Quantitative estimation of Silybin in Iraqi Silybum marianum by HPLC

Milk thistle's extract by an organic solvent is known as silymarin, which is a mixture of about 75% flavolignan and trace content of flavonoid (mainly taxifolin), other ingredients involve fatty acids and polyphenolic compounds. The major flavolignans are silybin, isosilybin, silydianin, and silychristin, quantitatively silybin is considered as the major compound followed by isosilybin, and both are represent approximately 60% of silymarin. This study was designed to determine the concentration of silybin in different parts of the plant and to study the effect of extraction methods, solvents and the time of collection on silybin concentration, which have been done by HPLC instrument. The results show that the higher yield of silybin is obtained from the seed using ethanol in the soxhlet apparatus and that all other parts of the plant which are produced in huge amount by the plant also contain a good concentration of silybin when compared with seeds, So they can be used as an alternative source for silybin production.

Aim of the study was to analyze the phytochemical constituents and estimation of total flavonoids and phenolic compounds of leaves and roots of the medicinal plant Lagascea mollis. Methanol extract of dried leaves and roots of L. mollis was prepared by using soxhlet apparatus. The extract prepared was tested forpreliminary qualitative phytochemical screening, followed by the quantitative estimation of total flavonoids and phenols by spectroscopy. The selected plant parts were found to contain alkaloids, flavonoids and terpenoids in both parts and saponin is present only in leaf. In quantitative estimation the reports revealedthe presence 0.017 and 0.013 mgRE/g extract and 458.91 and 704.50 mgGAE/g extract total flavonoids and phenolic compounds in leaf and root extract respectively. These major phytoconstituents present in this species may be accounted as factors for the medicinal importance of L. mollis.

The present enquiry was intended to analyze the phytochemicals qualitatively and quantitatively from flowers of Spanish cherry tree. Flower powder was extracted using polar and nonpolar solvent by soxhlet apparatus. Percentage yield of crude extracts was determined and further the extracts were subjected to analyze the phytochemicals qualitatively and quantitatively by standard procedure. Qualitative analysis showed the absence of alkaloids while presences of tannins, saponins, terpenoids, steroids, glycosides, flavonoids, phenols. Quantitative estimation of phytochemicals was determined using standard curve. Result revealed that the tannin content was 4.3±0.01 (mgTAE /gm), flavanols content was 0.28±0.05 (mgQE/gm). Saponins content was 3.6±0.7 % and terpenoids content was 1.47±0.37 %. A well conducted studies on phytochemicals revealed that they are vital for humans because they provide protection against a variety of ailments. "The history of medicinal plants is associated with the evolution of civilization. In all regions of the world, the history of nations shows that these plants have always occupied an important place in medicine, in cosmetic products, and culinary preparations. The paper aims to determine the total polyphenols in different parts of the species Lysimachia nummularia L. In our study, we focused on the extraction of polyphenolic compounds in different solvents. The solvents used in the extraction were: 40% ethanol, concentrated methanol, in water. The total polyphenol content was determined by spectrophotometric methods, a method from the European Pharmacopoeia 10.0, with minor modifications. The total polyphenol content of different extracts varied depending on the extraction process. Different parts of the plant and different solvents were used in the determinations carried out to establish the optimal extraction method for the organs of Lysimachia nummularia L."

Introduction. Plants are a valuable source of biologically active compounds, including antioxidants, which could prevent the development of oxidative stress. The members of Rosaceae family such as Sorbus aucuparia L. and Cydonia oblonga Mill. are the examples of plants contained antioxidants. Aim. The aim of the study was to evaluate the antioxidant potential of leaves and fruits extracts of rowan and quince, taking into account influence of the extrahent as well as extraction method on this activity. Material and methods. The extracts were obtained by classic extraction methods – shaking and extraction in Soxhlet apparatus. Methanol, acetone as well as 70 and 96% (v/v) ethanol were used as solvents. Antioxidant activity was estimated by DPPH, FRAP, Folin-Ciocalteu (F-C) and ABTS methods. Results. The highest antioxidant activity was found in leaf extracts of both plants. In most cases, rowan extracts showed higher potential than appropriate extracts of quince. The most effective extraction method was extraction in Soxhlet apparatus using methanol (in case of rowan and quince) as well as 96% (v/v) ethanol (quince) and 70% (v/v) ethanol (rowan). The highest results were obtained with FRAP method, whereas the lowest – with DPPH. Conclusions. The presented in vitro results confirmed, that the leaves and fruits extracts of selected plants belonging to the Rosaceae family showed antioxidant activity and could be applied in various industry branches.

Extraction of DNA is very important nowadays in bio-molecular researches. Extracted DNA should be purified and the quality of DNA should also be very high. The objective of the study was to develop a simple efficient method to isolate DNA from the rice varieties in an open laboratory environment, and to eliminate the usage of expensive chemicals and tools. The DNA extraction methods developed by the DNeasy plant kit method supplied by QIAGEN, Cheng et al., Doyle et al. and Michiels et al. were applied to five different rice varieties grown in different parts of Sri Lanka. Based on the quantity and quality of the extracted DNA tested by measuring the absorbance of DNA at 260 nm using Nanodrop® ND-1000 spectrophotometer and measuring the ratio of A260 / A280 and gel running on agarose, the efficiency of the extraction method chosen varied among rice varieties. Among the methods used, the methods introduced by DNeasy plant kit method supplied by QIAGEN and Cheng et al, yielded good and amplifiable quality DNA with satisfactory concentration for all the rice varieties tested. Therefore the modified method of Cheng et al, 1987 could be used to extract DNA from rice varieties instead of the commercially available expensive and hazardous DNeasy plant kit method supplied by QIAGEN.Int J Appl Sci Biotechnol, Vol 3(3): 398-401

The extraction of secondary metabolites by water, MeOH:water (8:2) containing NaF, methanol, ethanol and acetone (all of them diluted (7:3) in water)from the different parts (leaves, flowers, stems and roots) of Passiflora caerulea L., Physalis peruviana L. and Solanum muricatum Aiton via decoction and maceration methods was studied. The highest extraction yields were recorded by methanol for decoction and acetone for maceration. The total polyphenol content (TPC) obtained by decoction had the highest TPC contents, and MeOH containing NaF was the best solvent for the extraction of TPC. Maceration was suitable for flavonoid extractions, with ethanol and acetone being the best solvents. In general, the highest levels of TPC and flavonoids were obtained from Passiflora leaves regardless of the solvent or extraction method applied. Furthermore, the roots of Physalis and Solanum showed important levels of these compounds in consonance with the total antioxidant activity (TAA) evaluated in the different organs of the plant in the three species. In this study, the solvents and extraction methods applied were tools that determined significantly the level of extraction of bioactive compounds, showing a different impact on plant organs for each medicinal species studied.

Edible insects represent an interesting alternative source of protein for human consumption but the main hurdle facing the edible insect sector is low consumer acceptance. However, increased acceptance is anticipated when insects are incorporated as a processed ingredient, such as protein-rich powder, rather than presented whole. To produce edible insect fractions with high protein content, a defatting step is necessary. This study investigated the effects of six defatting methods (conventional solvents, three-phase partitioning, and supercritical CO2) on lipid extraction yield, fatty profiles, and protein extraction and purification of house cricket (Acheta domesticus) and mealworm (Tenebrio molitor) meals. Ethanol increased the lipid extraction yield (22.7%–28.8%), irrespective of the insect meal used or the extraction method applied. Supercritical CO2 gave similar lipid extraction yields as conventional methods for Tenebrio molitor (T. molitor) (22.1%) but was less efficient for Acheta domesticus (A. domesticus) (11.9%). The protein extraction yield ranged from 12.4% to 38.9% for A. domesticus, and from 11.9% to 39.3% for T. molitor, whereas purification rates ranged from 58.3% to 78.5% for A. domesticus and from 48.7% to 75.4% for T. molitor.

In the paper separation methods of organic part from oil sands were investigated at experimantally. For the determination of organic part (liquid fraction) of oil sands the extraction methods was used in Soxhlet apparatus by solvent. Thermal processing was carried out with a monotonic heating of oil sands from room temperature to 560 0C. As showing results, organic part of Beke oil sands studied by extraction method were 12.0 wt.%. Thermal processing results showed, that the organic part of Beke oil sands were 9.6 wt.%. Physical and mechanical characteristics of liquid fraction are established by standard methods.

Helichrysum graveolens (Bieb) Sweet possesses various medicinal applications such as diuretic, treatment of the sand and stone in kidney, regulation of digestion problems and strenghtening of the immune system. The investigation was carried out to reveal ethanol extracts of flower and stem parts of H. graveolens for antimicrobial, antioxidant, anti-tyrosinase and anti-urease activities. Ethanol extract of stem part of H. graveolens exhibited more antibacterial activity against growth of bacteria except for K. pneumoniae. Stem part of H. graveolens has higher copper reducing antioxidant capacity (CUPRAC) and anti-urease activities than flower part of H. graveolens. Anti-tyrosinase activity (IC50 values) of stem and flower parts of H. graveolens was found to be 0.01549 ± 0.0006 μg/ml and 0.01515 ± 0.0016 μg/ml, respectively. This survey suggests that ethanol extracts of different parts of H. graveolens is an alternative source for natural antioxidant and antimicrobial compounds, urease and tyrosinase inhibitors that could be utilized in drug and food industries.
