The paper presents the results of an experimental study of the dose-dependent nature of functional changes in the body systems under chronic administration of uranyl acetate dihydrate in doses of 0.5 and 5.0 mg/kg per element for 18 weeks. The study was performed on 45 male outbred rats. It has been shown that uranyl acetate dihydrate in a dose of 0.5 mg/kg had no significant effect on hematological parameters. At the same time, activation of bactericidal activity of neutrophils, a decrease in the immunoregulatory index, and an increase in the blood concentration of tumor necrosis factor (TNF-α) have been revealed. The toxicant administered to rats in a dose of 5 mg/kg led to a decrease in the absolute number of erythrocytes, hemoglobin, hematocrit, platelets, the release of myelocytes into the blood, basophilia, monocytosis, the appearance of leukolysis cells and plasmatization of lymphocytes. On the part of the immune system, an increase in the biocidal capacity of neutrophilic granulocytes, TNF-α production, an increase in the number of CD8+ cells, and a reduction in the CD4+/CD8+ ratio have been found. Uranyl acetate dihydrate had a dose-dependent effect only on the number of cytotoxic T-lymphocytes, T-cells with the CD4+CD8+ phenotype, on the immunoregulatory index, and on the level of TNF-α. Hyperglycemia and glucosuria were also dose-dependent. An increase in glucose in the blood and urine indicated a violation of carbohydrate metabolism and kidney function. There was a decrease in the concentration of thyroxine, testosterone and an increase in the level of insulin. Uranyl acetate dihydrate led to the development of insulin resistance. The level of hormones did not depend on the dose of the toxicant administered to the animals.

Alopecia areata (АA) is an autoimmune multifactorial disease characterized by increased hair loss as a result of morphological and functional changes in hair follicles. АA is divided into four main forms, among which the most severe is the universal form (UA), in which complete hair loss is possible throughout the body. Alopecia in the practice of a rheumatologist can occur with some systemic diseases of the connective tissue, with the use of high doses of chemotherapy drugs and, more recently, with the use of inhibitors of tumor necrosis factors alpha (TNF-α). The article presents 3 clinical cases of the development of UA during therapy with TNF-α. Possible mechanisms are discussed, as well as the role of pro-inflammatory cytokines in the development of this condition.

Abstract Objective To evaluate the quantity and functional changes of subsets of Th cells in the bone marrow of severe aplastic anemia (SAA) patients and the relationship between these changes and the patient hematopoietic function. Methods By FACS, the quantity and ratio of Th1 cells and Th2 cells, the percentage of CD3+CD8+ cells in the bone marrow were detected in 24 patients with SAA at active phase, 15 patients with SAA at recovery phase, and 16 normal controls respectively. By radioimmunoassay, the levels of cytokines of Th1 type (TNF-α), or Th2 type (IL-4) in the serum of 20 patients with SAA at active phase, 12 patients with SAA at recovery phase, 16 normal controls were measured respectively. The relationships between CD3+CD8+ cells, TNF-α and Ret, ANC were evaluated; the relationships between Th1 cells and CD3+CD8+ cells, TNF-α or Ret, ANC were evaluated; the relationships between IL-4, balance of Th1/Th2 and Ret, ANC were also evaluated. Results The percent of Th1 cells, Th2 cells, and ratio of Th1/Th2 in bone marrow of patients with SAA at active phase was: 4.87±2.64%, 0.41±0.26%, 21.22±5.07, were higher than those of normal controls: 0.42±0.30% (p<0.01), 0.24±0.17% (p<0.05), 1.57±0.93(p<0.01) respectively, and all of these decreased to normal levels at recovery phase (p>0.05); the percent of CD3+CD8+ cells decreased from 32.32±18.69% at active phase to 13.76±2.96% at recovery phase significantly (p<0.01); the serum levels of TNF-α, IL-4 at active phase was 4.29±3.15ng/ml and 1.24±0.73 ng/ml respectively, higher than those of normal controls(1.21±1.16 ng/ml, 1.18±0.97 ng/ml), but only the difference of TNF-α was significant (p<0.01); in recovery SAA patients, the serum levels of TNF-α significantly decreased to 1.46±1.41 ng/ml (p<0.01), and the levels of IL-4 increased continually markedly to 3.05±1.94 ng/ml; the CD3+CD8+ cells and TNF-α of patients correlated with Ret (p<0.05;p<0.05) and ANC (p<0.05;p<0.05) negatively, Th1 cells correlated with CD3+CD8+ cells and TNF-α positively (p<0.01;p<0.05), the Ret and ANC negatively(p<0.01;p<0.01), IL-4 and the balance of Th1/Th2 correlated with Ret and ANC positively (p<0.05;p<0.01) (p<0.01;p<0.01). Conclusion The formation of bone marrow failure in SAA might be caused not only by the increase of Th1 cells, Th1 type effective cells and cytokines, but also by unsufficient compensation of Th2 cells and Th2 type cytokines, which shifted the balance of Th1/Th2 to Th1.

Background: The experiment was conducted to evaluate the effect of different dosages of lead in the hematological parameters of mice and to observe the antitoxic role of Vitamin E and Selenium in induced lead toxicities. Moreover, the toxic effect of lead in the reproduction of female mice was also examined. Methods: A total of 72 (48 male and 24 female) Swiss albino mice were used in the experiment. After adaptation, 42 male mice were selected for hematological studies and divided into seven groups (n=6) where Group A represented healthy control mice and Group B, C, and D were treated with lead acetate at the rate of 0.5mg/kg, 1mg/kg and 2mg/kg respectively. Similarly, three other groups B+, C+, and D+ were treated with lead acetate plus Vitamin E and Selenium at the rate of 2ml per liter drinking water. A total of 24 female mice were divided into four groups (n=6), group E represented control mice and Group F, G, and H were treated with lead acetate at the rate of 0.5mg/kg, 1mg/kg, and 2mg/kg for three weeks followed by matting and treatment was continued for another one week of gestation. Blood sample was analyzed from the hematological study group. Result: The lead treatment caused a dose-dependent decrease in the value of Hb and PCV significantly whereas the value of TEC and TLC were significantly decreased in Group C and D in relation to Control. The value of ESR increased significantly in a dose-dependent manner in Group D in relation to Control whereas MCV and MCH values were significantly decreased than that of control. The value of TEC, Hb, PCV, ESR, and TLC improved in the Lead plus Vitamin E- Selenium treated group as compared to the Lead treated group but, only Group C+ showed significant improvement as compared to Group C. The value of neutrophil and monocyte were significantly decreased were as lymphocyte and eosinophil were significantly increased relative to control. There was a dose-dependent effect of lead in pregnancy of female mice with the highest effect (premature delivery and infant mortality) on high dose treated mice. Conclusion: It can be concluded that lead has a great impact on hematological parameters and has an effect on various systems of the body. Premature birth and abortion are major effects of lead toxicity. Our results suggest that hemolysis of RBC and or impairment of erythropoiesis may be caused by lead toxicity and the hematological values can be restored by the use of Vitamin E plus Selenium.

Research  background  and  hypothesis.  Traditional  time  series  analysis  techniques,  which  are  also  used  for the analysis of cardiovascular signals, do not reveal the relationship between the  changes in the indices recorded associated with the multiscale and chaotic structure of the tested object, which allows establishing short-and long-term structural and functional changes.Research aim was to reveal the dynamical peculiarities of interactions of cardiovascular system indices while evaluating the functional state of track-and-field athletes and Greco-Roman wrestlers.Research methods. Twenty two subjects participated in the study, their average age of 23.5 ± 1.7 years. During the study standard 12 lead electrocardiograms (ECG) were recorded. The following ECG parameters were used in the study: duration of RR interval taken from the II standard lead, duration of QRS complex, duration of JT interval and amplitude of ST segment taken from the V standard lead.Research  results.  Significant  differences  were  found  between  inter-parametric  connections  of  ST  segment amplitude and JT interval duration at the pre and post-training testing. Observed changes at different hierarchical levels of the body systems revealed inadequate cardiac metabolic processes, leading to changes in the metabolic rate of the myocardium and reflected in the dynamics of all investigated interactions.Discussion and conclusions. It has been found that peculiarities of the interactions of ECG indices interactions show the exposure of the  functional changes in the body at the onset of the workload. The alterations of the functional state of the body and the signs of fatigue, after athletes performed two high intensity training sessions per day, can be assessed using the approach of the evaluation of interactions between functional variables. Therefore the evaluation of the interactions of physiological signals by using time series analysis methods is suitable for the observation of these processes and the functional state of the body.Keywords: electrocardiogram, time series, functional state.

Introduction. The most susceptible to lead is the hematopoietic system of hematopoietic organs due to lead inhibition of heme and globin synthesis and cytotoxic effect on the membrane of Mature red blood cells. The aim of study was to evaluate the informative value of the study of erythrocyte and reticulocyte parameters determined on modern hematological analyzers in patients working in contact with lead during medical and biological monitoring. Materials and methods. 45 employees of the lead battery processing plant and 30 persons of control group were examined. The level of lead in the blood was determined by atomic absorption spectrometry, δ-ALA in the urine-by the reaction of pyrol formation with acetylacetone in terms of gram of creatinine, the study of hematological parameters was performed on a Sysmex HT-2000i analyzer. Statistical processing of the results was performed using the program STATISTICA 10.0. Results. Significant changes in erythrocytic (RDW) and reticulocytic (RET, IRF, LFR, MFR, HFR, RET-He) parameters, erythropoietin in workers in contact with lead compared to the control group, changes in MCV, MCH, RDW, RET indicators in the group working in dynamics after 2 years were revealed. Associations of hematological parameters with biomarkers of exposure and effect (lead level in blood and ALA in urine) were revealed. Conclusions. Assessment of erythrocyte (MCV, MCH, RDW) and reticulocyte parameters (RET% and their distribution by maturity) in dynamics during periodic medical examinations of workers in contact with lead allows us to detect the development of hematological disorders at early stages.

The assessment and dynamic control over students’ status is a very important task. It allows timely detection of prenosological status prior to pathology and health maintenance in students. The objective of the paper is to assess the adaptive abilities of the body, to analyze changes in heart rate variability indicators in students with various types of autonomic regulation, to identify prenosological status and precursory pathological symptoms. Materials and Methods. The study enrolled 302 students from India, aged 21.54±1.43. Programming complex «Psychophysiologist» was used to register the main HRV parameters within 5 minutes. Health status was evaluated according to the index of functional changes and the scale of functional states. Results. N.I. Shlyk (2009) distinguished two groups of students with different types of autonomic regulation: type 1 (53 %) with moderate and type 2 (5 %) with marked characteristics of central regulation profile, type 3 (35 %) with moderate and type 4 (7 %) with marked characteristics of autonomous regulation profile. Main parameters of HRV and adaptation potential were defined for each student.All the parameters characterized functional and health status. Conclusions. It was shown that 82 % of trial subjects (type 1), 53 % (type 2), 94 % (type 3) and 95 % (type 4) demonstrated satisfactory adaptation and their physiological processes were at an optimal level. 18 % of students (type 1) demonstrated reduced adaptive abilities of the body. Moreover, they were under moderate stress. 47 % of subjects (type 2) were also under a significant stress, which was proven by excessively high SI, low SDNN and TP, and an increased index of functional changes. 5 % of students (type 4) revealed dysfunctional characteristics in the heart rhythm, peculiar to pathology. Оценка состояния студентов и динамический контроль за ним является важной задачей, поскольку позволяет своевременно выявлять у студентов донозологические состояния, предшествующие патологии, и способствовать сохранению здоровья. Цель. Оценка адаптивных возможностей организма, анализ изменений показателей вариабельности сердечного ритма у студентов с различными типами вегетативной регуляции, выявление донозологических состояний и ранних признаков патологии. Материалы и методы. В исследовании участвовало 302 студента в возрасте 21,54+1,43 года из Индии. Регистрировались основные параметры ВСР в течение 5 мин с использованием программно-аппаратного комплекса «Психофизиолог». Состояние и уровень здоровья оценивались по индексу функциональных изменений и шкале функциональных состояний. Результаты. По способу, предложенному Н.И. Шлык, выделены группы студентов с различными типами вегетативной регуляции: I (53 %) и II типы (5 %) – с умеренным и выраженным преобладанием центрального контура регуляции соответственно, III (35 %) и IV типы (7 %) – с умеренным и выраженным преобладанием автономного контура регуляции соответственно. У каждого из студентов определены основные параметры ВСР и адаптационного потенциала, характеризующие функциональное состояние и уровень здоровья. Выводы. Показано, что для 82 % обследуемых с I типом, 53 % со II типом, 94 % c III типом и 95 % с IV типом регуляции характерно состояние удовлетворительной адаптации, физиологические процессы сохраняются на оптимальном уровне. В группе студентов I типа у 18 % студентов адаптивные возможности организма снижены, выявлено состояние умеренного напряжения. У 47 % обследуемых II типа также зафиксировано состояние резко выраженного напряжения, индикатором которого является чрезмерно высокое значение SI, низкие величины SDNN и ТP, повышенное значение индекса функциональных изменений. В группе студентов с IV типом у 5 % учащихсяв регуляции ритма сердца выявлены дисфункциональные признаки, характерные для патологии. The main purpose of this study is to investigate the role of citrus leaf extract in carbon tetrachloride-induced hepatic injury and its potential molecular mechanism. Carbon tetrachloride was used to construct hepatic injury animal model. To this end, rats were randomly divided into 4 groups: control, carbon tetrachloride-treated, and two carbon tetrachloride + citrus leaf extract-treated groups. The results show that citrus leaf extract treatment significantly reversed the effects of carbon tetrachloride on the body weight changes and liver index. Besides, treatment with citrus leaf extract also reduced the levels of serum liver enzymes and oxidative stress in a dose-dependent manner. H&E staining and western blotting suggested that citrus leaf extract could repair liver histological damage by regulating AMPK and Nrf-2.

Diabetes mellitus is a serious pathologic condition that is responsible for major healthcare problems worldwide and costing billions of dollars annually. Insulin replacement therapy has been used in the clinical management of diabetes mellitus for more than 84 years. The present mode of insulin administration is by the subcutaneous route through which insulin is presented to the body in a non-physiological manner having many challenges. Hence novel approaches for insulin delivery are being explored. Challenges to oral route of insulin administration are: rapid enzymatic degradation in the stomach, inactivation and digestion by proteolytic enzymes in the intestinal lumen and poor permeability across intestinal epithelium because of its high molecular weight and lack of lipophilicity. Liposomes, microemulsions, nanocubicles, and so forth have been prepared for the oral delivery of insulin. Chitosan-coated microparticles protected insulin from the gastric environment of the body and released intestinal pH. Limitations to the delivery of insulin have not resulted in fruitful results to date and there is still a need to prepare newer delivery systems, which can produce dose-dependent and reproducible effects, in addition to increased bioavailability.

The skin is an important organ in the human body that protects the body from environmentally hazardous substances. Reactive oxygen species (ROS) cause inflammatory reactions and degradation of the extracellular matrix leading to skin aging and various cutaneous lesions. This study evaluated the potential of isoflavones isolated from Maclura tricuspidata fruit to prevent TNF-α-induced skin inflammation in normal human dermal fibroblasts (HDFs). It focused on alpinumisoflavone (AIF) that suppressed the accumulation of ROS and nitric oxide (NO) in tumor necrosis factor-alpha (TNF-α)-treated HDFs. AIF inhibited the TNF-α-induced increase in matrix metalloproteinase-1, decreased procollagen I α1, and suppressed pro-inflammatory mediators and pro-inflammatory cytokines, including NO synthase, cyclooxygenase-2, interleukin (IL)-1β, IL-6, and IL-8 that trigger inflammatory responses. AIF inhibited nuclear factor-κB and activating protein 1 mitogen-activated protein kinases that were increased by TNF-α stimulation. These results suggest that AIF may protect skin from aging and various cutaneous lesions.
