Pengembangan formulasi ekstrak daun Salam (Syzygium polyanthum W.) sebagai antihiperlipidemia yang mampu hancur cepat setelah diletakkan di atas lidah sangat diperlukan untuk mempercepat onset obat dan memberikan kenyamanan terutama pada pasien hiperlipidemia usia lanjut yang sulit menelan obat. Tujuan dari penelitian ini untuk mengetahui komposisi perbandingan superdisintegrants crospovidone dan croscarmellose sodium yang mampu menghasilkan sifat fisik Fast Disintegrating Tablet (FDT) yang optimum. Pembuatan FDT menggunakan metode granulasi basah dengan variasi konsentrasi crospovidone dan croscarmellose sodium dalam rentang 2%-5%. Hasil uji sifat fisik FDT dianalisis menggunakan metode Simplex Lattice Design (SLD) program Design Expert 10.1.). Kombinasi kedua superdisintegrants dapat mempengaruhi respon sifat fisik yaitu mampu menurunkan kekerasan, waktu disintegrasi, waktu pembasahan dan meningkatkan kerapuhan FDT ekstrak daun salam. Formula optimum didapat pada kombinasi crospovidone dan croscarmellose sodium dengan perbandingan 25 mg : 10 mg dalam setiap 500 mg tablet. Formula optimum tersebut memiliki kekerasan 4,21 kg, kerapuhan 0,52%, waktu pembasahan 106,65 detik, dan waktu hancur 55,73 detik. Hasil analisis dengan one sample t-test menunjukkan persamaan SLD valid digunakan untuk menyusun formula yang memberikan parameter-parameter optimum FDT.

AbstractThe aim of this work was to optimize the mass ratio of three Curcuma plants’ rhizomes to obtain the highest curcuminoid content and antioxidant activity using the simplex lattice design. The selected Curcuma plants were C. longa, C. zedoaria and C. aromatica. The simplex lattice design was applied in the work. The individual curcuminoids (curcumin, demethoxycurcumin and bis-demethoxycurcumin) and total curcuminoid content were determined using high-performance liquid chromatography. Curcuma longa alone provided the highest content of bis-demethoxycurcumin and demethoxycurcumin. A mixture of C. longa and C. aromatica in the mass ratio of 72:28 % provided the highest curcumin content. The results showed that C. longa alone exhibited the highest antioxidant activity.

Purple sweet potato has the content of anthocyanin compounds that are efficacious as antioxidants. To deliver these efficacious compounds, formulations of effervescent tablets are required as antioxidants with a combination of citric acid and tartrate acid to accelerate the solubility of the drug. The research aims to optimize the combination of citric acid and tartrate acid as a sourgent on effervescent tablets of purple sweet potato extract using the simplex lattice design method. The manufacture of tablets using a direct pumping method with effervescent powder evaluation includes flow rate test, silent angle, compressibility, and powder moisture. Evaluation of tablets includes organoleptic tests, uniformity of tablet weight, hardness, fragility, and soluble time of tablets—data analysis using simplex lattice design software design expert version 10. The results of effervescent powder evaluation include a test of flow rate, quiet angle, compressibility, and moisture of powder in each formula that already meets the requirements. The physical evaluation results on the effervescent tablet weight uniformity test on F3, F4, and F5 have met the requirements. The evaluation of tablet hardness F1 and F2 also meet the requirements. The results of the evaluation of fragility and time dissolve tablets have met the requirements of all formulas. The simplex lattice design analysis results obtained F5 as the most optimal formula with a concentration of citric acid 103.25 mg and tartrate acid 44.25 mg. The combination of citric acid and tartaric acid affects the evaluation of hardness, fragility, and time-soluble effervescent tablets of purple sweet potato extract. The optimal concentration based on simplex lattice design analysis was obtained in F5 with a citric acid concentration of 103.25 mg and tartrate acid of 44.25 mg.

Furosemide is a potent diuretic drug that has low bioavailability. Furosemide can be formulated into nanoemulsion preparations using the SNEDDS method to increase its bioavailability as SNEDDS can form stable nanoemulsions with droplet sizes 200 nm. This study aims to identify the optimum formula for variations in the concentration of surfactant Tween 80 and cosurfactant PEG 400 based on the characterization tests of emulsification time, percent transmittance, and drug loading. The independent variables used in this study were Tween 80 and PEG 400. Seven furosemide SNEDDS formulas from the Simplex Lattice Design (SLD) method were tested for characterization in the form of emulsification time, percent transmittance, and drug loading. The characterization results were optimized using Simplex Lattice Design. The optimum formula was re-characterized, including emulsification time, percent transmittance, drug loading, particle size, zeta potential, and in vitro dissolution. The results were then compared with theoretical values and analyzed using the One-Sample T-test method. Optimization results showed Tween of 61.4922% and PEG 400 of 18.5078% with the characterization of emulsification time 15.25 seconds, percentage transmittance 94.20%, drug loading 50 100.2 ppm, particle size 12.18 nm. Furthermore, the zeta potential was -17.6 mV, and the in vitro dissolution rate reached 106.71% within 15 minutes.

Ekstrak kulit buah manggis (Garcinia mangostana L) mengandung senyawa xanton yang memiliki aktivitas farmakologis dimana salah satu turunan xanton  α-mangostin bekerja sebagai antioksidan. Penelitian ini bertujuan untuk mengetahui etanol yang menghasilkan ekstrak dengan jumlah xanton tertinggi, menguji aktivitas antioksidan dan tabir surya dari ekstrak terpurifikasi serta mengoptimasi formula sediaan losion tabir surya dari ekstrak terpurifikasi kulit manggis.Optimasi formula losion dilakukan menggunakan software Design Expert® versi 11 dengan kombinasi ekstrak terpurifikasi, asam stearat dan trietanolamin. 14 formula losion dievaluasi nilai SPF, pH dan viskositasnya untuk penentuan formula optimum. Formula optimum selanjutnya diuji nilai SPF, ph dan viskositas untuk membandingkan prediksi software dan hasil percobaan. Selain itu dilakukan uji daya sebar, daya lekat serta stabilitas mekanis terhadap formula optimum. Hasil penelitian menunjukkan kandungan xanton dalam ekstrak etanol 50% sebesar 20,42%, ekstrak etanol 70 % sebesar 29,88% dan ekstrak etanol 96% sebesar 64,57%. Ekstrak terpurifikasi kulit buah manggis memiliki aktivitas antioksidan dengan nilai IC50 sebesar 5,53 dan nilai AAI 14,46 serta memiliki nilai SPF 8,94 pada konsentrasi 50 µg/ml. Formula optimum yang diperoleh memiliki nilai SPF 7,49 ± 0,0297, kekentalan 21.166,666 cp ± 10,41 serta pH 6,490 ± 0,0404, daya sebar 7,833333 ± 0,15, daya lekat 2,03 ± 0,01.

Helminthostachys zeylanica extract has pharmacological activities such as antioxidant, antiinflamatory, and antihyerucemia. This extract is nontoxic substance from the acute and subchronic toxicity tests. This extract has a potency to be formulated into tablet dosage forms. This study aims to optimize a tablet formula from Helminthostachys zeylanica extract. Disintegrant and binder concentrations were independent variables, while physical properties and dissolution time of the tablets were dependent variables. The tablet was prepared by a wet granulation method. Formula was optimized by Simplex Lattice Design. Physicochemical propertiesof granule, physical properties and dissolution of tablet were then analyzed with One Way ANOVA (p = 0.05). Based on granule analysis, specification of physicochemical parameters, such as hausner’s ratio, compressibility index, flowability, repose angle, and water content, met standard British Pharmacopeia. In addition, the starch and PVA concentrations influenced thickness, weight variation, hardness, friability, disintegration time and dissolution of the tablets (p <0.05), except for friability (p> 0.05). Based on this study, the starch and PVA concentrations for the optimum tablet formula were 19.5% and 1.05%, respectively.

Ashitaba is known to have antioxidant activity and gram-positive antibacterial activity that causes body odor. This is the potential activity for an active substance to be developed as deodorant. The appropriate formula is needed, so it is necessary to optimize the formula using the right method. This study aimed was to determine the application of the simplex lattice design method on the optimization of a deodorant roll-on formula of ashitaba extract. Ashitaba was extracted by the maceration method. The formula optimization design was determined using the simplex lattice design method by Design Expert®7.5.1. The components for optimization were the concentration of carbopol and concentration of TEA, and the optimization parameters were the spreadability test, sticky power, and pH test. The optimum formula of deodorant consists of 0.45% carbopol and 2.05% TEA. The responses of optimum formula obtained spreadability test 6.32 ± 0.33 cm, sticky power 44.67 ± 3.94 seconds, and pH 7.73 ± 0.17. These results meet the criteria for good preparation but need further testing related to the effectiveness of the preparation and the level of acceptance of the preparation by the user.

Daun jambu biji (Psidium guajava L.) mengandung senyawa flavonoid yang berpotensi sebagai antioksidan yang dapat mencegah terjadinya kerusakan kulit karena radikal bebas sehingga produk kosmetik sediaan topikal dengan zat aktif ekstrak daun jambu biji perlu dikembangkan. Tujuan dilakukannya penelitian ini untuk menentukan formula optimum sediaan lotion ekstrak daun jambu biji (Psidium guajava L.) dengan variasi setil alkohol dan gliserin serta menentukan sifat fisik formula optimumnya. Metode ekstraksi yang digunakan yaitu maserasi dengan pelarut etanol 96%. Nilai rendemen yang diperoleh yaitu 17,64%. Hasil skrining flavonoid menunjukkan hasil positif yang ditandai dengan terjadinya perubahan warna larutan menjadi merah. Optimasi formula dilakukan dengan metode Simplex Lattice Design yang diolah menggunakan software Design Expert dan sediaan lotion diformulasi dengan meleburkan bahan fase minyak (asam stearat, setil alkohol, propil paraben) dan fase air (trietanolamin, gliserin, metil paraben, aquades) secara terpisah pada suhu 70-75oC kemudian disatukan dan diaduk hingga homogen. Dari software tersebut diperoleh formula optimum dengan konsentrasi setil alkohol dan gliserin masing-masing sebesar 3,93% dan 13,07%. Hasil evaluasi sifat fisik formula optimum yaitu memilki warna hijau kecoklatan, bau khas daun jambu biji, konsistensi yang kental dan pH 7,942, daya sebar 6,350 cm dan daya lekat 0,975 detik.

Rosella flower (Hibiscuss sabdariffa L.) in traditional medicine can provide protection against various diseases, such as coronary heart disease, cancer, lowering cholesterol, and liver disorders. This study aims to obtain the optimum formula of tablet of rosella flower extract with a mixture of Avicel PH 101 and lactose as a filler using wet granulation method. Rosella flower extract was obtained by maceration of rosella flower powder using 70% ethanol. The optimization formula for tablets using the Simplex Lattice Design method with Design Expert software version 10. The parameters used in the optimization are weight variation, hardness, friability and disintegration time. Verification of the optimal formula using the SPSS 18 program with the One Sample T-test method with a confidence level of 95%. The results showed that the optimal formula of tablet of rosella flower extract was the Avicel PH 101 97.82%: 2.17% lactose. The optimal formula has weight variation 1.53% ± 0.0073; hardness 5.087 kg ± 0.5135; the friability is 0.3% ± 0.041. The resulting disintegration was 22.99 minutes ± 2.218. The results of the verification of the optimal formula show that there is no significant difference between the predictions of the Simplex Lattice Design method and the results of the tests performed so that it can be said that the method is valid.

A modified Scheffe's simplex lattice design method is proposed to study the properties of multicomponent materials. This modified Scheffe's method allowed determining the optimal compositions of cordierite and corundum based ceramic materials that are used as catalyst carrier for gas purification equipment. The obtained material (0.63-1.25 mm weight fraction of cordierite of 0.35 mass% fraction; < 0.63 mm weight fraction of cordierite of 0.35 mass% fraction; < 0.06 mm weight fraction of corundum of 0.2 mass% fraction; 1.25-2.5 mm weight fraction of cordierite of 0.2 mass% fraction) was used successfully for the manufacturing of catalytic neutralizers of gas emissions.
