Effect of Tween 80 and salinomycin on ruminal fermentation and nutrient digestion in steers fed a diet containing 70% barley

Ten Jersey steers (484 ± 55 kg) were fed a basal diet of 70% rolled barley grain and 30% alfalfa silage [dry matter (DM) basis] with or without additives, in a replicated 5 × 5 Latin square experiment. Treatments (additives per kilogram dietary DM) were: C, no additives (control); T, Tween 80 (2 g kg−1); SH, salinomycin (13 mg kg−1); TSM, Tween 80 (2 g kg−1) plus salinomycin (6.5 mg kg−1); and TSL, Tween 80 (2 g kg−1) plus salinomycin (3.25 mg kg−1). Ruminal pH, concentrations of ammonia, total free amino acids, reducing sugars and total volatile fatty acids (VFA), and fluid phase viscosity were unaffected (P > 0.05) by treatment. Ruminal carboxymethylcellulase, xylanase and amylase activities, numbers of protozoa, and outflow rates of the liquid and solid phases of ruminal contents did not differ (P > 0.05) among treatments. Ruminal lactic acid bacteria populations tended (P < 0.1) to be smaller with SH compared with C, but counts were unaffected (P > 0.05) by T, TSL and TSM. Partitioning of ruminal digesta and microbial protein flow were similar (P > 0.05) among treatments. Compared with C, the ruminal rate of degradation (in situ) of alfalfa DM tended to be higher (P < 0.10) with SH, and the rate of barley grain DM degradation was higher (P < 0.05) with T. Intake and apparent digestibilities of DM, neutral detergent fibre (NDF), acid detergent fibre (ADF) and crude protein were unaffected (P > 0.05) by treatment, as were blood glucose and urea levels. Tween 80 and salinomycin did not affect ruminal fermentation or nutrient digestibility in steers fed a barley grain/alfalfa silage diet. Background and Aim: Crude glycerin is changed to propionate in the rumen, while chitosan can be used as a feed supplement to increase propionic acid concentration and decrease methane (CH4) production. We hypothesized that supplementation with a combination of a high level of crude glycerin with chitosan could have a beneficial effect on ruminal fermentation and mitigate CH4 production. This study aimed to explore the combined effects of crude glycerin and chitosan supplementation on nutrient digestibility, rumen fermentation, and CH4 calculation in native Thai bulls. Materials and Methods: Four 2-year-old native Thai bulls, weighing 150±20 kg, were kept in a 2×2 factorial arrangement in a 4×4 Latin square design. Factor A represented the incorporation of crude glycerin at 10.5% and 21% of the dry matter (DM) of a total mixed ration (TMR), and factor B represented the supplementation of chitosan at 1% and 2% DM of a TMR. Results: Increasing levels of crude glycerin at 21% decreased DM intake by 0.62 kg/day compared with 10.5% crude glycerin (p<0.05), whereas nutrient digestibility did not change (p>0.05). The incorporated crude glycerin and supplemented chitosan levels did not affect the pH, temperature, concentrations of ammonia-nitrogen, microbial population, and blood urea nitrogen (p>0.05). Supplemented chitosan and incorporated crude glycerin did not show any interaction effects on the molar portions and total volatile fatty acids (VFAs), except estimated CH4. Increasing the incorporated crude glycerin levels increased propionate and decreased the ratio of acetate to propionate ratio, whereas levels of butyrate, acetate, and total VFAs were unchanged. The combination of crude glycerin at 21% in the TMR with chitosan at 2% reduced CH4 estimation by 5.08% compared with the other feed treatment. Conclusion: Increasing incorporated crude glycerin levels in a TMR significantly elevated the propionate concentration, whereas combining 21% crude glycerin in the TMR diet with 2% chitosan supplementation could depress CH4 estimation more effectively than adding one of these supplements alone.

Incubation trials were carried out with the rumen simulation technique (RUSITEC) to study the effects of four forms of N on the growth of ruminal micro-organisms and the fermentation variables when an all-fibre basal diet was incubated. The basal diet consisted of 10 g neutral-detergent fibre (NDF) from grass hay plus 2 g NDF from sugarbeet pulp. N forms were isolated soyabean protein, soyabean peptides, amino acids blended to profile soyabean protein and NH3 as NH4Cl. Half of the daily N supply was infused as NH4Cl and the other half was infused as each of the four treatments described. Non-NH3 N (NAN) forms increased NDF (P = 0·006), acid-detergent fibre (P = 0·003) and cellulose (P = 0·015) disappearance after 48 h incubation, CO2 (P < 0·001), CH4 (P = 0·002) and total volatile fatty acids production (P < 0·001), as well as the molar percentages of isobutyrate, isovalerate and valerate, which reflected the fermentation of amino acid C skeletons. NAN treatments also increased microbial N flow (P < 0·001) compared with NH3, with peptides and protein supporting more (P = 0·036) than amino acids. The proportion of microbial N derived from NH3 decreased successively (P < 0·05) with NH3 > amino acids > peptides > protein treatments, indicating preferential uptake of peptides without passage through the NH3 pool. Microbial efficiency (g microbial N/kg organic matter apparent disappearance) was greater (P = 0·002) for the NAN forms than for the NH3 treatment, with peptides and protein treatments supporting higher (P = 0·009) values than amino acid treatment. These results indicate that N forms other than NH3 are required for optimal fibre digestion and microbial growth.

This study evaluated the effects of precision processing (PP; processing based on kernel size) barley grain on ruminal fermentation and productivity of lactating dairy cows. Twenty multiparous Holstein cows, including eight ruminally cannulated cows, were used in a replicated 4×4 Latin square design with 21-d periods. Diets contained light barley grain processed precisely using a narrow roller setting (LB), heavy barley processed precisely using a wide roller setting (HB), processed HB and LB mixed at equal proportions (PP), or equal parts of light and heavy barley grain processed at a single narrow roller setting (CON). All diets consisted of 40% barley grain, 40% barley silage, and 20% of a supplement premix. Comparisons were made between LB and HB to evaluate the effect of barley quality, and between PP and CON to evaluate the effect of precision processing. Dry matter intake, sorting index, ruminal fermentation characteristics, and nutrient digestibility were not affected by diet. In addition, milk yield and concentrations of milk fat, protein, and lactose were not different, although milk urea nitrogen concentration was greater for PP vs. CON and for LB vs. HB. These results suggest that precision processing barley grain based on kernel size may not drastically affect ruminal fermentation and milk production in lactating dairy cows.

The objective of this study was to investigate the effects of the forage shrub tedera (Bituminaria bituminosa) on nutrient digestibility, rumen microbial fermentation and furanocoumarins degradation in the rusitec. The variables were measured in fermentation liquid on Days 13 and 17 and were compared with a control (lucerne hay). Overall, tedera had greater (P < 0.05) neutral detergent fibre and acid detergent fibre digestibility than lucerne hay on both days of the experiment, but on Day 17 it had lower (P < 0.01) dry matter and crude protein digestibility than lucerne hay. There were no significant differences in concentration of NH3-N and pH between treatments, but NH3-N concentrations in both treatments were lower (P < 0.05) on Day 17 than on Day 13. The concentration of total volatile fatty acids in vessels were not affected by treatments, but the concentration of acetate was lower and acetate-to-propionate ratio higher (P < 0.05) in tedera than lucerne hay on Day 13 of the experiment. Furanocoumarins were detected in the tedera treatment only. Their concentration in the fermentation liquid increased immediately after the addition of the plant material to the fermenter, reaching highest concentrations after 2 h. These concentrations gradually declined over the next two sampling times, but 6 h after the ‘feeding’, they were still detectable in the fermentation liquid. It was concluded that (i) tedera had in vitro digestibility and fermentability variables comparable to lucerne; (ii) furanocoumarins were degraded in the fermentation fluid, and (iii) furanocoumarins from tedera did not seem to impede microbial fermentation. Tedera may provide an alternative feed source to hay and grain for filling the summer–autumn feed gap without negatively affecting nutrient digestibility and rumen microbial fermentation.

Abstract This experiment evaluated the influence of protein supplementation frequency (SF) and amount offered on intake, nutrient digestibility, and ruminal fermentation by rumen-fistulated beef steers consuming low-quality [2.9% crude protein (CP); dry matter (DM) basis], cool-season forage. Seven Angus × Hereford steers (300 ± 27 kg) fitted with ruminal cannulas were randomly assigned to 1 of 7 treatments in an incomplete 7 × 4 Latin square. Treatments, in a 2 × 3 factorial design plus a non-supplemented control (CON), consisted of 2 levels of supplemental soybean meal, 100% (F) or 50% (H) of the estimated rumen-degradable protein requirement, provided daily (D), once every 5 d (5D), or once every 10 d (10D). Experimental periods were 30 d and dry matter intake (DMI) was measured from d 19 to 28. On d 21 (all supplements provided) and 30 (only daily supplements provided; day immediately prior to supplementation for 5D and 10D treatments) ruminal fluid was collected for ruminal pH, ammonia-N (NH3), volatile fatty acids (VFA), and determination of ruminal fermentation variables. Forage and total DM, organic matter (OM), and nitrogen (N) intake increased with supplementation (P ≤ 0.04). However, a linear effect of SF × amount of supplement interaction was observed for forage and total DM, OM, and N intake (P ≤ 0.04), with each variable decreasing as SF decreased, but the decrease being greater with F vs. H. Apparent total tract DM, OM, and neutral detergent fiber (NDF) digestibility was not affected by supplementation or amount of supplement provided (P ≥ 0.10). In contrast, N digestibility increased with supplementation and for F vs. H (P &lt; 0.01). Digestibility of DM, OM, and N increased linearly as SF decreased (P ≤ 0.03). When all supplements were provided, ruminal NH3, total VFA, and molar proportions of all individual VFA increased with supplementation (P ≤ 0.04), whereas acetate:propionate ratio decreased (P &lt; 0.01). When only daily supplements were provided, none of the aforementioned fermentation parameters were affected (P ≥ 0.09). In summary, reducing the amount of supplemental CP provided to ruminants consuming low-quality forages, when supplementation intervals are greater than 5 d, can be a management tool to maintain acceptable levels of DMI, nutrient digestibility, and ruminal fermentation while reducing supplementation cost.

Forages play an important role in ruminant animal production worldwide. Unlocking the nutritional potential of poor-quality tropical forages with fibrolytic enzymes would improve forage digestibility and utilization. Using in vitro and in vivo methods this study investigated the effect of pre-treating Smutsfinger hay for 24 hours with a mixture of fibrolytic enzyme (100% cellulase; 75% cellulase: 25% xylanase; 50% cellulase: 50% xylanase; 25% cellulase: 75% xylanase; 100% xylanase and a control with no enzyme) on ruminal fermentation and digestibility of nutrients by sheep. For in vitro fermentation, dry matter, neutral detergent fibre (NDF) degradability and volatile fatty acids (VFA) were determined with standard procedures. The same treatments were used for an in vivo digestibility trial using Merino sheep in a 6 x 6 Latin square design. Feed intake and total tract digestibility were recorded. Rumen fluid samples were collected daily, preserved, and analysed for VFA. The addition of 100% cellulase enzyme to Smutsfinger hay in vitro increased (P <0.05) NDF degradability and gas production compared with the control and inclusion of 100% xylanase enzyme. Both 100% cellulase and xylanase enzymes significantly reduced in vitro end time fermentation pH. A 50:50 mixture of cellulase and xylanase plus enzyme in vivo, increased acetate, total VFA concentration, and higher NDF and ADF digestibility of the test feed compared with the control. Inclusion of a 50-75% mixture of cellulase and 50-25% xylanase enzymes treatment led to higher gas production and butyrate concentration, decreased ruminal pH and improved nutrient digestibility.

Abstract Fermentable fiber may increase endogenous losses of P and AA, thereby reducing apparent nutrient digestibility. Acacia gum fiber with medium-to-high fermentability and low viscosity was used to investigate its effect on apparent ileal digestibility (AID) and apparent total tract digestibility (ATTD) of nutrients, and standardized total tract digestibility (STTD) of P in grower pigs. A P-free basal diet (49% corn starch; 18% bovine plasma protein) was formulated to measure basal endogenous P losses (EPL). Three diets were formulated to include 2.5, 5.0, or 7.5% acacia gum fiber at the expense of corn starch in the P-free basal diet. Diets contained 16.1–17.4% CP and 0.31–0.33% total P (DM-basis). The 4 diets were fed to 8 ileal-cannulated barrows (initial BW, 54.6 kg) for four 9-d periods in a double 4 × 4 Latin square. Apparent hindgut fermentation (AHF) was ATTD minus AID. Increasing inclusion of acacia gum quadratically decreased (P &lt; 0.01) AID of DM (∆ 11.1%), linearly decreased (P &lt; 0.05) ATTD of DM (∆ 1.7%) CP (∆ 1.2%), and quadratically increased (P &lt; 0.05) AHF of DM (∆ 9.4%). Basal EPL were 391 and 377 mg/kg DM intake (DMI) for ileum and total tract, respectively. Increasing inclusion of fiber linearly increased (P &lt; 0.05) ileal EPL (∆ 184 mg/kg DMI), and tended to linearly increase (P &lt; 0.10) EPL for total tract (∆ 243 mg/kg DMI). Dietary inclusion of acacia gum tended to linearly decrease (P &lt; 0.10) AID of P, but did not affect (P &gt; 0.10) ATTD, or STTD of P. In conclusion, increasing inclusion of fermentable, low viscous acacia gum fiber decreased diet digestibility of DM and ATTD of CP, but did not affect total tract P digestibility, indicating that increasing fermentable fiber did not increase specific endogenous losses of P in the total tract.

Feeding goats with calcium salts of fatty acids (CSFA) can supply ruminants with lipids, with minimal effects on ruminal fermentation and fiber digestibility. However, there is a shortage of information on the effect of CSFA on characteristics of rumen fermentation in grassland goats. Thus, the present study aimed to assess the addition of CSFA to concentrate on the parameters of rumen fermentation of grazing goats. Five rumen cannulated goats were distributed in a Latin square 5x5 design (treatments: 0%, 1.5%, 3.0%, 4.5% and 6.0% CSFA. The pH, ammonia N and volatile fatty acids (VFA) content were analyzed in the ruminal fluid at 0, 2, 4, 6 and 8 hours after concentrate supplementation. The pH and ammonia N concentration showed a linear effect with the addition of CSFA. There was no effect observed for the VFA molar concentration after grazing goats were fed with the experimental diet. In conclusion, further research is needed to investigate the addition of CSFA to goat diets because there is evidence that CSFA increases ruminal pH and decreases excess ruminal ammonia without changing the VFA concentration in the rumen fluid.

Objective: The experiment was conducted to study the effect of rambutan (Nephelium lappaceum) fruit peel powder (RP) on feed consumption, digestibility of nutrients, ruminal fermentation dynamics and microbial population in Thai breed cattle.Methods: Four, 2-year old (250±15 kg) beef bull crossbreds (75% Brahman×25% local breed) were allotted to experimental treatments using a 4×4 Latin square design. Four dietary supplementation treatments were imposed; non-supplementation (control, T1); supplementation of RP fed at 2% of dry matter intake (DMI) (low, T2); supplementation of RP fed at 4% of DMI (medium, T3) and supplementation of RP fed at 6% of DMI (high, T4). All cattle were given a concentrate supplement at 1% of body weight while Napier grass was provided as a free choice.Results: The findings revealed that RP supplementation did not negatively affect (p>0.05) DMI of Napier grass, while RP intake and total DMI were the greatest in the RP supplementation at 4% and 6% DMI. Nevertheless, the nutrients (dry matter, organic matter, crude protein, neutral detergent fiber, and acid detergent fiber) digestibilities were not changed in the RP supplementation groups. Rumen fermentation parameters especially those of total volatile fatty acids, acetate and butyrate were not significantly changed. However, the propionate concentration was remarkably increased (p<0.05) in the RP supplementation. Notably, the ratio of acetate to propionate, the number of protozoa, as well as the methane estimation were significantly reduced in the RP supplemented groups (4% and 6% of DMI), while the counts of bacteria was not altered.Conclusion: Supplementation of RP (4% of DMI) improved rumen propionate production, reduced protozoal population and methane estimation (p<0.05) without a negative effect on feed consumption and nutrients total tract digestibilities in beef cattle. Using dietary rambutan fruit peel powder has potential promise as a rumen regulator.

Abstract The goal of this study was to evaluate the effects of different levels (10 and 13% crude protein) and protein source on ruminal fermentation of feedlot Nellore steers fed high-concentrate diets. The protein sources tested were urea, dried distillers grains (DDG) and corn gluten meal. The DDG and gluten were used as RUP sources. We used 6 cannulated Nellore steers, rumen, duodenum, and ileum arranged in 6×6 Latin square designed in factorial (2 nitrogen levels and 3 protein sources) balanced for residues. The pH was measured immediately after rumen fluid sampling at 0, 2, 4, 6, 8 and 10h after feeding. The NH3-N concentrations were determined with micro-Kjeldahl apparatus. The concentration of volatile fatty acids (VFA) was assessed by gas chromatography. Data were evaluated using the MIXED procedure of SAS, with repeated measures over time. There was no interaction between sampling time and treatments for NH3-N and pH (P &gt; 0.05). Animals fed diets containing Urea had a higher concentration of NH3-N (20.6 mg/dL) than animals supplemented with Gluten (16.2 mg/dL) or DDG (12.7 mg / dL). There was an increase in the NH3-N concentration (P = 0.049) with the increase of the protein level 10 to 13% CP with 14.7 and 18.3 mg/dL, respectively. The ruminal pH was affected by the dietary protein level (P = 0.016), in which animals fed a level of 10% CP had a higher pH (6.80) compared to the level of 13% CP (6.43). Total and individual VFA concentration was not affected by protein levels and sources (P &gt; 0.05). The reduction of N level and the use of RUP did not adversely affect the ruminal fermentation. Although significant differences were observed for pH, they were small and adequate for microbial protein synthesis in the rumen.
