Objective. Preeclampsia (PE) is a severe complication in pregnancy and a leading cause of maternal and infant mortality. However, the exact underlying etiology of PE remains unknown. Emerging evidence indicates that the cause of PE is associated with genetic factors. Therefore, the aim of this study is to identify susceptibility genes to PE. Materials and Methods. Human Exome BeadChip assays were conducted using 370 cases and 482 controls and 21 loci were discovered. A further independent set of 958 cases and 1007 controls were recruited for genotyping to determine whether the genes of interest ROS1 and PTPRK are associated with PE. Immunohistochemistry was used for localization. Both qPCR and Western blotting were utilized to investigate the levels of PTPRK in placentas of 20 PE and 20 normal pregnancies. Results. The allele frequency of PTPRK rs3190930 differed significantly between PE and controls and was particularly significant in severe PE subgroup and early-onset PE subgroup. PTPRK is primarily localized in placental trophoblast cells. The mRNA and protein levels of PTPRK in PE were significantly higher than those in controls. Conclusion. These results suggest that PTPRK appears to be a previously unrecognized susceptibility gene for PE in Han Chinese women, and its expression is also associated with PE, while ROS1 rs9489124 has no apparent correlation with PE risk.

Understanding the biochemical mechanisms underlying bud dormancy and bloom time regulation in deciduous woody perennials is critical for devising effective strategies to protect these species from spring frost damage. This study investigated the accumulation profiles of carbohydrates, ROS and antioxidants during dormancy in ‘Cripps Pink’ and ‘Honeycrisp’, two apple cultivars representing the early and late bloom cultivars, respectively. Our data showed that starch levels generally declined during dormancy, whereas soluble sugars increased. However, the present study did not record significant alternations in the carbohydrate accumulation profiles between the two cultivars that could account for the differences in their bloom dates. On the other hand, H2O2 accumulation patterns revealed an apparent correlation with the dormancy stage and bloom dates in both cultivars; peaking early in the early-blooming cultivar, sustaining high levels for a longer time in the late-blooming cultivars, and fading by the time of bud burst in both cultivars. Also, the redox balance during dormancy appeared to be maintained mainly by catalase and, to a lesser extent, by glutathione (GSH). Overall, the present study concludes that differences in ROS and the bud redox balance could, at least partially, explain the differences in dormancy duration and bloom date among apple cultivars.

The physical features of the stinger are compared in 51 species of vespid wasps: 4 eumenines and zethines, 2 stenogastrines, 16 independent-founding polistines, 13 swarm-founding New World polistines, and 16 vespines. The overall structure of the stinger is remarkably uniform within the family. Although the wasps show a broad range in body size and social habits, the central part of the venom-delivery apparatus—the sting shaft—varies only to a modest extent in length relative to overall body size. What variation there is shows no apparent correlation with social habits. This is consistent with the hypothesis that stinger size is constrained by the demands of a flight-worthy body. The sting lancets bear distinct, acute barbs in all examined species except in members of the Stenogastrinae. Barbs vary considerably among species in number, their summed lengths, and the relative degree of serration (summed length relative to lancet width). Where they are numerous and strong, it increases the likelihood of the stinger remaining fatally embedded in the skin of a vertebrate adversary (sting autotomy). Although an index that combines the number and strength of barbs is a more natural measure of overall serration, the number of barbs alone is almost as good a predictor of the likelihood of sting autotomy. Across the family as a whole, the tendency to sting autotomy is concentrated in the swarm-founding New World polistines.

To better understand the potential factors contributing to genome instability and phenotypic diversity, we conducted mutation accumulation (MA) experiments for 120 days for 7 diploid cryptococcal hybrids under fluconazole (10 MA lines each) and non-fluconazole conditions (10 MA lines each). The genomic DNA content, loss of heterozygosity (LOH) rate, growth ability, and fluconazole susceptibility were determined for all 140 evolved cultures. Compared to that of their ancestral clones, the evolved clones showed: (i) genomic DNA content changes ranging from ~22% less to ~27% more, and (ii) reduced, similar, and increased phenotypic values for each tested trait, with most evolved clones displaying increased growth at 40 °C and increased fluconazole resistance. Aside from the ancestral multi-locus genotypes (MLGs) and heterozygosity patterns (MHPs), 77 unique MLGs and 70 unique MPHs were identified among the 140 evolved cultures at day 120. The average LOH rates of the MA lines in the absence and presence of fluconazole were similar at 1.27 × 10−4 and 1.38 × 10−4 LOH events per MA line per mitotic division, respectively. While LOH rates varied among MA lines from different ancestors, there was no apparent correlation between the genetic divergence of the parental haploid genomes within ancestral clones and LOH rates. Together, our results suggest that hybrids between diverse lineages of the human pathogenic Cryptococcus can generate significant genotypic and phenotypic diversities during asexual reproduction.

A possible correlation between the impact of Covid-19 and universal immunization program against tuberculosis with Bacillus Calmette-Guérin (BCG) vaccine was suggested previously, based on apparent correlation between lower impact of the epidemics and a record of national BCG immunization program. In this work a time-adjusted dataset of Covid-19 statistical data by national and subnational health jurisdictions at the time point of six months after the local arrival of the epidemics was used to perform a statistical analysis of the significance of the correlation hypothesis between universal BCG immunization and a milder early phase Covid-19 scenario. With the data accumulated up to the point of the analysis, the significance of the correlation hypothesis was evaluated both qualitatively and quantitatively with the conclusion that it has achieved statistically significant level of confidence. The conclusions of this research can be used in development of epidemiological policy as well as the rationale to investigate the origin and mechanisms of a broad immunity protection that can be associated with an early-age exposure to BCG vaccine. Doi: 10.28991/SciMedJ-2021-03-SI-8 Full Text: PDF

Previous research has shown that hops contain enzymes able to hydrolyze unfermentable dextrins into fermentable sugars when added during the dry-hopping process. In the presence of live yeast, these additional fermentable sugars can lead to an over-attenuation of the beer; a phenomenon known as “hop creep”. This study attempts to analyze the effect of different Saccharomyces yeast species and strains on hop creep, with the intent to find an ability to mitigate the effects of dry-hop creep by using a specific yeast. Thirty different yeast species and strains were chosen from commercial and academic collections and propagated for pilot fermentations. Brews were performed at the Anheuser-Busch Research Brewery (1.8 hL, 10 °P, 20 IBU) at UC Davis and split to 40 L cylindroconical fermenters, with one fermenter in each yeast pair receiving 10 g/L Centennial hop pellets towards the end of fermentation. Standard analytical measurements were performed over the course of fermentation, with real degrees of fermentation (RDF) and extract measured on an Anton Paar alcolyzer. In order to preemptively determine the amount of hop creep to be experienced with each unknown fermentation, bench-top fermentations with 20 g/L dry-hops were performed concurrently and compared to the pilot scale fermentations. RDF was significantly higher (p < 0.01) on dry-hopped than non-dry-hopped fermentations beginning two days post dry-hopping to the end of fermentation, with the exceptions of SafAle™ BE-134, a S. cerevisiae var. diastaticus, and UCDFST 11-510, a S. mikatae. No apparent correlation between flocculation and increased RDF was shown in dry-hopped treatments. pH was significantly different between the dry-hopped and non-hopped fermentations (p < 0.05 one day post dry-hop, p < 0.01 for all subsequent days); this may have impacted on additional attenuation. No yeasts in this study indicated their use for mitigation of dry-hop creep, but this is a first look at beer fermentation for some of the chosen yeasts. The results also present a new perspective on how hop creep varies in fermentation.

AbstractObjectivesSARS-CoV-2 infection induces the formation of different antibodies. However, not all of which might prevent the virus from entering the cell, although their concentrations correlate with the titers of viral neutralization tests (NTs). Antibodies against the viral nucleocapsid (NC), e.g., can be classified as such. We aimed to prove the hypothesis that the apparent correlation between NC-antibody levels and NT-titers is mediated by simultaneously occurring antibodies against viral spike-protein components.MethodsWe included 64 individuals with previous SARS-CoV-2 infection (>14d after symptom onset). SARS-CoV-2 antibodies against the NC (Roche total antibody ECLIA, Abbott IgG CMIA) and spike-protein (Technozym RBD ELISA, DiaSorin S1/S2 CLIA) were measured, and neutralization tests were performed. The effect of spike-protein antibodies on the correlation between NC-antibodies and NT-titers was evaluated by partial correlation and mediation analyses.ResultsBoth tested assays assessing antibodies against the NC correlated significantly with NT titers: Abbott ρ=0.742, P<0.0001; Roche ρ=0.365, P<0.01. However, when controlling the rank correlations for the presence of RBD or S1/S2 antibodies, correlation coefficients dropped to ρ=0.318/ρ=0.329 (P<0.05/P<0.01), respectively for Abbott and vanished for Roche. As a result, only a maximum of 11% of NT titer variability could be explained by NC-antibody levels.ConclusionsOur data suggest that the apparent correlation between NC antibodies and NT titers is strongly mediated by co-occurring RBD antibody concentrations. To avoid falsely implied causal relationships, all correlation analyses of non-spike-associated antibody assays and neutralization assays should include a partial correlation analysis to exclude a possible mediator effect of spike-associated antibodies.

ABSTRACT We observed 51 sources in the Q-U-I JOint TEnerife (QUIJOTE) cosmological fields that were brighter than 1 Jy at 30 GHz in the Planck Point Source Catalogue (version 1), with the Very Large Array at 28–40 GHz, in order to characterize their high-radio-frequency variability and polarization properties. We find a roughly lognormal distribution of polarization fractions with a median of 2 per cent, in agreement with previous studies, and a median rotation measure (RM) of ≈1110 rad m−2 with one outlier up to ≈64 000 rad m−2, which is among the highest RMs measured in quasar cores. We find hints of a correlation between the total intensity  flux density and median polarization fraction. We find 59 per cent of sources are variable in total intensity, and 100 per cent in polarization at 3σ level, with no apparent correlation between total intensity variability and polarization variability. This indicates that it will be difficult to model these sources without simultaneous polarimetric monitoring observations and they will need to be masked for cosmological analysis.

Abstract It is often assumed that checks and balances are effective in curbing corruption, in part because checks and balances are so often assumed to be synonymous with the separation of powers. We argue that checks and balances are only one of several potential manifestations of the separation of powers. We suggest that the apparent correlation between checks and balances and control of corruption is driven by a variety of conditions antecedent to both. Using examples from Western democracies, we demonstrate that the concept of checks and balances is by itself an empty vessel, made effective only by “hard” factors such as the balance of political forces and “soft” factors such as the adherence of elites to particular behavioral norms. This does not mean that checks and balances cannot be useful, but rather that our assumptions about their precise utility may be misinformed: the relationship between checks and balances and curbing corruption is at best indirect.
