The activity and content of aspartate aminotransferase (AST) isoforms in bovine ejaculates due to sperm survival were studied. Ejaculates of bulls of the Ukrainian black-spotted dairy breed (n = 22) were selected for research. In freshly obtained and incubated semen the activity and isozymes of AST was studied in connection with spermatozoa survival at a temperature of 2-4 ° C (on the first, second, third and fourth days) until the cessation of rectilinear translational movement. AST activity depended on the duration of sperm survival. When survival was more than 100 hours - AST activity was the highest - 65.2 ± 1.7 nmol / min × mg of protein. When survival was lower - up to 100 hours, enzymatic activity lower by 26.8% (P <0.001). Two enzymes of the enzyme (AST1 and AST2) were found in the semen of the fetuses, which differ in electrophoretic mobility and intensity of staining in 7.5% polyacrylamide gel. The established correlation with sperm survival time has a strong straight line for AST1 (η2АSТ1 = 0.88) and inverse - for AST2 (η2АSТ2 = 0.87) isozymes. During sperm incubation, the ratio of AST isozymes changes - the content of AST1 increases and decreases - AST2. The correlation ratio for sperm survival for enzyme activity and isozymes is up to 100 hours, respectively. - η2АSТ = 0.83; η2АSТ1 = 0.68 and η2АSТ2= 0.69 and more than 100 hours - η2АSТ = 0.75; η2АSТ1= 0.92 and η2АSТ2= 0.69. Therefore, ejaculates of bulls with reduced sperm survival are characterized by lower AST activity and, accordingly, the speed of the amino acid transamination process. Increased supply of substrates from the cytosol in the mitochondria of germ cells ensures high survival of sperm. Changes in the activity and content of AST isozymes, which characterize the energy supply of germ cells, can serve as a criterion for the physiological quality of sperm of freshly obtained sperm.

Desiccation tolerance in seeds depends on the species, development stage and drying conditions, especially the water removal rate. Coffea seeds are considered of intermediate performance, because they tolerate relative dehydration compared to orthodox seeds and are sensitive to low temperatures. The objective of this study was to verify the effect of different drying rates on the viability and storability of Coffea canephora seeds. A complete randomized experimental design was used, in a factorial 3 x 5 x 2 design, with three drying rates (fast, intermediate and slow), five final mean water contents after drying (51, 42, 33, 22 and 15 %) and two storage temperatures (10 and 20°C). The germination and seed vigor assessments, using radicle protrusion, cotyledon leaf opening, seedling emergence and emergence speed index, were performed shortly after drying and after two and four months storage. It was observed that with reduction in the water content there was reduction in the germination values and seed vigor, for all the drying rates. The greatest reductions in physiological quality occurred when the seeds were dried quickly and the best results were obtained at the intermediate drying rate. There was an effect of drying rate and storage temperature on the physiological quality of the seeds, and lower germination and vigor values were observed in seeds with lower water content stored at 20°C. C. canephora seeds were tolerant to desiccation down to 15 % water content and can be stored for four months at 10°C. A temperature of 20ºC can be used to store C. canephora seeds, as long as the water content is not reduced to values below 22 % water content.

The aim of this work was to correlate macronutrient content of soybean seeds to physiological quality of different cultivars. The work was developed in the Federal University of Pelotas, in the facilities of the Seed Science and Technology Graduate Program. The experimental design was randomized blocks in arranged in four replicates. The following soybean cultivars were used: BMX Apolo RR (12 lots), BMX Ativa RR (13 lots), BMX Energia RR (26 lots), BMX Força RR (24 lots), BMX Impacto RR (35 lots), BMX Magna RR (16 lots), BMX Turbo RR (44 lots), BMX Potência RR (82 lots) and NA 5909 RR (28 lots), with seeds produced in northwest Rio Grande do Sul. The determination of nutritional contents found in plant tissues of soybean seeds, were measured: Nitrogen (N), Phosphorus (P), Potassium (K), Magnesium (Mg), Calcium (Ca), Sulfur (S). The macronutrient contents of the seeds vary according to genetic characteristics of the cultivars, with higher oscillations of nitrogen, phosphorus, magnesium and sulfur contents present in soybean seeds. Potassium and calcium are defined as the most stable nutrients for the cultivars and seed lots analyzed. High concentrations of nitrogen, phosphorus and calcium are determinants for the physiological quality of soybean seeds.

The physiological quality is based on the genotype and may be accompanied since the first stages of selection through the study of genetic diversity. The purpose of this study was to evaluate the parental genetic contribution in the seed physiological quality in corn intervarietal crossbreeds. The intervarietal crossbreeds were performed using landraces genotypes, open pollinated varieties and commercial hybrids. The seeds obtained from the crossbreed, their equivalents and parentals were evaluated regarding their water percentage, germination, germination speed rate, accelerated aging, field emergence and electrical conductivity. The seeds obtained in the crossbreed are viable and vivid with heterosis when compared with the parentals. The accelerated aging test was more effective for selecting the strongest genotypes. The canonical variables were useful to group the genotypes that presented similar physiological characteristics. The genotypes MPA 01 x Pixurum 05, MPA 01 x AS1565, MPA 01 x SJC5886, SJC5886 x Pixurum 05 and Fundacep 35 x SCS 154 Fortuna were the best crossbreeds regarding the production of better physiological quality seeds.

Seed treatment is a practice that helps the initial establishment of the crop without the effects caused by pests and diseases. The association of diazotrophic bacteria with grasses has been used in the supply of nitrogen to plants; however, these microorganisms produce growth-promoting substances, which promote benefits in the growth and development of the crops. Thus, the objective of this study was to evaluate the compatibility of Azospirillum brasilense associated with the fungicide difenoconazole and the insecticide thiamethoxam by observing the effects on the quality of seed emergence of three wheat cultivars (Triticum aestivum L.). Three wheat cultivars, arranged in a 4 x 2 factorial system with four replicates, were tested. The treatments were the control; difenoconazole (Spectro® at a dose of 150 mL per 100 kg of seed); thiamethoxam (Cruiser® FS 350 at a dose of 200 mL per 100 kg seed) and difenoconazole + thiamethoxam, applied on wheat cultivars TBIO Mestre, TBIO Itaipu and TBIO Sinuelo. Bacteria from the Azospirillum genus were used in the inoculation. The wheat seed retains its quality when it checks the germination, vigor and independent accelerated aging, whether or not fungicide, insecticide and A. brasilense were used. The insecticide thiamethoxam increased the length of shoots and roots and provided compatibility with A. brasilense, and fungicide inhibited the length of shoots and roots and was antagonistic to the bacterium A. brasilense.

Studies of seed physiological processes represent the starting point for the sustainable utilization of native or cultivated plant species from the caatinga biome for which germination studies are still scarce. In order to determine adequate methods for the conservation of Gliricidia sepium seeds, the physiological quality of propagules exposed to different conditions and times of storage were examined. Recently collected seeds of G. sepium were analyzed immediately, while the other sample was stored for three, six, nine, or twelve months in plastic or paper bags under refrigeration or at room temperature for further analysis. The germination rates of seeds from each storage regime were determined and these rates compared to their water content. It was observed that germination capacity was directly related to changes in seed humidity. Storing seeds in plastic bags yielded the best conservation, mainly when they were stored at low temperatures. However, even at temperatures near 25°C it was possible to prolong seed viability when their water content was controlled through the use of impermeable packing.

Differences in levels of lignin in the plant between conventional and transgenic cultivars RR has been reported by several authors, however, there are few studies evaluating the influence of spraying of glyphosate on the lignin in the plant and RR soybean seeds. The aim of this study was to evaluate the physiological quality of RR transgenic soybean seeds and the lignin contents of plants sprayed with the herbicide glyphosate. The assays were conducted both in greenhouse and field in the municipality of Lavras, MG, in the agricultural year 2007/08. The experiment was arranged in a splitplot design with four replicates, considering the treatments hand weeding and herbicide glyphosate as plots, and five RR soybean cultivars (BRS 245 RR, BRS 247 RR, Valiosa RR, Silvânia RR and Baliza RR) as splitplots. In the greenhouse, the cultivars tested were BRS 245 RR and Valiosa RR in a randomized block design with four replicates. The sprayings were carried out at stages V3, V7 and early R5 (3L/ha). The 1000 seed weight, mechanical injury, germination and germination velocity index, emergence velocity index, accelerated aging, electrical conductivity and water soaking seed test, lignin content in the seed coat, in the stem and legumes were determined. The spraying of glyphosate herbicide, in greenhouse and field, did not alter the physiological quality of seeds and the lignin contents in the plant.

ABSTRACT: Processing of particles in a moving bed, such as a fluidized bed or a spouting bed, is commonly used in the operations of drying, coating, and granulation of particulate systems. This process has applications in the chemical, pharmaceutical and, presently, agronomical industries, especially for seed treatment/coating. This research aimed to analyze the fluid-dynamic behavior of fluidized and spouting beds with different air temperatures and loads of flaxseeds (Linum usitatissimum L.), with estimates of the fluid-dynamic parameters correlated to each process. The parameters were compared with the values obtained from classical correlations in the literature, with indications of associated percentages of deviation. Influence of fluid dynamics on the physiological quality of seeds was assessed by germination tests and the germination speed index. An analysis of the results indicated that seed processing was adequate for processing in dynamically active beds; however, temperatures above 50ºC in both beds caused significant reductions in the physiological quality of the seeds. Processing in a fluidized bed presented a smaller reduction of the physiological properties of the flaxseed.
