied between 300.2 and 557.8 mg kg-1 fw. Antioxidant activity ranged from 0.81 mmol Trolox kg-1 fw to 1.74 mmol Trolox kg-1 fw and it was significantly correlated to total phenolics content (r = 0.91; p< 0.05). The cocktail type cultivar ‘Tiger’ and the cherry type cultivar ‘Belle’ proved to be the most powerful in antioxidant activity and phenolic compounds while the rectangular plum shaped cultivar ‘Porto’ recorded the highest average lycopene content. A valuable cultivar proved to be the brownish red ‘Sacher’ which registered among the highest contents of phenolics, lycopene and ascorbic acid. Mineral and trace elements contents were also significantly affected by cultivars. Values recorded for K, Ca and Mg ranged from 2139.6 to 3056.9 mg kg-1, 137.7 to 325.8 mg kg-1 and 27.3 to 168.7 mg kg-1 respectively.

Objective: The present study deals with the antioxidant assays of the different leaf extracts of two medicinal plants, Sphaeranthus indicus and Psophocarpus tetragonolobus. Methods: Dried leaves of S. indicus and P. tetragonolobus were packed in separate round bottom flasks for sample extraction using ethanol, methanol, hexane, and distilled water as solvents for 72 h, and the extracts were collected after evaporating the solvents. Antioxidant studies of the various extracts were performed by 1-diphenyl-2-picrylhydrazyl and Ferric Reducing Ability of Plasma assays.Results: Among the two plants studied, S. indicus showed better 2-diphenyl-1-picryl-hydrazyl (DPPH), scavenging activity than P. tetragonolobus with IC50 values of 174.380 and 262.313, respectively, as compared to that of the standard, ascorbic acid, IC50 value of which being 111.16. The FRAP assay results for both the plants indicated that the methanol fractions showed closer results when compared with standards, ascorbic acid and quercetin. The IC50 value of S. indicus, P tetragonolobus, ascorbic acid, and quercetin was 70.065, 151.953, 85.162, and 79.647, respectively. These results clearly indicate that S. indicus methanol fraction had better antioxidant activity when compared to both standards.Conclusion: It is concluded that S. indicus and P. tetragonolobus have excellent antioxidant activities which could be the major contributing factors for their medicinal roles. Further studies in this direction are being carried on.

Extracts of the leaves, stem, roots, and seeds of <em>Mitracarpus scaber</em> plants have been thoroughly investigated in an attempt to determine their phytochemical and antioxidant activities.<em> </em>Phytochemical screening carried out by thin-layer chromatography (TLC) revealed the presence of several secondary metabolites in all the selective extracts of the plant. The levels of phenolic compounds showed that the leaves of <em>M. Scaber</em> are the richest in polyphenols with an average value of 488.291 ± 1.205 μg GAE/g (microgram of gallic acid equivalent per gram of the dry matter of the powdered plant). The total flavonoid assay revealed a high content in the leaves (1.624 ± 0.032 %). Also, studies of the antioxidant activities by DPPH<sup>● </sup>(2,2-diphenyl-1'-picrylhydrazyl) method showed a significant effect compared to vitamin C (96.62 %). The percentages of inhibition are respectively 89.82 % and 82.3 % for ethyl acetate extracts of the leaves and seeds. Furthermore, the inhibitory percentages of n-butanol in the leaves and seeds are 90.57 % and 83.67 %, respectively. Calculated IC<sub>50</sub> showed that the n-butanol fraction of the seeds exhibited the highest activity with an IC<sub>50</sub> of 0.293 mg/mL less than ascorbic acid with an IC<sub>50</sub> of 0.387 mg/mL.

The presence of natural antioxidant in plants is well known. Plant phenolics constitute one of the major groups of components that act as antioxidant and free radical terminator. Hence, this study focused on investigating the antioxidant activity of Celery plant (Apium graveolens L). The fresh leaves were collected, crushed and extracted with ethanol and acetone by maceration. The radical scavenging properties of the extracts were determined by measuring changes in absorbance of DPPH radical at a wave lenght of 517 nm by UV and ascorbic acid is used as the standard. It showed that the crude ethanolic extract has higher antioxidant activity compared to ascorbic acid and acetone extract with less scavenging activity. The values were (IC50 114.6 µg/mL) for ascorbic acid, (IC50 112 µg/mL) for the crude ethanolic extract and (IC50172 µg/mL) for crude acetone extract. The result shows that Celery plant grown in Jos possess good antioxidant properties which may be linked to the presence of phenolics and flavonoids in the plant, which justifies its use as a medicinal plant. This can be further investigated for the isolation and identification of active compounds of medicinal utilities.

Environmental and occupational exposure to chromium compounds, especially hexavalent chromium, is widely recognized as potentially nephrotoxic in humans and animals. The present study aimed to assess the efficacy of cactus (Opuntia ficus-indica) against sodium dichromate-induced nephrotoxicity, oxidative stress, and genotoxicity. Cactus cladodes extract (CCE) was phytochemically studied and tested in vitro for its potential antioxidant activities. Additionally, the preventive effect of CCE against sodium dichromate-induced renal dysfunction in a Wistar rat model (24 rats) was evaluated. For this purpose, CCE at a dose of 100 mg/kg was orally administered, followed by 10 mg/kg sodium dichromate (intraperitoneal injection). After 40 days of treatment, the rats were sacrificed, and the kidneys were excised for histological, lipid peroxidation, and antioxidant enzyme analyses. The phenol, flavonoid, tannin, ascorbic acid, and carotenoid contents of CCE were considered to be important. Our analyses showed that 1 mL of CCE was equivalent to 982.5 ± 1.79 μg of gallic acid, 294.37 ± 0.84 μg of rutin, 234.78 ± 0.24 μg of catechin, 204.34 ± 1.53 μg of ascorbic acid, and 3.14 ± 0.51 μg of β-carotene. In vivo, pretreatment with CCE was found to provide significant protection against sodium dichromate-induced nephrotoxicity by inhibiting lipid peroxidation, preserving normal antioxidant activities, and protecting renal tissues from lesions and DNA damage. The nephroprotective potential of CCE against sodium dichromate toxicity might be due to its antioxidant properties.

The lipid peroxidation-inhibiting activities of aqueous methanolic extracts of eleven Salvia species (Fam. Lamiaceae) were evaluated in an enzyme-independent biological system. The total polyphenol contents and the amounts of the most abundant phenoloids of the genus, caffeic and rosmarinic acids, were also determined. The EC50 values of the extracts displayed substantial differences. All of the investigated species except S. jurisicii (EC50 191.2 μg/mL) exhibited higher activities than that of ascorbic acid (EC50 123.8 μg/mL), the reference compound. Among the studied species, S. scabiosifolia (EC50 5.4 μg/mL) demonstrated the highest effect, followed in sequence by S. dumetorum, S. transsylvanica, S. officinalis l albiflora, S. nemorosa l albiflora and S. recognita (EC50 6.5 – 10.2 μg/mL). The close correlation was confirmed between the antioxidant activities and the total phenol contents of the extracts. For caffeic and rosmarinic acids, the correlation was much weaker, indicating the important role of other polyphenols in the antioxidant activity.

AbstractGarcinia kola (GK) stem bark, Uvaria chamae (UC) root, and Olax subscorpioidea (OS) root are components of various indigenous/traditional anticancer regimens. It is, therefore, possible that they might combat oxidative stress and impair cellular proliferation linked to carcinogenesis. In this study, we investigated the antioxidative, mito-depressive, and DNA-damaging activities of the three plant extracts in order to provide further mechanistic insights into their potential anticancer roles in documented cancer remedies that include them. Antioxidative properties were investigated in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and nitric oxide (NO) radical scavenging assays and an animal model of drug (cisplatin)-induced oxidative stress. The Allium cepa assay and the single cell gel electrophoresis (SCGE) assay were used to assess mito-depressive and DNA-damaging activities. GK and OS showed significantly higher antioxidant activities in the DPPH assay than ascorbic acid; OS had the lowest IC50 of the three plants in the NO assay, comparable to that of ascorbic acid. Pretreatment with the extracts produced an ameliorative and protective effect against the cisplatin-induced oxidative stress as shown by inhibition of lipid peroxidation and improved or restored reduced glutathione and superoxide dismutase levels. In the Allium test, the three extracts produced significant decreases in root growth and also significant cytotoxicity as evidenced by decreased mitotic index. Each of the extracts also showed significantly increased tail DNA (%) in the SCGE assay, indicating the significant DNA-damaging effect. Taken together, this study demonstrates the possible chemopreventive and chemotherapeutic potentials of the three study extracts, which may explain the roles of their source plants in traditional remedies in the therapy of cancers.
