Circadian clocks schedule biological functions at a specific time of the day. Full comprehension of the clock function requires precise understanding of their entrainment to the environment. The phase of entrained clock is plastic, which depends on different factors such as the period of endogenous oscillator, the period of the zeitgeber cycle (T), and the proportion of light and darkness (day length). The circadian clock of fruit fly Drosophila melanogaster is able to entrain to a wide range of T-cycles and day lengths. Here, we investigated the importance of the neuropeptide Pigment-Dispersing Factor (PDF) for entrainment by systematically studying locomotor activity rhythms of Pdf 0 mutants and wild-type flies under different T-cycles (T22 to T32) and different day lengths (8, 12, and 16 hour [h]). Furthermore, we analysed PERIOD protein oscillations in selected groups of clock neurons in both genotypes under T24 and T32 at a day length of 16 h. As expected, we found that the phase of Drosophila’s evening activity and evening neurons advanced with increasing T in all the day lengths. This advance was much larger in Pdf 0 mutants (~7 h) than in wild-type flies causing (1) pronounced desynchrony between morning and evening neurons and (2) evening activity to move in the morning instead of the evening. Most interestingly, we found that the lights-off transition determines the phase of evening neurons in both genotypes and that PDF appears necessary to delay the evening neurons by ~3 h to their wild-type phase. Thus, in T32, PDF first delays the molecular cycling in the evening neurons, and then, as shown in previous studies, delays their neuronal firing rhythms to produce a total delay of ~7 h necessary for a wild-type evening activity phase. We conclude that PDF is crucial for appropriate phasing of Drosophila activity rhythm.

The quality of the question can describe the development of a person's thinking level. The questioning activity is also an indication that a student is actively involved in the learning process. The purpose of this study was to describe the quality profile of the students' questions from a natural science family in terms of their learning experience. This study uses a descriptive method because it aims to describe the profile of research subjects. The research instrument was a learning experience questionnaire and a question matrix. Learning experience questionnaires are used to reveal a picture of student learning experiences during lectures. Meanwhile, the question matrix is used to measure the level of thinking on each question made by students. The population of this study were all students in the Mathematics and Natural Sciences Education Department who had attended Basic Biology courses. Data analysis using simple regression shows that student learning experience does not significantly influence the quality of student questions. Each study program has a t score of 0.120 (chemistry education); 1,341 (Physics education); and 0.578 (Biology education). The entire t score is less than the t table. This condition is strongly suspected because the core activity phase of the lecture process did not go well. Based on the data analysis, it was found that the core activity phase had the lowest score of 67.33 which was significant.

The particulate nature of matter (PNM) is central to learning science and is a difficult concept for both children and adults. The purpose of this study is to examine how teaching materials from an integrated science and literacy curriculum on the PNM affects communication between pre-service teachers. We were especially interested in examining communication during an activity phase and whether and how participants used PNM models. The interactions between participants were recorded with headcams and analysed using a framework developed by Mortimer and Scott in 2003. The findings revealed that the participants mainly described what they observed directly using scientific words and concepts, but they did not necessarily use PNM models or theoretical knowledge spontaneously. Research indicates that the ability to use knowledge at a theoretical level is key to understanding chemical concepts, so our study underscores the importance of explicitly asking participants to use models and theoretical knowledge.

This study investigates whether a chronotherapeutic treatment of hepatocellular carcinoma (HCC) may improve treatment efficacy and mitigate side effects on healthy liver (HL). HCC was induced in Per2::luc mice which were irradiated at four time points of the day. Proliferation and DNA-double strand breaks were investigated in irradiated and non-irradiated organotypic slice culture (OSC) and ex vivo samples by detection of Ki67 and γ-H2AX. OSC proved useful to determine dose-dependent effects on proliferation and DNA damage but appeared unsuited to test the chronotherapeutic approach. Irradiation of ex vivo samples was most effective at the proliferation peaks in HCC at ZT02 (early inactivity phase) and ZT20 (late activity phase). Irradiation effects on HL were minimal at ZT20. Ex vivo samples revealed disruption in daily variation and down-regulation of all investigated clock genes except Per1 in non-irradiated HCC as compared with HL. Irradiation affected rhythmic clock gene expression in HL and HCC at all ZTs except at ZT20. Irradiation at ZT20 had no effect on total leukocyte numbers. Our results indicate ZT20 as the optimal time point for irradiation of HCC in mice. Translational studies are now needed to evaluate whether the late activity phase is the optimal time point for irradiation of HCC in man.

Background: Professional rodeo is a sport with a high risk of injuries for which research is needed to support interventions. To date, there have been no epidemiological studies performed specifically on hip conditions sustained during rodeo. Purpose: To describe the epidemiology of hip conditions in professional rodeo. Study Design: Case series; Level of evidence, 4. Methods: Deidentified hip injury data from electronic medical records of the Justin Sportsmedicine Team from 2011 to 2014 were analyzed for risk, frequency, type, location, and mechanism, as well as activity phase, of hip injuries. Results: A total of 84 hip injuries among 82 adult male and female athletes were reported, resulting in an overall hip injury density of 0.41 injuries per 1000 competitor-exposures (95% CI, 0.0003-0.0005) and a risk probability of 0.04%. Rough stock athletes sustained 83.3% of hip injuries, with bull riders sustaining 50.0% of the injuries. Contusions (45.2%), impingement (15.5%), and hip strains (13.1%) were the most common injuries. Athletes were most likely to be injured during the dismount (36.1%), and 36.9% of injuries were due to contact with the ground. Conclusion: Rough stock athletes have the greatest risk for hip injury in professional rodeo, with bull riders sustaining the most hip injuries. Athletes are most likely to be injured during the dismount. Common hip injuries in professional rodeo are contusions, impingement, and strains. The majority of contusions result from collision with the ground. Applicable measures to prevent or reduce the severity of injuries to the hip, such as protective padding, proprioceptive training, and eccentric strength training, should be implemented.

Electrophoretic displays (EPDs) have excellent paper-like display features, but their response speed is as long as hundreds of milliseconds. This is particularly important when optimizing the driving waveform for improving the response speed. Hence, a driving waveform design based on the optimization of particle activation was proposed by analyzing the electrophoresis performance of particles in EPD pixels. The particle activation in the driving waveform was divided into two phases: the improving particle activity phase and the uniform reference grayscale phase. First, according to the motion characteristics of particles in improving the particle activity phase, the real-time EPD brightness value can be obtained by an optical testing device. Secondly, the derivative of the EPD brightness curve was used to obtain the inflection point, and the inflection point was used as the duration of improving particle activity phase. Thirdly, the brightness curve of the uniform reference grayscale phase was studied to set the driving duration for obtaining a white reference grayscale. Finally, a set of four-level grayscale driving waveform was designed and validated in a commercial E-ink EPD. The experimental results showed that the proposed driving waveform can cause a reduction by 180 ms in improving particle activity phase and 120 ms in uniform reference grayscale phase effectively, and a unified reference grayscale can be achieved in uniform reference grayscale phase at the same time.

Growing evidence shows that theta-band (4-7Hz) activity in the auditory cortex (AC) phase-locks to the rhythm of overt speech. However, does theta activity in AC also phase-lock to inner speech? Previous research established that silent reading of direct speech quotes (e.g., Mary said: “This dress is lovely!”) elicits more vivid inner speech than indirect speech quotes (e.g., Mary said that the dress was lovely). Using EEG, the present study found that direct (vs. indirect) quote reading was associated with increased theta phase-locked activity (phase and power) over trials at 250-500 ms during quotation reading with sources estimated in the speech processing network. An eye-tracking control experiment confirmed that increased theta phase-locked activity in direct quote reading could not be driven by reading patterns and may reflect the onset of inner speech. These findings provide novel evidence for a functional role of theta activity in reading-induced inner speech.
