Antioxidant and Antimutagenic (Anticlastogenic) Effect of Solanum xanthocarpum Seed Extracts

The aim of the present study was to investigate the in-vitro antioxidant and in-vivo Antimutagenic properties of Solanum xanthocarpum seed extracts the preliminary qualitative phytochemical screening was to reveal presence of polyphenols, flavonoids, glycoside, alkaloids, carbo-hydrates, and reducing sugar etc. Based preliminary qualitative phytochemical screening, Quantitative estimation of polyphenols was performed, quantitative estimation alcoholic extract found significant amounts of polyphenols as compare to aqueous extract. In-vitro antioxidants was performed by two method DDPH and superoxide radical scavenging method, the alcoholic extract shows significant antioxidant properties as compare to aqueous extract, based on polyphenols and antioxidant properties alcoholic extracts was used for the antimutagenic (clastogenic) test. Alcoholic extract produced significant result in antimutagenic activity.

Background: Research on medicinal plant antioxidants has emerged as a potential therapeutic to prevent free radical generated damage in the human body. Hammada elegans Botsch (popularly known as “Ajram”) is a xerophytic plant widely found in Laghouat region, but there are only a few reports about the biological or chemical properties of these species. Hence, the aim of this study is to investigate the antioxidant and the antihemolytic activities of hexanic, acetonic, methanolic and aqueous extracts of aerial parts of Algerian Hammada elegans Botsch by employing different in vitro assay systems. Methods: The total phenolic content, the flavonoid content and the condensed tannin amount were analyzed using Folin-Ciocalteu, aluminum chloride and vanillin assays, respectively. The in vitro antioxidant capacity of extracts was assessed by CUPRAC, iron chelating, ABTS•+and antihemolytic assays, and was expressed as EC50 values. Results: Among the analyzed extracts, the aqueous extract had the highest phenolic, flavonoid and tannin contents. Also, this extract displayed the highest antioxidant capacities compared to the other extracts and standards. Its EC50 value for ABTS radical-scavenging activity was 0.265 ± 0.003 mg/L. Moreover, this extract showed high iron (II) chelating ability (EC50 = 0.958 ± 0.001 mg/L), and good antioxidant activity in the cupric ion reducing activity (CUPRAC) in a concentration dependent manner (EC50 were 0.709 ± 0.002 mg/L). Additionally, this extract had the best antihemolytic activity against AAPH-induced hemolysis (EC50=0.090 ± 0.004 mg/L). Conclusion: Our study revealed that the aqueous extract of Hammada elegans Botsch, is a potential source of antioxidants which possess a high protective effect of membrane against free radical.

Aim: This study investigated the phytochemical constituents and in vitro antioxidant properties of methanol and aqueous leaf extracts of Geophila obvallata using standard methods. Materials and Methods: The in vitro antioxidant assays carried out were 1, 1‐diphenyl‐2‐picrylhydrazyl (DPPH) radical scavenging ability, Nitric oxide (NO•) radical scavenging activity assay, 2, 2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS•+) radical cation scavenging assay, ferric reducing properties and hydroxyl radical scavenging assays. Results: Phytochemical analysis revealed the presence of alkaloids, flavonoids, phenolic compounds, steroids, saponins, terpernoids and cardiac glycosides in both extracts. Relative to the aqueous extract, the methanol extract contained a higher amount of the secondary metabolites. However, both extracts exhibited appreciable and dose-dependent capacities for quenching DPPH, ABTS•+ and NO• free radicals, and potent ferric reducing ability to levels comparable to those of ascorbic acid. The crude methanol extract showed significantly increased (P<0.05) antioxidant activity than the aqueous extract. Conclusion: It was concluded that the extract possesses strong antioxidant properties due to its content of phytochemicals, and provides scientific basis for its ethno medicinal applications.

Diarrhea is a common health complaint occurring with mild, temporary conditions to a potentially life-threatening condition. <i>Cucurbita maxima</i> (Cucurbitaceae) seed is reportedly used traditionally for the treatment of diarrheal and thus this study aimed to evaluate the in vitro antioxidants, total tannins, phenolics contents, and antidiarrheal potentials of <i>n</i>-hexane and aqueous <i>C. maxima</i> seed extracts in castor oil induced diarrheal rats. The <i>n</i>-hexane extract mainly contains oils while the aqueous extract was thick, brown solid. The aqueous and <i>n</i>-hexane seed extract of <i>C. maxima</i> expressed significant 2,2, diphenyl-1-picrythdrazyl (DPPH) scavenging activities at 6.25 – 1000 mg/mL with IC<sub>50</sub> values of 104.01 mg/mL, 29.27 mg/mL and 26.78 mg/mL for <i>n</i>-hexane, aqueous and vitamin C respectively. The hydroxyl radical scavenging activities of the <i>n</i>-hexane and aqueous seed extract of <i>C. maxima</i> were significantly lower at higher concentrations compared with that of vitamin C. Furthermore, the total antioxidant capacities of <i>n</i>-hexane (22.08mg/mL) and aqueous seed extract (11.03 mg/mL) of <i>C. maxima</i> were found to be higher than that of vitamin C (134.46 mg/mL). The aqueous extracts (658.33±380.08 mg QE/g) total tannins were not significantly different from the <i>n</i>-hexane extract (468.33±102.55 mg QE/g) while the <i>n</i>-hexane extract contains significantly higher total phenolics (2.93±1.25 mg GAE/g) compared with the aqueous extract (0.19±0.04 mg GAE/g). Percentage inhibition of stooling was found to be 57±22%, 41±16%, and 46±11% for loperamide, <i>n</i>-hexane, and aqueous respectively. The study concludes that aqueous and n-hexane seed extracts of <i>C. maxima</i> (pumpkin) possess in vitro antioxidant activities and antidiarrheal properties.

Dietary intake of foods rich in antioxidant properties is suggested to be cancer protective. Foods rich in antioxidant properties include grape (Vitis vinifera), one of the world’s largest fruit crops and most commonly consumed fruits in the world. The composition and cancer-protective effects of major phenolic antioxidants in grape skin and seed extracts are discussed in this review. Grape skin and seed extracts exert strong free radical scavenging and chelating activities and inhibit lipid oxidation in various food and cell modelsin vitro. The use of grape antioxidants are promising against a broad range of cancer cells by targeting epidermal growth factor receptor (EGFR) and its downstream pathways, inhibiting over-expression of COX-2 and prostaglandin E2 receptors, or modifying estrogen receptor pathways, resulting in cell cycle arrest and apoptosis. Interestingly, some of these activities were also demonstrated in animal models. However,in vivostudies have demonstrated inconsistent antioxidant efficacy. Nonetheless, a growing body of evidence from human clinical trials has demonstrated that consumption of grape, wine and grape juice exerts many health-promoting and possible anti-cancer effects. Thus, grape skin and seed extracts have great potential in cancer prevention and further investigation into this exciting field is warranted.

Aims: Antioxidant and Antimutagenic (Anticlastogenic) activity of alcoholic extract of Bauhinia variegata (Linn.) root. Place: C. U. Shah College of Pharmacy and Research, Wadhwan, Surendranagar, Gujarat, India. Methodology: Shade dried Bauhinia variegata (Linn.) root, extraction was carryout by isolation extract were subjected to primary and secondary Phytochemical investigation. Then In-vitro antioxidant properties were estimated by reducing power and nitric oxide free radical scavenging method. Based on Phytochemical constituent and antioxidant properties In-vivo Antimutagenic (Anticlastogenic) activity was performed. Results: Preliminary phytochemical investigation revealed the presence of carbohydrates, free amino acids, and secondary metabolites like tannins, phenolic compounds and flavonoids, then polyphenol estimation found ALBV contains 86.38% phenolic compounds. In antioxidant properties determination IC50 respectively found 55.27±2.57 µg/ml, 125.52±8.15 µg/ml against of Ascorbic acid and Curcumin. Then In-vivo Antimutagenic (Anticlastogenic) activity ALBV shows significant reeducation in % MNPCE, % MNNCE and P/N ratio at 24 h, 48 h and 72 h against the cyclophosphamide-induced mutagenicity. Conclusion: Therefore, from the present study, it is concluded that alcoholic extract of Bauhinia variegata root (ALBV) can prove to be a very good antioxidant and effective chemopreventive against cyclophosphamide-induced mutagenesis.

We have investigated effect ofMoringa oleiferaleaf and fruit extracts on markers of oxidative stress, its toxicity evaluation, and correlation with antioxidant properties usingin vitroandin vitroassays. The aqueous extract of leaf was able to increase the GSH and reduce MDA level in a concentration-dependent manner. The ethanolic extract of fruit showed highest phenolic content, strong reducing power and free radical scavenging capacity. The antioxidant capacity of ethanolic extract of both fruit and leaf was higher in thein vitroassay compared to aqueous extract which showed higher potentialin vivo. Safety evaluation studies showed no toxicity of the extracts up to a dose of 100 mg/kg body weight. Our results support the potent antioxidant activity of aqueous and ethanolic extract ofMoringa oleiferawhich adds one more positive attribute to its known pharmacological importance.

The present study evaluates the in vitro antioxidant free radical scavenging capacities of aqueous and alcoholic extracts of Glycyrrhiza glabra since this medicinal plant has been traditionally used in Ayurvedic system of medicine as a tonic, rejuvenator, demulcent and expectorant. Total phenolic content of alcoholic extract and aqueous extract was found as 18.70 and 23.26 Gallic acid equivalents/mg while the total flavonoid content was assessed as 15.03 and 6.62 Quercetin equivalents/mg respectively. IC50 (µg/ml) of alcoholic, aqueous extract and Ascorbic acid was found to be 65.064±3.538, 73.423±9.057 and 18.156±1.356 in Hydrogen Peroxide Radical Scavenging, 1.424±0.013, 1.632±0.052 and 0.879±0.273 in DPPH radical scavenging, and 335.948±29.040, 251.891±6.330 and 6.142±0.061 during FRAP essay. Similarly, during ABTS radical scavenging, IC50 (µg/ml) was assessed as 575±26.694, 683.9±49.220 and 6.337±0.167 for alcoholic, aqueous extract and Trolox respectively. While both the extracts showed substantial and significant free radical scavenging activity during the study which is compared to that of standard drug, the alcoholic extract of Glycyrrhiza glabra exhibits higher in vitro antioxidant activity than the aqueous extract in all assessment methods except FRAP, possibly due to higher concentrations of flavonoids in it, validating and confirming the therapeutic properties ascribed to it in Ayurvedic texts.

Avian coccidiosis is probably one of the most expensive parasitic diseases with major economic impact on poultry industries worldwide. The purpose of this study was to evaluate the ethanolic and aqueous extracts of Conyza aegyptiaca in terms of phytonutrients, in vitro oocysts sporulation inhibition and antioxidant properties. The extraction process of plant leaf powder (100 g) pulverized using a clean manual grinder was carried out in ethanol and hot water and the yields were calculated as a percentage ratio of extract mass on plant powder mass after solvent evaporation. Phytochemical analysis procedures were performed to determine the presence of phytonutrients. The in vitro oocysticidal sporulation inhibition was determined at five different concentrations (0.25; 0.5; 1; 2 and 4 mg/ml) of each extract in petri dishes each containing 3000 unsporulated oocysts and examined after 24 and 48 hours under a microscope. In vitro antioxidant capacity of extracts was estimated using different assays. Quantitative aqueous extract (11.72%) was higher than ethanolic extract (4.34%). In terms of qualitative yields, ethanolic extract revealed higher phytonutrients investigated (100%) than aqueous extract (42.86%). The sporulation inhibition of ethanolic extract was generally higher than the aqueous extract after 24 and 48 h and varied according to the different tested concentrations. In all the antioxidant assays, ethanolic extract exhibited significant free radical scavenging activity with inhibitory concentration (IC50=26.10±1.09) close to that of ascorbic acid at the probability level of 5% error (p<0.05). The ethanolic extract with higher free radical scavenging activities and ferric reducing effect also showed significant higher content of both phenols (127.01±3.99 mgGAE/g) and flavonoids (108.66±3.49 mgCE/g) than aqueous extracts, suggesting correlation between phenolic content and antioxidant activity. Data from this study could be used for developing bioactive elements for natural anticoccidials and antioxidants of health promoting activities

Pergularia daemia belongs to the family Asclepiadaceae, known to have anticancer, anti-inflammatory activity. Aim of the present study was to evaluate qualitative and quantitative phytochemical and antioxidant properties of ethanolic extracts of leaf, stem and root parts of P. daemia . Preliminary phytochemical analysis and in vitro antioxidant properties were evaluated by standard methods. The qualitative phytochemical analysis of P. daemia showed presence of flavonoids, tannins, alkaloid, phytosterol, carbohydrate, phenol, saponin, glycosides, terpenoids, steroids proteins and reducing sugars. Quantitative analysis showed polyphenol, flavonoid, flavonone, flavone and flavonol in P. daemia leaves, stem and root in considerable quantity. The in vitro antioxidant activity of P. daemia clearly demonstrated that leaf, stem and root parts have prominent antioxidant properties and was effective in scavenging free radicals.

Introduction: This study aimed to evaluate the antioxidant property of silymarin (SM) extracted from the seed of Silybum marianum and its anticancer activity on KB and A549 cell lines following 24, 48, and 72 h of treatment. Methods: Ten grams of powdered S. marianum seeds were defatted using n-hexane for 6 hours and then extracted by methanol. The silymarin extracted of extraction components The extracted components of silymarin were measured by spectrophotometric assay and HPLC analysis. 2, 2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, phenol content, total flavonoid content, and total antioxidant capacity were measured to detect the antioxidant properties of SM. The anticancer activity of the SM on cell lines evaluated by MTT. Results: In HPLC analysis, more than 50% of the peaks were related to silibin A and B. SM was reducedDPPH (the stable free radical) with a 50% inhibitory concentration (IC50) of 6.56 μg/ ml in comparison with butylated hydroxyl toluene (BHT), which indicated an IC50 of ~3.9 μg/ ml.The cytotoxicity effect of SM on the cell lines was studied by MTT assay. The cytotoxicity effect of the extracted silymarin on KB and A549 cell lines was observed up to 80 and 70% at 156 and 78 µg/ml, respectively. The IC50 value of the extracted SM on KB and A549 cell lines after 24 hours of treatment was seen at 555 and 511 µg/ml, respectively. Conclusion: Due to the good antioxidant and anticancer properties of the isolated silymarin, its use as an anticancer drug is suggested.
