Interest in developing food, feed, and other useful products from farmed insects has gained remarkable momentum in the past decade. Crickets are an especially popular group of farmed insects due to their nutritional quality, ease of rearing, and utility. However, production of crickets as an emerging commodity has been severely impacted by entomopathogenic infections, about which we know little. Here, we identified and characterized an unknown entomopathogen causing mass mortality in a lab-reared population of Gryllodes sigillatus crickets, a species used as an alternative to the popular Acheta domesticus due to its claimed tolerance to prevalent entomopathogenic viruses. Microdissection of sick and healthy crickets coupled with metagenomics-based identification and real-time qPCR viral quantification indicated high levels of cricket iridovirus (CrIV) in a symptomatic population, and evidence of covert CrIV infections in a healthy population. Our study also identified covert infections of Acheta domesticus densovirus (AdDNV) in both populations of G. sigillatus. These results add to the foundational research needed to better understand the pathology of mass-reared insects and ultimately develop the prevention, mitigation, and intervention strategies needed for economical production of insects as a commodity.

AbstractEngineered micro- and nanomechanical resonators with ultra-low dissipation constitute a promising platform for various quantum technologies and foundational research. Traditionally, the improvement of the resonator’s performance through nanomechanical structural engineering has been driven by human intuition and insight. Such an approach is inefficient and leaves aside a plethora of unexplored mechanical designs that potentially achieve better performance. Here, we use a computer-aided inverse design approach known as topology optimization to structurally design mechanical resonators with optimized performance of the fundamental mechanical mode. Using the outcomes of this approach, we fabricate and characterize ultra-coherent nanomechanical resonators with, to the best of our knowledge, record-high Q ⋅ f products for their fundamental mode (where Q is the quality factor and f is the frequency). The proposed approach - which can also be used to improve phononic crystals and coupled-mode resonators - opens up a new paradigm for designing ultra-coherent micro- and nanomechanical resonators, enabling e.g. novel experiments in fundamental physics and extreme sensing.

AbstractPosttraumatic stress disorder can be viewed as a disorder of fear dysregulation. An abundance of research suggests that the prefrontal cortex is central to fear processing—that is, how fears are acquired and strategies to regulate or diminish fear responses. The current review covers foundational research on threat or fear acquisition and extinction in nonhuman animals, healthy humans, and patients with posttraumatic stress disorder, through the lens of the involvement of the prefrontal cortex in these processes. Research harnessing advances in technology to further probe the role of the prefrontal cortex in these processes, such as the use of optogenetics in rodents and brain stimulation in humans, will be highlighted, as well other fear regulation approaches that are relevant to the treatment of posttraumatic stress disorder and involve the prefrontal cortex, namely cognitive regulation and avoidance/active coping. Despite the large body of translational research, many questions remain unanswered and posttraumatic stress disorder remains difficult to treat. We conclude by outlining future research directions related to the role of the prefrontal cortex in fear processing and implications for the treatment of posttraumatic stress disorder.

The introductory course at Penn State Brandywine--Earth in the Future: Predicting Climate Change and Its Impacts Over the Next Century--utilizes a scaffolded instructional approach and faculty from several campus units to develop multiple literacies (information, digital, science) and professional skills (writing and speaking) in students. The audio narrative assignment has provided additional outcomes beyond improved knowledge in climate science.

This chapter reviews the research that set the stage for Joshua Lederberg’s surprising discovery of bacterial conjugation. While the foundational research of Gregor Mendel and his principles of inheritance had been effectively combined with Darwinian evolution, producing the Modern Synthesis in the mid-forties, bacteria did not fit into this grand synthesis. Most biologists believed that bacteria were too primitive to have real genes. But Delbruck, Hershey and Luria organized the Phage School, leading a novel approach to discovering the molecular biology of the gene by studying bacteriophages. Microbiologists like Tracy Sonneborn and Carl Lindegren turned to alternative microorganisms—protists, fungi, and yeast—to develop new model systems that offered advantages over the classical genetics organisms of animals and plants. The research of Edward Tatum and Jacques Monod indicated that mutations seemed to explain variation in bacteria. For many years, however, bacteriologists had known that bacteria reproduced by fission. The lack of any genetic hybridization seemed to argue against using bacteria to study basic genetic processes.

Abstract Background Phony peach disease (PPD) is caused by the plant pathogenic bacterium Xylella fastidiosa subsp. multiplex (Xfm). Historically, the disease has caused severe yield loss in Georgia and elsewhere in the southeastern United States, with millions of PPD trees being removed from peach orchards over the last century. The disease remains a production constraint, and management options are few. Limited research has been conducted on PPD since the 1980s, but the advent of new technologies offers the opportunity for new, foundational research to form a basis for informed management of PPD in the U.S. Furthermore, considering the global threat of Xylella to many plant species, preventing import of Xfm to other regions, particularly where peach is grown, should be considered an important phytosanitary endeavor. Main topics We review PPD, its history and impact on peach production, and the eradication efforts that were conducted for 42 years. Additionally, we review the current knowledge of the pathogen, Xfm, and how that knowledge relates to our understanding of the peach—Xylella pathosystem, including the epidemiology of the disease and consideration of the vectors. Methods used to detect the pathogen in peach are discussed, and ramifications of detection in relation to management and control of PPD are considered. Control options for PPD are limited. Our current knowledge of the pathogen diversity and disease epidemiology are described, and based on this, some potential areas for future research are also considered. Conclusion There is a lack of recent foundational research on PPD and the associated strain of Xfm. More research is needed to reduce the impact of this pathogen on peach production in the southeastern U.S., and, should it spread internationally, wherever peaches are grown.

Abstract Background The EU’s 2006 Paediatric Regulation aims to support authorisation of medicine for children, thus effectively increasing paediatric research. It is ethically imperative to simultaneously establish procedures that protect children’s rights. Method This study endeavours (a) to evaluate whether a template consent form designed by the Standing Working Group of the German-Research-Ethics-Committees (AKEK) adequately informs adolescents about research participation, and (b) to investigate associated phenomena like therapeutic misconception and motives for research participation. In March 2016 a questionnaire study was conducted among 279 pupils (mean age 13.1 years) of a secondary school in northern Germany. Results A majority of participants showed a general good understanding of foundational research ethics concepts as understood from the AKEK consent form. Nevertheless, our data also suggests possible susceptibility to therapeutic misconception. Own health concerns and pro-social considerations were found to be significant motivational factors for participating in research, while anticipation of pain lessens likelihood of participation. Advice from trusted others is an important decisional influence, too. Furthermore, data security was found to be a relevant aspect of adolescents’ decision-making process. Conclusion Bearing in mind adolescents’ generally good understanding, we infer the lack of knowledge about medical research in general to be one source of therapeutic misconception. To further improve the quality of consent we propose a multi-staged approach whereby general research education is completed before an individual becomes a patient or potential participant. To the best of our knowledge this is the first German questionnaire-study addressing issues of informed consent in a large under-age sample.

Simulation-based training systems should be tailored to the actual work environment of the envisioned audience of learners. After building an augmented reality-based training system for combat medics, we began to explore strategies for adapting the training to medical students. While people who provide medical care on the battlefield and people who practice medicine in a hospital need to know how to treat traumatic injuries, there are vast differences in how they are trained. Foundational research into the work environment has implications for training design, especially with regards to training content and delivery.

An exciting development in the sociology of medical education has been its recent return as a distinct scholarly conversation in medical sociology. During the 1980s and 1990s, the sociology of medical education, an historically prominent subfield in sociology, seemed to disappear from the scholarly conversation despite ongoing development in this area. In this narrative review I describe this “missing period” of sociology of medical education, discussing complementary explanations for why it receded and describing what research activity did take place during those decades. In reviewing this work, I argue that articulating theoretical advances made within sociology of medical education research during these decades allows us to link foundational research from the 1950s and 1960s with the renaissance of this subfield in the early 2000s. Fundamentally, understanding the intellectual history and development of this subfield supports a broader movement to understand the import of studies of medical training for exploring questions of interest in general sociology.

AbstractCommemorating the 40 th anniversary of the Diagnostic and Statistical Manual (DSM) III, the purpose of this commentary is to describe school-based and school-relevant interventions and instructional approaches for children and youth with autism that have been developed and employed during that time period. The commentary begins with a brief description of foundational research that provides an historical context. Research themes shaped by science, ethics, social policy, and the changes in the DSM provide an organization for describing the evolution of intervention and instructional practices over the four previous decades. The commentary concludes with a discussion of school-contextual variables that influence implementation and the promise of the “iSciences” for closing the research to practice gap in the future.
