Abstract: Endophytic Fungi are microorganisms that live in the plant tissue systems such as seeds, leaves, flowers, twigs, stems, and roots. Endophytic fungi used in this study was isolated from Mangrove Rhizophora apiculata leaves. R. apiculata is widely grown in coastal areas. This study aimed to determine whether there was an antibacterial effect of endophytic fungi that was isolated from leaves of Rhizophora apiculata against the sample bacteria Pseudomonas aeruginosa and Staphylococcus aureus. This study was conducted in the Pharmacology and Therapeutics Laboratory of Sam Ratulangi University by using experimental methods. The results showed two types of fungi, tusky white endophytic fungi and white filamentous endophytic fungi, isolated from Mangrove Rhizophora apiculata leaves. Both types of fungi showed the presence of bacterial growth inhibition, but the white filamentous endophytic fungi had greater antibacterial effect than the tusky white endophytic fungi.Keywords: endophytic fungi, antibacterial, rhizophora apiculata leavesAbstrak: Jamur endofit merupakan mikroorganisme yang hidup di dalam sistem jaringan tumbuhan seperti biji, daun, bunga, ranting, batang, dan akar. Jamur endofit yang digunakan pada penelitian ini merupakan hasil isolasi dari daun mangrove Rhizophora apiculata. Tanaman mangrove R.apiculata ini banyak tumbuh di kawasan pesisir pantai. Penelitian ini bertujuan untuk mengetahui apakah terdapat efek antibakteri jamur endofit yang diisolasi dari daun Rhizophora apiculata terhadap bakteri uji Pseudomonas aeruginosa dan Staphylococcus aureus. Penelitian ini dilakukan di Laboratorium Farmakologi dan Terapi Universitas Sam Ratulangi Manado dengan menggunakan metode eksperimental. Hasil penelitian ini didapatkan dua jenis jamur yaitu jamur endofit berwarna putih gading dan jamur endofit berwarna putih berserabut yang diisolasi dari daun mangrove Rhizophora apiculata. Kedua jenis jamur menunjukan adanya daya hambat pertumbuhan kedua bakteri uji, namun jamur endofit berwarna putih berserabut memiliki efek antibakteri lebih besar dari pada jamur endofit berwarna putih gading.Kata kunci: jamur endofit, antibakteri, daun rhizophora apiculata

Abstract: Endophytic fungi is a species of fungi which grows on plant tissue that are harmless for the host plant it self. Endophytic fungi can produce a substance potencial to be antibacteria. This research aimed to test the presence of antibacterial effect on an isolated root of mangrove plant Bruguiera gymnorrhiza to Escherichia coli and Staphylococcus aureus. The methods that are being used as test to antibacterial was sandwich method, done by putting mycelia of endophytic fungi in agar combination media that has been smear with Escherichia coli and Staphylococcus aureus. Obtained from research that has been made, it conducted two species of endophytic fungi that isolated from mangrove plant root Bruguiera gymnorrhiza. Both endophytic fungi has effect as antibacteria against Escherichia coli and Staphylococcus aureus, but the second type of endophytic fungi has more effective and stronger antibacterial effect compare to endophytic fungi type I and positive control. Jamur endofit dapat menghasilkan senyawa yang berpotensi sebagai antibakteri. Penelitian ini bertujuan untuk menguji ada tidaknya efek antibakteri jamur endofit yang diisolasi dari akar tumbuhan bakau Bruguiera gymnorrhiza terhadap bakteri Staphylococcus aureus dan Escherichia coli. Metode yang digunakan untuk uji antibakteri yaitu metode sandwich, dilakukan dengan cara menempelkan miselia jamur endofit pada media agar kombinasi yang telah dioleskan bakteri uji. Dari penelitian yang dilakukan diperoleh dua jenis jamur endofit yang diisolasi dari akar tumbuhan bakau Bruguiera gymnorrhiza. Kedua jamur endofit memiliki efek sebagai antibakteri terhadap kedua bakteri uji akan tetapi jamur endofit tipe II memiliki efek antibakteri yang lebih baik dibandingkan dengan jamur endofit tipe I dan kontrol positif. ABSTRACTStaphylococcus aureusandPseudomonas aeruginosaare common nosocomial pathogens responsible for biofilm-associated infections. Proton pump inhibitors (PPI), such as esomeprazole, may have novel antimicrobial properties. The objective of this study was to assess whether esomeprazole prevents sessile bacterial growth and biofilm formation and whether it may have synergistic killing effects with standard antibiotics. The antibiofilm activity of esomeprazole at 0.25 mM was tested against two strains each ofS. aureusandP. aeruginosa. Bacterial biofilms were prepared using a commercially available 96-peg-plate Calgary biofilm device. Sessile bacterial CFU counts and biomass were assessed during 72 hours of esomeprazole exposure. The killing activities after an additional 24 hours of vancomycin (againstS. aureus) and meropenem (againstP. aeruginosa) treatment with or without preexposure to esomeprazole were also assessed by CFU and biomass analyses.P. aeruginosaandS. aureusstrains exposed to esomeprazole displayed decreased sessile bacterial growth and biomass (P< 0.001, each parameter). After 72 h of exposure, there was a 1-log10decrease in the CFU/ml of esomeprazole-exposedP. aeruginosaandS. aureusstrains compared to controls (P< 0.001). After 72 h of exposure, measured absorbance was 100% greater inP. aeruginosacontrol strains than in esomeprazole-exposed strains (P< 0.001). Increased killing and decreased biomass were observed for esomeprazole-treated bacteria compared to untreated controls exposed to conventional antibiotics (P< 0.001, each parameter). Reduced biofilm growth after 24 h was visibly apparent by light micrographs forP. aeruginosaandS. aureusisolates exposed to esomeprazole compared to untreated controls. In conclusion, esomeprazole demonstrated an antibiofilm effect against biofilm-producingS. aureusandP. aeruginosa.

ABSTRACTEndophytic fungi are fungi that live in plant tissues and do not harm the plants. Endophytic fungi can produce compounds that have the potential to be antibacterial. This study aims to examine the antibacterial activity of endophytic fungi isolated from the leaves and stems of the cat's whiskers plant (Orthosiphon aristatus) against Escherichia coli and Staphylococcus aureus bacteria. Cat's whiskers contain essential oils which are used as antibacterial against some pathogenic bacteria such as Staphylococcus aureus and Escherichia coli. The method used is experimentally in the laboratory to test the antibacterial activity of endophytic fungi isolates derived from cat's whiskers (Orthosiphon aristatus) on Escherichia coli and Staphylococcus aureus bacteria using the hole / well method. In the research results obtained four samples of endophytic fungi extract isolated from the leaves and stems of the cat's whiskers plant (Orthosiphon aristatus) . The results of antibacterial research showed that endophytic mushroom extract isolated from cat's whiskers were more effective in inhibiting Escherichia coli bacteria compared to Staphylococcus aureus. In Escherichia coli bacteria the average value of inhibition is 5 mm whereas in Staphylococcus aureus bacteria the average value of inhibition is only 2 mm. So it can be seen that the endophytic fungus from the leaves of cat's whiskers has a better antibacterial effect than the endophytic fungus from the stems of the cat's whiskets. Conclusion, etidophytic fungi isolated from leaves and stems of cat's whiskers (Orthosiphon aristatus) have an antibacterial effect on the growth of E. coli bacteria and are not effective against S. aureus bacteria.Keywords: Cat's whiskers (Orthasiphan aristatus), antibacterial activity, Escherichia coli and Staphylococcus aureus. ABSTRAKJamur endofit ialah jamur yang hidup di dalam jaringan tumbuhan dan tidak membahayakan tumbuhan tersebut. Penelitian ini bertujuan untuk menguji aktivitas antibakteri jamur endofit yang diisolasi dari daun dan batang tumbuhan kumis kucing (Orthosiphon aristatus) terhadap Bakteri Escherichia coli dan Staphylococcus aureus. Kumis kucing mengandung minyak atsiri yang digunakan sebagai antibakteri terhadap beberapa bakteri patogen seperti Staphylococcus aureus dan Escherichia coli. Metode yang dipakai yakni secara eksperimen di laboratorium untuk menguji adanya aktivitas antibakteri dari isolat jamur endofit  yang diberasal dari tumbuhan kumis kucing (Orthosiphon aristatus)  pada bakteri  Escherichia coli dan Staphylococcus aureus dengan menggunakan metode lubang/sumuran. Pada hasil penelitan didapatkan empat sampel ekstrak jamur endofit yang diisolasi dari daun dan batang  tumbuhan kumis kucing (Orthosiphon aristatus). Hasil penelitian antibakteri menunjukkan bahwa ekstrak jamur endofit yang diisolasi dari tumbuhan kumis kucing lebih efektif menghambat  bakteri Escherichia coli dibandingkan dengan bakteri Staphylococcus aureus. Pada bakteri  Escherichia coli nilai rata-rata daya hambat yaitu 5 mm sedangkan pada bakteri Staphylococcus aureus nilai rata-rata daya hambat yaitu hanya 2 mm. Sehingga dapat diketahui bahwa jamur endofit dari daun kumis kucing memiliki efek antibakteri yang lebih baik dibandingkan dengan jamur endofit dari batang kumis kucing. Kesimpulan, Jamur endofit yang diisolasi dari daun  dan batang kumis kucing (Orthosiphon aristatus) memiliki efek antibakteri terhadap pertumbuhan bakteri E. coli dan tidak efektiv terhadap bakteri S. aureus.Kata Kunci:  Tumbuhan kumis kucing (Orthosiphon aristatus), aktivitas antibakteri, Escherichia coli dan Staphylococcu saureus.

The study involved the selection of two isolates from Bacillus subtilis to investigate their inhibitory activity against some bacterial pathogens. B sub-bacteria were found to have a broad spectrum against test bacteria such as Staphylococcus aureus and Pseudomonas aeruginosa. They were about 23-30 mm and less against Klebsiella sp. The sensitivity of some antibodies was tested on the test samples. The results showed that the inhibitory ability of bacterial growth in the test samples using B. subtilis extract was more effective than the antibiotics used.

<p><em>Metil merah merupakan salah satu zat warna golongan azo yang sering digunakan dalam industri dan laboratorium. Penggunaan metil merah dapat menimbulkan efek terhadap kesehatan dan lingkungan. Oleh sebab itu dilakukan metode fotodegradasi dengan menggunakan semikonduktor dan radiasi sinar tampak. Semikonduktor yang digunakan yaitu berbahan dasar tembaga sulfat hidrat dan perak nitrat. Prekusor tembaga sulfat hidrat dibuat dari pengolahan limbah logam tembaga hasil pemotongan tembaga yang ada di bengkel Lembaga Ilmu Pengetahuan Indonesia (LIPI) Bandung. Bahan semikonduktor juga memiliki kemampuan dalam menghambat pertumbuhan bakteri. Hasil optimum yang didapatkan dalam proses fotodegradasi dan antibakteri merupakan gabungan antara kedua prekusor tembaga sulfat hidrat dan perak nitrat dengan bantuan penyinaran. Kemampuan dalam menghambat pertumbuhan bakteri didapatkan persentase kematian 100 % untuk masing-masing bakteri, yaitu Escherichia coli dan Staphylococcus aureus. Aktifitas fotokatalitiknya dengan konsentrasi semikonduktor 10 ppm untuk mendegradasi zat warna metil merah 5 ppm, selama 23 jam, dimana persentase degradasi yang didapatkan dengan penyinaran lebih tinggi dibandingkan dengan tanpa penyinaran. Pengaruh pH larutan terhadap degradasi metil merah yaitu optimum pada pH 12 (basa).</em></p><p><em><br /></em></p><p><em>Methyl red is one of the azo group dyes that is often used in industry and laboratories. The use of methyl red can have an effect on health and the environment. Therefore photodegradation method is done by using semiconductor and visible light radiation. The semiconductor used is based on copper sulfate hydrate and silver nitrate. The copper sulphate hydrate precursor is made from the processing of copper-cut copper metal waste in the workshop of the Indonesian Institute of Sciences (LIPI) in Bandung. Semiconductor materials also have the ability to inhibit bacterial growth. The optimum results obtained in the photodegradation and antibacterial process are a combination of both copper sulfate hydrate precursor and silver nitrate with the help of irradiation. The ability to inhibit bacterial growth obtained 100% mortality for each bacterium, namely Escherichia coli and Staphylococcus aureus. Photocatalytic activity with 10 ppm semiconductor concentration to degrade methyl red dye 5 ppm, for 23 hours, where the percentage of degradation obtained by irradiation is higher than without irradiation. The effect of pH of the solution on the degradation of methyl red is optimum at pH 12 (base).</em></p>

Sulfaquinoxaline (SQX) has been detected in environmental water samples, where its side effects are still unknown. To the best of our knowledge, its oxidation by Fenton and photo-Fenton processes has not been previously reported. In this study, SQX oxidation, mineralization, and toxicity (Escherichia coli and Staphylococcus aureus bacteria) were evaluated at two different setups: laboratory bench (2 L) and pilot plant (15 L). The experimental design was used to assess the influence of the presence or absence of radiation source, as well as different H2O2 concentrations (94.1 to 261.9 mg L−1). The experimental conditions of both setups were: SQX = 25 mg L−1, Fe(II) = 10 mg L−1, pH 2.8 ± 0.1. Fenton and photo-Fenton were suitable for SQX oxidation and experiments resulted in higher SQX mineralization than reported in the literature. For both setups, the best process was the photo-Fenton (178.0 mg L−1 H2O2), for which over 90% of SQX was removed, over 50% mineralization, and bacterial growth inhibition less than 13%. In both set-ups, the presence or absence of radiation was equally important for sulfaquinoxaline oxidation; however, the degradation rates at the pilot plant were between two to four times higher than the obtained at the laboratory bench.
