Background and Aims The diabetic animal model was developed as a type for studies in vivo because of the death rates in many countries associated with the prevalence of diabetes, this disease is a respecter of no person. This study assessed and compared total protein and albumin concentration, basal fasting blood sugar and post induction fasting blood sugar levels in rat model. Methods Qualitative and Quantitative test and analysis were carried to assess the levels of albumin and total protein and their absorbance levels were read and recorded in g/dL, while the fasting blood sugar was determined using the glucometer strip method at intervals and recorded in mg/dL. Group A served as the negative, non-diabetic control for the study and they were fed normally with water without inducing diabetes while Group B served as the positive control, diabetic untreated group where Diabetes mellitus was induced using 150 mg/kg body weight of alloxan and left untreated. Results Plasma total protein in diabetic Wistar rat (8.44 ±4.39 g/dl) did not change significantly (P>0.05) compared with the control Wistar rat (10.04 ± 4.14). However, the level of 1-week post-induction FBS in diabetic Wistar rat (475.75 ± 66.74 mg/dl) was significantly (P<0.05) higher compared to that of the control (85.0 ± 6.7 mg/dl). Conclusions Protein levels may not change within a week after developing diabetes mellitus and as such should not be the sole biomarker to test for diabetes.

Sinom drinks mixed with lime and honey are made from rhizome of turmeric and young tamarind leaves which are added with lime juice and honey. The aim of the study was to determine the effect of antioxidant compounds of ethyl acetate on sinom drinks mixed with lime juice and honey (SCJM) various doses on fasting blood sugar spraque dawley (SD) white rats diabetes mellitus. Experimental research using RAK, ANOVA analysis with further BNT testing consisted of two stages: 1) test the antioxidant capacity of various solvents of SCJM yielding 29691.1 µg AAEAC / g ingredients namely the highest number of ethyl acetate fractions of n- hexane, chloroform, and water. Phase 2 Ethyl acetate fraction of SCJM was further tested in vivo to SD white rats with diabetes mellitus with various doses, namely: control (-) normal mice, 50, 100, 150, 200 mg/dl BW, control (+) of diabetic rats. The results of the study of the effect of the antioxidant compound SCJM  ethyl acetate fraction of various doses significantly affected (p <0.01) in decreasing fasting blood sugar (GDP) white SD diabetes mellitus rats, a dose that can reduce the lowest GDP based on statistical tests is 150 mg/kg BW of rats, 168.4 mg/dl from other doses. Diabetes mellitus type 2 (T2DM) results from beta cell dysfunction or reduced action of insulin responsive. The objective of this study was to examine the relevance between blood sugar, the activity of aspartate aminotransferase (AST) and alanine aminotransferase (ALT) in fasting women diabetic patients in different durations. A total of sixty-eight women were divided into three groups: first a healthy group – non-diabetic (twenty-six women), second and third groups (twenty-one) were diabetic patients of age 35 – 50 and 51 – 69 years respectively. Serum fasting blood sugar was significantly (P < 0.05) elevated to 181.60 mg/dl in female patients with 35 – 50 years. The same effect happened in activity of AST to 32.91 u/L in 51 – 69 years and ALT was 28.43 u/L in 35 – 50 years. No significant differences were found between the aged and fasting blood sugar, AST and ALT in diabetic patients. The correlation factor (r) between fasting blood sugar and the activity of ALT was highly significant.

The present study was designed to explore whether exosomal lncRNA-KLF3-AS1 derived from human mesenchymal stem cells (hMSCs) can serve as a positive treatment for osteoarthritis (OA). hMSCs and MSC-derived exosomes (MSC-exo) were prepared for morphological observation and identification by transmission electron microscopy and flow cytometry. IL-1β-induced OA chondrocytes and collagenase-induced rat model of OA were established for the further experiments. Lentivirus-mediated siRNA targeting KLF3-AS1 was transfected into MSCs for silencing KLF3-AS1. The real-time quantitative PCR and western blotting analysis were performed to examine the mRNA and protein levels of type II collagen alpha 1 (Col2a1), aggrecan, matrix metalloproteinase 13 and runt-related transcription factor 2. Cell proliferation, apoptosis and migration were evaluated by CCK-8 assay, flow cytometry and transwell assay. HE (hematoxylin and eosin) staining and immunohistochemistry were used for histopathological studies. MSC-exo ameliorated IL-1β-induced cartilage injury. Furthermore, lncRNA KLF3-AS1 was markedly enriched in MSC-exo, and exosomal KLF3-AS1 suppressed IL-1β-induced apoptosis of chondrocytes. Further in vivo investigation indicated that exosomal KLF3-AS1 promoted cartilage repair in a rat model of OA. Exosomal KLF3-AS1 promoted cartilage repair and chondrocyte proliferation in a rat model of OA, which might be an underlying therapeutic target for OA.

The increasing prevalence of diabetes mellitus (DM) in Indonesia requires participation of all parties including pharmacists. This study aims to determine the impact of the implementation of home pharmacy service guidelines for patients with type 2 diabetes mellitus on the level of knowledge, medication adherence, therapeurical satisfaction and glycemic control. This study used a quasi-experimental design with one group pre-test and post-test design involving 37 patients with type 2 diabetes mellitus who met the requirements of inclusion and exclusion criterias. The Diabetes Knowledge Questionnaire (DKQ-24) and the Diabetes Medication Satisfaction Tool (DMSAT) were used for collecting the data. Patient adherence was measured using a pillcount method and fasting blood sugar levels (FSB) data were obtained from the laboratory results. The study was conducted in two community health centers and patient’s home in the Yogyakarta Municipality area. The data were analysed using Wilcoxon and Spearman test. The results showed the change of the level of knowledge, medication adherence, therapeutic satisfaction and glycemic control before and after intervention of pharmacy services at home. The average decreasing of fasting blood sugar levels 17.09 mg/dL ± 1.43

We have investigated twenty five patients with type-2 diabetes mellitus aged (35-60) years and fifteen healthy persons as control group to detect Anti-Helicobacter pylori IgG antibody. All studied groups were carried out to measure fasting blood sugar, anti- Glutamic acid decarboxylase (GAD), anti-? islets cells antibody by IFAT, Anti-H. pylori IgG antibody by ELISA technique. There was significant elevation in the concentration of fasting blood sugar than in control group (P < 0.05), the patients had negative results for anti-GAD antibody and anti- ? islets cells antibody, there were significant differences (P < 0.05) of anti-H. pylori IgG antibody in 28 % of patients had type-2 diabetes than control group. This lead to suggestion that type -2 diabetes mellitus patients are more susceptible to H. pylori .

Objective: To evaluate the hyperglycemic effects of Kursi Ziyabet (KZ) tablets on the rat models of diabetes mellitus. Methods: In total, 58 male SD rats were assigned randomly to six groups. All except the normal group were transformed into experimental diabetes mellitus rat models by injecting streptozocin. The hyperglycemic effect and the mechanism of Ziyabet were evaluated by body weight, food and water intake, fasting blood sugar, and related parameters by measuring the oxidative stress-related factors and lipid metabolism indicator level by the corresponding kits using the rat experimental models. Results: Compared with the model group, body weight markedly increased after 3–6 weeks of intragastric administration of Ziyabet tablets ([Formula: see text]), while the water intake significantly decreased in the same period of time ([Formula: see text]). Food intake and fasting blood sugar level also decreased with the high dosage of Ziyabet tablets ([Formula: see text]). There is no significant difference in pancreas’ MDA content of the Ziyabet groups when compared to the model group ([Formula: see text]), while significant increase in SOD level was observed in high-dosage KZ group ([Formula: see text]). The blood serum insulin and free fatty acid level also decreased in the high-dosage KZ group compared with the model group ([Formula: see text]). Conclusion: We conclude that Ziyabet tablets demonstrated protective effects on the diabetic rat models.

Aims: To investigate the effect of the methanolic extracts of Caryota no (CN) seeds negative geotaxis, fecundity and acetylcholinesterase (AChE) using Drosophila melanogaster (DM).                                                                                                      Study Design: Experimental design. Place and Duration: Sample: African Centre of Excellence for Phytomedicine Research and Development, University of Jos, Jos Plateau State Nigeria between June 2018 and February 2019                                                                                                  Methodology: 50 flies were exposed in each vial to the following concentrations: 300 mg, 350 mg, 400 mg, 500 mg and 600 mg of methanolic extracts in 5 replicates for 7 days with daily recording of mortality. Total protein assays were carried out by Randox method from the supernatant from homogenized whole flies. In vivo antioxidant activity study was conducted by measuring level of acetylcholinesterase (AChE) enzyme activity from supernatants of whole fly homogenates using a spectrophotometer at specific wavelengths over a 2 minute duration. The values were derived as part of the total protein value. Negative geotaxis was done by the climbing assay and fecundity was examined by rate of emergence of larva after exposure of the flies to treatment. The statistical difference among test groups was presumed at P < .05. Results: The methanolic extract of CN caused nonsignificant (P = .33) decrease in total protein levels compared to the control. There were also nonsignificant decreases in AChE (P = .30) activity, negative geotactic (P = .80) behaviour and nonsignificant increase in fecundity (P = .17) in the methanolic extract-treated flies compared to the controls. Conclusion: It can therefore be concluded that the methanolic extract of Caryota no nonsignificantly improved fertitity and reduced negative geotaxis and AChE activity in DM.

The screening of perfect diuretics for non –clinical utility in Diabetes mellitus is a relatively novel approach which gain insight into underlying the pathophysiological processes. This study aim to evaluate the diuretic effect of crude aqueous & alcoholic extract of M. charantia Linn. using Albino Wistar Rat model. The methodology of animal study includes the spectral analysis of Na+, K+, Cl-concentration against the body weight, this was done by spectrophotometry. In this study, the comparative observation of Diuretic activity with standard and extracted compound has shown that the estimated 24‐hour urine contains the Na+-3.82 g, 3.82 g, 3.92 g  and K+-1.35 g, 1.39 g, 1.48 gwt. For Vehicle control, Standard drug, and Extracted compound respectively. Which possess the favouring result means from the spot urine were 10.7±7.0 g/24 hand 3.9±2.1 g/24 h, respectively. Coefficients were 0.035, 0.022, 0.046 at (d±2SD=7.07 g, 4.42 gand 8.92 g) for sodium chloride and 0.068, 0.031, 0.046 at (d ± 2SD = 4.92 g, 2.31 g and 3.34 g) for potassium chloride. The present study guide formulation of non-clinical trials with statistical study to further measuring the claimed efficacy of M. charantia as a natural remedy for diabetes mellitus.
