Heavy metals are one of the most persistent pollutants in waters, and molecular biomarkers as bioindicators could uncover early warning signals of stress suffered by organisms exposed to these pollutants. This research was conducted to explore the impact of lead contaminant on expression levels of hsp70 gene as well as protein pattern and innate immune indicators of skin mucus in Common carp as a biological model. The fish were exposed to different concentrations of Pb(NO3)2 as a source of lead (0, 7.875, 15.75, and 31.5 mg/L of lead) for 14 days. After the completion of the experiment, the liver and gill tissue plus mucus samples were collected from the fish. Next, the effects of exposure to sublethal doses of lead were investigated on expression levels of hsp70 gene, protein pattern, and innate immune indicators of skin mucus. The results indicated that the expression level of hsp70 gene significantly increased (at 7.5% & 30% LC50) in the liver tissue compared to the control group (P<0.05). The protein pattern of treatment samples was different from that of the control group. Also, in the mucus innate immune parameters, the levels of alkaline phosphatase increased significantly at high concentrations of lead contamination in comparison with the control group (P<0.05). The protein levels first revealed an ascending trend (P<0.05), and then a descending trend (at 30% LC50), though it was not a significant downward trend compared with the control group (P>0.05). The results suggested that these physiological indicators in Cyprinus carpio could be used as molecular and mucosal biomarkers for ecotoxicological studies.

AbstractThe present study investigated possible effects of dietary malic acid on the expression of immunity, antioxidant and growth related genes expression as well as skin mucus immune parameters in common carp. Common carp (Cyprinus carpio) fingerlings were fed diets supplemented with different levels (0 [control], 0.5%, 1%, 2%) of malic acid (MA) for 60 days. The results revealed highest expression levels of immune-related genes (tnf-alpha, il1b, il8 and lyz) in skin of common carp fed 2% MA (P < 0.05). Regarding 1% MA treatment comparison with control group, significant difference was noticed just in case of lyz (P < 0.05). Evaluation of growth related genes expression revealed no significant difference between treatments (P > 0.05). The study of antioxidant related genes (gsta and gpx) in common carp skin fed with MA, showed significant difference between treated groups and control (P < 0.05). Carps fed with 2% MA had highest alkaline phosphatase activity in skin mucus compared other treated groups and control (P < 0.05). There were no significant difference between 0.5% and 1% and control (P > 0.05). The study of total protein and total immunoglobulin (Ig) in common carp skin musus revealed no alteration following MA treatment (P > 0.05). The present data demonstrated that feeding with MA altered immune and antioxidant genes expression in skin mucus of common carp.

AbstractIn this study, the effect of artemisia (Artemisia annua) leaves extract (ALE) on the immune system, growth performance and antioxidant capacity of common carp (Cyprinus carpio) was investigated. Fish with initial weight (90.32 ± 1.12 g) were fed various levels of ALE (0, 0.5, 1 and 1.5 g/kg) for 8 weeks. The results showed that the growth performance (FW, WG, SGR, PER, and LER) in fish fed 1 g/kg ALE was increased significantly (P<0.05) and the feed conversion ratio (FCR) was improved in fish fed 1 and 1.5 g/kg ALE in comparison to the control group (P<0.05). Skin mucus total protein was higher in fish fed diet containing 0.5 g/kg ALE than the other experimental groups (P<0.05), while the lysozyme, immunoglobulin and protease was increased by increasing of ALE in diets. Respiratory burst activity and serum immune parameters were higher in fish fed 0.5 g/kg ALE than the other treatments (P<0.05). Increasing of catalase (CAT) and decreasing of malondialdehyde (MDA) were observed in fish fed ALE, while glutathione peroxidase (GPx) increased only in fish fed 0.5 g/kg ALE. Hence, it can be concluded that ALE has considerable potential as a natural immunostimulant and growth promotor supplement for the common carp’s aquaculture.

The aim of this study was to analysis the effect of SA intake on these factors using common carp (Cyprinus carpio) as model organism. Two hundred and forty healthy common carp (mean weight = 15 ± 0.9 g) supplied and randomly stocked into 12 fiberglass tanks 200 L (20 fish per tank) assigned to four treatments and triplicates. The study was performed in a completely randomized design. The treatments were feeding carps with experimental diets containing different levels (0.0 [control], 0.5, 1 and 2%) of SA. The skin mucus total immunoglobulin and total protein levels in fish fed 2% SA showed significant increase compared to the control group (P < 0.05). Results showed a significant increase in the GH gene expression in 1 and 2% SA treatments (P < 0.05). The carps were fed with diet content 2% SA showed significantly increase in IGF-1 expression (P < 0.05). The expression of GSTa, and GPX (antioxidant genes) revealed a significant increase in the GSTa (fish fed SA at 1% and 2% levels) and GPX gene expression with fish fed 2% SA (P < 0.05). Supplementation of fish diet with SA induce slight elevation in the intestine of all immune-related genes (TNF-α, IL-1β, IL8 and Lyz) compared to the control group (P > 0.05). However, Lyz gene significantly up-regulated in 1 or 2% SA treatments. These results confirmed beneficial effects of SA as a feed additive in common carp culture.

AbstractCommon carp (Cyprinus carpio L.) is a dominant fish species in aquaculture, and as it is a stomachless species, absorption and digestion of nutrients take place in the intestine. The aim of the study was to evaluate the effects of a prebiotic on the content of selected minerals found in the meat, gills, and skeleton of common carp. The research applied trans-galactooligosaccharide (GOS) prebiotic produced by enzymatic transgalactosylation of milk lactose by whole cells of Bifidobacterium bifidum. The following diets have been applied: control diet without feed additives (C), diet 2 (B1) with 1% of GOS, and diet 3 (B2) with 2% of GOS. In the freeze-dried samples, concentrations of the analyzed metals were determined using atomic absorption spectroscopy (AAS). The content of phosphorus was determined using colorimetric method. The analyses confirmed that the highest level of Mg was detected in the skeleton of fish fed with 1% GOS (2.51 g kg−1) and was significantly higher compared the control treatment (2.11 g kg−1) (P < 0.05). Zn content in fish meat fed with 1% GOS (35.41 mg kg−1) was significantly higher (P < 0.05) than in the control group (24.59 mg kg−1). The tissue that accumulated the greatest amount of Zn was the gills. GOS had a positive effect on Fe accumulation in the meat, gills, and skeleton. It has been concluded that supplementation of feed with 2% GOS significantly influenced the positive correlations between Mg and P in the meat and skeleton, Fe–Ca correlation in gills, and Fe–Zn correlation in the skeleton.

This study was conducted to compare the effects of commercially available (C) and green synthesized (GS) Zinc oxide nanoparticles (ZnO-NPs) on immunological responses of common carp (Cyprinus carpio) skin mucus. GS ZnO-NPs were generated using Thymus pubescent and characterized by UV–vis diffuse reflectance spectroscopy (DRS), Fourier-transform infrared spectroscopy (FTIR), X-ray powder diffraction (XRD), scanning electron microscope (SEM), and energy-dispersive X-ray spectroscopy (EDX). Fish (n = 150) were randomly allocated into five groups in triplicate and received a waterborne concentration of 0% (control), 25%, and 50% of LC50 96 h of commercially available (C1 and C2) and green synthesized ZnO-NPs (GS1 and GS2) for 21 days. Results from XRD displayed ZnO-NPs with 58 nm in size and UV-vis DRS, EDX, and FT-IR analysis showed that some functional groups from plant extract bonded to the surface of NPs. The SEM images showed that ZnO-NPs have conical morphology. Acute toxicity study showed a higher dose of LC5096h for green synthesized ZnO-NPs (78.9 mg.L−1) compared to the commercial source (59.95 mg.L−1). The highest activity of lysozyme and alternative complement activity (ACH50) were found in control and GS1 groups. A significant decrease in alkaline phosphatase activity (ALP) was found in C1 and C2 groups compared to other treatments. Protease activity (P) was significantly decreased in the C2 group compared to the control and GS groups. Total immunoglobulin (total Ig) content was the highest in the control. In addition, total Ig in the GS1 group was higher than GS2. The exposure to ZnO-NPs lowered total protein content in all experimental groups when compared to control. Present findings revealed lower induced immunosuppressive effects by green synthesized ZnO-NPs on key parameters of fish skin mucus.

The aim of presented work was to analyze the impact of particular polymorphic changes in the promoter regions of the -1607 1G/2GMMP1, -1562 C/TMMP9, -82 A/GMMP12, -511 C/TIL-1β, and 372 T/CTIMP1genes on their expression level in POAG patients. Blood and aqueous humor samples acquired from 50 patients with POAG and 50 control subjects were used for QPCR and protein levels analysis by ELISA.In vivopromoter activity assays were carried on HTM cells using dual luciferase assay. All studied subjects underwent ophthalmic examination, including BCVA, intraocular pressure, slit-lamp examination, gonioscopy, HRT, and OCT scans. Patients with POAG are characterized by an increased mRNA expression ofMMP1,MMP9,MMP12, andIL-1βgenes as compared to the control group (P<0.001). Aqueous humor acquired from patients with POAG displayed increased protein expression of MMP1, MMP9, MMP12, and IL-1βcompared to the control group (P<0.001). Allele -1607 1G ofMMP1gene possesses only 42,91% of the -1607 2G allele transcriptional activity and allele -1562 C ofMMP9gene possesses only 21,86% of the -1562 T allele. Increased expression levels of metalloproteinases can be considered as a risk factor for the development of POAG.

The aim of the present study was to assess the effect of a commercial ß-glucan against acute toxic effect of copper sulfate on common carp Cyprinus carpio L. behavioral, hematological parameters and biochemical tests. A total of 250 Fish (150 ± 2 g) were fed a basal control diet or the basal diet plus a ß-glucan (1 g kg-1 diet) for 60 days. LC50 was calculated for 72 h which was 10.83 mg L-1.  At the end of the trial, CuSO4.5H2O was added to all treatments  at a dose  of  5 mgL-1  for  T1  and  T2, 7mg L-1  for  T3  and  T4, 9  mgL-1 for T5  and  T6   for  96h. Fish behavioral changes were recorded during 96h of exposure to different Cu concentrations with and without β-glucan, which showed abnormalities such as increase swimming activity, jerky movement, loss of equilibrium, respiratory stress, increase operculum movement, frequent jumping, erratic swimming and swimming near the water surface. On the other hand, ß-glucan supplemented groups showed significant increase (P≤0.05) in Hb concentration, PCV%, RBC and WBC count in comparison with groups without ß-glucan. The results of dietary groups without ß-glucan showed a significant decrease (P≤ 0.05) in Hb concentration, hematocrit value, RBC count, but there were significant increase (P≤ 0.05) in WBC count with an increase of Cu concentration compared to control groups. Results showed significant increase (P≤ 0.05) in dietary groups without ß-glucan in values of ALT, AST and ALP enzymes activity with elevation of Cu levels compared to control group. But, ß-glucan supplemented groups showed significant decrease (P ≤ 0.05) in values of ALT, AST and ALP enzymes activity in comparison with dietary groups without ß-glucan. These results suggested that β-glucan has protective effect against CuSO4 toxicity and future studies should be pursued particularly immune response and molecular studies
