Abstract The paper is devoted to the optimization of one of the factors influencing the success of micropropagation – the choice of the optimal part of vine to take explant, size of the explant and the method of its exposure in a test tube. The studies have confirmed that the place, where micro-cutting was taken, its size and its orientation during planting can affect the speed of growth processes and the survival rate of plants. The experiment was put on the Purple Early grapevine variety. The part of vine for taking micro-cutting, best size of explant and the method of its orientation in the test tube are established.

The paper presents the technological study of red wine variety Augusta bred in Ya.I. Potapenko Institute for Viticulture and Winemaking. The research was carried out in the laboratory of winemaking technology in the conditions of micro-winemaking and semi-reproduction. The mass of one batch of grapes was 50–350 kg. Samples of natural dry, semi-sweet and liqueur wines were pre-pared. The main physical and chemical pa-rameters of wort and wine are determined. All of them corresponded to the Standard indicators. The organoleptic evaluation of wine materials showed that all the samples (dry, semi-sweet and liqueur) were distin-guished by a dark ruby color, had a bright, complex aroma, with tones of berries, and light shades of tea rose, and the high organo-leptic properties of the wines were increased when using bottle holding. According to the results of the research, conclusions were made about the feasibility of using the technical grade Augusta for the preparation of various types of wines (dry, semi-sweet and liqueur).

An agrobiological and techno-logical study of the Pamid grape variety was carried out during the realization of different variants of loading with winter eyes during pruning. An agrobio-logical and technological study of the Pamid grape variety was carried out during the realiza-tion of different loading variants with winter eyes during pruning. It has been established that the phenophases of development during the vegetation period occur and take place al-most simultaneously in the same time periods, and the grapes ripen during the first ten days of September. With the increase in the number of knots of a vine and with their lengthening, there is a well-defined tendency of reducing the percentage of developed winter eyes, fruit-ing shoots, weight per 1 cluster, less of reduc-ing the fertility rate. The vines grow well under the pruning loads of the four variants and the grapes manage to ripen. The best combination of sugar and titratable acid content for wine production is in the grapes from the variants with a load of 6 and 7 knots of a vine. The number of knots has a proven strong or moder-ate negative effect on the percentage of devel-oped eyes, fruiting shoots, bunches, fertility rate, weight of one cluster and 100 grains, dur-ing both years of the study. The increase in the number of knots is in a moderate, positive cor-relation with the yield of one vine.

Making raisins (dried grapes) is a laborious process and grape growers around the world are increasingly interested in adopting mechanized methods, especially dry-on-vine (DOV). The DOV method entails severing canes bearing mature fruit, causing them to DOV, after which they may be harvested by machine. The overhead arbor is an important DOV raisin trellis system, but basic agronomic performance of raisin varieties on that system are scant. Therefore, a three-year experiment was conducted to compare the performance of several raisin grape varieties (Vitis vinifera L.), on an overhead arbor trellis, for production of DOV raisins. Grapevine variety, ‘DOVine’, ‘Fiesta’, ‘Selma Pete’, or ‘Thompson Seedless’, was the main-plot factor, and number of canes, six or eight, was the sub-plot. ‘Fiesta’ produced the highest yields, averaging about 12.24 Mg/ha raisins. However, ‘Fiesta’ generally had the lowest soluble solids and among the lowest raisin grades. The later ripening of ‘Fiesta’ fruit delayed harvest pruning to initiate fruit drying compared to the other varieties. Therefore, its raisins did not dry sufficiently to meet industry standards. ‘Selma Pete’, ‘DOVine’, and ‘Thompson Seedless’ had similar yields, generally about 20% less than ‘Fiesta’, but ‘Selma Pete’ produced fruit that were the largest in size, with the highest soluble solids, and that produced raisins of the highest grades, compared to the other varieties. Vines pruned to eight canes produced higher yields without sacrificing raisin quality. In conclusion, ‘Fiesta’ was notable for its high productivity, and ‘Selma Pete’ for its earliness and high quality, and both varieties had sufficient capacity to carry eight 15-node canes.

The aim of this paper is to analyzed climatic influence on quality of yield of Merlot grapevine variety, in Trebinje region (Entity of Republic of Srpska, Bosnia and Herzegovina) vineyards, during the vegetation 2016 and 2017. We investigated the mechanical properties of bunch and berry (bunch weight, number of berries on the bunch, weight of 100 berries, weight of 100 berries flesh, weight of the berries skin, weight of seeds in 100 berries ) and quality properties of grapes (sugar content and total acid content in the must). The highest bunch weight, as well as the number of berries on the bunch was achieved in 2017 (276.84 g, i.e. 183.03), while the lowest bunch weight (193.6 g) and number of berries on the bunch (158.53) were measured in 2016. Weight of 100 berries, weight of 100 berries flesh, weight of the berries skin, weight of seeds in 100 berries were larger in 2017. The highest sugar content was measured during 2017 and the highest level of acid in must was measured during 2016. During the years 2016 and 2017 a significant influence of climatic factors on the quality characteristics of the studied variety was observed. Although both considered years had above average temperature and less precipitation, further analysis of differences in monthly values of climatologically parameters could provide an explanation for differences in mechanical and quality properties of grapes.

Molecular genotyping of native varieties of Vitis vinifera L. from different winegrowing areas is a current trend in the grapevine genetic diversity research. Abkhazia is among the world cradles of tamed grape, and its indigenous gene pool is of particular interest. Avasirkhva is a native Abkhasian grapevine variety mainly grown in the Gudauta District. Te research aimed to obtaining a genetic passport of Avasirkhva grapevine using microsatellite polymorphism data. Te study sampled grape plants from private farmsteads of the Gudauta District. Te plant phenotype corresponded to the variety’s ampelographic description. DNA was isolated from young shoot tip leaves with a CTAB-based protocol. Genotyping was performed with polymerase chain reaction (PCR) followed by capillary fragment separation. High-polymorphic SSR loci (VVS2, VVMD5, VVMD7, VVMD25, VVMD27, VVMD28, VVMD32, VrZAG62, VrZAG79) recommended for grapevine varietal identification were used as markers. Te amplicon size was estimated with an ABI Prism 3130 automated genetic analyser using the GeneMapper and PeakScanner soſtware and Pinot Noir as a reference variety genotype. Four samples exhibited an identical microsatellite profile. Te microsatellite assay-based genetic passport of the Avasirkhva variety is as follows: VVS2141-145 VVMD5234-242 VVMD7239- 249, VVMD25239-249, VVMD27184-190, VVMD28234-248, VVMD32248- 262, VrZAG62200-204, VrZAG79251-257. Te obtained passport is unique with respect to the known genotypes in the Vitis International Variety Catalogue (VIVC). A Principal Coordinate Analysis of microsatellite data was used to infer the genetic relationships between Avasirkhva and the Abkhasian varieties Kachich and Azhizhkvakva genotyped in our earlier studies, as well as nine native grapevines of Georgia, the nearest viticultural area. Te Avasirkhva genetic passport can be used in grapevine genotyping studies to clarify the varietal identity.
