Objective: The study was conducted to evaluate the affect of Dimethyl Sulfoxide (DMSO) concentration on the viability of H9C2 cells under the different cultural conditions. Methods: H9C2 cells were cultured under normal conditions and subjected to hypoxia/reoxygenation model. DMSOat the doses of 0,001÷10% (v/v) was added to the cultural medium during normal culture period and reoxygenation period. Cellular viability of the experimental groups was assessed by using CCK-8 kit. Results: The results indicated that the viability of H9C2 cardiomyocytes was stable in the different culture media supplied with DMSO at the doses of0,001÷0,5% (v/v). Meanwhile, supplementation of DMSO at the doses of1% and 2% significantly decreased the survival rate of H9C2 cells (v/v,p<0,05). Conclusion: The affect of DMSO on H9C2 cardiomyocytes is dose-dependant maner.

Dyes are usually difficult to be decolorized due to their complex chemical structures. In this work, a bacterium which had the ability of decoloration on synthetic dyes was isolated from Yancheng printworks and was identified as Pseudomonas aeruginosa. The effects of concentration of the dye concentration, cultural time, cultural temperature and initial pH of cultural medium on the efficiency of decoloration were investigated. The result showed that the optimal cultural conditions was: dye concentration was 50mg.L-1, cultural time was 72 h, cultural temperature was 28°C, initial pH of cultural medium was 7.0.

Background/Aims: Ganoderic acid A (GAA) isolated from Ganoderma lucidum, shows various benefit activities, such as anti-tumor activity, anti-HIV activity and hepatoprotective activity. However, the potential effects of GAA on hypoxia-induced injury of cardiomyocytes are still unclear. In this study, we aimed to reveal the effects of GAA on hypoxic-induced H9c2 cell injury, as well as potential underlying molecular mechanisms. Methods: Rat H9c2 cardiomyocytes were cultured in hypoxia condition with different doses of GAA. Cell viability and apoptosis were detected by CCK-8 assay and flow cytometry, respectively. qRT-PCR was performed to assess the expression levels of microRNA-182-5p (miR-182-5p) and phosphatase and tensin homologue (PTEN). Cell transfection was conducted to change the expression levels of miR-182-5p and PTEN in H9c2 cells. Finally, protein levels of key factors involved in cell proliferation, cell apoptosis and PTEN/PI3K/AKT pathway were evaluated using western blotting. Results: Hypoxia treatment significantly induced H9c2 cell viability loss and apoptosis. GAA incubation remarkably protected H9c2 cells from hypoxia-induced viability loss, proliferation inhibition and apoptosis. In addition, GAA obviously enhanced the expression level of miR-182-5p in H9c2 cells. Suppression of miR-182-5p notably alleviated the protective effects of GAA on hypoxia-treated H9c2 cells. Furthermore, miR-182-5p negatively regulated the mRNA and protein levels of PTEN in H9c2 cells. GAA attenuated hypoxia-induced inactivation of PI3K/AKT pathway in H9c2 cells by up-regulating miR-182-5p and then down-regulating PTEN. Conclusion: GAA protected rat H9c2 cardiomyocytes from hypoxia-induced injury might via up-regulating miR-182-5p, down-regulating PTEN and then activating PI3K/AKT signaling pathway.

Pratama et al, 2018. The Effect of Different C/N Ratio in for Biofloc of Formation Culture Media to the Growth and Survival Rate of Juvenile Climbing Perch (Anabas testudineus). JLSO 7(1): Climbing perch fish resources still have a problem due to this fish is still obtained only through capture and also the growth of climbing perch fish is slow. The solutions to increase the growth of the climbing perch fish is by applying biofloc technology for climbing perch fish culture. The purpose of this research was to know the effect of different C/N ratio on the growth of climbing perch fish (Anabas testudineus) juvenille with biofloc farming system. This research used a completely randomized design consist of four treatments and three replications. The treatments were without molase (P0), molase with C/N ratio 15 (P1), C/N 20 (P2) and C/N ratio 25 (P3). The results showed that C / N ratio 20 was the best treatment with absolute length of growth of 1.33 cm, absolute growth of weight 1.92 g, survival rate of 99.17% and feed efficiency 138.06%.

pH is one of the important water quality parameters in aquaculture. This study aimed to observe the growth performance and survival rate of common carp (Cyprinus carpio) Rajadanu strain reared in culture media with different pH levels. Fish (length: 3.60 ± 0.18 cm; weight: 1.68 ± 0.18 g) were stocked in nine aquariums (40 cm x 40 cm x 30 cm) for 10 days with a stocking density of 25 fish per aquarium. The treatments used were (A) pH 4-5, (B) pH 5-6, and (C) pH 6-7, with three replications. A commercial feed containing 28% protein was given daily and as much as 3% of the total biomass. The observed data were analyzed using variance analysis (ANOVA), followed by Duncan test. The results showed that the pH treatments did not affect the growth (length and weight) and survival rate of common carp Rajadanu strain (P>0.05). The best growth was achieved by the fish group reared in water with a pH range of 6-7 (length = 0.38 cm; weight = 0.17 g). The highest survival was attained by the fish group reared in water with a pH range of 5-6 (90.66%).

Chronic heart failure (CHF) is a complex clinical syndrome caused by a variety of heart problems, with a high incidence. The 5-year survival rate of patients with clinical symptoms is similar to that of malignant tumors. Wenyang Zhenshuai granules are a safe and effective granule of traditional Chinese medicine components, including aconite, dried ginger, licorice, and red ginseng. In contemporary clinical applications, it is widely used in acute and chronic heart insufficiency, coronary heart disease, and arrhythmia. This research cultured H9C2 cardiomyocytes and divided them into the normal control group, LncRNA-MiR143HG overexpression group, LncRNA-MiR143HG silence group, Adriamycin (ADR) group, ADR + medicated serum group, ADR + LncRNA-MiR143HG overexpression + medicated serogroup, and ADR + LncRNA-MiR143HG silence + medicated serogroup. The cells of each group were treated differently, and the survival rate of each group of cells and the expression levels of LncRNA-MiR143HG/miR-143 and ERK5 were detected at the end of the experiment, and the expression of LncRNA-MiR143HG/miR-143 in H9C2 cardiomyocytes was regulated by Wenyang Zhenshuai granules’ impact. The results of this study showed that, in the doxorubicin-induced H9C2 cardiomyocyte injury model, the expression of miR-143 was upregulated, and the expression of LncRNA-MiR143HG and ERK5 was significantly downregulated. Wenyang Zhenshuai granules can downregulate the expression of miR-143 to promote ERK5 protein expression and phosphorylation. The process is regulated by LncRNA-MiR143HG/miR-143, which may be one of its important mechanisms for the treatment of chronic heart failure.

Imagining the Mulatta: Blackness in U.S. and Brazilian Media demonstrates how mixed-race women of African and European descent are harnessed in popular media as a tool to uphold white supremacy and discipline people of African descent to uphold state policies of antiblackness. Uncovering the racialized and gendered paradigms of U.S. and Brazilian media, the book uses case studies of texts from a broad range of popular culture media—film, telenovelas, television shows, music videos, magazines, newspapers, and Olympic ceremonies—to elucidate how the U.S. mulatta and Brazilian mulata figures operates within and across the United States and Brazil as a response to racial anxieties and notions of white superiority. These shared concepts of race, gender, and sexuality crystallize in the mulatta/mulata figure as representative of interlinked racial projects in Brazil and the United States. Focusing on popular culture and political events of the 2000s, the book demonstrates how the mulatta and mulata figures facilitated multicultural and postracial discourses. Exploring representations, definitions, and meanings of blackness in the context of the Americas, the book traverses the cultural conditions of racializations in the United States alongside Brazil to unveil the workings of pervasive racial and gender inequalities.

Sepsis-induced myocardial depression has high mortality and is very common in intensive care units. Previous studies have found that microRNAs play an important role in regulating sepsis-induced myocardial depression. miR-150-5p is involved in many biological processes; however, the mechanism underlying its role in sepsis-induced myocardial depression is still unclear. In this study, we generated rat models of septic shock induced by lipopolysaccharide. Whole genomic RNA sequencing was performed on 12 left ventricles collected after LPS treatment to identify miRNAs. Most of the target genes of the differently expressed microRNAs were involved in apoptosis, according to Gene Ontology. We also observed apoptosis in the heart tissue and in H9C2 cardiomyocytes stimulated with lipopolysaccharide, indicating that cell apoptosis may be an important mechanism in sepsis-induced myocardial depression. Furthermore, the expression of miR-150-5p was reduced, and overexpression of miR-150-5p with mimics resulted in a decrease in apoptosis, decreased expression of cleaved caspase3 and Bax, and increased expression of Bcl-2. Additionally, after H9C2 cells were transfected with miR-150-5p mimics or an inhibitor, the expression of Akt2 decreased or increased, respectively. These findings suggest that miR-150-5p can alleviate apoptosis and may be a novel therapeutic target for sepsis-induced myocardial depression.

Effects of different culture media (Bd-2, Bd-3, Bd-4 and IFP), different culture vessels (conical flasks, plastic bowls and earthen pots) and different cultural conditions (stock, pilot plant and shade culture) on coiled and straight filament characteristics of Spirulina were studied. Stock culture maintained under special conditions, retained coiled filament characteristics even after 17 years, while Spirulina filaments in pilot ponds lost their coiled characteristics. Food value of the straight filaments was acceptable and their survival capacity was more than that of coiled filaments. The effects of dimethyl sulfoxide (DMSO) on creatine kinase (CK) conformation and enzymatic activity were studied by measuring activity changes, aggregation, and fluorescence spectra. The results showed that at low concentrations (<65% v/v), DMSO had little effect on CK activity and structure. However, higher concentrations of DMSO led to CK inactivation, partial unfolding, and exposure of hydrophobic surfaces and thiol groups. DMSO caused aggregation during CK denaturation. A 75% DMSO concentration induced the most significant aggregation of CK. The CK inactivation and unfolding kinetics were single phase. The unfolding of CK was an irreversible process in the DMSO solutions. The results suggest that to a certain extent, an enzyme can maintain catalytic activity and conformation in water–organic mixture environments. Higher concentrations of DMSO affected the enzyme structure but not its active site. Inactivation occurred along with noticeable conformational change during CK denaturation. The inactivation and unfolding of CK in DMSO solutions differed from other denaturants such as guanidine, urea, and sodium dodecyl sulfate. The exposure of hydrophobic surfaces was a primary reason for the protein aggregation.Key words: creatine kinase, dimethyl sulfoxide, denaturation, activity, conformation.

Recently, there has been development of an antifreeze polyamino acid (carboxylated poly-l-lysine: PLL) as new cryoprotectants (CPA). This compound counts as amphoteric macromolecular cationic and anionic substituents (polyampholyte) by chemically modifying poly-lysine. In addition, PLL is highly safe and frequently used as a food additive in substitution for other CPA. Other common CPA have high toxicity and caused physiological damage. In vitro-produced blastocysts were randomly assigned into 3 groups: (1) vitrified embryos with PLL vitrification solution [PLL-vit-1: 15% PLL + 15% ethylene glycol (EG); PLL-vit-2: 30% PLL + 30% EG + 0.5 M sucrose], (2) vitrified embryos with Vajta et al. solution (Conv-vit-1: 7.5% dimethyl sulfoxide + 7.5% EG; Conv-vit-2: 16.5% dimethyl sulfoxide + 16.5% EG + 0.5 M sucrose), and (3) nonvitrified blastocysts (control). All embryos were frozen by droplet vitrification method. First, the PLL-vitrified embryos were exposed to 5, 10, and 15 min in the PLL-vit-1 and then putted for 30 to about 60 s in the PLL-vit-2. Then, we compared with each group regarding exposure time of Vit-1. Post-thaw survival rate of each exposure time did not significantly differ among the 3 groups (100 v. 100 v. 100%). However, hatching rate of the 10-min exposure group was higher than that of 5- and 15-min groups (75.0 v. 25.0 v. 66.7; P < 0.05). Therefore, we confirmed that exposure time of Vit-1 was exposed for a minimum of 10 min. The post-thawed survival rate of each vitrification method was not significantly different between PLL-vit and Conv-vit groups (97.7 v. 86.4%). The total cell numbers of blastocyst did not significantly differ among groups. However, the apoptotic cell numbers of blastocyst was significantly different between the control and Conv-vit groups (0.4 ± 0.6 v. 4.4 ± 3.9; P < 0.05) but was not different in control v. PLL-vit (0.4 ± 0.6 v. 2.1 ± 2.4) and Conv-vit v. PLL-vit (4.4 ± 3.9 v. 2.1 ± 2.4). In conclusion, PLL-vit for bovine blastocyst could reduce toxicity and osmotic shock and showed high efficiency on the quality of post-thawed bovine blastocysts compared with that of Conv-vit.This work was partly supported by a grant from the Next-Generation BioGreen 21 Program (No. PJ009587022014), IPET (Grant No. 112020-3), and a scholarship from the BK21 plus program.
