<p><strong>Abstract</strong><strong></strong></p><p>The present work examined the <em>in vitro</em> antioxidant activity of sequentially extracted, solvent extracts (Petroleum ether, chloroform and Methanol) of stem bark, leaves and callus from <em>Amoora rohituka</em>. The antioxidative capacity of the extracts were examined by using DPPH (1,1-diphenyl,2-picryl hydrazyl), ABTS (2,2’-azinobis-3-ethyl-benzothiozoline-6-sulphonic acid) and NOS (Nitric oxide scavenging) assays by using Gallic acid as standard control. Among the tested extracts, methanol extract of stem bark showed better antioxidative capacity. The total antioxidant activity was expressed as µg equivalent of Gallic acid per gram on dry weight basis. IC<sub>50</sub> values for methanol extract of stem bark were 7.53 µg/ml; 34.92 µg/ml and 56.31 µg/ml in ABTS, DPPH and NOS assays respectively. Whereas, leaves and callus extracts showed moderate antioxidative properties. </p>

The present study aimed to determine the total polyphenol content and total antioxidant activities of ten selected seasonal fruit extracts grown in Sri Lanka. The aqueous refluxed fruit extracts (4h) were used at the initial concentration of 0.05 g/mL. The total polyphenol content was determined according to the Folin-Ciocalteu method. The total antioxidant activity was evaluated by DPPH (2, 2-diphenyl-2-picrylhydrazyl), FRAP (ferric reducing power) and TBA (thiobabituric acid assay) assays with L-ascorbic acid as the reference compound. The total polyphenol content of the fruit extracts varied from 0.81±0 to 17.54±0.50 mg GAE (gallic acid equivalents) per gram of dry weight. The antioxidant activities ranged in IC50 of 46.60±0.60 to 367.90 ± 4.90 μg/mL, 3.41±0.02 to 50.46±1.02 µM, 30.62±0.50 to 182.64±0.90 mg AAE (L-ascorbic acid equivalents) per gram of dry weight for DPPH, FRAP, TBA assays, respectively. All aqueous fruit extracts exert dose dependent in vitro antioxidant activities in different degrees. Among the selected fruit extracts S. carophylatum, P. zeylanica, A. comosus exert relatively high total antioxidant activity together with high total polyphenol content. Hence, the selected fruit extracts are deserved to be as potent sources for antioxidant lead compounds in the development of nutraceuticals using the above edible fruits.

Twelve halophyte species belonging to different families, widely represented along French Atlantic shoreline and commonly used in traditional medicine, were screened for protective activities against mycotoxins, in order to set out new promising sources of natural ingredients for feed applications. Selected halophytic species from diverse natural habitats were examined for their in vitro anti-mycotoxin activities, through viability evaluation of Madin-Darby Bovine Kidney (MDBK) and intestinal porcine enterocyte (IPEC-J2) cell lines. Besides, the in vitro antioxidant activities of plant extracts were assessed (total antioxidant and 2,2-diphenyl-1-picrylhydrazyl (DPPH)-scavenging bioassays). Of the 12 species, Galium arenarium, Convolvulus soldanella and Eryngium campestre exhibited the most protective action on MDBK and IPEC-J2 cells against zearalenone (ZEN) or T2 toxin contamination (restoring about 75% of cell viability at 10 μg·mL−1) without inflammation response. They also had strong antioxidant capacities (Inhibitory concentration of 50% (IC50) < 100 μg·mL−1 for DPPH radical and total antioxidant capacity (TAC) of 100 to 200 mg Ascorbic Acid Equivalent (AAE)·g−1 Dry Weight), suggesting that cell protection against intoxication involves antioxidant action. A bio-guided study showed that fractions of G. arenarium extract protect MDBK cells against T2 or ZEN toxicity and several major compounds like chlorogenic acid and asperuloside could be involved in this protective effect. Overall, our results show that the halophytes G. arenarium, C. soldanella and E. campestre should be considered further as new sources of ingredients for livestock feed with protective action against mycotoxin intoxication.

A phenolic acid had been isolated from chloroform soluble fractions of a methanol extract of stem bark of Syzygium litorale, Fam. Myrtaceae. The structure of the isolated compound was elucidated and established as gallic acid through extensive spectroscopic studies (UV-Vis, FTIR, and NMR) and by comparison with literature data and authentic sample. This is the first report of the isolation of compound from this plant, although it has previously been found in Myrtaceae family such as S. aromaticum, S. cumini, S. polyanthum, S. cordatum, etc. The chloroform fraction, isolated compound, and vitamin C showed very strong antioxidant activity against 2,2-diphenyl-1-picrylhydrazyl (DPPH) with IC50 value of 23.2, 7.5, and 12.5 mg/mL, respectively.

Introduction: The immune-modulatory effects of synbiotics and their ability to reduce free radical levels may be useful for functional food that is able to be active throughout whole period of colonization of the gastrointestinal tract.The aim of the present study was to investigate the immune-modulatory and antioxidant effects of the synbiotic product "NАR," a probiotic beverage.Methods: The presence of IL-2, IL-4, IL-6, IL-8, IL-10, αTNF, γIFN, Ig A, Ig M, and Ig E was studied in vitro using a solid immunosorbent analysis. The total antioxidant activities of superoxide dismutase and glutathione reductase were determined by a spectrophotometry using the Sigma-Aldrich sets.Results: Studies of the immune-modulatory properties of the synbiotic product NAR showed 1.7 fold increase of γINF levels (p<0.01) in blood after consumption of the synbiotic product “NAR” in comparison to control values, whereas the concentrations of IL-4 and Ig E decreased 2.0 times (treatment: 9.3; control: 18.7; p<0.01) and 1.3 times (p<0.1), respectively. The consumption of the synbiotic product “NAR” caused an increase in the proportion of γINF/IL 4 (treatment: 15.4; control: 4.4; p<0.01), which indicates a reduction in functional activity of Th2-type lymphocytes in comparison with the function of Th1 cells.Our study showed a high level of the total antioxidant activity of the synbiotic product (67.4 mmol/ml). The antioxidant activity of the intact cells of consortium (15.3 mM/ml), which was the basis for the preparation of the symbiotic product, is several times lower than the activity observed in the symbiotic samples.Expression of SOD is one of the mechanisms of antioxidant stress radicals inactivation by bacteria. The analysis identified a superoxide dismutase activity of synbiotic product (1.42 U/mg protein). A glutathione reductase activity of the synbiotic product was elevated (0.06 U/ml). Conclusion: The majority of the inflammatory mediators found in the blood after the consumption of symbiotic product NAR were inflammatory mediators that activate a cellular component of the resistance. Moreover, the symbiotic product has a high antioxidant activity.

<p>In the present work methanolic and water extracts of 19 medicinal plants, commonly used in Sudanese folk medicines against gastrointestinal tract, infectious diseases were investigated for their antioxidant activity. Antioxidant assays were carried out by using different in vitro models such as DPPH (2, 2-diphenyl-1picrylhydrazyl) radical scavenging and Iron chelating activity. Methanolic and water extracts of <em>Punica granatum </em>showed the highest total antioxidant activity of 71±0.03 and 73±0.01 respectively. The antioxidant activities of water extracts were poor when compared to the methanol extract. The iron chelating power of methanol and aqueous extracts of <em>Commiphora myrrha</em> 58±0.01 and <em>Azadirachta indica</em> 57±0.20 respectively.</p>

Background Amaranthus hybridus L. is an annual, erect or less commonly ascending herb that is a member of the Amaranthaceae family. Polysaccharides extracted from traditional Chinese medicines may be effective substances with antioxidant activity. Methods In this study, we isolated crude polysaccharides from A. hybridus (AHP-M) using microwave-assisted extraction. Then, the AHP-M was purified by chromatography with DEAE-32 cellulose, and two fractions, AHP-M-1 and AHP-M-2, were obtained. The structural characteristics of AHP-M-1 and AHP-M-2 were investigated, and their antioxidant activities were analyzed in vitro. Results We found that the monosaccharide composition of AHP-M-1 was different from that of AHP-M-2. The molecular weights of AHP-M-1 and AHP-M-2 were 77.625 kDa and 93.325 kDa, respectively. The results showed that the antioxidant activity of AHP-M-2 was better than that of AHP-M-1. For AHP-M-2, the DPPH radical scavenging rate at a concentration of 2 mg/mL was 78.87%, the hydroxyl radical scavenging rate was 39.34%, the superoxide anion radical scavenging rate was 80.2%, and the reduction ability of Fe3+ was approximately 0.90. The total antioxidant capacity per milligram of AHP-M-2 was 6.42, which was higher than that of Vitamin C (Vc). Conclusion The in vitro test indicated that AHP-M-1 and AHP-M-2 have good antioxidant activity, demonstrating that A. hybridus L. polysaccharide has immense potential as a natural antioxidants.

Background: Tetracarpidium conophorum (African walnut) is an African plant with ethnobotanical uses. Objectives: The purpose of this study was to evaluate the phytochemical screening and in vitro antioxidant activities of methanol extract and fractions (F) [n-hexane (HEX-F), dichloromethane (DCM-F) and, ethyl acetate (EA-F)] of T. conophorum seeds. Methods: Phytochemical screening and in vitro antioxidant activity study were carried out using DPPH, ABTs radical scavenging assays, nitric oxide inhibitory and reducing potential assays. Results: Methanol extract and its fractions contain phenols, flavonoids, saponins, tannins, terpenoids, and alkaloids. The concentrations of total phenols and flavonoids content were significantly higher in EA-F and crude methanol extract compared to other fractions. Crude methanol and EA-F contain higher concentrations of tannin while hexane fraction had the lowest tannin content but relatively higher proanthocyanidin content compared to other fractions. The antioxidant activity study showed that both methanol crude extract and fractions of T. conophorum seeds have significant activities for DPPH radical scavenging, reducing power, ferric reducing antioxidant potential, nitric oxide inhibitory activities, ABTS and hydroxyl radical scavenging for. DPPH radical scavenging activities of EA-F showed the lowest IC50 of 33.11 µg/mL, followed by Hex-F, DCM-F and crude methanol extract with IC50 of 33.43, 42.09 and 45.44 µg/mL, respectively, when compared to ascorbic acid with IC50 of 17.08 µg/mL. Conclusion: The study showed that T. conophorum seed is a rich source of secondary metabolites, which may be responsible for its antioxidant activities.

Objective: The aim of this study was focussed on phytochemical analysis, in vitro antioxidant activities of different solvent extracts of Hypoestes phyllostachya leaf. Methods: The leaves were washed efficiently, shade dried and coarsely powdered. The powdered leaf material of Hypoestes phyllostachya was successively extracted with hexane, chloroform, methanol and water using Soxhlet apparatus. Preliminary phytochemical screenings for polyphenolics, proteins, phytosteroids, flavonoids, tannins, glycosides, alkaloids carbohydrates, polyphenolics, saponins were done by following standard procedure. In vitro antioxidant activities of various solvent extracts were assessed using DPPH, ABTS, Hydroxy radical, total antioxidant capacity and reducing power assay. Polyphenolics and flavonoids were also estimated. Results: The results showed that most of the phytochemical compounds such as carbohydrates, proteins, steroids, fixed oils and fatty acids, flavonoids, alkaloids, saponins, tannins, polyphenolics and glycosides present in the methanolic and aqueous extract of Hypoestes phyllostachya. The in vitro antioxidant activity of Hypoestes phyllostachya leaf clearly showed that it has better antioxidant activity, which was confirmed by different assays. Conclusion: From this work, it can be concluded that Hypoestes phyllostachya has the potential to be a good antioxidant but have only a lesser amount of flavonoids and polyphenolic contents. This was the first report on the pharmacological activity of the Hypoestes phyllostachya.

In this study, hydrogen peroxide ( H 2 O 2)-mediated Caco-2 cytotoxicity was employed to investigate the potential antioxidant activity of the methanol extract from the lotus leaf (Nelumbo nucifera Gertn.). A dose-dependent protective effect against reactive oxygen species (ROS)-induced cytotoxicity was observed when Caco-2 cells were treated with 10 mM H 2 O 2 in combination with the methanol extract of the lotus leaf (0.1–0.3 mg/ml). However, no significant effect was found when co-treating Caco-2 cells with 10 mM H 2 O 2 and α-tocopherol. In vitro assay revealed that the extract exhibited scavenging activities on free radicals and hydroxyl radicals, and metal binding ability as well as reducing power, which may explain in part the mechanism behind the extract's ability to protect cells from oxidative damage. In addition, the extract also exhibited concentration-dependent antioxidant activities against hemoglobin-induced linoleic acid peroxidation and Fenton reaction-mediated plasmid DNA oxidation.

Background: Stilbene phytalexis (1,2-diphenyloethylen) and benzamide are beneficial for human health. To increase the stilbene ring activity, a new series of its derivatives containing benzamide structure was synthesized and evaluated for their in vitro antioxidant power. Methods: 1H nuclear magnetic resonance, mass spectroscopy, and chromatographic analyses were used to confirm the successful synthesis. The antioxidant properties were determined by the elimination of , HO , DPPH , ABTS+ radicals, total antioxidant status (TAS) and the ferric reducing antioxidant activities (TAC) measurements. Results: Stilbenebenzamide compounds showed a wide spectrum of antioxidant ability, however their total antioxidant power was weaker than those of butylated hydroxytoluene (BHT), ascorbic acid, and resveratrol. The highest antiradical activity towards and HO was shown by the compounds with structures containing amine group (SBEBA, SBA) ( : 37.7 – 38.0% and 40.8 – 43.5%, HO : 29.8%, 28.7% inhibition, respectively) at1.25 mM concentration. The antiradical power of SBEBA (0.29) in DPPH assay was lower than those of resveratrol (1.83), ascorbic acid (3.63) and BHT (4.09). The TAS values of the synthesized compounds ranged from 152.9±5.3 to 240.2±6.7µM trolox equivalent/gram (TE/g) and were much lower than those of BHT (1304±43.0), reservatrol (1360±29.0) and ascorbic acid (2782±39.7) µM TE/g. Similarly, the TAC values ranging from 29.7±0.9 to 41.5±1.6 µM TE were weaker than that of resveratrol (239.2 ±6.7 µM TE/g). Conclusion: The results suggest that the presence of hydroxyl group in stilbene ring should be considered in further design of stilbenebenzamide compounds to enhance their antioxidant activity.
