To assess nephropathic potential of Caladium bicolor methanolic extract within the renal parenchyma of experimental Wistar rats. Twenty four albino Wistar rats (weighing between 180-195 g) were divided into four groups which include Group I treated with distilled water (2 ml/kg b.w.), Group II treated with methanolic extract of C. bicolor (100 ml/kg b.w.), Group III treated with methanolic extract of C. bicolor (200 ml/kg b.w.), Group IV treated with methanolic extract of C. bicolor (300 ml/kg b.w.). All administrations were done orally and once daily for a period of thirty five days. Body weight of animals was recorded during days 0, 7, 14, 21, 28 and 35 of study. After the study period, kidney tissue of study animals was harvested, weighed and processed for histopatho logical study. Staining of renal tissue sections was done using H & E technique, examined under microscope for observable histopathological changes that were scored using image-J software. Results of this study showed that sub-chronic exposure to methanolic extract of C. bicolor caused reduction in body and renal tissue weight. Moreover, exposure to the extract increases significantly (p < 0.05) renal histopatho logical changes including inflammation, necrosis, glomerular congestion and tubular dilatation within the renal parenchyma of study animals. Therefore, methanol extract of C. bicolor exhibited dose-independent nephropathic effect on the renal parenchyma following a sub-chronic exposure in experimental Wistar rats.

Chemotherapy of trypanosomosis has been adversely affected by widespread drug resistance and toxicity. Attention has now been shifted towards the search for ethnobothanical means of controlling the disease. The aimof this study was to determine the effect of methanol extract of the fruit pod of Acacia nilotica on experimental Trypanosoma brucei infection inWistar rats. TwentyWistar rats were randomly alloted into four groups of 5 rats each. Rats in groups I to III were experimentally infected with an isolate of T. brucei while group V served as uninfected control. At day 2 post infection (PI), rats in groups I were orally treated with extract of fruit pod of A nilotica at a dose of 500 mg/kg for 5 days while rats in group II were treated with diminazene aceturate at 3.5mg/kg subcutaneously once. Rats in group III served as infected untreated control. Parasitaemia, PCV and survival rates were monitored during the course of the study. Parasitaemia begins 3 days PI and progressively increased in groups I and III but was not observed in group II. There was decrease in PCV of rats in groups I and III compared to that of groups II and IV. The differences observed in parasitaemia and PCV between the groups compared were not statistically significant (P>0.05).Mortalitywas observed in group I from day 8PIwith 2 rats (40%) surviving up to day 10 PI. All rats in group III died within 7 days post infection while mortalities were not recorded in groups II and IV. It was concluded that the methanol extract of the fruit pod of A. nilotica had no In vivo trypanocidal effect at the dose of 500mg/kg, but demonstrated some ameliorative effect on the severity of the infection in Wistar rats.

The continued use of potassium bromate in some food and cosmetic products in Nigeria against regulatory provision has continually exposed Nigerians to its deleterious effect. The mechanism of bromate induced damage in the liver and kidney is through oxidative stress. Phytochemicals have antioxidant capacity and have gained research interest in the management of oxidative stress. In this study, the effect of the ethanol extract of Aframomum angustifolium seed on kidney and liver damage induced by exposure to an acute dose of potassium bromate was measured using appropriate biomarkers. Eighteen male Wistar rats, randomly divided into 3 groups were used for the study. Group I (normal control) received distilled H2O; Group II (positive control) received 400 mg/kgbw bromate once while Group III (test) received same acute dose as Group II but thereafter treated with an oral dose of ethanol extract (750 mg/kgbw) for 10days. Half the populations of each group were sacrificed on day 3 while the remaining was sacrificed on day 10 of treatment. Liver and kidney tissues as well as blood samples were collected for analysis. Biochemical parameters; malondialdehyde, creatinine, electrolytes (Na+ and K+) concentration as well as catalase, alanine aminotransferase and alkaline phosphatase activities were measured using standard spectrophotometric procedures. In the kidney tissue, malondialdehyde concentration increased significantly in all groups from day 3 to day 10 but was highest in Group II. A similar trend was observed in the liver tissue but only group III increased significantly. Catalase activity was inhibited in Group II in both kidney and liver tissues and was significantly lower than test group. Serum Na+ decreased and K+ increased respectively but Group III were comparable to the Group I. Creatinine concentration increased in all groups but was highest in Group II though it was not significant. ALP activity was significantly higher (p<0.05) in Group III on day 3 compared to Group I but was not significantly different (p>0.05) on day 10. The results of this study suggests that ethanol extract of Aframomum angustifolium seeds can accelerate the in vivo repair of bromate induced hepato- and reno- toxicity in Wistar rats. Abstract: Gentamycin, a frequently used aminoglycoside antibiotics, has a nephrotoxic effect to human beings and animals. The purpose of this research was to find out the microscopic changes of wistar rat kidneys after gentamycin induction. This was an experimental study, using five adult wistar rats, divided into three groups. Group I was the control group; group II consisted of two rats, injected with gentamycin 0,3 ml/day (dose of 60 mg/kg body weight/day) intraperitoneally for seven days; and group III consisted of two rats, injected with gentamycin 0,3 ml/day intraperitoneally for 10 days. Group I and II were terminated at day-8, and group III at day-11. Their kidneys were processed for microscopic slides, stained with hematoxylin eosin and Periodic Acid Schiff. In microscopic evaluation, group II and III showed oedema, necrosis, apoptosis, and basal membrane destruction of tubular epithelial cells. Group III also showed fat vacuoles in these epithelial cells (macrovesicular fatty changes). Conclusion: wistar rats injected with gentamycin 60 mg/kg body weight/day for 7 and 10 days showed oedema, necrosis, apoptosis, and basal membrane destruction of tubular epithelial cells; and macrovesicular fatty changes after 10 days of gentamycin.Key words: gentamycin, necrosis tubular epithelial cells, fatty changesAbstrak: Gentamisin termasuk antibiotik golongan aminoglikosida berspektrum luas yang bersifat nefrotoksik terhadap manusia dan hewan. Tujuan penelitian ini untuk melihat perubahan mikroskopik struktur ginjal tikus Wistar setelah diberikan gentamisin. Metode penelitian eksperimental dengan menggunakan lima ekor tikus Wistar dewasa yang dibagi atas tiga kelompok. Kelompok I tanpa perlakuan; kelompok II terdiri dari dua ekor tikus perlakuan yang diinjeksi dengan gentamisin 0,3 ml/hari (dosis 60 mg/kgBB/hari) secara intraperitonial selama tujuh hari; dan kelompok III terdiri dari dua ekor tikus perlakuan yang diinjeksi dengan gentamisin 0,3 ml/hari secara intraperitonial selama 10 hari. Tikus Wistar kelompok I dan II diteminasi hari ke-8, sedangkan kelompok III diterminasi hari ke-11. Ginjal tikus kelompok I -III kemudian dibuat preparat histopatologik dengan pengecatan rutin hematoksilin eosin dan Periodic Acid Schiff (PAS). Hasil penelitian menunjukkan tikus Wistar perlakuan yang diberikan gentamisin 0,3 ml/hari selama 7 sampai 10 hari secara mikroskopik memperlihatkan pembengkakan, nekrosis, apoptosis, dan destruksi membrana basalis sel epitel tubulus; dan pada hari ke-10 terlihat vakuol-vakuol lemak pada sel epitel sehingga inti terdesak ke tepi (perlemakan makrovesikuler). Simpulan: pemberian gentamisin pada tikus Wistar dengan dosis 60 mg/kg BB/hari selama 7-10 hari menunjukkan pembengkakan, nekrosis, apoptosis sel epitel tubulus, dan membrana basalis tubulus rusak; dan setelah hari ke-10 juga terlihat perlemakan makrovesikuler.Kata kunci: gentamisin, nekrosis sel epitel tubulus, perlemakan makrovesikuler

This study evaluated the ameliorative and prophylactic effects of 2 different doses of polyphenolic-rich fraction of Garcinia kola (PPRF Gk) seeds on the histology and hormones of pituitary–testicular axis of male Wistar rats. Thirty-five male Wistar rats (150-200 g) were divided into 7 groups of 5 rats each. Groups I and II were given distilled water (0.5 mL/day) for 8 days followed by propylene glycol (0.2 mL/d) and 600 mg/kg of PPRF Gk, respectively, for 21 days. Group III received sodium arsenate (8 days), left untreated for 21 days. Groups IV and V received sodium arsenate (20 mg/kg) for 8 days followed by PPRF Gk (300 and 600 mg/kg, respectively) for 21 days. Groups VI and VII received PPRF Gk (300 and 600 mg/kg, respectively) for 21 days followed by sodium arsenate (20 mg/kg) for 8 days. Rats were killed by cervical dislocation 24 hours after the last dose and their blood collected through cardiac puncture. Blood sera were assayed for the levels of follicle-stimulating hormone (FSH), luteinizing hormone (LH), and testosterone using immunoassay techniques. Histology of the pituitary gland and testes was carried out. A significant reduction was observed in the concentration of FSH in groups IV, V, VI, and VII in comparison with groups I and II. The concentrations of both LH and testosterone showed significant decreases in groups IV, V, VI, and VII in comparison with group I. Group III presented with the lowest serum hormonal concentrations. Photomicrographs of the pituitary gland revealed greatly reduced basophils in group III and mildly reduced basophils in groups IV, VI, and VII in comparison with groups I and II. Group V revealed hypercellularized and distorted basophils. Photomicrographs of the testes showed detachment of the seminiferous tubules from the basement membrane and disruption of the interstitial space which was worse in group III, moderate in groups V and VI, and mild in group VII. In conclusion, PPRF Gk effected a dose-dependent reversal and prevention of the perturbations caused by arsenate in rats.

The effects of the ischemia and reperfusion on the lens metabolites and the effect of a phytotherapeutic commercial product called “Enoant” (mixed polyphenol content) on the selected lens metabolites were investigated. For this aim, 30 Wistar rats were divided into three groups according to their diet and being subjected to ischemia. 10 of the rats as Group I were fed on dry diet; the other 10 (Group II) were fed on dry diet and drinking water with Enoant. At the end of 15 days period, both groups of rats were subjected to ischemia for 2 hours and reperfused. After another 15 days with their same diet, the rats were decapitated. The remaining 10 rats, which were not subjected to ischemia (Group III), were fed on dry diet only. 1HNMR spectroscopy was used for the determination of lens metabolites of each group of rats. The results obtained from the three groups have been compared statistically. The differences of metabolites were significant except pyruvate and succinate. Oral administration of Enoant revealed effects on increasing membrane stabilization, the antioxidant capacity, osmotic regulator molecule capacity, and sorbitol content of lens disturbed by ischemia. Enoant can be used where oxidative or osmotic stress is formed.

Summary The effect of aqueous matured stem extract of Opuntia dillenii on selected biochemical parameters in Male Wistar rats was explored. Standard analytical methods were applied. Forty Wistar rats (80-100g) were used in the animal studies, separated into four groups. The control group was solely administered normal feed and saline, group I was administered 100mgkg−1 of the extract, group II received 300mgkg−1 of the extract and group III received 500 mg/kg−1 of the extract. A significant increase (p<0.05) in the activities of alanine aminotransferase (ALT) and alkaline phosphatase was observed in group II and III rats, as compared with the controls. A significant decrease in urea and creatinine concentrations was found only in group III rats against the controls. Also, a significant (p<0.05) decrease in triglyceride, total cholesterol, and low density lipoprotein (LDL)-cholesterol was seen in group II and group III rats when compared with the control. The hematological evaluation revealed a significant (p<0.05) decrease in red blood cell and hemoglobin levels in group III rats when compared with the control. The findings showed both beneficial and toxicological effects of the plant. Hence, for optimal therapeutic benefits, a further toxicological survey could still be carried out perhaps at higher doses.

Background: This study was designed to evaluate the effect of Berberis asiatica root extract (BAE) against streptozotocin induced elevated blood glucose level and other liver and kidney functions changes in adult male Wistar rats.Methods: Thirty male Wistar rats were divided into five groups of six rats in each (Groups I-V). Group I and Group II served as normal control and disease control, respectively. Group III received standard anti-diabetic drug glibenclamide (5mg/kg), while Group IV and Group V received the low dose (250mg/kg) and high dose (500mg/kg) of BAE. Serum blood glucose, SGOT, SGPT, ALP, total bilirubin, BUN, serum creatinine, TC, TG, HDL-C, LDL-C, and VLDL-C were estimated using standard methods. After collection of samples for biochemical evaluation, the pancreas from each animal was isolated and examined for histological changes.Results: BAE and glibenclamide treated disease rats showed significant (p <0.05) decrease in blood glucose concentration. Treatment with BAE at 250mg/kg and 500mg/kg in Group IV and V and standard drug glibenclamide in Group III showed significant (p <0.05) reduction in the level of liver function substances such as SGOT, SGPT, ALP and total bilirubin as compared to disease group, as well as showed significant (p <0.05) decrease in renal panel. Liver function parameters were significantly (p <0.05) improved in groups treated with BAE. Histopathological analysis revealed the protective effect of BAE against streptozotocin induced damage to islets of Langerhans.Conclusions: This study showed the destruction of islets of Langerhans and elevation in blood glucose level as well as alteration in other biochemical parameters were ameliorated by the effect of Berberis asiatica extract.

Abstract: Liver is the largest organ in the abdominal cavity. As the center of metabolism in the body, liver is potentially damaged by exposure of toxic substances, inter alia carbon tetrachloride (CCl4). Metabolism of carbon tetrachloride (CCl4) produces CCl3 free radicals that can damage the liver. In Indonesia, there are a lot of natural ingredients that have antioxidant properties, such as tomato. Lycopene in tomatoes contains antioxidant compounds that can prevent damages due to free radical. This study aimed to obtain liver histopathological changes of wistar rats fed with tomato juice after being induced of carbon tetrachloride (CCl4). This was an experimental study, using 10 wistar rats which were divided into 4 groups. Group I was the negative control; group II was induced with CCl4 0,05 cc/day and was terminated on day 6; group III was induced with CCl4 0,05 cc/day and was given tomato juice 3 ml/day, and terminated on day 13; group IV was induced by CCl4 0,05 cc/day, given regular pellets, and terminated on day 13. The results showed that group II had histopathological changes of the liver indicating fatty liver, meanwhile group III showed regeneration of nearly all liver cells. Conclusion: Administration of tomato juice after the induction of 3 ml carbon tetrachloride (CCl4) for 7 day showed regeneration of almost all liver cells. Keywords: histopathological changes of the liver, carbon tetrachloride, tomato juice.   Abstrak: Hati merupakan organ terbesar dalam rongga abdomen, dan pusat metabolisme tubuh dengan fungsi yang sangat kompleks dan sangat berpotensi mengalami kerusakan akibat terpapar oleh bahan-bahan toksik, salah satunya yaitu karbon tertraklorida (CCL4). Metabolisme CCl4 menghasilkan radikal bebas CCl3 yang dapat merusak hati. Di Indonesia terdapat  banyak sekali bahan-bahan alami yang mempunyai kandungan antioksidan, salah satunya yaitu tomat. Tomat mengandung senyawa likopen sebagai antioksidan yang dapat mencegah kerusakan jaringan akibat radikal bebas. Penelitian ini bertujuan untuk mendapatkan gambaran histopatologi hati tikus wistar yang diberi jus tomat pasca induksi karbon tetraklorida (CCl4). Metode penelitian ialah eksperimental. Sampel sebanyak 10 ekor tikus wistar yang dibagi dalam 4 kelompok. Kelompok I sebagai kontrol negatif; kelompok II diinduksi CCl4 0,05cc/perhari dan diterminasi hari ke-6; kelompok III diinduksi CCl4 0,05 cc/hari kemudian diberikan jus tomat 3ml/hari;  dan kelompok IV diinduksi CCl4 0,05 cc/hari kemudian diberikan pelet biasa dan diterminasi hari ke-13. Hasil penelitian menunjukkan pada pemberian CCl4 pada tikus wistar selama 5 hari terdapat gambaran morfologik perlemakan sel hati. Pemberian jus tomat dosis 3 ml pasca induksi karbon tetraklorida (CCl4) menunjukkan terjadinya regenerasi pada hampir seluruh sel-sel hati. Simpulan: Pemberian jus tomat dosis 3 ml pasca induksi karbon tetraklorida (CCl4) selama 7 hari menunjukkan regenerasi pada hampir seluruh sel-sel hati. Kata kunci: gambaran histopatologi hati, karbon tetraklorida, jus tomat.

The prevalence of parasitic infections such as malaria, which leads to decrease in hematological indices, the major cause of anemia, constitutes a serious health challenge in many developing countries such as Nigeria. This study investigated the effect of methanol seed extract of Aframomum melegueta on selected hematological indices of 2,4-dinitrophenylhydrazine (2,4-DNPH)-induced anemic rats model. The toxicity study and qualitative phytochemical screening of the extract were carried out using standard procedure. Twenty Wistar rats were grouped into five of four rats each (n = 4). Group I: Normal control; Group II: Negative control; Group III: administered 20 ml/kg b.w. of Astifer (Standard drug); Group IV and V were administered 200 and 400 mg/kg b.w. of the extract, respectively. The animals of Groups II to V were induced with 2,4-dinitrophenylhydrazine (20 mg/kg b.w.) once daily for seven consecutive days; their blood samples were collected by ocular puncture into heparinized capillary tubes for hematological analysis and animals with packed cell volume (PCV) ≥ 30% reduction were considered anemic for the study. The result of the qualitative phytochemical analysis showed that the methanol extract tested positive to alkaloids, carbohydrate, saponins, flavonoids, steroids, terpenoids, and anthraquinones. Acute toxicity and lethality studies on methanol extract showed an oral LD50 equal or less than 5000 mg/kg b.w. in mice. The rats administered 20 ml/kg b.w. of Astifer showed significant (P < 0.05) increase in PCV, hemoglobin, red blood cell (RBC) count, white blood cell (WBC) count, and neutrophils count compared with the normal control animals. The rats administered 200 mg/kg b.w. of A. melegueta showed significantly (P < 0.05) higher PCV and WBC count; non significant (P > 0.05) decrease in hemoglobin count, RBC, platelet, neutrophils and lymphocyte count compared with the normal control animals. The rats administered 400 mg/kg b.w. of A. melegueta showed significant (P < 0.05) increase in hemoglobin, platelet, erythrocyte sedimentation rate (ESR) and neutrophils count; non significant (P > 0.05) lower PCV, RBC, WBC count, and lymphocyte count compared with anemic rats administered with 0.3 ml of normal saline. It can be concluded that Aframomum melegueta seed has beneficial immunological and hematological properties in Wistar rats and possessed erythropoietic potentials at minimal dose that support its use for treating anemia.

Background: Drug induced liver injury (DILI) is known as the damage of liver cells due to chronic administrations of drug. The chronic administration of paracetamol could be trigger the damage of liver cells.The hepatoprotector agents are still limited worldwide.  Gambier(Uncaria gambir Roxb) is an Indonesia’traditional medicine which have many benefits as antioxidant, antiseptic, antidiarrhoea, etc  that commonly used in society.  Method: The present study was conducted to investigate the hepatoprotector effect ofgambier in wistar rats induced by paracetamol.  The wistar rats were divided into seven groups and received the treatment orally for 12 days. Group I (aquadest), II(curcuma,400 mg/kgBW),III (gambier,26 mg/200gr), IV(gambier, 53 mg/200gr), V(gambier,106 mg/200gr),VI(gambier,212 mg/200gr) and VII(gambier,424mg/200gr). Termination, blood and liver organ collection were done after all group induced by paracetamol for two days. Histopatology changes of liver were examined using Hematoxycilline (HE) staining. AST and ALT levels were analyzed. Results: There were significant differentiation of AST levels among the groups, especially between group I and group IV and between group II and group IV. The ALT levels were statistically significant between group II and group V using Mann-Whitney test (p<0,05). In histopatology examination, there were significant differentiation between group I with another group, not only group II but also group III-VII (p<0,05). In the treatment group, group III and IV had been showed the improvement of liver cells damage than group I by using One-way Annova, post hoc Bonferroni (p<0,05). Conclusion: Uncaria gambir Roxb has hepatoprotector activity start at dose 53 mg/200grBWin rats.  The hepatoprotector activity was not superior than curcuma.  Keywords: hepatoprotector, Uncaria gambir Roxb, AST,ALT,histopatology
