ABSTRACTThis study aims to determine the antioxidant activity and free radical antidote from fraction of bark sago baruk (Arenga microcarpha Beccari). This study initiated by extracting the powder of bark sago baruk using the maceration method for 3 days with ethanol 80%. The extract then partitioned using a series of solvent such as petroleum ether, ethyl acetate, buthanol, aquadest. The results showed that the ethyl acetate fraction had the highest free radical antidote content followed by the aquadest fraction, buthanol fraction, petroleum ether fraction. The content of free radical antidote respectively was 86,25%; 66,30%; 65,32%; 43,43%. Based on this study, the ethyl acetate fraction was the best fraction can act as an antidote to free radicals better than other fractions. Keywords: Bark sago baruk, fraction, free radical antidote ABSTRAKPenelitian ini bertujuan untuk menentukan Aktivitas penangkal radikal bebas dari fraksi kulit kayu sagu baruk (Arenga microcarpha beccari). Penelitian ini dimulai dengan mengekstraksi serbuk kulit kayu sagu baruk menggunakan cara maserasi selama 3 hari dengan pelarut etanol 80%. Ekstrak kemudian dipartisi menggunakan pelarut petroleum eter, etil asetat, butanol, dan aquades. kemudian ditentukan aktivitas antioksidan dan penangkal radikal bebas. Hasilnya menunujukan bahwa fraksi etil asetat memiliki kandungan penangkal radikal bebas tertinggi diikuti fraksi aquades, fraksi butanol, dan fraksi petroleum eter. Kandungan penangkal radikal bebas berturut-turut adalah 86,25%; 66,30%; 65,32%; 43,43%. Berdasarkan penelitian ini, dapat disimpulkan bahwa fraksi etil asetat dapat berperan sebagai penangkal radikal bebas lebih baik dibandingkan dengan fraksi lainnya. Kata Kunci:  Kulit Kayu Sagu Baruk, Fraksi, Penangkal Radikal Bebas

The antibacterial activity of methanol extract from the root bark of Akanda (Calotropis gigantea L.) and its petroleum ether, chloroform and ethyl acetate fractions were investigated. Both of methanol extract and its chloroform fraction showed activity against Sarcina lutea, Bacillus megaterium and Pseudomonas aeruginosa. Petroleum ether fraction showed activity against Bacillus subtilis and Shigella sonnei whereas ethyl acetate fraction showed activity against Pseudomonas aeruginosa and Escherichia coli at 20ìg/disc, 30ìg/disc and 40ìg/disc doses. Among the tested materials, methanol extract and its chloroform fraction showed comparatively better results. Minimum inhibitory concentration (MIC) for methanol extract and each fraction were also determined by serial dilution technique. The characteristics of anti-fungal activity to Alternaria alternate of three fractions of water, ethyl acetate, petroleum ether of sunflower discs and stalks have been studied. The fractions of water, ethyl acetate, petroleum ether of sunflower disc and petroleum ether fraction of sunflower stalk have good anti-fungal activity to A. alternate. The best concentrations of anti-fungal activity to A. alternate of water, ethyl acetate, petroleum ether fractions and petroleum ether fraction of sunflower stalk were 320 mg/mL, 240 mg/mL, 160 mg/mL and 160 mg/mL, respectively. The anti-fungal activities to A. alternate of these four kinds of extract are increased with the increase in concentration and arrive at stability after the best concentration of anti-fungal activity to A. alternate. The ethyl acetate fraction and petroleum ether fraction of sunflower stalk had no anti-fungal activity to A. alternate.

Free radicals are molecules containing unpaired electrons so that they are not stable and very reactive to other molecules. ROS/RNS radicals have physiological function, but the overproduction of free radicals can initiate oxidative/nitrosative stress that contributes to a high number of diseases. Body has an ability to neutralize the free radicals by forming the endogenous antioxidant. Environmental changes, living style, certain pathological conditions can cause the shift of prooxidant-antioxidant equilibrium. Thus, endogenous antioxidant intake is needed, particularly that originating from natural materials. One of the plants believed to have antioxidant activity is sesewanua (Clerodendrum fragrans [Vent.] Willd.) leaf. This study was aimed to evaluate the antioxidant activity of ethanol extract, hexane fraction, ethyl acetate fraction, and water fraction of the sesewanua leaf using DPPH and nitrate-oxide free radical scavenging method. The study is a laboratory experiment. The sample was sesewanua (Clerodendrum fragrans) obtained from East Malalayang I village, Malalayang district, Manado city, North Sulawesi. The antioxidant activity testing utilized 1,1-Diphenyl-2-Picrylhydrazyl (DPPH) and nitrate-oxide free radical scavenging method. Data included percent inhibition of free radicals and were analyzed using linear regression to determine 50% inhibition concentration (IC50) of DPPH and nitrate-oxide free radicals. As conclusion, the ethanol extract, hexane fraction, ethyl acetate fraction, and water fraction of the sesewanua leaf had antioxidant activity through DPPH free antiradical activity, but not active as antiradical NO.

Antioxidants are compounds that can inhibit oxidation reactions by binding to free radicals and highly reactive molecules. One form of reactive oxygen compounds is free radicals, these compounds are formed in the body and are triggered by various factors. Noni fruits have been used by the community as a medicine for high blood pressure, beriberi, urinating, inflammation of bile, inflammation of the intestine, dysentery, constipation, spleen pain, liver pain, diabetes and lumbago. Nevertheless, research on the identification of the active compounds of noni fruit which act as antioxidants yet. This study aims to extract and fractionation of Noni fruit, determine the antioxidant activity of Noni fruit extract, and identify the types of bioactive compounds that act as antioxidants. Noni fruit was extracted used maceration technique with 96% ethanol and then fractionated using n-hexane, chloroform and ethyl acetate solvents. Antioxidant activity from the results of noni fruit fractionation was measured by the DPPH (1.1 diphenyl-2-picrylhydrachyl) method and identification of bioactive compounds was carried out by GCMS. The results showed the greatest antioxidant activity was found in chloroform fraction with a percentage of inhibition of 78.19%. For the first time this study revealed the types of bioactive compounds from noni fruit ethyl acetate fraction which act as antioxidants are n-hexadecanoic acid, squalene, pyridin-3-carboxamide, oxime, n- (2-trifluoro methyl phenyl), and beta-sitosterol

Abstract Background Bergenia ciliata is a medicinal plant used for the treatment of diarrhea, vomiting, fever, cough, diabetes, cancer, pulmonary disorders and wound healing. Methods In this study, Bergenia ciliata crude extract, subfractions, and isolated compounds were evaluated for their antioxidant and anticholinesterase potential. The free radical scavenging capacities of the extracts determined using DPPH and ABTS assays. The anticholinesterase potentials were determined using acetylcholine esterase and butyryl choline esterase enzymes. To determine the phytochemical composition, the extracts were subjected to HPLC analysis and silica gel column isolation. Based on HPLC fingerprinting results, the ethyl acetate fraction was found to have more bioactive compounds and was therefore subjected to silica gel column isolation. As a result, three compounds; pyrogallol, rutin, and morin were isolated in the pure state. The structures of the isolated compounds were elucidated using spectroscopic techniques like 1H-NMR, IR and UV-Visible. Results The crude extract showed maximum anticholinesterase (acetylcholinesterase = 90.22 ± 1.15% and butyrylcholinesterase = 88.22 ± 0.71%) and free radical scavenging (87.37 ± 2.45 and 83.50 ± 0.70% respectively against DPPH and ABTS radicals) potentials. The total phenolic contents (expressed as equivalent of gallic acid; mgGAE/g) were higher in ethyl acetate fraction (80.96 ± 1.74) followed by crude extract (70.65 ± 0.86) while the flavonoid contents (expressed as quercetin equivalent; mgQE/g) and were higher in crude extract (88.40 ± 1.12) followed by n-butanol fraction (60.10 ± 1.86). The isolated bioactive compounds pyrogallol, rutin, and morin were found active against ABTS and DPPH free radicals. Amongst them, pyrogallol was more active against both free radicals. Reasonable anticholinesterase activities were recorded for pyrogallol against selected enzymes. Conclusion The extracts and isolated compounds showed antioxidant and acetylcholinesterase inhibitory potentials. It was concluded that this plant could be helpful in the treatment of oxidative stress and neurological disorders if used in the form of extracts.

Column chromatography of the dichloromethane fraction from an aqueous methanolic extract of fruit peel of Citrus pyriformis Hassk. (Rutaceae) resulted in the isolation of seven compounds including one coumarin (citropten), two limonoids (limonin and deacetylnomilin), and four sterols (stigmasterol, ergosterol, sitosteryl-3-β-D-glucoside, and sitosteryl-6ʹ- O-acyl-3-β-D-glucoside). From the ethyl acetate fraction naringin, hesperidin, and neohesperidin were isolated. The dichloromethane extract of the defatted seeds contained three additional compounds, nomilin, ichangin, and cholesterol. The isolated compounds were identified by MS (EI, CI, and ESI), 1H, 13C, and 2D-NMR spectral data. The limonoids were determined qualitatively by LC-ESI/MS resulting in the identification of 11 limonoid aglycones. The total methanolic extract of the peel and the petroleum ether, dichloromethane, and ethyl acetate fractions were screened for their antioxidant and anti-inflammatory activities. The ethyl acetate fraction exhibited a significant scavenging activity for DPPH· free radicals (IC50 = 132.3 μg/mL). The petroleum ether fraction inhibited 5-lipoxygenase with IC50 = 30.6 μg/mL indicating potential anti-inflammatory properties. Limonin has a potent cytotoxic effect against COS7 cells [IC50 = (35.0 ± 6.1) μM] compared with acteoside as a positive control [IC50 = (144.5 ± 10.96) μM]

AbstrakTelah dilakukan penelitian uji aktivitas senyawa antioksidan dari ekstrak metanol kulit pisang raja (Musa paradisiaca Sapientum). Ekstraksi dilakukan dengan cara maserasi menggunakan pelarut metanol. Ekstraksi bertingkat dilakukan terhadap ekstrak metanol dengan n-heksan dan etil asetat. Dari uji fitokimia terhadap fraksi metanol menunjukkan positif flavonoid. Pengujian aktifitas antioksidan dilakukan dengan reaksi oksidasi asam linoleat dengan metoda feritiosianat (FTC) 0,05%. Sebagai standar antioksidan digunakan Butil hidroksianisol (BHA). Hasil penelitian ini menunjukkan ekstrak metanol, fraksi n-heksan dan etil asetat memiliki aktifitas antioksidan yang tidak jauh berbeda dengan BHA. The antioxidant activities were tested by using ferric thiocyanate method 0,05% (FTC) on linoleic acid and buthyl hydroxyanisole (BHA) as antioxidant standard. Metanol extract, n-hexan and ethyl acetate fraction exhibited antioxidative activity that was not significantly different from BHA, on the other hand, the ethyl acetate fraction exhibited significant antioxidative activity, which is better than BHA.Keywords : antioxidant, ferric thiocyanate, Musa paradisiaca Sapientum

Sargassum sp. adalah salah satu jenis rumput laut paling banyak di Indonesia dan memiliki manfaat sebagai antioksidan, obat penyakit jantung, stroke, dll. Penelitian ini bertujuan untuk menentukan kandungan antioksidan fraksi etil asetat dari ekstrak metanol Sargassum sp. Sampel diambil dari Pantai Sundak, Gunung Kidul, Yogyakarta. Metode yang digunakan adalah eksploratif diskriptif. Sampel dikeringkan dalam suhu ruangan selama 7 hari dan dimaserasi dengan pelarut metanol, lalu dievaporasi dengan rotary evaporator. Ekstrak metanol di fraksinasi menggunakan etil asetat. Pengujian aktivitas antioksidan dilakukan dengan menggunakan 2 metode, yaitu metode penangkapan radikal bebas DPPH dan total kapasitas antioksidan fosfomolibdat. Vitamin C digunakan digunakan sebagai standar penangkapan radikal bebas DPPH dan total kapasitas antioksidan fosfomolibdat. Kadar total fenolat diuji menggunakan larutan Folin-Ciocalteu dengan standar asam galat. Hasil penelitian menunjukkan bahwa kadar total fenolat fraksi etil asetat Sargassum sp. sebesar 64,42 mg GAE/g sampel. Aktivitas antioksidan dengan metode penangkapan radikal bebas DPPH memiliki nilai IC50 1.289 ppm, vitamin C memiliki IC50 sebesar 122,71 ppm, sedangkan total kapasitas antioksidan adalah 39,52 mg AAE/g sampel. Kesimpulannya, yaitu kandungan aktivitas antioksidan fraksi etil asetat dari ekstrak metanol Sargassum sp. dengan metode penangkapan radikal bebas DPPH diduga sangat lemah dan kandungan aktivitas antioksidan dengan menggunakan metode fosfomolibdat diduga tinggi.  Sargassum sp. is one of the many types of seaweed in Indonesia and has benefits as an antioxidant, a drug for heart disease, stroke, etc. This study aims to determine the antioxidant content of ethyl acetate fraction of methanol extract of Sargassum sp. Samples were taken from Sundak, Gunung Kidul, Yogyakarta. The method used is descriptive explorative. Samples were dried at room temperature for 7 days and macerated with methanol, then evaporated at a rotary evaporator. The methanol extract fractionation using ethyl acetate. Testing the antioxidant activity using two methods, the method of catching free radicals DPPH and total antioxidant capacity fosfomolibdat. Vitamin C is used as a standard arrest DPPH free radical and total antioxidant capacity fosfomolibdat. Levels of total phenolics were tested using the Folin-Ciocalteu solution with gallic acid standard. The results showed that levels of total phenolic fraction of ethyl acetate Sargassum sp. amounting to 64,42 mg GAE/g sample. The antioxidant activity with catching free radicals DPPH methods have IC50 values 1.289 ppm, vitamin C has an IC50 of 122,71 ppm, while the total antioxidant capacity was 39,52 mg AAE/g sample. In conclusion, the content of the antioxidant activity of ethyl acetate fraction of methanol extract of Sargassum sp. with catching free radicals DPPH methods allegedly extremely weak and the content of the antioxidant activity using methods fosfomolibdat predictably high.

ABSTRAKPala adalah tanaman yang termasuk dalam jenis tanaman obat. Penelitian ini bertujuan untuk menganalisis total kandungan fitokimia dan uji efek sedatif  dari ekstrak etanol, fraksi petroleum eter, etil asetat dan air daging buah pala. Ekstraksi dilakukan dengan cara maserasi, kemudian ekstrak etanol yang diperoleh difraksinasi dengan cara partisi. Analisis fitokimia dilakukan dengan metode spektrofotometri UV-Vis sehingga diperoleh hasil kandungan total fenolik ekstrak etanol, fraksi petroleum eter, etil asetat dan air daging buah pala secara berturut-turut adalah 26,284 μg/mL; 29,203 μg/mL; 28,725 μg/mL; 24,904 μg/mL. Total flavonoid secara berturut-turut adalah 9,999 μg/mL; 13,095 μg/mL; 13,729 μg/mL; 10,475 μg/mL. Total tanin secara berturut-turut adalah 16,642 μg/mL; 16,571 μg/mL; 16,357 μg/mL; 10,428 μg/mL. Uji efek sedatif dilakukan dengan metode traction test  dan diperoleh hasil ekstrak etanol dan ketiga fraksi memiliki pengaruh efek sedatif pada dosis 300 mg/kgBB dan 500 mg/kgBB dan dosis yang paling baik dalam memberikan pengaruh efek sedatif adalah 500 mg/kgBB. ABSTRACT Nutmeg is a plant that is included in a type of medicinal plant. This study aimed to analyze the total phytochemical content and test the sedative effect of ethanol extract, petroleum ether fraction, ethyl acetate and nutmeg pulp. Extraction was carried out by maceration, then the ethanol extract obtained was fractionated by partitioning. Phytochemical analysis was carried out using UV-Vis spectrophotometry method so that the total phenolic content of ethanol extract, petroleum ether fraction, ethyl acetate and nutmeg water was obtained, respectively, 26.284 μg/mL; 29,203 μg/mL; 28.725 μg/mL; 24,904 μg/mL. The total flavonoids were 9.999 μg/mL, respectively; 13.095 μg/mL; 13,729 μg/mL; 10,475 μg/mL. The total tannins were 16,642 μg/mL, respectively; 16,571 μg/mL; 16,357 μg/mL; 10,428 μg/mL. The sedative effect test was carried out by the traction test method and the results of the ethanol extract and the three fractions had a sedative effect at a dose of 300 mg/kgBB and 500 mg/kgBB and the best dose in giving a sedative effect was 500 mg/kgBB.

Baccaurea racemosa (Reinw. ex Blume) Müll. Arg. is a fruit widely grown in Indonesia locally known as ‘kepundung’. Some of genus Baccaurea has potential to be developed as a natural antioxidant. However, the antioxidant activity of B. racemosa pulp has not been published. This study is aimed to (i) explore the antioxidant activity, (ii) measure the total phenolic and flavonoid contents, and (iii) investigate the correlation of antioxidant activity of extract and fractions of B. racemosa pulp with their total phenolic and flavonoid contents. Fresh B. racemosa pulp was macerated with methanol to obtained methanolic extract. The methanolic extract was partitioned using n-hexane followed dichloromethane and ethyl acetate. This study revealed that ethyl acetate fraction has the highest antioxidant activity with the scavenging activity of free radical 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2’-Azinobis(3-etylbenzthiazoline)-6-sulfonic acid (ABTS) with values IC50 127.155±4.311ug/mL and 108.155±6.455 mM trolox equivalent/100 mg dry extract respectively. However, the highest antioxidant activity using β-carotene bleaching (BCB) assay is the methanolic extract with value 36.898±13.240%. Furthermore, the ethyl acetate fraction has the highest total phenolic content (TPC) and total flavonoid content (TFC) with values 42.975±1.978 mg gallic acid equivalent/g dry extract (mg GAE g-1 dry extract) and 122.813±1.604 mg rutin equivalent/g dry extract (mg RE g-1 dry extract) respectively. TPC and TFC of B. racemosa pulp have a significant correlation with ABTS free radical scavenging activity. In this current study, it was discovered that ethyl acetate fraction of B. racemosa pulp can be developed to be a natural antioxidant.
