Among abiotic stresses, both drought and UV-B radiation effectively trigger the accumulation of secondary metabolites, and can be widely applied in plant factories. The objectives of this study were to investigate antioxidant accumulation under drought stress alone, or in combination with UV-B radiation near harvest, and to determine an optimal treatment time for maximum antioxidant production. Kale (Brassica oleracea L. var. acephala) plants were grown in a plant factory and harvested at 42 days after transplanting. The single and combination treatments lasted for 7 to 1 days and 4 to 2 days before harvest, respectively. The results of both Fv/Fm (maximal photochemical efficiency in photosystem II) and leaf water potential could ensure the function of photosynthesis and maintain normal leaf moisture in single drought treatments of less than 4 days. The total phenolic and flavonoid contents and antioxidant activities were significantly increased in both single and combination treatments for 3 to 4 days, compared to other treatments. The supplementary UV-B treatments showed no extra formation of antioxidants compared to the single drought treatments. As a result, drought for 3 days before harvest could achieve the highest potential value of kale as a source of natural antioxidants.

Honey is a well-known natural sweetener and is rich in natural antioxidants that prevent the occurrence of oxidative stress, which is responsible for many human diseases. Some of the biochemical compounds in honey that contribute to this property are vitamins and phenolic compounds such as phenolic acids and flavonoids. However, the extent to which these molecules contribute towards the antioxidant capacity in vitro is inconsistently reported, especially with the different analytical methods used, as well as other extrinsic factors that influence these molecules’ availability. Therefore, by reviewing recently published works correlating the vitamin, total phenolic, and flavonoid content in honey with its antioxidant activities in vitro, this paper will establish a relationship between these parameters. Based on the literature, vitamins do not contribute to honey’s antioxidant capacity; however, the content of phenolic acids and flavonoids has an impact on honey’s antioxidant activity.

ABSTRACT Th e aim of this study was to examine the phenolic and flavonoid contents and antioxidant activities of methanol and chloroform extracts of leaves and twigs of Daphne blagayana. Th e total phenolic content in the chloroform extract of plant twigs (90.26 ± 0.69 mg GA/g) was higher than that of the other extracts (from 76.56±0.89 to 77.45±0.43 mg GA/g). In the case of flavonoids, a greater value was also obtained for the chloroform extract of twigs (35.24±0.55 mg RU/g). Several diff erent methods were used to determine the antioxidant activity of the tested extracts, including total antioxidant capacity, metal chelating activity, hydroxyl radical scavenging activity and inhibitory activity against lipid peroxidation. Our results showed that although secondary metabolites of the plants may contribute significantly to their antioxidant activities, those antioxidant activities were not directly related to the phenolic and flavonoid amounts. Th e results of the present analysis demonstrated, for the first time, that Daphne blagayana leaves and twigs possess high phenolic and flavonoid contents, as well as potential antioxidant activity. Th is study suggests that Daphne blagayana twigs and leaves may potentially be used as an accessible source of natural antioxidants.

Ophiopogon japonicustubers have been widely used as food and traditional Chinese medicine in China. However, their nutritional composition has not been fully reported yet. This study aimed to analyze the nutritional composition ofO. japonicustubers. Theα-glucosidase inhibitory and antioxidant activities of the extracts obtained fromO. japonicustubers were also evaluated byin vitroassays. The results indicated thatO. japonicustubers are rich in carbohydrates, proteins, minerals, and amino acids. Among four extracts, the n-butanol fraction (nBF) and chloroform/methanol extract (CME) ofO. japonicustubers had high amounts of total phenolic and flavonoid contents and exhibited goodα-glucosidase inhibitory and antioxidant activities. Theα-glucosidase inhibition of nBF was higher than acarbose. Overall,O. japonicustubers are full of nutritional compounds and have goodα-glucosidase inhibitory and antioxidant activities.

Olive pomace (OP) is a valuable food byproduct that contains natural phenolic compounds with health benefits related to their antioxidant activities. Few investigations have been conducted on OP from the United States while many studies on European OP have been reported. OP of Arbequina, the most common cultivar from California, was collected and extracted by water, 70% methanol and 70% ethanol, followed by purification using macroporous absorbing resin. Results showed that the extractable total phenolic content (TPC) was 36–43 mg gallic acid equivalents (GAE)/g in pitted, drum-dried defatted olive pomace (DOP), with major contributions from hydroxytyrosol, oleuropein, rutin, verbascoside, 4-hydroxyphenyl acetic acid, hydroxytyrosol-glucoside and tyrosol-glucoside. Macroporous resin purification increased TPC by 4.6 times the ethanol crude extracts of DOP, while removing 37.33% total sugar. The antioxidant activities increased 3.7 times Trolox equivalents (TrE) by DPPH and 4.7 times TrE by ferric reducing antioxidant power (FRAP) in the resin purified extracts compared to the ethanol crude extracts. This study provided a new understanding of the extraction of the bioactive compounds from OP which could lead to practical applications as natural antioxidants, preservatives and antimicrobials in clean-label foods in the US.

In this study, the total phenolic and flavonoid contents and antioxidant activities of hexane and methanol extracts of Cotoneaster transcaucasicus Pojark. fruits in the Rosaceae family were investigated. Using the Folin-Ciocaltaeu method, the amount of phenolic substances was determined as 50.34 ± 2.70 mg of GAE/g in methanol extract and 38.06 ± 1.32 mg of GAE/g in hexane extract. The total amount of flavonoids was calculated as 52.01 ± 3.78 mg of QE/g in the methanol extract and 60.54 ± 6.82 mg of QE/g in the hexane extract, using the aluminum nitrate method. Antioxidant activities of C. transcaucasicus hexane and methanol extracts were evaluated by DPPH and FRAP methods. IC50 values of the extracts were calculated as 0.411 mg/mL for hexane extract and 0.237 mg/mL for methanol extract. In addition, it was determined that the reducing antioxidant power of Fe3+-Fe2+ was higher in hexane extract. As a result, it has been observed that the phenolic and flavonoid content and antioxidant activities of C. transcaucasicus fruits are high. This study is the first study in C. transcaucasicus species and it is thought that it will lead the future studies.

Two new prenylated isoflavonoids, namely Erinasone A and Erinasone B along with 19 known secondary metabolites, including twelve isoflavonoids, three polyphenols, one flavonol, two flavanones and three steroids, were isolated from the whole plant (roots and aerial parts) of Erinacea anthyllis. Structures of all isolated compounds were elucidated by spectroscopic analysis, including 1D and 2D NMR (1H, 13C, COSY, HSQC, TOCSY, HMBC and NOESY), mass spectrometry (ESI-MS), UV-Vis, measurement of optical rotation [α]D and by comparison with the literature data. The total phenolic and flavonoid contents were determined in this study. Furthermore, the antioxidant and antibacterial activities of the EtOAc and n-BuOH extracts of E. anthyllis were evaluated. These extracts exhibited moderate antibacterial and antioxidant activities. Their IC50 values were approximately 0.04 mg/mL ( n-BuOH) and 0.037 mg/mL (EtOAc). Consequently, Erinacea anthyllis is a rich source of polyphenolic compounds particularly isoflavonoids used as chemotaxonomic markers for the subfamily Papilionoideae of the family Fabaceae.

  Objectives: The goals of this research were to observe antioxidant properties from different parts of Simana lagi apple ([Malus domestica Borkh.] “Simana lagi”) using two antioxidant testing methods which were 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2’-azino-bis(3-ethylbenzthiazoline-6- sulfonic acid (ABTS) and correlation of total phenolic and flavonoid content with their inhibitory concentration 50 (IC50) of DPPH and IC50 of ABTS.Methods: Each sample was extracted by reflux using different polarity solvents. The extracts were evaporated using rotary evaporator. Antioxidant activities were tested using DPPH and ABTS assays, determination of total phenolic, and flavonoid content were carried out by ultraviolet-visible spectrophotometry and correlation with their IC50 of DPPH and IC50 of ABTS activities were analyzed by Pearson’s method.Results: The ethanolic leaves extract (LV3) of Simana lagi apple also exposed the highest total phenolic content (TPC) (13.88 g gallic acid equivalents/100 g), while the highest total flavonoid content was presented by ethyl acetate extract (LV2) (7.21 g QE/100 g). The lowest IC50 of DPPH scavenging activity 0.19 μg/ml, and the lowest IC50 of ABTS scavenging activity 0.15 μg/ml was given by ethanolic flesh extract (FL3) of Simana lagi apple. There were significantly negative correlation between TPC in all different parts extracts of Simana lagi apple with their IC50 of DPPH and IC50 of ABTS.Conclusions: All different parts extracts of Simana lagi apple were categorized as very strong antioxidant by DPPH and ABTS method (except n-hexane peels extract and n-hexane LV by DPPH method, and n-hexane FE by ABTS method). The major contributor in antioxidant activities of peels, flesh and leaves extracts of Simana lagi apple by DPPH and ABTS methods were phenolic compounds. Antioxidant activities of peels, flesh and leaves extracts of Simana lagi apple showed linear results by DPPH and ABTS methods.

Objective: The present study was undertaken to investigate the antioxidative activity of fruit pulp, seed and bark of T. bellerica. Methods: Methanol (70%) and water were used for the extraction, and the respective extracts were analyzed for total phenolic contents along with the antioxidant activities through different assays. Results: Amongst the tested extracts, methanolic extracts were found to be better than aqueous with the highest total phenolic contents (TPC). Methanolic fruit pulp showed lower IC50 for free radical (118.7 µg/ml), superoxide anion radical (77.65 µg/ml) and hydroxyl radical (73.76 µg/ml) scavenging activities. Methanolic fruit pulp also showed lower IC50 for lipid peroxidation (115.6 µg/ml) and ferric thiocyanate (184.98 µg/ml) assay than aqueous extracts in comparison to standard quercetin. Further, correlation between TPC and antioxidant studies revealed that phenolics are mainly responsible for antioxidant activity of T. bellerica. Conclusion: The current study suggests that the methanolic extract of T. bellerica could be a potential source of natural antioxidants for food and pharmaceutical companies.

Hypochaeris laevigata var. hipponensis (Asteraceae) is an endemic plant from Algeria. In the current study, we analyzed for the first time its chemical composition, especially phenolic constituents of dichloromethane (DCM), ethyl acetate (EA), and n-butanol (BuOH) fractionsof the aerial parts of Hypochaeris laevigata var. hipponensis by liquid chromatography-mass spectrometry (LC-MS/MS). The number of phenolic compounds detected in DCM, EA, and BuOH fractions were found to be 9, 20, and 15, respectively. More specifically, 12 phenolic acids were detected. Among them, quinic acid, chlorogenic acid, and caffeic acid were the most abundant ones. Meanwhile, only seven flavonoids were detected. Among them, rutin, apigetrin, and isoquercitrin were the major ones. We also determined the total phenolic and flavonoid contents, and fraction EA showed the highest values, followed by BuOH, and DCM fractions. Furthermore, the antioxidant action was dictated by five methods and the tested plant fractions demonstrated a noteworthy antioxidant action.
