Aims: This study was aimed at investigating the antihyperglycemic effect of a combined extract of Vernonia amygdalina and croton zabensicus compare with a hypoglycemic drug, glibenclamide. Methodology: Twenty 20 experimental animals were used (albino rats); the rats were divided equally into four groups of five rats each; namely A (control), B (glibenclamide 10 mg/kg body weight), C (synergetic treatment 1000 mg/kg body weight), D (synergetic treatment 500 mg/kg of body weight). Diabetes was induced intraperitoneal using Alloxan Monohydrate to all the animals and their blood glucose rise above 200 mg/dl. Results: It was observed that group B and group C treated with glibenclamide (10 mg/kg body weight) and synergetic aqueous extract (1000 mg/kg body weight) show significant decrease in the blood glucose level from 451.75 mg/dl to 64.50 mg/dl and 339.50 mg/dl to 182.50 mg/dl respectively compared with group D with 278.25 mg/dl to 194.75 mg/dl. However, a change was also observed in the body weight of the groups; Group A (Normal control) showed a continuous increase in the body weight, Group B, C and D were observed to have decreased in body weight from induction period, but a steady increase was observed as treatment commences. Conclusion: Hence this combined extract can be used as antihyperglycemic; only that it is slower in remediation compared with the glibenclamide; but without side effect, as may be in the case of most standard drug.

Diabetes is a chronic disease which characterized by hyperglycemia (elevated or abnormally high blood sugar levels) and other metabolic disturbances, including metabolism of lipids and haemostasis. Caesalpinia pulcherrima has previously showed strong anti-diabetic and hepatoprotective potential. The present research work was to investigate the anti-diabetic activity and hepatoprotective activity Caesalpinia pulcherrima in streptozotocin-induced (STZ) diabetic rats. The dose-dependent effects of 45days oral treatment with methanol extract of plant (200 and 300mg/kg) of CPAE on body weight, blood glucose level, total protein, albumin, liver marker enzymes and carbohydrate metabolizing enzymes were evaluated in STZ-induced diabetic rats. Oral administration methanolic extract of Caesalpinia pulcherrima of showed significant restoration of the body weight and decrease in the blood glucose level, liver marker enzymes (ALT, AST ALP) and carbohydrate metabolizing enzymes were observed in diabetic rats. These results suggest that fruit extract of Caesalpinia pulcherrima has valuable anti-diabetic activity in STZ-induced diabetic rats which is comparable to the standard drug metformin and hence might be of use in the management of diabetes.

Accumulating evidences have reinforced the use of medicinal plants in the treatment of various ailments as a result of negative side effects associated with conventional drugs. Plant components such as phenols and flavonoids with antioxidant potential have confirmed protective roles against oxidative stress-induced degenerative diseases like diabetes mellitus (DM). The current study was carried out to investigate the effect of seed pod ethanol extract from Copaifera salikounda (SPEECS) in alloxan-induced diabetic rats. SPEECS was obtained by maceration of seed pod powder in absolute ethanol for 72 h, filtered, concentrated and dried in-vacuo. Gas chromatography-mass spectrometry (GC–MS) technique was used to quantitatively elucidate the chemical constituents of SPEECS. Twenty-four male albino rats were randomly allocated into four groups (n=6): normal control, DM control, DM + 200 mg/kg SPEECS and DM + 400 mg/kg SPEECS groups. DM was induced in the Wistar albino rats through intraperitoneal injection of 200 mg/kg body weight of alloxan. After 14 days of treatment, the body weight changes and the fasting blood glucose level were determined in the different groups. Also, serum biochemical parameters such as alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP), albumin (ALB), total protein (TP), malondialdehyde (MDA), superoxide dismutase (SOD) and catalase (CAT) were estimated. The GC-MS results confirm nine bioactive compounds with 9-octadecenoic acid (55.75%) being most abundant. SPEECS (200 and 400 mg/kg) administration significantly (P 0.05) caused gain in weight, decreased fasting blood glucose and reversed the elevated liver function enzymes (ALT, AST, ALP) while total TP and ALB were markedly elevated relative to DM control group. Furthermore, SPEECS attenuated the activities of SOD and CAT while the level of MDA was significantly (P 0.05) decreased in dose dependent manner in comparison to the DM control. This study indicated that SPEECS can alleviate hyperglyceamia of DM. Key words: Copaifera salikounda; oxidative stress; medicinal plants; diabetes mellitus; phytochemicals; orthodox ANTIDIABETIČNI UČINEK EKSTRAKTA ETANOLA Copaifera salikounda (HECKEL) NA SLADKORNO BOLEZEN, SPROŽENO Z ALLOXAN-om, PRI PODGANAHIzvleček: Obstaja vedno več dokazov, ki poudarjajo uporabnost zdravilnih rastlin pri zdravljenju različnih bolezni, tudi zaradi različnih negativnih stranskih učinkov, povezanih s konvencionalnimi zdravili. Rastlinske sestavine kot so fenoli in flavonoidi z antioksidativnim potencialom, imajo po nekaterih raziskavah zaščitno vlogo pred degenerativnimi boleznimi, ki jih povzroča oksidativni stres, kot je sladkorna bolezen diabetes mellitus (DM). Študija je bila izvedena z namenom raziskovanja učinka etanolnega semenskega ekstrakta iz rastline Copaifera salikounda (SPEECS) pri podganah s sladkorno boleznijo, ki jo je povzročil alloxan. SPEECS je bil pridobljen z maceracijo praška semen v prahu v absolutnem etanolu 72 ur ter nadaljnjo filtracijo, koncentracijo in sušenjem v vakuumu. Za kvantitativno ugotavljanje kemijskih sestavin SPEECS je bila uporabljena tehnika plinske kromatografije in masne spektrometrije (GC-MS). Štiriindvajset samcev podgan Wistar je bilo naključno razporejenih v štiri skupine (n=6): normalna kontrola, kontrola DM, DM + 200 mg/kg SPEECS in DM + 400 mg/kg SPEECS. DM je bil pri podganah sprožen z intraperitonealno injekcijo 200 mg/kg telesne mase alloxana. Po 14 dneh zdravljenja so bile pri različnih skupinah določene spremembe telesne teže in nivo glukoze v krvi (na tešče). Poleg tega so avtorji raziskave izmerili še nekatere serumske biokemične parametre kot so ravni alaninske aminotransferaze (ALT), aspartatne aminotransferaze (AST), alkalne fosfataze (ALP), albumina (ALB), skupnih proteinov (TP), malondialdehida (MDA), superoksiddismutaze (SOD) in katalaze (CAT). Rezultati GC-MS so v izvlečku SPEECS pokazali devet bioaktivnih spojin, v katerih je največ 9-oktadecenojske kisline (55,75%). SPEECS (200 in 400 mg/kg) je povzročil znatno (P 0,05) povečanje telesne mase, znižanje glukoze v krvi na tešče in znižal raven encimov pokazateljev jetrne funkcije (ALT, AST, ALP), medtem ko je bila raven TP in ALB pri podganah, ki so prejemale SPEECS izrazito povišana v primerjavi z DM kontrolno skupino. Zdravljenje s  SPEECS je tudi oslabilo aktivnosti SOD in CAT, medtem ko se je raven MDA znatno zmanjšala (P 0,05) v primerjavi s kontrolno skupino DM. Objective: To determine the anti-hyperglycemic effects of interleukin-1 inhibitor (diacerein) in alloxan induced diabetic albino wistar rats. This experimental study was performed at the Department of Animal Husbandry and Veterinary Sciences, Sindh Agriculture University, Tando Jam within 6 months from April 2016 to September 2016. Total of 160 adult Albino Wistar Rats having an average of 200 to 300 grams body weights were selected. Animals were categorized into 4 groups as; Group A (n=15): Control rats – receive 0.9% normal saline as placebo Experimental Groups Group B (n=15): Experimental Control (Diabetic rats) - Alloxan50 mg/kg body weight intraperitoneal. Group C (n=15): Diabetic rats + Diacerein (30 mg/kg/day) orally daily. Group D (n=15): Diabetic rats + Diacerein (50 mg/kg/day) orally daily. Animals were kept and treated as per the NIH Guideline for Use and Care of Laboratory Animals. Diabetes mellitus was induced via a single intraperitoneal injection of 50 milligram/kg alloxan monohydrated dissolved in aseptic 0.9% saline. After 72 hours, blood specimens were taken from the caudal vein of the rats and glucose level>200 mg/dL was taken as diabetes. Experimental rats were given diacerein approximately 30 and 50 mg orally for 6 weeks. At the completion of experiment the body weight was measured of each animal by electronic measuring balance and blood sample was taken from each animal of all groups to assess the blood glucose level and HbA1c level. Data were recorded via self-made proforma and analysis was done by using SPSS version 20. Results: Average body weight of Diabetic control (Group B) was 193.33±22.50 grams, which was lower in contrast to Diacerein treated group C 202.47±25.70 grams and significantly lower as compared to Diacerein treated group D as  212.6±23.43 grams. A significant increase in blood glucose levels 182.07±10.63 mg/dl was noted in the Diabetic control (Group B) compared to Diacerein treated group C (110.13± 8.54 mg/dl) and group D (85.87±8.41 mg/dl) (P=0.001). HbA1c was markedly raised in the Group B- diabetic controls, while diacerein treated diabetic rats (groups C and D) showed a significant decrease in HbA1c (P=0.001). Conclusion: It was concluded that Diacerein achieves the Euglycemic state by reducing the levels of blood glucose and glycated hemoglobin (HbA1c) in Alloxan-Induced diabetes mellitus in Wistar Albino Rats.

 Objective: The objective was to study the evaluation of antidiabetic and antioxidant activity of different doses ethanolic extract of Mikania scandens leaves.Methods: The rats were divided into five groups of six animals each (non-diabetic control, diabetic control, standard drug-treated, and plant treated groups (250 and 500 mg/kg body weight)). Diabetes was induced in the healthy male Wistar rats (150–200 g body weight, 4–6 weeks old) by the administration of alloxan monohydrate (150 mg/kg, i.p.). On the 0th, 3th, 7th, 11th, and 15th days, the blood samples were analyzed for blood glucose. The antioxidant and antidiabetic parameters were evaluated by standard protocol. The liver tissue was used for histopathological assessment of liver damage.Results: The ethanolic extract of leaves of M. scandens showed a significant reduction (p<0.001) in the blood glucose levels and the antioxidant levels, with 500 mg/kg of body weight in the alloxan-induced diabetic rats as compared to the controls. Histopathological studies provided evidence that the leaf extract possesses antidiabetic activity.Conclusions: The ethanolic extract of leaves of M. scandens showed potent antidiabetic and antioxidant abilities against alloxan-induced rats, which indicate the presence of hypoglycemic constituents in the plant.

Nerunjil kudineer/Neermulli Kudinner is an official Siddha polyherbal formulation used for the various ailments related to the kidney. Cisplatin is an alkylating agent used in chemotherapy for the treatment of various cancers. Its use is limited due to their nephrotoxicity. In this study nephroprotective effect of Nerunjil kudineer (NK) on cisplatin-induced nephrotoxicity in Wistar albino Rats was studied. Male Wistar albino rats were used for this study. Animals were divided into 5 groups (n=6) as control, Cisplatin control (Single dose 7 mg/Kg i.p on the 7th day), Cisplatin with Cystone (p.o), NK200 and NK400 mg/Kg (p.o) for 10 days. At the end of the study, animals were weighed and sacrificed to estimate the relative kidney weight, serum creatinine and Urea. Kidney tissue was estimated for oxidant-antioxidant parameters (MDA, GSH & SOD) and histology study was carried out. Cisplatin reduced body weight and increased kidney weight significantly (P<0.0001). It deprived the renal function by elevating serum creatinine & urea, MDA and reduction in endogenous antioxidants GSH and SOD significantly (P<0.0001). Cisplatin group exhibited focal tubular necrosis and congested blood vessels in histology study. The standard drug Cystone and NK400 significantly increased the body weight, reduced the kidney weight, normalized the kidney function parameters (Serum Cr and Urea), bolstered antioxidant status and showed a trend towards the recovery of histological alterations. NK showed a dose-dependent activity and higher dose, NK400mg /Kg possessed strong nephroprotective activity, which may be due to the efficient antioxidant potential, which reduces lipid peroxidation and oxidative stress induced by cisplatin. The results strongly suggest that Nerunjil kudineer is an effective nephroprotective drug against Cisplatin induced nephrotoxicity.

Objective: To observe the absolute, relative weight of kidney and body weightof albino rats on celecoxib induced kidney with protection by lycopene. Study Design:Experimental study. Place and Duration of study: This study was conducted in BMSI (Anatomydepartment), JPMC, Karachi, from 4th May 2015 to 3rd June 2015. Materials and Methods: Fortyhealthy adult, male Albino rats, 90-120 days old, weighing 200-220gm was taken for the study.The rats were divided into 4 groups, Group A was control group, Group B receive Celecoxib 50mg/kg body weight orally, Group C receive Celecoxib 50 mg/kg body weight orally along withlycopene50 mg/kg body weight orally and Group D receive lycopene 50 mg/kg body weightorally for 30 days. At the end of study rats were sacrificed and renal tissue sections were stainedwith hematoxylin and eosin. Results: Markedly decreased weight was observed in rats takingcelecoxib. Slides which were stained with hematoxylin and eosinshowed general architecture ofrenal parenchyma, shape and arrangement of epithelial cells. Apoptosis, hemorrhage, necrosisand vacuolation seen in Celecoxib group, whereas renal architecture were ameliorated andreverted back in celecoxib along with lycopene receiving group. Conclusion: This studyconcludes that lycopene restored the body weight, absolute and relative kidney weight incelecoxib treated group.

The effect of the ethanolic extract of Myristica fragrans was evaluated on cafeteria diet induced body weight, glucose and lipid elevations in albino rats. 30 rats were taken randomly and divided into five groups and six each. Group-1 normal control and Group 2-5 were give cafeteria diet for 6 weeks to induce obesity and treatment period was 10 weeks. After 70 days of treatment, the extract, at doses of 200 and 400mg/kg, significantly reduced the body weight, glucose and lipid levels (p < 0.001) dose dependently. The standard drug Orlistat at 50mg/kg effectively prevented the body weight, glucose and lipid levels when compared with control and test groups. With these observations and previous data, the study concludes that Myristica fragrans extract can stimulate AMP-Kinase enzyme system and can reduce glucose and lipid concentrations. This may be useful for obesity treatment.

Background: Azathioprine is an immunosuppressant drug which is used to inhibit the body's rejection of transplanted tissues, autoimmune diseases, and cancers treatment. It may causes reduction of the body and organ weights and toxicity can be made better by the anti-oxidant zinc chloride. Objective: This study was planned to find out the effects of azathioprine on the body and testes weights and the ameliorative role of zinc chloride.Material and Methods: This experimental study was conducted in the department of Anatomy, Basic Medical Sciences Institute, Jinnah Postgraduate Medical Centre, Karachi, from January 2016 till January 2017. Sixty male adult albino rats of three to four months of age were chosen for this study and distributed into three equal groups A, B and C. They were further divided into four subgroup for 1st, 3rd, 6th and 8th weeks of the treatment. Group A served as control, received only injection 0.9 % normal saline, group B received inj. Azathioprine 15mg/kg body weight and group C received inj. azathioprine plus inj. zinc chloride 1mg/kg body weight. The route of injection was intraperitoneally daily. At the end of respective period of treatment initial and final body and testes weights were recorded.Results: Rats of control groups gained statistically significant average body and testes weights throughout experimental procedure. Azathioprine treated showed overall decrease in the body and testes weights as compared to control. Azathioprine plus zinc chloride treated group showed increase weights but comparatively less as compared to control.Conclusion: Zinc chloride played an ameliorative role against azathioprine-induced changes in body and testes weight of the albino rats.

Background: Alloxan-induced diabetes model is used as a “study tool” to elucidate the pathophysiology of the disease and much more as a “search engine” for antidiabetic compounds with better therapeutic characteristics. It was the first agent used in the category of chemically induced diabetes to create a model of insulin dependent diabetes mellitus. Other chemicals being streptozocin, dexamethasone, insulin antibodies-induced diabetes.Methods: Albino rats were divided into four groups with ten rats in each group. Alloxan monohydrate 2%, solution which was dissolved in 0.9% of sodium chloride (normal saline) as a diluent and given intraperitoneally to rats and blood glucose estimation made by using glucometer. Total 40 albino rats were taken and divided into 4 groups. 10 rats receiving normal saline were grouped as Group A, 10 rats received alloxan at a dose of 150 mg/kg as Group B, 10 rats received alloxan at a dose of 160 mg/kg as Group C and 10 rats received alloxan at a dose of 170 mg/kg as Group D.Results: Highest rate of mortality and alopecia were noted in group D receiving alloxan at a dose of 170 mg/kg whereas highest percentage of fluctuation in fasting blood glucose range was seen in group C receiving alloxan at a dose of 160 mg/kg.Conclusions: Such unpredictable response shows that alloxan is not ideal drug for induction of diabetes in experimental animal. Mortality, fasting blood glucose returning to non-diabetic range and alopecia are the chief drawbacks.

The effects of  methanolic leaf extracts of Vernonia amygdalina (VA) (Bitter leaf) and Gongronema latifolium (GL) (Utazi) and metformin on the histology and the DNA of the pancreas of streptozocin (STZ) induced diabetic rats were investigated in this study. 24 male albino rats which were divided into four groups of six rats each were used for this work: A (Normal control) and B (Diabetic control) received tap water, C received 5mg/kg of Metformin daily, D received combined extract of VA and GL, 400mg/kg twice daily. Groups B – D animals were induced for diabetes by intraperitoneal injection of 45mg/kg of Streptozocin, Following sacrifice, the pancreas and blood were collected for histopathological and biochemical studies. The results revealed normal exocrine and endocrine pancreas in group A. In group B, the pancreatic islets cells were shrunken with signs of degeneration and inflammation. In the group that received metformin, the islet appeared shrunken. In group D, the pancreas showed prominent and normal acinar and islet cells, suggestive of a possible regeneration. Feulgen’s reaction showed damage to the DNA of group B which was almost completely reversed in group D. The blood glucose in group B was significantly higher (P<0.05) compared to group A. In all the treatment groups, the blood glucose was significantly reduced (p<0.05) compared to group B.  Administration of methanolic leaf extracts of Vernonia amygdalina and Gongronema latifolium especially in combination restores pancreatic beta cells damaged by STZ compared to metformin.
