Gamete fusion is the climax of fertilization in all sexually reproductive organisms, from unicellular fungi to humans. Similarly to other cell-cell fusion events, gamete fusion is mediated by specialized proteins, named fusogens, that overcome the energetic barriers during this process. In recent years, HAPLESS 2/GENERATIVE CELL-SPECIFIC 1 (HAP2/GCS1) was identified as the fusogen mediating sperm-egg fusion in flowering plants and protists, being both essential and sufficient for the membrane merger in some species. The identification of HAP2/GCS1 in invertebrates, opens the possibility that a similar fusogen may be used in vertebrate fertilization. HAP2/GCS1 proteins share a similar structure with two distinct families of exoplasmic fusogens: the somatic Fusion Family (FF) proteins discovered in nematodes, and class II viral glycoproteins (e.g., rubella and dengue viruses). Altogether, these fusogens form the Fusexin superfamily. While some attributes are shared among fusexins, for example the overall structure and the possibility of assembly into trimers, some other characteristics seem to be specific, such as the presence or not of hydrophobic loops or helices at the distal tip of the protein. Intriguingly, HAP2/GCS1 or other fusexins have neither been identified in vertebrates nor in fungi, raising the question of whether these genes were lost during evolution and were replaced by other fusion machinery or a significant divergence makes their identification difficult. Here, we discuss the biology of HAP2/GCS1, its involvement in gamete fusion and the structural, mechanistic and evolutionary relationships with other fusexins.

It has been exactly 20 years since the common grounds for the design of track access charges (TAC) were laid for the European railways by the publication of Directive 2001/14/EC. However, these grounds were defined broadly, thus resulting in significant divergence both in the models applied by countries and during the model redesign within one country over the course of time. The participants in the process of charge system redesign includes all stakeholders from a country’s railway sector (infrastructure manager, train operating companies, the ministries responsible for transport, finance and economy, government, and regulatory bodies). Their opinions and requirements are often opposed, and they all need to be acknowledged simultaneously. This paper aims to solve the issue of ensuring continuity in the charge model redesign while achieving a balance between the requirements of all stakeholders. Moreover, it tackles the issue of producing a sustainable long-term TAC model by using survey methods and statistical analysis. The proposed approach was tested in practice during the access charge model redesign for the railways of Montenegro. The results show the importance of continual enhancement in TAC model development as one of the challenges and key precursors for the harmonization of all stakeholders’ requirements.

The black soldier fly (BSF), Hermetia illucens (Diptera: Stratiomyidae), has become one of the most economically important insects that can be a sustainable replacement for traditional feed for swine, poultry, and aquaculture. Despite its emerging importance, studies examining the nutritional requirement of BSF have been scarce. Here we used nutritional geometry approach to determine the separate and combined effects of dietary protein and carbohydrate content on multiple life-history traits and body composition linked to fitness in BSF. BSF larvae were reared on one of 32 chemically defined diets that varied in protein-to-carbohydrate ratio (P:C=1:16, 1:8, 1:4, 1:2, 1:1, 2:1, 4:1, or 8:1) and in protein plus carbohydrate concentration (P+C=60, 120, 180, or 240 g/l). Females developed more slowly into larger adults than males, but two sexes responded to dietary variation in protein and carbohydrate in a similar manner. The topography of the nutritional performance landscapes mapped for all measured traits differed significantly from one another, with the optimal P:C ratio being 1:1.71 for preadult survivorship, 1:1.56 – 1:1.11 for development time, 1:3.36 – 1:3.16 for body weight at adult emergence, and 1:1.83 – 1:1.40 for growth rate. Significant divergence in nutritional optima among these components of fitness indicates that the occurrence of fundamental life-history trade-off can be mediated by macronutrient intake. The index representing the preadult performance of BSF had a nonlinear relationship with dietary protein, with the peak centred at the intermediate P:C ratios of 1:1.43 – 1:1.37. The optimal P:C ratio was 1:2.12 – 1:1.70 for lean body weight and 1:14.14 – 1:10.82 for lipid content. Our results highlight the importance of the balanced composition of dietary protein and carbohydrate for optimising BSF performance and have implications for enhancing the mass production of this beneficial species.

Abstract When studying the influence of excavator bucket capacity on haul truck loading time, it is necessary to assess the technical and economic performance of the complex. If the cost of the machine shift of the complex will not cover the increase in productivity, then it is inexpedient to use this complex. This issue requires further, more detailed study and performance of detailed technical and economic analysis. Also, it is necessary to consider classical recommendations about ratio of excavator bucket capacity and haul truck body. It should be noted that at present, there is a significant divergence in the size range of excavators: powerful, with a large capacity bucket, rope shovels are used for mining the coalless zone; small maneuverable hydraulic shovels – for mining the coal-bearing zone. In this paper, we will try to combine the first and the second: consider hydraulic excavators with maximum bucket capacity, which is reasonable to use when mining overburden. In addition, a detailed study of the options of mutual arrangement of excavator and haul truck has not been performed until recently, this work is intended to fill this gap.

Urban sprawl is one of the most common landscape alterations occurring worldwide, and there is a growing list of species that are recognised to have adapted to urban life. To be successful, processes of urban colonization by wildlife require a broad spectrum of phenotypic (e.g., behavioural or physiological) adjustments, but evidence for genetic adaptations is much scarcer. One hypothesis proposes that different pathogen-driven selective pressures between urban and non-urban landscapes leads to adaptations in host immune genes. Here, we examined urbanization-related differentiation at the key pathogen-recognition genes of vertebrate adaptive immunity-the major histocompatibility complex (MHC)-in a common waterbird, the Eurasian coot (Fulica atra). Samples were collected from an old urban population (established before the 1950s), a new urban population (established in the 2000s), and two rural populations from central Poland. We found strong significant divergence (as measured with Jost’s D) at the MHC class II between the old urban population and the remaining (new urban and rural) populations. Also, there was a moderate, but significant divergence at the MHC between the new urban population and two rural populations, while no divergence was found between the two rural populations. The total number of MHC alleles and the number of private (population-specific) MHC alleles was lower in old urban populations, as compared to the rural ones. These patterns of differentiation at the MHC were not consistent with patterns found for neutral genetic markers (microsatellites), which showed few differences between the populations. Our results indicate that MHC allele composition depended on the level of anthropogenic disturbance and the time which passed since urban colonization, possibly due to the processes of genotype sorting and local adaptation. As such, our study contributes to the understanding of genetic mechanisms associated with urbanization processes in wildlife.

This gene contains a (GGC)13, spanning its core promoter and 5′ untranslated region (RASGEF1C-201 ENST00000361132.9). Here we sequenced the (GGC)-repeat in a sample of human subjects (N = 269), consisting of late-onset NCDs (N = 115) and controls (N = 154). We also studied the status of this STR across various primate and non-primate species based on Ensembl 103. The 6-repeat allele was the predominant allele in the controls (frequency = 0.85) and NCD patients (frequency = 0.78). The NCD genotype compartment consisted of an excess of genotypes that lacked the 6-repeat (divergent genotypes) (Mid-P exact = 0.004). A number of those genotypes were not detected in the control group (Mid-P exact = 0.007). The RASGEF1C (GGC)-repeat expanded beyond 2-repeats specifically in primates, and was at maximum length in human. We conclude that there is natural selection for the 6-repeat allele of the RASGEF1C (GGC)-repeat in human, and significant divergence from that allele in late-onset NCDs. STR alleles that are predominantly abundant and genotypes that deviate from those alleles are underappreciated features, which may have deep evolutionary and pathological consequences.

The COVID-19 pandemic is having a significant impact on tourism, and emotion projection is one way to understand the extent of destination image resiliency during the crisis. Therefore, this research captured emotions expressed in social media during a peak pandemic month to compare to the prior year period. Toronto and New York were selected due to their tourism importance within their countries but to also compare the effects of different policy approaches used during the pandemic. This study found resiliency of the destination images although there was a significant increase in projections of fear for both cities. Additionally, there was a significant divergence observed for the two cities with a decrease in joy and an increase in sadness projections for New York versus Toronto. This implies that tourism destination marketers have a stable basis of emotions to use in communications, but there are weaknesses to address.

Abstract The link between biodiversity and ecosystem functioning has been the topic of considerable research, but it remains unclear how biodiversity decline is compromising ecosystem functionality, particularly in the pelagic realm. Here, we explore how pelagic fish species diversity relates to functional diversity by sampling two locations, which, on the basis of biogeography, environmental conditions, and human pressures, were expected to host pronounced differences in species composition and abundances, and therefore functionality. Strings of five drifting mid-water Baited Remote Underwater Video Systems were used to survey pelagic vertebrate diversity and abundance in two isolated oceanic island systems, the Malpelo Fauna and Flora Sanctuary – a large, 25-year-old marine protected area – and an unprotected area in Cape Verde. Functional diversity, which offers insight into a community’s resilience against disturbance, was analysed using six key functional traits of marine fishes. Cape Verde showed high overall abundance (Total MaxN 873) and low biomass (3,559 kg), with a predominance of smaller fishes. Malpelo showed high biomass (7,839 kg) but lower abundance (Total MaxN 465), with a predominance of large species. Species and functional diversity were marginally different between locations. Multivariate analysis of species relative abundances showed significant divergence between locations, although community functional traits overlapped strongly, suggesting that both communities share a similar structure and vulnerability. The existence of a common functional ‘backbone’ in diverging species communities across the oceans, under different productivity regimes, and under different protection levels, suggests that although pelagic communities may differ considerably in terms of species composition, this does not translate into a differing functional structure and resilience potential. Whether this vulnerability is a common feature of pelagic communities and how this contrasts with benthic systems warrants further research.
