The purpose of this study was to evaluate aortic wall thickness after periodontal disease and/or obesity induction in a Wistar rat model.Sixty male Wistar rats were randomly divided into four groups: control (CT), periodontal disease (PD), obesity (OB), and obesity plus periodontal disease (OB+PD). Groups OB and OB+PD received cafeteria diet for 17 weeks. After they had acquired obesity (week 12), periodontal disease was induced by placing a silk ligature on the maxillary right second molar of groups PD and OB+PD. During the experimental period, body weight and Lee index were assessed. Mean alveolar bone loss (ABL) was evaluated, and aortas were prepared for histometric analysis of the aortic wall by ImageJ software. Body weight and Lee index increased in rats exposed to cafeteria diet. Mean ABL was higher in Groups PD and OB+PD than in control and OB (p<0.05). ABL was 18% higher in Group OB+PD than in Group PD, with statistically significant difference (p<0.001). Aortas were thicker in Groups OB and OB+PD than in control and PD groups, respectively (2.31mm ± 0.28 and 2.33 ± 0.29 vs. 2.18 ± 0.26 and 2.14 ± 0.27). Group OB differed significantly from the control group (p=0.036), and OB+PD and OB differed significantly from PD (p=0.004 and p= 0.001, respectively). Obesity alters aortic wall thickness in Wistar rats. However, the presence of periodontal disease did not affect the aortic wall thickness under the conditions of the present study.

PURPOSE: To compare body weight and length, heart weight and length, heart-to-body weight ratio, glycemia, and morphometric cellular data of offspring of diabetic rats (ODR) and of normal rats (control). METHODS: Diabetes was induced in 3 pregnant Wistar rats, bearing 30 rats, on the 11th day after conception by intraperitoneal injection of 50 mg/kg of streptozotocin. Six normal pregnant Wistar rats, bearing 50 rats, made up the control group. Morphometric data were obtained using a scale for the weight, length, heart and body measurements. Morphometric cellular data were obtained by a computer assisted method applied to the measurements of myocytes. Statistical analysis utilized Student's t-test, ANOVA and Levene test. RESULTS: Control offspring had greater mean body weight and length than offspring of diabetic rats (p < 0.001). Heart weight and length and heart-to-body ratios of newborn rats differed between groups at birth (p < 0.001), but showed no difference at 21 days. Mean nuclei area and perimetric value of the myocytes decrees throughout the first 21 days of life (p < 0.01) in the diabetic group. CONCLUSIONS: Heart hypertrophy on the offspring of diabetic rats at birth was demonstrated by the significant difference between the groups. After the eleventh day, no difference was found, which confirmed regression of cardiomegaly. The significant difference between the first and the 21th day of life, for nuclei area feature, demonstrate regression of cardiac hypertrophy in the offspring of diabetic rats.

The aim of this study was to evaluate the influence of ligature-induced periodontal disease in pregnant rats on their newborn's health parameters. Twenty-four female adult Wistar rats were divided into two groups: the control group (G1) and the group that was submitted to dental ligatures around second upper molars (G2). After the four week period of development of periodontitis, the female animals were mated with male adult Wistar rats. There were no differences in the body weight of females between the two groups during mating and pregnancy. No differences were observed among the groups in relation to the viable newborn index. However, there were differences in newborn birth weight, explained by the diverse size of the litters. In this study, ligature-induced periodontal disease did not promote changes during pregnancy that resulted in low birth weight in newborn Wistar rats.

Background. Herbal medication is a worldwide and ancient practice, mostly in developing countries, where a large part of the population is involved in this practice. Hence, studies must be conducted to evaluate their safety and efficiency to avoid or prevent toxicological risks due to their usage. In Togo, Carissa spinarum is a medicinal plant belonging to Apocynaceae family, used as an aphrodisiac or to heal some ailments including malaria, sickle cell anemia, hypertension, pain, and asthma. Notwithstanding its several ethnomedicinal benefits, just a few toxicological data associated with its chronic use are available. Objective. Therefore, this study aims to assess the toxicity of an ethanolic root extract of Carissa spinarum in Wistar rats. Methods. The 90-day oral toxicity process following OECD TG 408 guidelines is used. Male Wistar rats received Carissa spinarum root hydroethanolic extract at 500 and 1000 mg/kg for 90 days by oral gavage. Body weight changes, hematological and blood biochemical parameters, organ weight changes, malondialdehyde as a lipoperoxidation marker expressed according to tissue proteins, and histopathology of vital organs were assessed. Results. No signs of toxicity or mortality were observed during the 90 days experiment. Hematological parameters have not shown any treatment-related abnormalities. According to biochemical parameters, an increase in the chloride ion level was observed at 1000 mg/kg p < 0.01 . There was no significant difference between the treated groups and the control group concerning the malondialdehyde concentration, body weight, and organ relative weight. No changes in necropsy and histopathology of vital organs associated with extract treatment were observed. Conclusion. The results indicated that an ethanolic root extract of Carissa spinarum does not cause adverse effects, which can lead to Wistar rats’ death after 90-day oral administration at 500 and 1000 mg.

Abstract Aim To examine alveolar bone level (ABL) changes before (T1) and after (T2) orthodontic treatment (OT) in subjects with periodontal disease. Methods The study included 50 subjects with periodontal disease. All patients received subgingival debridement following baseline examination. Control group patients received final periodontal treatment before the start of OT. For the test group patients final periodontal treatment was performed simultaneous to OT. OT was performed with a straight-wire appliance. Micro-implants or temporary crowns on implants were used for posterior anchorage when needed. ABL measurements of 3821 tooth surfaces were performed on cone beam computed tomography images. Results No difference was observed between mean ABL at T1 and T2. ABL remained unchanged on 69 per cent of surfaces. A mean of 15.6 (SD 7.4) per cent of surfaces experienced ABL gain, and a mean of 15.1 (SD 7.5) per cent was found with ABL loss. Small significant median ABL difference was observed on mesial and distal surfaces (P < 0.001). A significant difference was found between median ABL changes on mesial/distal in comparison to buccal/lingual surfaces (P < 0.01). Significantly more buccal (17.9 %) and lingual (18.5 %) surfaces experienced ABL loss when compared with mesial (11.3 %) and distal (12.0 %) surfaces (P < 0.001). Significant difference was found in the median ABL change of intruded (0.5 (IQR 0.94) mm) and non-intruded (−0.4 (IQR 1.07) mm) maxillary incisors (P = 0.04). Significant median ABL gain was found on the lingual surface of maxillary incisors, which were retroclined more than 8.6 degree and intruded more than 1.6 mm. Conclusions ABL changes after periodontal–orthodontic treatment in patients with periodontal disease were small. ABL gain was more observed on mesial and distal surfaces and ABL loss on buccal and lingual surfaces. Larger orthodontic movements of maxillary incisors influenced ABL gain.

Introduction: Monosodium glutamate (MSG) is one of the most widely employed food enhancers. Although the umami compound, controversy persists regarding the effects of MSG intake on body weight. Chronic MSG intake may result in excessive body weight gain and obesity. Consumption of MSG result in organ damage, cardiovascular disease, oxidative stress, and also risk factors for obesity. This study aims to determine the effect of oral MSG on obesity in adult male Wistar rats (Rattus norvegicus).Methods: This true experimental study used the post-test control group design. Twenty-four adult male Wistar rats were randomly divided into four groups: control (received distilled water), Group 1 (MSG 0.378 mg/gr BW), Group 2 (0.756 mg/gr BW) and Group 3 (1.512 mg/gr BW). The obesity parameter was obtained by the Lee index. Kruskal-Wallis test follows by Mann-Whitney test were used to compare the Lee index between groups.Results: Lee’s index mean for each group was 358.4%, 314.1%, 287.8%, and 320.9%, respectively. The Kruskal Wallis test showed a significant difference in the Lee index between groups (p = 0.043). A follow-up test using Mann-Whitney found a significant difference between group 2 and the control group (p = 0.043, p <0.05). The mean of Lee index of group 2 was 70.51% lower than the control group.Conclusion: This study concluded that Lee index was not increased in MSG-treated rats than in the control group after oral MSG intervention for 21 days.

The aim of this experiment was to determine the effect of meniran herbs (Phyllantus niruri L.) extract to blood triglycerides level in Wistar rats (Rattus norvegicus). This experiment was a laboratory experiment with pre and post-test with a control group design. The samples were 30 male Wistar rats with ±200 gram of body weight and ±2 months old. The samples were divided into five groups by using a purposive random sampling technique and each group consisted of six rats. Group I as a negative control, group II as a positive control, and groups III, IV and V as meniran herbs extract groups with first dose, second dose, and third dose. All rats were simultaneously given with hyperlipemic feed. Group II was simultaneously administered with Cholvastin 0.5 mg/200 grams body weight/day. Groups III, IV and V were simultaneously treated with meniran herbs extract at dose of 25 mg, 50 mg and 100 mg/200 grams body weight/day. The total period of this experiment was 21 days. At the day 1st and 21st, blood was collected from rat orbital venous plexus. Serum was analyzed for their pre and post-test blood triglycerides. Statistical analysis was performed by one-way analysis of variance (ANOVA) followed by a post-hoc test. The value of p=0.002 (p<0.05) showed there was a significant difference between pre-test and post-test in delta means of blood triglycerides level among groups. Meniran herbs extract had a significant triglycerides lowering effect against blood triglycerides level of Wistar rats. The most potential of meniran herbs extract dose was 100 mg/200 gram body weight/day.

Objectives: The objectives of this research is to evaluate the effects of aqueous extract of T. indica on  weight, lipid parameters, some electrolytes and urea of Wistar rats Design & Methods: Twenty-five Male  Adult  rats were grouped into four  consisting of six rats each. Group I served as control group, group II was administered 100mg/kg of the Aqueous extract, group III was administered 150mg/kg of the Aqueous extract, and Group IV was administered 200mg/kg of the Aqueous extract. The administration took place for seven days, and on the eight day, The Animals were sacrificed. The blood samples was collected via cardiac puncture and used for analysis using a visible spectrophotometer. Results: From the result, there was no significant difference (p<0.05) in body weight of Wistar rats. In the Test Groups; Serum cholesterol, low density lipoprotein (LDL), High density lipoprotein (HDL), Very Low-density Lipoprotein and Triglycerides revealed a significant increase (p<0.05) compared to Group 1 which is the control Group but had a significant decrease (p<0.05) compared within test groups. For electrolytes concentrations, potassium, Sodium, Chloride and Urea concentrations was significant at p<0.05. Conclusion: the results above ,T. indica extract may help to maintain healthy weight, may have hypolipidemic properties, may help in body homeostatic and fluid balance and may also prevent renal damages.

The aim of the present study was to evaluate the effect of alcohol and/or tobacco exposure on spontaneous alveolar bone loss in Wistar rats. Twenty-four, male, 60 day-old, Wistar rats were assigned to 4 groups: Group 1 received 10 mL/kg of glucose solution (5%). Group 2 received 2 g/kg alcohol (20%). Group 3 was exposed to tobacco smoke (6 cigarettes/60 min). Group 4 received both interventions of groups 2 and 3. Alcohol was given by gastric gavage and cigarette exposure was performed using a forced ventilation chamber. After 30 days, animals were sacrificed and the upper maxillae removed and defleshed. Morphometric analysis of alveolar bone loss (ABL) around the second molar was performed in standardized digital photographs. Statistical analysis was conducted using paired t-test, one-way ANOVA and occurrence of spontaneous periodontal disease (ABL ≥ 0.39 mm) was analyzed by Fisher's exact test. Significant differences in body weight were observed between all groups. Group 2 presented higher body weight as compared to the 3 other groups at 4 weeks (p≤0.05). Mean ABL values were 0.31 mm (±0.08), 0.29 mm (±0.07), 0.33 mm (±0.10), and 0.33 mm (±0.08) for groups 1, 2, 3, and 4, respectively. No significant differences were found among groups. In the analysis of occurrence of periodontal breakdown, alcohol exposure decreased the occurrence of ABL and cigarette exposure increased ABL. The combination of alcohol and cigarette exposure did not differ from the control group. Alcohol consumption decreased the occurrence of periodontal breakdown, while tobacco increased this rate.

Backgound: Non Alcoholic Fatty Liver Disease (NAFLD) occurs when the intake and free fatty acid synthesis occurs more frequently than its oxidation and resecretion in the blood. Melatonin is a powerful antioxidant that can boost the synthesis of endogenous antioxidants in the body, suppress the inflammatory response and inhibit the formation of steatosis.Objective: To analyze the effect of melatonin supplementation in reducing body weight andTNF-α levels in male Wistar rats were fed by waste cooking oil.Methods: True experimental studyusing post-test only control group design. This study was done on 18 male wistar rats were divided into 3 groups : the positive control group (P0) was administrated waste cooking oil, the treated group 1 (P1) was administrated waste cooking oil and 5mg/kgBW melatonin, and the treated group 2 (P2) was administrated waste cooking oil and 10mg/kgBW melatonin for 28 days.Data analysis using One Way ANOVA test and followed by Tukey test to determine the most effective dose of melatonin.Results: There was significant difference in body weightbetween P2group and K0 group (p=0,019) with the mean body weight difference was 19,167g lower than K0 group. There was no difference in TNF-α levels between the three groups (p=0,155). Conclusion: Melatonin dose of 10mg/kgBWloses body weight male Wistar rats have given by waste cooking oil for 28 days, but does not reduce TNF-α levels.

Abstract This study aimed to assess the effects of low molecular weight heparin (LMWH) on alveolar bone loss (ABL), blood count, and counting of megakaryocytes and adipocytes in male Wistar rats. Forty male 60-day Wistar rats were randomly divided into four groups: Control (C), Periodontal Disease (PD), Heparin (Hp) and Heparin + Periodontal Disease (Hp+PD). LMWH was applied for 60 days at doses of 1 ml/kg/day. Blood samples were collected at baseline, 30 and 60. On day-49, PD and Hp+PD groups were subjected to ligature-induced periodontitis around second upper right molar. The left side was assessed as spontaneous alveolar bone loss. Mean ABL in the side with ligature showed significantly different between C (0.35±0.07 mm) and Hp+DP (0.49±0.09 mm) groups (p<0.001), between PD (0.55±0.11 mm) and Hp (0.32±0.06 mm) groups (p<0.001) and between Hp and Hp+DP groups (p<0.001). No significant differences were found among groups for ABL in the side without ligature. Animal weight, food intake, and water consumption showed no statistically significant difference among groups. Megakaryocytes and adipocytes were counted using optical microscopy and no statistically significant differences were found. Within-groups, there were an increase and a decrease, respectively, in the counting of lymphocytes (p=0.005 for C and p=0.009 for Hp+PD groups only) and leukocytes (p=0.003 for C, p=0.001 for PD, p=0.002 for Hp, and p<0.001 for Hp+PD groups). There was no decrease in the number of platelets in the three collection periods. LMWH was not able to affect ABL, but it may change the blood counting, especially increasing lymphocytes.
