Dengue Hemorrhagic Fever (DHF) is a disease caused by the dengue virus which is transmitted through the bite of the Aedes mosquito. Aedes aegypti, which is known as the primary vector of dengue virus, is naturally not infected by Wolbachia sp. endosymbiont, while Ae. albopictus which is a secondary vector naturally infected with Wolbachia sp. The Wolbachia sp. known to inhibit the transmission of Dengue virus, to study the mechanism, it is necessary to eliminate Wolbachia sp. from Ae. albopictus, then infects the Ae. albopictus with the Dengue virus. The aim of the study was to determine the ability of tetracycline antibiotics to eliminate Wolbachia sp. from the Ae. albopictus mosquito. Ae. albopictus eggs was obtained in the Ciamis area by survey method using ovitrap which was installed outside the house. The Ae. albopictus eggs are then incubated in the laboratory and reared until they become adult mosquitoes. Mosquitoes were treated with sugar feeding which had been given tetracycline 0.25mg/ml every two days alternated with blood feed. Detection of the presence of Wolbachia sp. on mosquitoes carried out in first to third generations by the polymerase chain reaction (PCR) method using Wsp-specific primers. The results showed that the first to third generation mosquitoes were still infected with Wolbachia sp. This shows that the dose of tetracycline antibiotics used has not been able to eliminate Wolbachia sp. from the Ae. albopictus mosquito.

Abstract Background Sugar feeding is a fundamental behaviour of many mosquito species. For Aedes albopictus, an important vector of dengue virus and chikungunya virus, little is known about its sugar-feeding behaviour, and no studies have been conducted on this in the southern hemisphere. This knowledge is pivotal for determining the potential of attractive targeted sugar baits (ATSBs) to control this important vector. Methods The prevalence of sugar was assessed in 1808 Ae. albopictus from Masig Island, Torres Strait, Australia collected between 13 and 25 March 2020. Fructose presence and content in field-collected Ae. albopictus were quantified using the cold anthrone assay. Results Significantly more male (35.8%) than female (28.4%) Ae. albopictus were sugar fed. There was a significant interaction between sex and time of day on the probability of capturing sugar-fed Ae. albopictus. For both sexes, fructose prevalence and content were higher in mosquitoes caught in the morning than in the afternoon. Female Ae. albopictus collected in the residential habitat were significantly more likely to be sugar fed than those collected in the woodland habitat. Conclusions These findings provide baseline information about the sugar-feeding patterns of Ae. albopictus and provide essential information to enable an assessment of the potential of ATSBs for vector suppression and control on Masig Island, with relevance to other locations where this species occurs. Background Aedes aegypti mosquitoes are globally distributed vectors of viruses that impact the health of hundreds of millions of people annually. Mating and blood feeding represent fundamental aspects of mosquito life history that carry important implications for vectorial capacity and for control strategies. Females transmit pathogens to vertebrate hosts and obtain essential nutrients for eggs during blood feeding. Further, because host-seeking Ae. aegypti females mate with males swarming near hosts, biological crosstalk between these behaviors could be important. Although mating influences nutritional intake in other insects, prior studies examining mating effects on mosquito blood feeding have yielded conflicting results. Methodology/Principal findings To resolve these discrepancies, we examined blood-feeding physiology and behavior in virgin and mated females and in virgins injected with male accessory gland extracts (MAG), which induce post-mating changes in female behavior. We controlled adult nutritional status prior to blood feeding by using water- and sugar-fed controls. Our data show that neither mating nor injection with MAG affect Ae. aegypti blood intake, digestion, or feeding avidity for an initial blood meal. However, sugar feeding, a common supplement in laboratory settings but relatively rare in nature, significantly affected all aspects of feeding and may have contributed to conflicting results among previous studies. Further, mating, MAG injection, and sugar intake induced declines in subsequent feedings after an initial blood meal, correlating with egg production and laying. Taking our evaluation to the field, virgin and mated mosquitoes collected in Colombia were equally likely to contain blood at the time of collection. Conclusions/Significance Mating, MAG, and sugar feeding impact a mosquito’s estimated ability to transmit pathogens through both direct and indirect effects on multiple aspects of mosquito biology. Our results highlight the need to consider natural mosquito ecology, including diet, when assessing their physiology and behavior in the laboratory.

As mosquito females require a blood meal to reproduce, they can act as vectors of numerous pathogens, such as arboviruses (e.g. Zika, dengue and chikungunya viruses), which constitute a substantial worldwide public health burden. In addition to blood meals, mosquito females can also take sugar meals to get carbohydrates for their energy reserves. It is now recognised that diet is a key regulator of health and disease outcome through interactions with the immune system. However, this has been mostly studied in humans and model organisms. So far, the impact of sugar feeding on mosquito immunity and in turn, how this could affect vector competence for arboviruses has not been explored. Here, we show that sugar feeding increases and maintains antiviral immunity in the digestive tract of the main arbovirus vector Aedes aegypti. Our data demonstrate that the gut microbiota does not mediate the sugar-induced immunity but partly inhibits it. Importantly, sugar intake prior to an arbovirus-infected blood meal further protects females against infection with arboviruses from different families. Sugar feeding blocks arbovirus initial infection and dissemination from the gut and lowers infection prevalence and intensity, thereby decreasing the transmission potential of female mosquitoes. Finally, we show that the antiviral role of sugar is mediated by sugar-induced immunity. Overall, our findings uncover a crucial role of sugar feeding in mosquito antiviral immunity which in turn decreases vector competence for arboviruses. Since Ae. aegypti almost exclusively feed on blood in some natural settings, our findings suggest that this lack of sugar intake could increase the spread of mosquito-borne arboviral diseases.

Background The red imported fire ant is one of the notorious species of ants all over the world. Sugar is one of the most important components of food and necessary for the survival of ants. Because more than 70% food of fire ants is honeydew produced by Homopteran insects such as aphids and scales. Methodology It is well known that beetles, flies, and honey bees can recognize the sugar taste through their legs and antennae, but in the case of fire ants, no records regarding gustatory sense were published. In the current study, considering the importance of sugar bait, we investigated the gustatory sense of the fire ant workers to sucrose via behavioral sequence and gustatory behavior. First, the feeding sequence (ethogram) of the fire ant workers on most preferred sugar (sucrose) solution was observed and categorized. Secondly, the gustatory behavior of treated fire ant workers (without flagellum and foreleg tarsi treated with HCL solution) was observed on the sucrose solution. In addition, using scanning electron microscopy (SEM) techniques, we identified the possible porous sensilla types on antenna flagellum and foreleg tarsi of fire ant workers. Results Based on the results of feeding sequence, foreleg tarsi of workers were the main body appendages in the detection of the sucrose droplet as compared to antennae flagellum and palps. Feeding time of treated workers with HCL solution was significantly decreased on sucrose solution as compared to those workers having no flagellum. While both types of treated workers have less feeding time in comparison to normal workers. Based on the results of feeding sequence analysis and feeding time, it is indicating that the foreleg tarsi of workers play a more important role in the detection of sucrose solution as compared to antennae flagellum. Based on the SEM results, sensilla chaetic, trichoid II, and basiconic I and II have a clear pore at their tip. This study provides a substantial basis for elucidating the gustatory function of antennal and tarsal sensilla on appendages of fire ant workers to sugars and further baits improvement for the management of fire ants.

AbstractDespite its epidemiological importance, the time Plasmodium parasites take to achieve development in the vector mosquito (the extrinsic incubation period, EIP) remains poorly characterized. A novel non-destructive assay designed to estimate EIP in single mosquitoes, and more broadly to study Plasmodium–Anopheles vectors interactions, is presented. The assay uses small pieces of cotton wool soaked in sugar solution to collect malaria sporozoites from individual mosquitoes during sugar feeding to monitor infection status over time. This technique has been tested across four natural malaria mosquito species of Africa and Asia, infected with Plasmodium falciparum (six field isolates from gametocyte-infected patients in Burkina Faso and the NF54 strain) and across a range of temperatures relevant to malaria transmission in field conditions. Monitoring individual infectious mosquitoes was feasible. The estimated median EIP of P. falciparum at 27 °C was 11 to 14 days depending on mosquito species and parasite isolate. Long-term individual tracking revealed that sporozoites transfer onto cotton wool can occur at least until day 40 post-infection. Short individual EIP were associated with short mosquito lifespan. Correlations between mosquito/parasite traits often reveal trade-offs and constraints and have important implications for understanding the evolution of parasite transmission strategies.

(1) Background: Bees are the primary animal pollinators in most ecosystems, and honey bees (Apis mellifera L.) are important providers of pollination ecosystem services and products. Climate change is one of the major threats for honey bees. (2) Objectives and methods: Qualitative research using focus group discussions was carried out in northwestern Italy, to investigate the beekeepers’ perceptions of climate change effects, the relevant management adaptations, and the main issues affecting the sector. (3) Results: Beekeepers reported several consequences related to severe weather events (weakening or loss of colonies; scarcity of nectar, pollen, and honeydew; decrease or lack of honey and other bee products; greater infestation by varroa; decline in pollination), making it necessary to provide supplemental sugar feeding, intensive transhumance, more effective and sustainable techniques for varroa control, and increased production of nuclei. A strengths, weaknesses, opportunities, and threats (SWOT) analysis was completed, displaying the factors able to strengthen or weaken the resilience of the beekeeping sector to climate change. (4) Conclusions: Thanks to their strong motivation and collaborative attitude, beekeepers succeed in adopting farm and bee hive adaptation strategies that are able to limit the climatic adverse effects. However, these findings highlight how the institutional and financial support for the beekeeping sector should be strengthened and better targeted.
