Free radicals are molecules that lose electrons in their outer orbitals so that the number of electrons becomes odd and unstable. Free radicals can damage various macromolecule cells including proteins, carbohydrates, fats. Antioxidants are compounds that can donate one or more electrons to free radicals. Avocado leaves contain high bioactive components that can be used as a source of natural antioxidants. Avocado leaves that have been dried, mashed and then shifted with methanol. The extracts were tested by phytochemical screening, including flavonpoid, phenolic, tannin and antioxidant activity using the DPPH (2,2-diphenyl-1-picrylhidrazyl) method. This method is based on the reduction of 2,2-diphenyl-1-picrylhydrazyl (DPPH) against free radicals which causes a change from purple to yellow 2,2-diphenyl-1-picrylhydrazyl) (DPPH). From the research, the IC50 values ​​in avocado leaves were 118.8056 µg / mL and 7,276 µg / mL on ascorbic acid as a positive control. From the IC50 value obtained, it is known that the methanol extract of avocado leaves has strong antioxidant activity to fight free radicals.

The aim of this study is determine the IC50 extracts values of white peel and flesh of red watermelon, white peel and flesh of yellow watermelon as antioxidants. To determine antioxidant activities DPPH reagent was used as well as UV-Vis Spectrophotometry was used to measure vitamin C as a positive control measurement. The samples mixed with ethanol in various concentration of 5, 15, 25 and 35 mg/L. The results showed that the IC50 values for each extract of red watermelon flesh, yellow watermelon flesh, white peel of red watermelon, and white peel of yellow watermelon were 16.619 mg/L, 16.575 mg/L, 14.729 mg/L, and 16.782 mg/L, respectively. Vitamin C as the positive control has IC50 value of 9.526 mg/L. These IC50 values showed that vitamin C had higher antioxidant activity than watermelon extracts, but the extracts of watermelon still categorized in strong natural antioxidants. This study concluded that watermelon is good to be consumed because it is a very strong antioxidant.

Free radicals play a role in the occurrence of various degenerative diseases that require free-radical scavengers or antioxidants. Buni fruit (Antidesma bunius (L.) Spreng) has the bioactive components are Anthocyanins (flavonoids) that serves to the free radicals. This study aimed to measure the antioxidant activity of theethanol extract contained 70% fruit Buni obtained by using the method of nitric oxide. Simplicia buni macerated dried fruit with 70% ethanol. Extracts were obtained in the test antioxidant activity against nitric oxide radicals. The antioxidant activity against free radical absorbance measured by means of UV-Vis spectrophotometry at a wavelength of 546 nm and calcul ated IC50 values. The results shows that the fruit buni has potential as a free radical with IC50 value of 2.28 µg/mL and a comparison of quercetin with IC50 value of 5.88 µg/mL.

Methanol extract from Lippia graveolens (Mexican oregano) was studied in order to identify inhibitory bioactives for protein tyrosine phosphatase 1B (PTP1B). Known flavone as lutein (1), and another flavone glycoside such as lutein-7-o-glucoside (2), 6-hydroxy-lutein-7-ohexoside (3) and lutein-7-o-ramnoide (4) were isolated from methanol extract of aerial parts of the Lippia graveolens. All isolates were identified based on extensive spectroscopic data analysis, including UV, IR, NMR, MS and compared with spectroscopic data previously reported. These flavones were evaluated for PTP1B inhibitory activity. Among them, compounds 1 and 3 displayed potential inhibitory activity against PTP1B with IC50 values of 7.01 ± 1.25 μg/ml and 18.4 μg/ml, respectively. In addition, compound 2 and 4 showed moderate inhibitory activity with an IC50 value of 23.8 ± 6.21 and 67.8 ± 5.80 μg/ml respectively. Among the four compounds, luteolin was found to be the most potent PTP1B inhibitor compared to the positive control ursolic acid, with an IC50 value of 8.12 ± 1.06 μg/ml. These results indicate that flavonoids constituents contained in Lippia graveolens can be considered as a natural source for the treatment of type 2 diabetes.

Free radicals and reactive species are widely believed to contribute to the development of several diseases by causing oxidative stress and eventually oxidative. Vernonia amygdalina (Astereacea) is a small shrub or tree between 1 and 5m high growing throughout tropical Africa. Plants are generally known as bitter leaves is well cultivated and is a general market for merchandise in several countries. The purpose of this study was to determine the antioxidant activity of hexane fraction from ethanol extract od Frican leaves (Vernonia amygdalina Del.). The method used in this study was the DPPH (1,1-Diphenil-2-Picrylhydrazyl) method. The result of phytochemical screening showed that ethanolic extract of African leaves contained a composition of secondary metabolites of alkaloids, flavonoids, tannins, steroids/triterpenoids and saponins. The antioxidant activity of the extract of n-hexane fraction was classified as very weak with an IC50 value of 317.98 ppm.

Antioxidants are substances that can slow down the oxidation process of free radicals. Limpasu plant (Baccaurea lanceolata (Miq) Muell. Arg), an indigenous plant of Borneo, is a natural antioxidant source. The purpose of this study was to determine the antioxidant activity of the limpasu pericarpium extract. The extraction of the limpasu pericarpium was done by maceration method using solvents with increasing polarity ranging from n-hexane, ethyl acetate, and methanol. Antioxidant activities of the three extracts were measured by the DPPH and FRAP methods. The IC50 values of n-hexane, ethyl acetate, and methanol extracts, as well as quercetin using the DPPH method were 517,45 µg/mL, 530,64 µg/mL, 10,63 µg/mL and 6,83 µg/mL, respectively. Meanwhile, the IC50 values obtained from FRAP method were 198,96 µg/mL, 190,07 µg/mL, 661,36 µg/mL, and 7,09 µg/mL, respectively. The results revealed that the methanol extract is more potent than other extracts tested for antioxidant activity.

Objective: The aim of this study is to investigate antioxidant activity and phytochemical screening of ethanol extract, fractions of water, ethyl acetate, and n-hexane from mistletoe tea (Scurrula atropurpurea Bl. Dans).Methods: Simplicia extracted using soxhlet equipment with 96% ethanol. Fractionation was conducted using liquid-liquid extraction using a solvent of water, ethyl acetate and n-hexane. Screening of phytochemical and antioxidant activity was performed against these fractions. Antioxidant activity was determined by 2,2-diphenyl-1-picrylhydrazyl method using ultraviolet-visible spectrophotometry with ascorbic acid as standard. Phytochemical screening was conducted based on the method of Farnsworth.Results: The IC50 values of ethanol extract, water fraction, fraction of ethyl acetate, and n-hexane fraction were 21.92 ppm, 89.57 ppm, 14.08 ppm, and 162.09 ppm, respectively, whereas for ascorbic acid was 4.41 ppm. The ethanol extract and ethyl acetate fraction contained compounds were the same group, polyphenolic, tannins, flavonoids, monoterpenoid, steroids, triterpenoids, and quinones. Fraction of water contained compounds such as polyphenolic group, flavonoids, monoterpenoids, sesquiterpenoids, steroids, and triterpenoids. n-hexane fraction compounds contained steroids and triterpenoids.Conclusion: The ethanol extract, water fraction, ethyl acetate fraction, and n-hexane fraction showed antioxidant activities. The ethanol extract, fractions of water, and ethyl acetate fraction contained flavonoids and polyphenolic potential as antioxidants.Keywords: Mistletoe tea, Scurrula atropurpurea, Antioxidant, 2,2-diphenyl-1-picrylhydrazyl, Ascorbic acid.

Searsia burchellii finds therapeutic applications in traditional medicine. Methanolic extracts, hexane, chloroform, ethyl acetate and methanol/water fractions of methanolic extracts and water extracts were obtained separately from the roots and leaves of Searsia burchellii by the combination of maceration, hot solvent extraction and solvent-solvent partition techniques. These extracts were evaluated for their antioxidant activity using 2,2- diphenyl-1-picrylhydrazil radical scavenging assay (DPPH). The extracts from roots and their fractions showed radical scavenging activity ranging from 6.60±4.50 to 63.27±1.93% at various concentrations. Similarly, the extracts from leaves and their fractions showed radical scavenging activity ranging from 3.32±0.95 to 64.91±0.15% at various concentrations. Ascorbic acid served as positive control which showed radical scavenging activity ranging from 53.62±2.80 to 60.82±0.62% at various concentrations. The IC50 values of these extracts and fractions were found to be < 200 to > 3000 µg/mL. The IC50 value of ascorbic acid was found to be <200 µg/mL. From this study, we concluded that extracts and their fractions from S. burchellii showed promising radical scavenging activity.

Antioxidant are compounds that are able to inactivate the development of the oxidation reactions of the molecules or neutralize free radicals. Mahogany bark (Swietenia mahagoni Jacq.) has properties as a medicinal plant. Through the phytochemical screening, the mahogany bark (Swietenia mahagoni Jacq.) contains antioxidant compounds such as alkaloids, saponins and phenolics. The aim of this study was to determine the presence of antioxsidant activity in the extract of mahogany bark (Swietenia mahagoni Jacq.). using the DPPH method with a concentration of 250 mg / L, 200 mg / L, 150 mg / L and 100 mg / L and Vitamin C p.a as a positive control. Each sample made three repetitions of the test. The test uses a UV-Vis Spectrophotometer. The result of the study showed that mahogany bark (Swietenia mahagoni Jacq.) showed antioxidant activity with the percentage of 21,50% at the concentration at 250 mg / LKeywords : Swietenia mahagoni Jacq., Antioxidant, Ethanol, DPPHABSTRAKAntioksidan ialah senyawa yang mampu menginaktivasi berkembangnya reaksi oksidasi molekul lain atau menetralisir radikal bebas. Mengandung senyawa antioksidan seperti alkaloid, saponin dan fenolik. Penelitian ini bertujuan untuk mengetahui adanya aktivitas antioksidan didalam ekstrak tanaman Kulit Batang Switenia mahagoni Jacq. menggunakan metode DPPH dengan konsentrasi 250 mg/L, 200 mg/L, 150 mg/L dan 100 mg/L dan Vitamin C p.a sebagai kontrol positif. Masing-masing sampel dibuat tiga kali pengulangan uji. Pengujian menggunakan alat Spektrofotometer UV-Vis. Hasil penelitian menunjukkan bahwa ekstrak Kulit Batang Swietenia mahagoni Jacq. memiliki aktivitas antioksidan dengan presentase sebesar 21,50% pada konsentrasi 250 mg/L Kata Kunci : Swietenia mahagoni Jacq., Antioksidan, Etanol, DPPH

Aging is a natural process in human life and is triggered by the presence of free radicals (ROS). The use of antioxidants from natural ingredients is one of the breakthroughs to overcome aging and counteract the harmful effects caused by the free radicals. This study aimed to determine and compare the antioxidant activity of H2O2 scavenging and hyaluronidase inhibition of red dragon fruit peel extract (DFPE) and kaempferol-3-o-rutinoside (KOR) compounds. Dragon fruit peel extract (DFPE) is obtained through extraction by maceration method using 70% ethanol solvent. The design of this study included antioxidant and anti-aging activity assay of EKBN and KOR at the series concentration of 15.63; 31.25; 62.50; 125; 250; 500 µg/mL through H2O2 scavenging, as well as the DFPE and KOR hyaluronidase inhibition assay at the series concentration of 5.21; 10.42; 20.83; 41.7; 83.33; 166.67 µg/mL. EKBN shows that the average activity of H2O2 scavenging is lower than KOR. In addition, the IC50 values of KOR for H2O2 scavenging is lower (351.46±2.30ug/mL) than DFPE (409.64±23.17ug/mL). While, KOR also has higher values of inhibitory activity than of the DFPE. However, the IC50 value of KOR for hyaluronidase inhibition activity was 84.07±10.46µg/mL, equivalent to the IC50 value of DFPE (85.32±10.24µg/mL). The presence of antioxidant and anti-aging activity in the EKBN is probably caused by betalain and the KOR compound itself contained in red dragon fruit. The results of the paired-samples T-test on antioxidant activity and anti-aging of DFPE and KOR showed non-significant difference. Thus, DFPE has an equivalent antioxidant and anti-aging through H2O2 scavenging and hyaluronidase activity as possessed by the KOR compound.
